Short Communication: Plasma Lymphocyte Activation Gene 3 and Subclinical Coronary Artery Disease in the Multicenter AIDS Cohort Study.

Chronic inflammation, including among people with HIV (PWH), elevates immune cell expression of lymphocyte activation gene 3 (LAG3); however, low plasma LAG3 predicts cardiovascular disease (CVD) events in the general population. The associations among LAG3 plasma levels, subclinical atherosclerosis, inflammation, and HIV infection have not been well described. We measured plasma LAG3 in 704 men with and without HIV from the multicenter AIDS cohort study, who underwent coronary computed tomography angiography. HIV serostatus was not independently associated with LAG3 after adjustment for socio

Binding domains of α-synuclein receptors with monomeric/oligomeric α-synuclein: Implications for Parkinson's disease.

Parkinson's disease (PD) and related synucleinopathies are characterized by the pathological aggregation and spread of alpha-synuclein (α-syn), a protein central to neurodegeneration. Currently three neuronal receptors -Family with sequence similarity 171 member A2 (FAM171A2), Lymphocyte Activation Gene 3 (LAG3), and Amyloid-Precursor-Like Protein 1 (APLP1) have been identified as critical mediators of α-syn preformed fibril (PFF) uptake, propagation, and neurotoxicity. Here we utilize AlphaFold 3 program to predict binding domains of respective α-syn receptors with monomer and oliogmer α-syn

Lymphocyte activation gene-3 (LAG-3) regulatory T cells: An evolving biomarker for treatment response in autoimmune diseases.

Regulatory T cells (Tregs) comprise a CD4+CD25+Foxp3+ T cell subset for maintaining immune tolerance, and their deficits and/or dysfunction are observed in autoimmune diseases. The lymphocyte activation gene 3 (LAG-3, also known as CD223), which is an immunoglobulin superfamily member expressed on peripheral immune cells, is recognized as an inhibitory regulator of Tregs. LAG-3+ T cells represent a novel protective Tregs subset that produces interleukin-10. Alterations in LAG-3+ Tregs have been reported in several autoimmune diseases, suggesting their potential pathogenic role. Recent studies

Genetic signature of CTLA-4, BTLA, TIM-3 and LAG-3 molecular expression in colorectal cancer patients: Implications in diagnosis and survival outcomes.

OBJECTIVE: Accumulating evidences suggest that immune checkpoints (ICs) inhibit immune response against cancerous cells and promote tumor cell survival. Up-regulation of ICs in tumor microenvironment is reported in patients with colorectal cancer (CRC). Thus, evaluating the peripheral blood expression of ICs may be used as non-invasive biomarkers for diagnosis and prognosis of CRC. METHODS: This study included 60 primary and treatment naïve CRC patients along with 15 age and sex matched healthy volunteers as a control group. Total RNA was extracted from peripheral blood samples and gene expre

A Bispecific Peptide-Drug Conjugate Targeting LAG-3 and PD-L1 Harnesses Antitumor Immunity of Macrophages and T Cells.

Programmed cell death protein 1 (PD-1) and lymphocyte-activation gene 3 (LAG-3) are identified as key immune checkpoints on tumor-infiltrating macrophages, beyond their roles in T-cell functions. Blockade of LAG-3 and PD-1 pathways skews M2 macrophage polarization and shows antitumor efficacy independent of T cells. A bispecific peptide-drug conjugate, BsPep-IMDQ, was developed to simultaneously block both pathways and deliver a Toll-like receptor 7/8 (TLR7/8) agonist, imidazoquinoline (IMDQ), via a matrix metalloproteinase (MMP)-cleavable linker. This conjugate demonstrated potent tumor suppr

Natural killer group 2D and CD28 receptors differentially activate mammalian/mechanistic target of rapamycin to alter murine effector CD8+ T-cell differentiation.

Memory CD8+ T cells are an essential component of anti-tumour and anti-viral immunity. Activation of the mammalian/mechanistic target of rapamycin (mTOR) pathway has been implicated in regulating the differentiation of effector and memory T cells. However, the mechanisms that control mTOR activity during immunity to tumours and infections are not well known. Activation of co-stimulatory receptors, including CD28 and natural killer group 2D (NKG2D), activate phosphatidylinositol-3 kinase and subsequently may activate the mTOR pathway in CD8+ T cells. This study compared the activation of the mT

Searching for Biomarkers in the Blood of Patients at Risk of Developing Parkinson's Disease at the Prodromal Stage.

Parkinson's disease (PD) is diagnosed many years after its onset, under a significant degradation of the nigrostriatal dopaminergic system, responsible for the regulation of motor function. This explains the low effectiveness of the treatment of patients. Therefore, one of the highest priorities in neurology is the development of the early (preclinical) diagnosis of PD. The aim of this study was to search for changes in the blood of patients at risk of developing PD, which are considered potential diagnostic biomarkers. Out of 1835 patients, 26 patients were included in the risk group and 20 p

The FGL1-LAG-3 axis attenuates melanoma-induced cachexia in mice.

