BACKGROUND: The mosquito vectors of Plasmodium spp. have largely been overlooked in studies of ecology and evolution of avian malaria and other vertebrates in wildlife.
METHODS: Plasmodium DNA from wild-caught Coquillettidia spp. collected from lowland forests in Cameroon was isolated and sequenced using nested PCR. Female Coquillettidia aurites were also dissected and salivary glands were isolated and microscopically examined for the presence of sporozoites.
RESULTS: In total, 33% (85/256) of mosquito pools tested positive for avian Plasmodium spp., harbouring at least eight distinct parasi

One of many uncertainties concerning the epidemiology of avian malaria in wild bird populations is the age at first infection. While nestlings, being naked and presumably immunologically naïve would seem a likely stage of first infection, most age-stratified prevalence studies have not examined the nestling cohort, whereas those that have use relatively insensitive blood smear examination to diagnose infection. In the study presented here, we used sensitive nested polymerase chain reaction methods to screen blood samples from 195, 14-day-old blue tit (Cyanistes caeruleus) nestlings for avian m

Interest in the schizontocidal activity of diaminodiphenylsulfone (DDS) and in its possible use in malaria chemoprophylaxis has recently been revived. Studies on its toxicity and action in avian and simian malaria show that its chronic toxicity in monkeys is similar to that of the established antimalarials and that it lacks causal prophylactic, gametocidal and sporontocidal activity against Plasmodium gallinaceum.DDS is also inferior to the established antimalarials in its schizontocidal activity against P. gallinaceum, while against P. knowlesi its activity is of the same order as that of pro

Infectious diseases increasingly play a role in the decline of wildlife populations. Vector-borne diseases, in particular, have been implicated in mass mortality events and localized population declines are threatening some species with extinction. Transmission patterns for vector-borne diseases are influenced by the spatial distribution of vectors and are therefore not uniform across the landscape. Avian malaria is a globally distributed vector-borne disease that has been shown to affect endemic bird populations of North America. We evaluated shared habitat use between avian malaria vectors,

BACKGROUND: Various ecological and physiological mechanisms might influence the probability that two or more pathogens may simultaneously or sequentially infect a host individual. Concurrent infections can have important consequences for host condition and fitness, including elevated mortality risks. In addition, interactions between coinfecting pathogens may have important implications for transmission dynamics.
METHODS: Here, we explore patterns of association between two common avian pathogens (West Nile virus and avian malaria parasites) among a suburban bird community in Chicago, IL, USA

BACKGROUND: Knowledge on the temporal dynamics of host/vector/parasite interactions is a pre-requisite to further address relevant questions in the fields of epidemiology and evolutionary ecology of infectious diseases. In studies of avian malaria, the natural history of Plasmodium parasites with their natural mosquito vectors, however, is mostly unknown.
METHODS: Using artificial water containers placed in the field, we monitored the relative abundance of parous females of Culex pipiens mosquitoes during two years (2010-2011), in a population in western Switzerland. Additionally, we used mol

Pathogens have traditionally been studied in isolation within host systems; yet in natural settings they frequently coexist. This raises questions about the dynamics of co-infections and how host life-history traits might predict co-infection versus single infection. To address these questions, we investigated the presence of two parasites, a gut parasite (Isospora coccidians) and a blood parasite (Plasmodium spp.), in House Finches (Haemorhous mexicanus), a common passerine bird in North America. We then correlated these parasitic infections with various health and condition metrics, includin

Escape behaviour is the behaviour that birds and other animals display when already caught by a predator. An individual exhibiting higher intensity of such anti-predator behaviour could have greater probabilities of escape from predators. Parasites are known to affect different aspects of host behaviour to increase their own fitness. Vector-transmitted parasites such as malaria parasites should gain by manipulating their hosts to enhance the probability of transmission. Several studies have shown that malaria parasites can manipulate their vectors leading to increased transmission success. How

