alpha2-macroglobulin in late-onset Alzheimer's disease.

alpha2-macroglobulin (alpha(2)M) is an abundant plasma protein similar in structure and function to a group of proteins called alpha-macroglobulins. alpha(2)M is also produced in the brain where it binds multiple extracellular ligands and is internalized by neurons and astrocytes. In the brain of Alzheimer's disease (AD) patients, alpha(2)M has been localized to diffuse amyloid plaques. alpha(2)M also binds soluble beta-amyloid, of which it mediates degradation. However, an excess of alpha(2)M can also have neurotoxic effects. Based on genetic evidence, is now recognized as one of the two conf

Expression of human alpha 2-macroglobulin cDNA in baby hamster kidney fibroblasts: secretion of high levels of active alpha 2-macroglobulin.

Human alpha 2-macroglobulin (alpha 2M) is a unique 720-kDa proteinase inhibitor with a broad specificity. Unlike most other proteinase inhibitors, it does not inhibit proteolytic activity by blocking the active site of the proteinase. During complex formation with a proteinase, alpha 2M entraps the proteinase molecule in a reaction that involves large conformational changes in alpha 2M. We describe the molecular cloning of alpha 2M cDNA from the human hepatoblastoma cell line HepG2. The cDNA was subcloned under control of the adenovirus major late promoter in a mammalian expression vector and

Effect of α2‑macroglobulin in the early stage of jaw osteoradionecrosis.

Advanced osteoradionecrosis (ORN) is one of the most serious complications in patients with head and neck cancer, resulting in poor prognosis. Numerous studies have therefore focused on the pathogenesis and interventions of ORN early stage. The present study aimed to investigate whether α2‑macroglobulin (α2M) could prevent early‑stage jaw osteoradionecrosis caused by radiotherapy (RT). Following local injection of α2M, a single dose of 30 Gy was delivered to rats for pathological exploration. For 28 days, the irradiated mandible and soft tissues were examined for potential changes. Furthermore

Alpha 2-macroglobulin and pregnancy zone protein. Serum levels, alpha 2-macroglobulin receptors, cellular synthesis and aspects of function in relation to immunology.

alpha 2-Macroglobulin, a 720 kDa tetramer, and Pregnancy Zone Protein, a 360 kDa dimer, are strongly homologous glycoprotein proteinase inhibitors of human plasma. alpha 2M and PZP monomers contain two regions of decisive functional importance i.e. the bait- and the thiol ester regions. The bait region serves as a substrate to a variety of proteinases, and cleavage in this domain causes a conformational change of the molecule. In addition, PZP forms tetramers when reacted with proteinase. As a result of the conformational change the thiol ester, which is hidden in the native structure, becomes

Isolation and Endopeptidase Inhibition of Human α 2-Macroglobulin.

The α2-macroglobulin (α2M) family comprises pan-peptidase inhibitors with shared structural characteristics and a common mechanism of action that play vital roles in various biological processes. This protocol describes the isolation and purification of the human homolog (hα2M) and outlines reaction conditions for evaluating its peptidase-inhibition activity in front of different substrates. It is valid for other homologues from both unicellular and multicellular organisms, providing a foundation for future research on physiological functions and potential biotechnological applications.

Human α2-Macroglobulin: Architecture, Mechanisms, and Functional Implications.

α2-Macroglobulin (α2M) is a large glycoprotein and one of the most abundant proteins found in the plasma of vertebrates and in the hemolymph of invertebrates. It performs several important functions in the innate immune system, serving as a pan-protease inhibitor and contributing to essential processes, such as cytokine and hormone transport. It also triggers diverse cellular responses, critical for the functioning of both eukaryotic and some prokaryotic biological systems. Here, we focus on the human α2-macroglobulin (hα2M) molecule, compiling relevant information from recent studies on its s

Molecular form and concentration of serum α2-macroglobulin in diabetes.

α2-Macroglobulin is a highly abundant serum protein involved in the development of atherosclerosis and cardiac hypertrophy. However, its circulating molecular form and exact concentrations in human health/diseases are not known. Blue native-polyacrylamide gel electrophoresis of human serum was used to confirm the native conformation of α2-macroglobulin. We created an enzyme-linked immunosorbent assay suitable for quantifying its circulating molecular form and undertook a cross-sectional study to measure its serum levels in 248 patients with diabetes mellitus and 59 healthy volunteers. The pred

Serum monomeric α2-macroglobulin as a clinical biomarker in diabetes.

OBJECTIVE: Despite the number of potential biomarker proteins for diabetes, very few of them have proven useful as clinically beneficial indicators, because of the technical difficulties associated with their identification among highly abundant serum proteins. We attempted to identify a protein with distinguishable expression in human diabetes. METHODS: We applied a highly efficient strategy for the purification of endogenous low abundance proteins from diabetic and non-diabetic serum samples. Extracted sera were fractionated by SDS-PAGE and protein bands were isolated and analyzed by mass s

Alpha-2-macroglobulin and hepatic fibrosis. Diagnostic interest.

