Succinate dehydrogenase inhibitors (SDHIs) are crucial fungicides, yet major commercial SDHIs exhibit suboptimal efficacy against the devastating rice blast pathogen Magnaporthe oryzae. This work introduces an alkene linker in the hydrophobic tail region of SDHIs through chain elongation and affords a library of 32 novel stereoisomeric compounds. This structural modification was predicated on the hypothesis that exploiting configurational isomerism (E vs Z) could optimize the spatial arrangement for enhanced binding. Indeed, the alkene-functionalized SDHIs demonstrated significantly enhanced a

Rice blast, caused by the fungal pathogen Magnaporthe oryzae, is one of the most devastating diseases for rice crops, significantly affecting crop yield and quality. During the infection process, M. oryzae secretes effector proteins that help in hijacking the host's immune responses to establish infection. However, little is known about the interaction between the effector protein AvrPik-D and the host protein Pikh, and how AvrPik-D increases disease severity to promote infection. In this study, we show that the M. oryzae effector AvrPik-D interacts with the zinc finger-type transcription fact

The Magnaporthe oryzae snodprot1 homolog (MSP1), secreted by M. oryzae, is a cerato-platanin family protein. msp1-knockout mutants have reduced virulence on barley leaves, indicating that MSP1 is required for the pathogenicity of rice blast fungus. To investigate the functional roles of MSP1 and its downstream signaling in rice, recombinant MSP1 was produced in Escherichia coli and was assayed for its functionality. Application of MSP1 triggered cell death and elicited defense responses in rice. MSP1 also induced H2O2 production and autophagic cell death in both suspension-cultured cells and r

This study presents the synthesis, characterisation, and photocatalytic antifungal performance of a Cu+/Cu2+-TiO2 nanocomposite designed to control rice blast disease caused by Magnaporthe oryzae. The heterostructure was fabricated through a chemical precipitation process in which TiO2 was first doped with Cu2+ ions, followed by partial reduction to Cu+ to form CuI. X-ray Photoelectron Spectroscopy (XPS) confirmed the coexistence of both Cu+ and Cu2+ oxidation states. A notable feature of this material is its Cu+/Cu2+ redox cycling, which enhances charge separation during photocatalysis and su

The global efficacy of succinate dehydrogenase inhibitors (SDHIs), a leading class of fungicides, is increasingly threatened by resistance, driven largely by their shared binding mode at the conserved ubiquinone-binding (Q) site. This challenge is particularly acute for rice blast, as no SDHI is registered for this disease and available compounds offer limited protection while remaining vulnerable to Q-site mutations. In this study, we identified a previously unrecognized, pathogen-adaptive pocket within the SDH complex of Magnaporthe oryzae, which is structurally distinct from the canonical Q

Rice blast, caused by Magnaporthe oryzae, is one of the most destructive fungal diseases in rice, resulting in major economic losses worldwide. Genetic and genomic studies have identified key genes and proteins, such as AvrPik variants and MAX proteins, that are crucial for the pathogen's virulence. These effector proteins interact with specific alleles of the Pik gene family on rice chromosome 11, modulating the host's immune response. In this study, we investigated 35 plant-derived metabolites known for their antifungal properties as potential fungicides against M. oryzae. Using molecular do

Magnaporthe grisea, the causal agent of rice blast, forms a dome-shaped melanized infection structure, an appressorium, to infect its host. Environmental cues that induce appressorium formation by this fungus include hydrophobicity and hardness of contact surface, and chemicals from the host. To determine if the calcium/calmodulin-dependent signaling systems are involved in appressorium formation in M. grisea, we tested the effects of the calcium chelator, calcium ionophore, diverse intracellular calcium modulators, and calmodulin antagonists on appressorium formation. Several calcium modulato

Appressorium is an infection structure of the phytopathogenic fungus Magnaporthe grisea. Analysis of gene expression profiles of appressorium development provides insight into the molecular basis of pathogenicity and control of this fungal plant disease. A cDNA array representing 2927 unique genes based on a large EST (expressed sequence tag) database of M. grisea strain Y34 was constructed and used to profile the gene expression patterns at mycelium and appressorium maturation stages. Compared with mycelia, 55 up-regulated and 22 down-regulated genes were identified in mature appressoria. Amo

The SUN family protein MoSun4 in Magnaporthe oryzae has been previously implicated in mitophagy and has potential as a target for reducing rice blast, but its role as a secreted protein remains poorly understood. In this study, signal peptide prediction and yeast secretion assays confirmed MoSun4 signal peptide function, and co-localization revealed the localization of the extra-invasive hyphal membrane (EIHM)-associated apoplastic compartment or matrix, establishing MoSun4 as a secreted protein. In addition, deletion of the MoSUN4 gene reduced the hyphal growth, conidiation and virulence of M

Endocytosis plays key roles during infection of plant-pathogenic fungi, but its regulatory mechanisms are still largely unknown. Here, we identified a putative endocytosis-related gene, PAL1, which was highly expressed in appressorium of Magnaporthe oryzae, and was found to be important for appressorium formation and maturation. Deletion of PAL1 significantly reduced the virulence of M. oryzae due to defects in appressorial penetration and invasive growth in host cells. The Pal1 protein interacted and colocalized with the endocytosis protein Sla1, suggesting it is involved in endocytosis. The

