Dehydroepiandrosterone and estrone 17-ketosteroid reductases in MCF-7 human breast cancer cells.

The identification of several steroid-transforming enzymes within human breast cancers has led to speculation that the growth of some hormone-responsive tumors might be mediated in part by intracellularly derived estrogens. Reports that MCF-7 human breast cancer cells can transform both estrone (E1)1 to estradiol (E2) and dehydroepiandrosterone (DHEA) to the estrogenic steroid 5-androstenediol (AED), have prompted us to investigate the 17-ketosteroid reductase activities (17-KSR's) which mediate these potentially important reactions. Enzyme assays were performed by quantifying the amounts of [

Chemicals in plastics differentially affect the transcriptome of MCF-7 breast cancer cells.

Humans are exposed to chemicals that leach from plastics, yet many remain data-poor and lack toxicological evaluation. High-throughput transcriptomics (HTTr) provides a scalable way to screen chemicals and generate mechanistic insights relevant to human health risk assessment. We applied HTTr in MCF-7 breast cancer cells to assess chemicals across several concentrations (0.001-50 µM) used in plastics and dyes. Transcriptomic points of departure (tPODs) were derived from general gene perturbations, including pathway-level and estrogen receptor α (ERα)-specific changes. We also used transcriptom

Antitumor activity of bimetallic silver/gold nanoparticles against MCF-7 breast cancer cells.

Breast cancer poses a global threat with rising incidence and high mortality. Conventional treatments, including chemotherapy, radiation, surgery, and immunotherapy, have side effects, such as resistance issues and adverse effects due to genetic mutations. Meanwhile, noble metal nanoparticles (NPs) synthesized using environmentally friendly methods offer alternative treatments. Bimetallic gold (Au) and silver (Ag) NPs, using natural compounds like starch as stabilizers, enhance biomedical applications, including breast cancer therapies. In this work, the optical properties, stability, and part

Brominated flame retardants effect in MCF-7 cells: Impact on vitamin D pathway.

Brominated flame retardants (BFRs) are persistent environmental pollutants, allowing a constant human exposure which carries several health risks, including the occurrence of breast cancer and vitamin D deficiency. Vitamin D inhibits cell growth and is negatively associated with breast cancer risk. The effect of BFRs in breast cancer and vitamin D pathway is still poorly understood. MCF-7 cells were treated with hexabromocyclododecane (HBCD), 2-ethylhexyl-2,3,4,5-tetrabromobenzoate (TBB), hexabromobenzene (HBB) and pentabromotoluene (PBT) using short and long-term exposure protocols. In vitro selection of aptamer S1 against MCF-7 human breast cancer cells.

Breast cancer is the most common female cancer. However, the known effective specific biomarkers for breast cancer are still scarce. Abnormal membrane proteins serve as ideal biomarkers for disease diagnoses, therapeutics and prognosis. Thus aptamers (single-stranded oligonucleotide molecules) with molecular recognition properties can be used as efficient tools to sort cells based on differences in cell surface architecture between normal and tumor cells. In this study, we aimed to screen specific aptamer against MCF-7 human breast cancer cells. Cell-SELEX process was performed to isolate apta

Estradiol preferentially enhances extracellular tissue plasminogen activators of MCF-7 breast cancer cells.

MCF-7 human breast cancer cells secrete two immunologic types of plasminogen activator, one related to urokinase, the other unrelated. We have now examined whether estrogen stimulation of secreted plasminogen activator activity reflects an increase in one or both types. Examined semiquantitatively by sodium dodecyl sulfate-polyacrylamide gel electrophoretic zymography, the conditioned media of control cells were seen to contain a major activator band (Mr approximately 54,000) immunologically related to urokinase and a barely discernible doublet (Mr approximately 64,000 and Mr approximately 68,

Effect of spider venom on cell apoptosis and necrosis rates in MCF-7 cells.

The purpose of this study was to examine the effects of antitumor activity of the venom from the spider Macrothele raven (Araneae, Hexathelidae) on the human breast carcinoma cell line, MCF-7. The spider venom affected cell viability in a dose- and time-dependent manner as observed by [(3)H]-methyl thymidine incorporation assay. Cytotoxicity changes in MCF-7 cells caused by the spider venom at concentrations of 10, 20, and 40 mug/mL were determined by lactate dehydrogenase release assay. Flow cytometry showed that the spider venom induced apoptosis and necrosis of MCF-7 cells at these concentr

Effect of bile acids and estradiol on thymidine incorporation into DNA in MCF-7 and MCF-10A breast cell lines.

Bile acid conjugates are found in human breast cyst fluid in average concentrations about 50-fold greater than those in blood. Because epidemiologic studies have linked colon and breast cancer and aberrant bile acid profiles are associated with colon cancer risk, we decided to study the influence of bile acid conjugates (glycochenodeoxycholic acid, glycodeoxycholic acid, glycocholic acid, and glycolithocholic acid) on thymidine incorporation into DNA in cancer (MCF-7) and noncancer (MCF-10A) human mammary cell lines. The two lines responded differently. In MCF-7, bile acids, except for glycoli

Identification of the aromatase in the breast carcinoma cell lines T47D and MCF-7.