Alpha-2-macroglobulin (A2M) is a proteinase inhibitor. Cells synthesizing A2M are in first-order hepatocytes and in second-order activated Ito cells (in culture starting at day 4-5 after seeding). This study was undertaken in 525 alcoholic patients with different histological stages of alcoholic liver disease to assess if the A2M could improve the diagnostic value of PGA index for detection of cirrhosis or fibrosis among drinkers, particularly in patients without clinical symptoms of liver failure and portal hypertension, and to assess the specific correlation of serum A2M with the score of li

Alpha2-macroglobulin deficiency in a patient with Ehlers-Danlos syndrome.

A new genetic defect, alpha2-macroglobulin deficiency, was found in a patient with Ehlers-Danlos syndrome (EDS). Other members of the family of five exhibiting this abnormality were the mother and one sister. All members, including the patient, had normal serum albumin and alpha1-antitrypsin levels. The deficiency, reported here for the first time, appears to be inherited by an autosomal co-dominant mode. Statistical evaluation of the dihybrid crosses for independent assortment between EDS and hypo-alpha2-macroglobulinemia showed a probability of 0.7 to 0.75. However, a possible link between E

The Role of Various Autoantibodies and Alpha2-Macroglobulin in Patients with Hashimoto Disease: Does the Presence of Elevated Antibodies Correlate with Alpha 2-Macroglobulin Levels in Hashimoto Disease?

INTRODUCTION: Autoimmune Thyroiditis (AIT) is caused by defects in the immune system in people with a genetic predisposition to the disease. The most prevalent type of autoimmune thyroiditis is Hashimoto's thyroiditis (HT). The present article reviews the possible relationship between α2-macroglobulin levels and autoantibodies in patients suffering from Hashimoto's disease. METHODS: A total of 170 patients with Hashimoto's disease, categorized into subclinical (96 patients) and manifest (74 patients) forms, were enrolled in the study. The control group comprised 65 individuals without thyroid

Alpha2-macroglobulin polymorphisms in Italian sporadic and familial Alzheimer's disease.

A 5-bp deletion and a Val1000 polymorphism at the alpha(2)-macroglobulin (A2M) gene have recently been reported to be associated with late onset Alzheimer's disease (AD). As recently it has been suggested that the effect of the A2M gene on AD susceptibility may be limited to certain populations or families, we analyzed the segregation of A2M and apolipoprotein E polymorphisms in Italian sporadic and familial AD. We analyzed the two polymorphisms in a total of 346 subjects including 98 controls by polymerase chain reaction-restriction fragment length polymorphism method. Our data do not confirm

Lack of association between alpha2-macroglobulin polymorphisms and Alzheimer's disease.

This study was undertaken to investigate the role of two alpha2-macroglobulin (A2M) polymorphisms, an intronic 5-bp deletion and Ile1000Val, in the development of Alzheimer's disease (AD) and to evaluate the interaction between the apolipoprotein E (APOE) and A2M polymorphisms. The A2M polymorphisms were screened by using polymerase-chain-reaction-based assays in 555 white late-onset AD cases and 446 controls. The gentoype distributions of the 5-bp deletion and Ile1000Val polymorphisms were comparable between cases and controls (P = 0.158 and P = 0.148, respectively). Likewise, there was no si

Alpha2-macroglobulin polymorphisms in Alzheimer's disease and dementia with Lewy bodies.

Dementia with Lewy bodies (DLB) is the second most common cause of dementia in the elderly after Alzheimer's disease (AD). The apolipoprotein E gene (APOE) is a major risk factor, but can only account for approximately 50% of AD cases. Whole genome scanning in late-onset AD families has suggested that a locus on chromosome 12 may contribute significantly to disease development. Recently the alpha2-macroglobulin gene (A2M) on chromosome 12 has been suggested as a candidate locus for AD. We therefore determined the influence of two polymorphisms in A2M, a pentanucleotide deletion 5' to the bait

Detection of alpha-macroglobulin in the heart of mice infected with Trypanosoma cruzi.

The aim of this work was to investigate the presence of alpha-macroglobulin (AM) in the heart of mice during acute experimental Chagas' disease and to study its localization as related to the presence of the parasite and/or to their antigens. Frozen heart tissue sections obtained from Swiss albino male mice at different days postinfection with Trypanosoma cruzi were examined for triple immunofluorescence in response to parasite antigen (green), AM (red), and nuclei (blue) from both cells. AM was found in the heart of all the infected animals studied. Parasites were seen arranged in nests insid

Mechanical unfolding of alpha2-macroglobulin molecules with atomic force microscope.

alpha2-Macroglobulin was derivatized with a sulfhydryl cross-linker and sandwiched between a mica substrate and a silicon nitride tip, both coated with gold, of an atomic force microscope and force curve measurement was carried out. An extensive downward deflection of the cantilever was observed in the retracting realm of the curve, when and only when the substrate was covered with the derivatized protein. The result was interpreted in terms of the mechanical stretching and unfolding of a single or a few protein molecules.
