Dark Classics in Chemical Neuroscience: Lysergic Acid Diethylamide (LSD).

Lysergic acid diethylamide (LSD) is one of the most potent psychoactive agents known, producing dramatic alterations of consciousness after submilligram (≥20 μg) oral doses. Following the accidental discovery of its potent psychoactive effects in 1943, it was supplied by Sandoz Laboratories as an experimental drug that might be useful as an adjunct for psychotherapy, or to give psychiatrists insight into the mental processes in their patients. The finding of serotonin in the mammalian brain in 1953, and its structural resemblance to LSD, quickly led to ideas that serotonin in the brain might b…

The impact of fluoxetine on the aversive effects of lysergic acid diethylamide.

Serotonergic antidepressant history can reduce the subjective effects, including therapeutic efficacy, of a number of psychedelics. A related issue is the impact of traditional antidepressants with serotonergic action on the adverse effects of psychedelics. Becker et al. (2025) reported that adverse effects induced by lysergic acid diethylamide (LSD) were reduced by pretreatment with the selective serotonin reuptake inhibitor paroxetine, while "good drug effects" induced by LSD were not. The present studies examined if exposure to the selective serotonin reuptake inhibitor fluoxetine impacts t…

Pharmacokinetics and Pharmacodynamics of Lysergic Acid Diethylamide in Healthy Subjects.

BACKGROUND AND OBJECTIVE: Lysergic acid diethylamide (LSD) is used recreationally and in clinical research. The aim of the present study was to characterize the pharmacokinetics and exposure-response relationship of oral LSD. METHODS: We analyzed pharmacokinetic data from two published placebo-controlled, double-blind, cross-over studies using oral administration of LSD 100 and 200 µg in 24 and 16 subjects, respectively. The pharmacokinetics of the 100-µg dose is shown for the first time and data for the 200-µg dose were reanalyzed and included. Plasma concentrations of LSD, subjective effect…

Comparative Pharmacological Effects of Lisuride and Lysergic Acid Diethylamide Revisited.

Lisuride is a non-psychedelic serotonin (5-HT) 2A receptor (5-HT2A) agonist and analogue of the psychedelic lysergic acid diethylamide (LSD). Lisuride also acts as an agonist at the serotonin 1A receptor (5-HT1A), a property known to counter psychedelic effects. Here, we tested whether lisuride lacks psychedelic activity due to a dual mechanism: (1) partial agonism at 5-HT2A and (2) potent agonism at 5-HT1A. The in vitro effects of lisuride, LSD, and related analogues on 5-HT2A signaling were characterized by using miniGαq and β-arrestin 2 recruitment assays. The 5-HT1A- and 5-HT2A-mediated ef…

Chromosome examinations after medically administered lysergic acid diethylamide and dextroamphetamine.

The results of the present study demonstrate once again that, on the average, the addition of LSD in vitro leads to chromosome damage in excess of that observed in cultures without such added LSD even though nearly all of the in vitro experiments were carried out on blood cultures derived from patients who had already been started on the drug regime in vivo. By contrast, the present data provide no evidence for a measurable detrimental effect of LSD and DA when administered to patients under medical supervision. It is conceivable that an effect might have emerged had we been able to monitor ch

Self-inflicted testicular amputation in first lysergic acid diethylamide use.

BACKGROUND: Genital self-mutilation is a well-known phenomenon in patients with schizophrenia and has occasionally been described in patients with personality disorders or transsexuality. However, literature just provides few cases of genital self-mutilation related to the use of psychotropic substances. CASE DESCRIPTION: A previously mentally healthy man (age, 32 years) performed manual amputation of both testes after first use of lysergic acid diethylamide in combination with alcohol consumption. Follow-up examination 6 month after the event did not reveal the development of a psychiatric d…

Chromosomal damage in human leukocytes induced by lysergic acid diethylamide.

Addition of lysergic acid diethylamide to cultured human leukocytes resulted in a marked increase of chromosomal abnormalities. The distribution of chromosome breaks deviated significantly from random, with an accumulation of aberrations in chromosome No. 1. Cytogenetic investigation of a patient extensively treated with this drug over a 4-year period for paranoid schizophrenia showed a similar increase in chromosomal damage.

Ocular effects of diacetyl morphine and lysergic acid diethylamide in rabbit.

Intravenous lysergic acid diethylamide (LSD) given to rabbits in doses from 1 to 100 mug per kilogram of body weight produced a dose-related increase in intraocular pressure and outflow facility. Minor changes in systemic blood pressure were observed, but respiration rate was accelerated, and mydriasis became pronounced at higher doses. Diacetyl morphine (heroin) was given intravenously in doses from 0.1 to 2 mg. per kilogram of body weight. A dose-related decrease in intraocular pressure and an increase in outflow facility was found. A dose-related miosis was observed and at higher doses resp…

Acute effects of lysergic acid diethylamide (LSD) on resting brain function.

Lysergic acid diethylamide (LSD) is a potent hallucinogenic substance that was extensively investigated by psychiatrists during the 1950s and 1960s. Researchers were interested in the unique effects induced by this substance, some of which resemble symptoms seen in schizophrenia. Moreover, during that period LSD was studied and used for the treatment of several mental disorders such as depression, anxiety, addiction and personality disorders. Despite this long history of research, how LSD induces its specific effects on a neuronal level has been relatively unclear. In recent years there has be…
