Potential roles of medicinal plants for the treatment of viral diseases focusing on COVID-19: A review.

The whole world is entangled by the coronavirus disease (COVID-19) pandemic caused by severe acute respiratory syndrome coronavirus 2 (SARS-CoV-2), people are dying in thousands each day, and without an actual medication, it seems not possible for the bringing this global health crisis to a stop. Natural products have been in constant use since ancient times and are proven by time to be effective. Crude extract or pure compounds isolated from medicinal plants and/or herbs such as Artemisia annua, Agastache rugosa, Astragalus membranaceus, Cassia alata, Ecklonia cava, Gymnema sylvestre, Glycyrr

Lycorine inhibits Ang II-induced heart remodeling and inflammation by suppressing the PI3K-AKT/NF-κB pathway.

BACKGROUND: Ang II induces hypertensive heart failure (HF) via hemodynamic and non-hemodynamic actions. Lycorine (LYC) is an alkaloid derived from Lycoris bulbs, and it possesses anti-cardiovascular disease-related activities. Herein, we explored the potential LYC-mediated regulation of Ang II-induced HF. METHODS: Over 4 weeks, we established a hypertensive HF mouse model by infusing Ang II into C57BL/6 mice using a micro-osmotic pump. For the final two weeks, mice were administered LYC via intraperitoneal injection. The LYC signaling network was then deduced using RNA sequencing. RESULTS: L

Narciclasine reduces proliferation and migration of neuroblastoma cells and decreases FAK/PI3K pathway activation.

Neuroblastoma is a highly aggressive childhood malignancy originating from neural crest-derived cells. Narciclasine, an Amaryllidaceae-derived alkaloid isolated from plants such as Lycoris radiata, has been reported to exhibit various pharmacological activities; however, its effects in neuroblastoma remain poorly understood. In this study, we investigated the effects of narciclasine on SH-SY5Y neuroblastoma cells, focusing on proliferation, migration, apoptosis, DNA damage, and FAK/PI3K-associated signaling. Cell viability was evaluated using the CCK-8 assay; proliferation by Ki67 immunofluore

Coclaurine N-methyltransferase-like enzymes drive the final biosynthetic reaction of the anti-Alzheimer's drug galanthamine in Amaryllidaceae.

Galanthamine, an isoquinoline alkaloid used to treat symptoms of Alzheimer's disease, is predominantly extracted from Amaryllidaceae plants, yet its supply remains limited. In this study, we identified, isolated, and characterized N-methyltransferases (NMTs) from three galanthamine-producing species: Leucojum aestivum, Lycoris radiata, and Hippeastrum papilio. The transcriptomic analysis identified five unique NMT isoforms, among which LaLrHpNMT1, an isoform highly conserved across all three species, exhibited the highest catalytic activity. Phylogenetic and structural analyses revealed that t

Inhibitory effects of herbal alkaloids on the tumor necrosis factor-α and nitric oxide production in lipopolysaccharide-stimulated RAW264 macrophages.

It is beneficial to treat chronic inflammatory condition in patients through diets that inhibit the production of proinflammatory cytokines and mediators such as tumor necrosis factor-α (TNF-α) and nitric oxide (NO). Since less attention has been paid to alkaloids in the diets than to polyphenols in this regard, we aimed at investigating anti-inflammatory activity of herb-derived alkaloids through suppression of TNF-α and NO production in lipopolysaccharide (LPS)-stimulated mouse RAW264 and/or human THP-1 cells. A harmala alkaloid, harmine, an opium alkaloid, papaverine, and Lycoris alkaloids,

The role of natural products in the discovery of new drug candidates for the treatment of neurodegenerative disorders II: Alzheimer's disease.

The present review is part II in a series (part I focuses on Parkinson's Disease) that addresses the value of natural product chemistry in the discovery of medicines for the treatment of neurodegenerative disorders. Data reviewed document that a host of products from plant species and derivatives have neuroprotectant effects in vitro and in vivo. In addition, besides neuroprotection, natural products also demonstrate biological effects that target biochemical pathways underlying associated symptoms of neurdegnerative disorders that include cognitive impairments, energy/fatigue, mood, and anxie

Narciclasine Exerts Anticancer Activity in Colorectal Cancer Cells in Association with HELLS Downregulation and DNA Damage-Associated Responses.

Background/Objectives: Colorectal cancer (CRC) remains a major cause of cancer-related mortality worldwide, largely owing to metastasis, recurrence, and resistance to conventional therapies. Natural-product-derived compounds represent an important source of anticancer drug candidates. This study investigated the anticancer activity of purified narciclasine, which was tentatively annotated as a major detected constituent of Lycoris sanguinea extract, and examined its association with HELLS-associated DNA damage responses in CRC cells. Methods: Human CRC cell lines HT29 and HCT116 were used to e

Lycorine suppresses DPP3-mediated Nrf2 signaling, thereby reducing SLC7A11 levels to trigger ferroptosis in colorectal cancer cells.

BACKGROUND: Colorectal cancer (CRC) exhibits high morbidity/mortality with poor late-stage survival, creating an urgent need for effective therapies. Although lycorine, a pyrrolophenanthridine alkaloid isolated from the traditional Chinese herb Lycoris radiata, has been confirmed to exhibit significant anti-CRC activity. the exact biological mechanism of its action remains unclear PURPOSE: The present study aimed to clarify whether lycorine exerts its anti-CRC effect by inducing ferroptosis in CRC cells (HCT-116 and RKO), and to elucidate its specific molecular mechanism. METHODS: Human CRC c

Integrating Narcissus-derived galanthamine production into traditional upland farming systems.

Alzheimer's disease (AD) is a disorder associated with progressive degeneration of memory and cognitive function. Galantamine is a licenced treatment for AD but supplies of the plant alkaloid that it is produced from, galanthamine, are limited. This three-year system study tested the potential to combine Narcissus-derived galanthamine production with grassland-based ruminant production. Replicate plots of permanent pasture were prepared with and without bulbs of Narcissus pseudonarcissus sown as lines into the sward. Two different fertiliser regimes were imposed. The above-ground green biomass
