The spread of antibiotic resistance genes (ARGs) in wastewater treatment systems poses a significant public health concern, yet the role of bacteriophages (phages), particularly temperate phages, in mediating horizontal gene transfer (HGT) of ARGs under antibiotic stress remains poorly understood. This study investigated the effects of escalating ciprofloxacin (CIP; 0-200 μg/L)-selected as a representative antibiotic due to its frequent occurrence and persistence in wastewater-on phage lysogeny-lysis dynamics and phage-mediated ARG transfer in a laboratory-scale activated sludge reactor. The co-selection of antibiotic resistance genes (ARGs) and metal resistance genes (MRGs) by heavy metals poses significant ecological risks. However, the contribution of bacteriophages (phages), particularly temperate phages, to this process via horizontal gene transfer (HGT) remains poorly understood. Here, we integrated metagenomics, metaviromics, and metatranscriptomics to investigate the impact of escalating Cu(II) concentrations (0.05-20.00 mg/L) on phage lifestyle dynamics and the dissemination of resistance genes in an activated sludge reactor. Our results revealed that phage-mediated H

Mycobacteriophage Mcgavigan could be a promising candidate for use as a preventative agent against infections with Mycobacterium avium subsp. paratuberculosis. Bioinformatic analysis of the Mcgavigan genome revealed the presence of an operon containing a "Bxb1-like" repressor. The operon may have been acquired by the phage through horizontal gene transfer with a Bxb1-like mycobacteriophage in its evolutionary past. We sought to investigate the function of the acquired repressor as a potential regulator of lysogeny or as a source of heterotypic superinfection immunity. Recombineering with CRISP

UNLABELLED: Temperate bacteriophages (phages) are common features of bacterial genomes and can act as self-amplifying biological weapons, killing susceptible competitors and thus increasing the fitness of their bacterial hosts (lysogens). Despite their prevalence, however, the key characteristics of an effective temperate phage weapon remain unclear. Here, we use systematic mathematical analyses coupled with experimental tests to understand what makes an effective temperate phage weapon. We find that effectiveness is controlled by phage life history traits-in particular, the probability of lys

A total of 61 bacteriophages were isolated from 100 strains of Salmonella senftenberg. Six bacteriophages were selected for typing purposes which were specific for S. senftenberg. Five phages, SasL1 to SasL5 were morphologically similar; phage SasL6 was morphologically different from the others. These phages fall into two morphological groups none of which correspond to the known tailed enterobacterial phage species. Hence, two new phage species represented by SasL1 and SasL6 are proposed.

Although phage lambda represents a well studied biological systems, it has certain features that remain obscure. Among these is the function of the roughly one third of the phage genome dispensable for growth in the laboratory, yet retained despite undoubted pressure to economize. Here we report that these 'accessory' sequences contain two genes which are expressed during lysogeny, and encode host-cell envelope proteins. One of these is lom, the product of which is found in the bacterial outer membrane, and is homologous to virulence proteins of two other enterobacterial genera. The other gene

The Patescibacteriota, also known as the Candidate Phyla Radiation (CPR), represent a large lineage of ultrasmall bacteria with highly reduced genomes and obligate dependence on bacterial hosts. Although genomic analyses have revealed CRISPR-Cas and restriction-modification systems in many CPR genomes, no cognate bacteriophages (phages) have been isolated, leaving CPR-phage interactions unexplored. Nanosynbacter lyticus TM7x, the first cultivated CPR bacterium, grows episymbiotically on its host, Schaalia odontolytica XH001, in the human oral microbiome. Here, we identify Xhp1, an inducible pr

Bacteria form groups composed of cells and a secreted polymeric matrix that controls their spatial organization. These groups-termed biofilms-can act as refuges from environmental disturbances and from biotic threats, including phages. Despite the ubiquity of temperate phages and bacterial biofilms, live propagation of temperate phages within biofilms has not been characterized on cellular spatial scales. Here, we leverage several approaches to track temperate phages and distinguish between lytic and lysogenic host infections. We determine that lysogeny within Escherichia coli biofilms initial

We apply mathematical modelling to explore bacteria-phage interaction mediated by condition-dependent lysogeny, where the type of the phage infection cycle (lytic or lysogenic) is determined by the ambient temperature. In a natural environment, daily and seasonal variations of the temperature cause a frequent switch between the two infection scenarios, making the bacteria-phage interaction with condition-dependent lysogeny highly complex. As a case study, we explore the natural control of the pathogenic bacteria Burkholderia pseudomallei by its dominant phage. B. pseudomallei is the causative