Cachexia is a debilitating condition characterized by the wasting of adipose and skeletal muscle tissues, resulting in significant weight loss and frailty. It is responsible for 20% of cancer-related mortalities. Despite its clinical significance, the precise mechanisms underlying cachexia remain inadequately understood. Our gene expression analysis of white adipose tissues has identified an upregulation of fibrinogen-like protein 1 (FGL1) in B16F10 tumor-bearing mice. In this study, we demonstrate that the intraperitoneal administration of FGL1 protein mitigates tumor-induced wasting of adipo

Regulatory B cells and SIVA1-LAG3 immune suppression predict poor prognosis in prostate cancer.

Prostate cancer (PCa) is among the most prevalent malignancies affecting men globally. The immunosuppressive characteristics of the tumor microenvironment significantly hinder the effectiveness of immunotherapeutic strategies. B cells have a dual role in immune responses, with regulatory B cells (Bregs) promoting immune tolerance through the secretion of immunosuppressive cytokines, facilitating tumor immune evasion. However, the specific role of Bregs in prostate cancer remains inadequately understood. In this study, we systematically identified Breg subpopulations in prostate cancer using si

Roles of LAG3 and EGR2 in regulatory T cells.

Regulatory T cells (Tregs) participate in the maintenance of tolerance to self-antigens and suppressive control of excessive immune responses to exogenous antigens. A lack or dysfunction of these cells is responsible for the pathogenesis and development of many autoimmune diseases. It is well known that CD4 Tregs play a major role in controlling immune responses and can be classified into two main populations: thymus-derived naturally occurring Tregs (nTregs) and induced Tregs (iTregs) generated from CD4(+)CD25(-) precursors in the peripheral lymphoid organs. The most extensively studied Tregs

Intracellular Ca2+ and related proteins in patients with oral lichen planus.

Oral lichen planus (OLP) is suggested to be a T cell-mediated chronic inflammatory oral mucosal disease. Gene expressed in the oligodendrocyte lineage-myelin basic proteins (Golli-MBP) and stromal interaction molecule 1 (STIM1) are important in the activation and function of T lymphocytes. This study aimed to analyze and compare the expression of Golli-MBP and STIM1 between OLP patients and healthy controls and to analyze the level of intracellular Ca(2+), which is involved in lymphocyte activation. The Ca(2+) fluorescent probe, Fluo-3/AM, was used to test the level of intracellular Ca(2+) in

Protein transport inhibitors downregulate the expression of LAG-3 on regulatory T cells.

Modern immunologic studies demand increasing complexity because of a need to improve our understanding of the relationship between a cell's phenotype and its function. Regulatory T cells (Tregs) have been defined by a narrow set of phenotypic markers, however their actual functional capacity has not been determined at the single-cell level. Although the lymphocyte activation gene 3 (LAG-3; CD223) is a key marker for the identification of exhausted T cells, it may be useful also in resolving Treg subpopulations by indicating distinct functional breadths. Here we define the experimental conditio

Early Growth Response Gene 2-Expressing CD4+LAG3+ Regulatory T Cells: The Therapeutic Potential for Treating Autoimmune Diseases.

Regulatory T cells (Tregs) are necessary for the maintenance of immune tolerance. Tregs are divided into two major populations: one is thymus derived and the other develops in the periphery. Among these Tregs, CD4+CD25+ Tregs, which mainly originate in the thymus, have been extensively studied. Transcription factor Foxp3 is well known as a master regulatory gene for the development and function of CD4+CD25+ Tregs. On the other hand, peripheral Tregs consist of distinct cell subsets including Foxp3-dependent extrathymically developed Tregs and interleukin (IL)-10-producing type I regulatory T (

The RIG-I pathway is involved in peripheral T cell lymphopenia in patients with dermatomyositis.

BACKGROUND: Peripheral T cell lymphopenia is a clinical phenomenon in some patients with dermatomyositis (DM). Patients with T cell lymphopenia are more susceptible to life-threatening infections. However, the pathogenesis of T cell lymphopenia remains unclear. In this study, we aimed to determine retinoic acid-inducible gene I (RIG-I) expression in peripheral T lymphocytes and explore the correlation between RIG-I and T cell lymphopenia in DM. METHODS: The mRNA and protein expression levels of RIG-I were determined in peripheral T lymphocytes of 26 treatment-naive DM patients by q-PCR and We

Resident memory T cells in the skin mediate durable immunity to melanoma.

Tissue-resident memory T (TRM) cells have been widely characterized in infectious disease settings; however, their role in mediating immunity to cancer remains unknown. We report that skin-resident memory T cell responses to melanoma are generated naturally as a result of autoimmune vitiligo. Melanoma antigen-specific TRM cells resided predominantly in melanocyte-depleted hair follicles and were maintained without recirculation or replenishment from the lymphoid compartment. These cells expressed CD103, CD69, and CLA (cutaneous lymphocyte antigen), but lacked PD-1 (programmed cell death protei

Distinct roles for talin-1 and kindlin-3 in LFA-1 extension and affinity regulation.

In inflammation, neutrophils and other leukocytes roll along the microvascular endothelium before arresting and transmigrating into inflamed tissues. Arrest requires conformational activation of the integrin lymphocyte function-associated antigen-1 (LFA-1). Mutations of the FERMT3 gene encoding kindlin-3 underlie the human immune deficiency known as leukocyte adhesion deficiency-III. Both kindlin-3 and talin-1, another FERM domain-containing cytoskeletal protein, are required for integrin activation, but their individual roles in the induction of specific integrin conformers are unclear. Here,