To probe salicylic acid (SA)-induced sequential events at translational level and factors associated with SA response, we conducted virulence assays and proteomic profiling analysis on rice resistant and susceptible cultivars against Magnaporthe oryzae at various time points after SA treatment. The results showed that SA significantly enhanced rice resistance against M. oryzae. Proteomic analysis of SA-treated leaves unveiled 36 differentially expressed proteins implicated in various functions, including defense, antioxidative enzymes, and signal transduction. Majority of these proteins were i

The rice blast fungus Magnaporthe grisea infects its host by forming a specialized infection structure, the appressorium, on the plant leaf. The enormous turgor pressure generated within the appressorium drives the emerging penetration peg forcefully through the plant cuticle. Hitherto, the involvement of cutinase(s) in this process has remained unproven. We identified a specific M. grisea cutinase, CUT2, whose expression is dramatically upregulated during appressorium maturation and penetration. The cut2 mutant has reduced extracellular cutin-degrading and Ser esterase activity, when grown on

Rice blast disease, caused by Magnaporthe oryzae, is a major threat to global rice production, necessitating sustainable disease management strategies. Compared to chemical pesticides, biocontrol agents, such as beneficial microbe antagonists, offer a sustainable approach to naturally inhibit plant pathogens. This study evaluates the biocontrol potential of Bacillus subtilis UD1022 against M. oryzae through both direct antagonism and volatile-mediated inhibition. In dual culture assays, UD1022 significantly inhibited fungal growth. Furthermore, a stacking plate assay demonstrated that UD1022 p

Histone acetylation and deacetylation play an essential role in the epigenetic regulation of gene expression. Histone deacetylases (HDAC) are a group of zinc-binding metalloenzymes that catalyze the removal of acetyl moieties from lysine residues from histone tails. These enzymes are well known for their wide spread biological effects in eukaryotes. In rice blast fungus, Magnaporthe oryzae, MoRPD3 (an ortholog of Saccharomyces cerevisiae Rpd3) was shown to be required for growth and development. Thus in this study, the class I HDAC, MoRpd3 is considered as a potential drug target, and its 3D s

Incorporation of plant defense activators is an innovative approach to development of an integrated strategy for the management of turfgrass diseases. The effects of salicylic acid (SA), benzothiadiazole (BTH, chemical analog of SA), jasmonic acid (JA), and ethephon (ET, an ethylene-releasing compound) on development of gray leaf spot in perennial ryegrass (Lolium perenne L.) caused by Magnaporthe oryzae were evaluated. Gray leaf spot disease incidence and severity were significantly decreased when plants were treated prior to inoculation with SA, BTH, and partially by ET but not by JA. Accumu

Rice blast, caused by Magnaporthe oryzae, severely threatens global rice production. This study deciphers the molecular basis of blast resistance through comparative transcriptomics of resistant (403) and susceptible (402) rice cultivars. Resistant plants exhibited limited symptom development, accompanied by targeted transcriptional reprogramming with only 393 differentially expressed genes (DEGs) versus 1,336 DEGs in susceptible plants. Kyoto Encyclopedia of Genes and Genomics (KEGG) analysis revealed preferential activation of defense-centric pathways in resistant cultivars, including diterp

Carbonic anhydrases (CAs) are a class of zinc-containing metalloenzymes widely present in the biological world, catalyzing the reversible hydration of CO2 to form HCO3- and H+. These enzymes play essential roles in pH homeostasis, gas exchange, metabolic regulation, and virulence expression in pathogens. In fungi, CAs mainly belong to the α- and β-classes and have undergone extensive diversification during evolution. In plant pathogenic fungi, the functions of CAs have extended beyond traditional metabolic roles, evolving into key "environmental adaptation and virulence regulatory factors." Th

Rice blast disease, caused by the ascomycete fungus Magnaporthe oryzae (syn. Pyricularia oryzae), poses a severe threat to global rice production. Southern China, a major rice-growing region characterized by diverse agroecological conditions, faces substantial challenges from blast disease, yet our understanding of the genetic structure of M. oryzae populations in this region remains limited. Here, we analyzed 885 M. oryzae strains from 18 nurseries across four rice ecological regions in Southern China using a panel of genome-wide SNP markers. Phylogenetic and principal component analyses reve

Biological control is a promising measure in the control of plant disease. In the present study, we isolated 13 endophytic strains from Angelica dahurica. Among them, an endophytic strain which was named GYLH001 exhibited remarkable activity against Magnaporthe oryzae. 16S rRNA sequence analysis, biochemical and physiological proved that it is Bacillus tequilensis. The sterilized culture filtrate of GYLH001 can inhibit the growth of M.oryzae, which suggests the presence of secondary metabolites. Proved by experiment, GYLH001 can produce cellulase, protease, gelatinase, indole-3-acetic acid and

Phyllosphere-microbe-plant interactions underlying nanoscale hydroxyapatite (nHAP)-mediated disease control remain poorly understood. Here, foliar application of nHAP (100-400 mg/L) was evaluated for controlling rice blast caused by Magnaporthe oryzae under greenhouse conditions. The optimal treatment (200 mg/L) reduced disease severity by 42.6% and outperformed ionic Ca and P controls, as well as a commercial fungicide. Mechanistically, nHAP exhibited superior foliar retention (6.88%), enhancing local Ca and P bioavailability, and reshaping the phyllosphere microbiome by enriching beneficial