Bacteriophages (phages) play a critical role in controlling bacterial populations, both in nature and as potential therapeutic agents. Their ability to replicate, compete against each other, and eradicate target cell populations is usually understood through a number of 'life history parameters', traditionally measured by population-level assays, which implicitly average the parameter's value across a large number of infection events. Recent experiments suggest that bacteriophage life history parameters are subject to considerable stochasticity, raising the question of whether experimental and

Temperate bacteriophages (phages) are ubiquitous viruses that co-evolve with their bacterial hosts. They are defined by their ability to undergo two distinct life cycles: the lytic cycle, in which the phage produces more viral copies and kills the host, and the lysogenic cycle, in which the temperate phage exists as a prophage in the host. Temperate phages have long served as fundamental models in microbiology, genetics and evolutionary biology research, and their life cycles are among the most thoroughly characterized in virology. Historically, the phage life cycle was viewed primarily throug

UNLABELLED: Quorum sensing is a cell-to-cell communication process bacteria use to orchestrate collective behaviors. Quorum sensing involves the production, release, and detection of extracellular signal molecules called autoinducers. Some temperate phages can monitor bacterial autoinducers, enabling them to track the abundance of potential host cells in the vicinity. Quorum-sensing-responsive phages can preferentially launch the transition from lysogeny to lytic replication at high cell density, presumably maximizing transmission. Once the phage lytic program is enacted, if nearby host cells

Temperate bacterial viruses are commonly thought to favor vertical (lysogenic) transmission over horizontal (lytic) transmission when the virion-to-host-cell ratio is high and available host cells become scarce. In P22-infected Salmonella Typhimurium populations, however, we find that host subpopulations become lytically consumed despite high phage-to-host ratios that would normally favor lysogeny. These subpopulations originate from the proliferation of P22-free siblings that spawn off from P22-carrier cells from which they cytoplasmically inherit P22-borne superinfection exclusion factors (S

Silver is commonly included in a range of household and medical items to provide bactericidal action. Despite this, the chemical fate of the metal in both mammalian and bacterial systems remains poorly understood. Here, we applied a metallomics approach using X-ray absorption spectroscopy (XAS) and size-exclusion chromatography hyphenated with inductively coupled plasma mass spectrometry (SEC-ICP-MS) to advance our understanding of the biochemical fate of silver ions in bacterial culture and cells, and the chemistry associated with these interactions. When silver ions were added to lysogeny br

In El Tor biotype strains of toxigenic Vibrio cholerae, the CTXϕ prophage often resides adjacent to a chromosomally integrated satellite phage genome, RS1, which produces RS1ϕ particles by using CTX prophage-encoded morphogenesis proteins. RS1 encodes RstC, an antirepressor against the CTXϕ repressor RstR, which cooperates with the host-encoded LexA protein to maintain CTXϕ lysogeny. We found that superinfection of toxigenic El Tor strains with RS1ϕ, followed by inoculation of the transductants into the adult rabbit intestine, caused elimination of the resident CTX prophage-producing nontoxige

Obligately lytic (virulent) phages always lyse host cells to release progeny viruses, while temperate phages can either lyse their hosts or integrate into host genomes as prophages, forming lysogens. There is a rich history of work studying the relative advantages and disadvantages of these two phage life history strategies, but little of this work has addressed the spatial constraints common to biofilm environments. We developed a live imaging system to track lytic infections, lysogenic infections, and uninfected cells at single-cell resolution within three-dimensional Escherichia coli biofil

Telomere phages are a small group of temperate phages, whose prophages replicate as a linear plasmid with covalently closed ends. They have been isolated from some Enterobacteriaceae and from bacterial species living in aquatic environments. Phage PY54 was the first Yersinia (Y.) enterocolitica telomere phage isolated from a nonpathogenic O:5 strain, but recently a second telomeric Yersinia phage (vB_YenS_P840) was isolated from a tonsil of a wild boar in Germany. Both PY54 and vB_YenS_P840 (P840) have a siphoviridal morphology and a similar genome organization including the primary immunity r

Currently, there is a lack of bacterial biomarkers indicative of exposure to ionizing radiation (IR). IR biomarkers have applications for medical treatment planning, population exposure surveillance, and IR sensitivity studies. In this study, we compared the utility of signals originating from prophages and the SOS regulon as biomarkers of IR exposure in the radiosensitive bacterium Shewanella oneidensis. Using RNA sequencing, we demonstrated that 60 min after exposure to acute doses of IR (40, 1, 0.5, and 0.25 Gy), the transcriptional activation of the SOS regulon and the lytic cycle of the T