The presence of estrogens in tumour cells is considered to be a critical factor for the development of the hormone-dependent forms of breast cancer. The last, rate-limiting step of estrogen biosynthesis is controlled by cytochrome P-450 type enzyme complex named aromatase. In the present study we determined and characterized the expression of aromatase mRNA in the breast carcinoma cell lines T47D and MCF-7. The expression was characterized by slot blot hybridization, reverse transcriptase-polymerase chain reaction technique and Northern hybridization analysis. Northern blotting revealed the pr

Estradiol esters can replace 17 beta-estradiol in the stimulation of DNA and esterase synthesis by MCF-7 cells: a possible role for the estrogen-sensitive MCF-7 cell esterase.

In this communication we extend our earlier observations on estrogen-sensitive carboxyl esterases in MCF-7 human breast cancer cells able to hydrolyze esters of estradiol. Using either estradiol acetate or p-nitrophenyl hexanoate as substrates, esterase activity was found to increase 2-3-fold in MCF-7 cells maintained in the presence of 10(-8) M estradiol. Following sucrose density centrifugation, over 85% of total esterase activity was found in the cytoplasmic fraction. No esterase activity was found in spent media from growing cells. By size exclusion chromatography, estradiol acetate estera

Transient peak of p53 induction persists at low radiation doses in MCF-7 cells.

BACKGROUND: The p53 protein is a central tumour suppressor regulating numerous genes involved in cell cycle control, DNA repair, and apoptosis. Mutations in p53 occur in over half of all cancers, disrupting these regulatory pathways and enhancing tumour progression. Many cell lines, including MCF-7 breast cancer cells, exhibit oscillatory p53 and MDM2 activity, a phenomenon linked to radiosensitivity and the cellular response to ionizing radiation. METHODS AND RESULTS: We investigated the dose-dependent dynamics of p53 activation in MCF-7 cells exposed to γ-radiation in the dose range of 0.1

Design, synthesis and anticancer evaluation of naphthalen-1-yloxyacetamide derivatives against MCF-7 cells.

Herein, a set of naphthalen-1-yloxyacetamide-tethered 2,3-disubstituted acrylamide conjugates was synthesized in a good yield via the reaction of naphthalene-1-yloxyacetohydrazide and an equimolar amount of respective ethyl 2,3-disubstituted acrylate ester in pure ethanol with a catalytic base of fused sodium acetate. All conjugates were assessed for their anti-proliferative activity against the MCF-7 Br Ca cell line. The results found that conjugate 5d with 3-(4-methoxyphenyl)-2-phenylacrylamide attached to the naphthalen-1-yloxyacetamide moiety demonstrated potent cytotoxic activity against

New curcumin derivative induces ferroptosis in MCF-7 cells through activating SLC7A11/GPX4 axis.

Previous experiments have revealed that curcumin exerts potential antitumor effect by inducing apoptosis and ferroptosis of tumor cells. However, its low solubility and bioavailability, as well as fast metabolism limit its clinical use. The structural modification of curcumin is beneficial for the discovery of potential candidate drugs for cancer treatment. Here, three new series of curcumin derivatives including 25 compounds were synthesized at active sites on benzene ring and β-diketone moiety. Further antiproliferative activities against five cancer cell lines (Hela, A549, HepG2, MCF-7 and

Oleuropein Induces Cytotoxicity and Peroxiredoxin Over-expression in MCF-7 Human Breast Cancer Cells.

BACKGROUND/AIM: As a fundamental staple of the Mediterranean diet, olive oil has long been recognized for its health benefits, including its ability to reduce cardiovascular and neurological disease. Oleuropein is the primary phenolic chemical found in all parts of the olive tree, especially in the leaves and fruit. Oleuropein exhibits anti-inflammatory and antioxidant properties, and has been associated with cancer inhibition in various animal and cell models. We investigated the effects of oleuropein on the MCF-7 human breast cancer cell line, and compared it to the non-cancerous MCF-10A bre

A Bacterial Metabolite, Compound K, Induces Programmed Necrosis in MCF-7 Cells via GSK3β.

Ginsenosides, the major active component of ginseng, are traditionally used to treat various diseases, including cancer, inflammation, and obesity. Among these, compound K (CK), an intestinal bacterial metabolite of the ginsenosides Rb1, Rb2, and Rc from Bacteroides JY-6, is reported to inhibit cancer cell growth by inducing cell-cycle arrest or cell death, including apoptosis and necrosis. However, the precise effect of CK on breast cancer cells remains unclear. MCF-7 cells were treated with CK (0-70 micrometer) for 24 or 48 h. Cell proliferation and death were evaluated by 3-(4,5-dimethylthi

Expression and inducibility of UDP-glucuronosyltransferase 1As in MCF-7 human breast carcinoma cells.

UDP-glucuronosyltransferases (UGTs) are conjugation enzymes, which are regulated in a tissue-specific manner by endogenous and environmental factors. In this study, we focused on UGT1A isoforms broadly expressed in hepatic and extrahepatic tissues and examined the expression and inducibility of UGT1As (UGT1A1 and UGT1A3-1A10) in MCF-7 cells (human breast carcinoma cell line). Reverse transcription polymerase chain reaction (RT-PCR) analysis demonstrated that UGT1A1, UGT1A6 and UGT1A9 mRNAs as well as the mRNAs of transcriptional regulators (AhR, aryl hydrocarbon receptor; Arnt, AhR nuclear tra