Infection and transmission of the avian malaria parasite Plasmodium gallinaceum in domestic chickens is associated with high economic burden and presents a major challenge to poultry industry in South East Asia. Development of drugs targeting both asexual blood stage parasites and sexual stages of the avian malarias will be beneficial for malaria treatment and eradication. However, current drugs recommended for treatment of the avian malaria parasites target specifically the asexual blood stage parasites, but have little or no impact to the gametocytes, the major target for development of tran

Avian malaria, caused by Plasmodium parasites, poses a significant threat to bird populations worldwide, particularly in vulnerable island ecosystems. Yet, progress in understanding avian malaria transmission dynamics has been hampered by the lack of molecular tools to quantify and sex the transmissible stages of the parasite: the male and female gametocytes. This challenge is compounded by the nucleated erythrocytes of avian hosts and the absence of an in vitro culture system, which have historically hindered the advancement of molecular approaches. Here, we develop and validate the first mol

Plasmodium relictum is the most widespread avian malaria parasite in the world. It is listed as one of the 100 most dangerous invasive species, having been responsible for the extinction of several endemic bird species, and the near-demise of several others. Here we present the first transcriptomic study focused on the effect of P. relictum on the immune system of its vector (the mosquito Culex quinquefasciatus) at different times post-infection. We show that over 50% of immune genes identified as being part of the Toll pathway and 30%-40% of the immune genes identified within the Imd pathway

One dead 6-wk-old male racing pigeon (Columba livia) was submitted for postmortem evaluation after presenting with weight loss, anorexia, dry shanks, dehydration, and lethargy. The bird belonged to a confined flock with 12 other pigeons raised by a hobbyist. Two pigeons in the flock reportedly had died with a history of similar clinical signs. On gross examination, the liver and the spleen were diffusely dark brown to black. Histopathology revealed moderate to large amounts of anisotropic, intracytoplasmic black pigment, compatible with hemozoin, in the spleen, liver, lung, and kidneys, with s

Haemosporidian parasites are increasingly being recognized as emerging threats to avian populations, with the potential to cause severe morbidity, leading to population decline, and even extinction in susceptible species. In February 2022, a Humboldt penguin (Spheniscus humboldti) housed at a zoo in northeastern Thailand died, later confirmed to be due to avian malaria infection. The same year, four more deaths occurred between July and November. This study aimed to (i) determine if these infections extended beyond the affected penguins to other avian species housed at the same or at other zoo

Two aspects of host infectiousness shape pathogen transmission and distribution but are underappreciated: the relationship between pathogen load and infectiousness, and variability in pathogen load within species. We quantified the relationship between host pathogen load (parasitemia) for avian malaria (Plasmodium relictum) and infectiousness for biting Culex quinquefasciatus mosquitoes with experimental infections in canaries (Serinus canaria). Using this relationship, we estimated the infectiousness of 17 bird species in 11 communities in Hawai'i and quantified the relative contributions of

Malaria parasites use vertebrate hosts for asexual multiplication and Culicidae mosquitoes for sexual and asexual development, yet the literature on avian malaria remains biased towards examining the asexual stages of the life cycle in birds. To fully understand parasite evolution and mechanism of malaria transmission, knowledge of all three components of the vector-host-parasite system is essential. Little is known about avian parasite-vector associations in African rainforests where numerous species of birds are infected with avian haemosporidians of the genera Plasmodium and Haemoproteus. H

Avian malaria (Plasmodium relictum) caused significant mortality in wild-caught Magellanic penguins (Spheniscus magellanicus) in 1986 at the Blank Park Zoo in Des Moines, Iowa (USA). In early winter, wild birds were captured off the southern coast of Chile and flown to Detroit, Michigan for a 38 day quarantine. After quarantine, 18 birds were dispersed to Lansing, Michigan, six to a facility in Maine, and 46 to Des Moines, Iowa. Upon arrival in Des Moines, several penguins became weak and inactive, had to be force-fed, and died after 2 days. Gross lesions at postmortem included splenomegaly, h

Research on the feeding preference of mosquitoes and the malaria parasites they transmit to vertebrate hosts is crucial for understanding the host-vector-parasite relationships in this complex system. In order to contribute to the growing data on these associations, and focusing on avian malaria, we collected mosquitoes at regular intervals between April and October for 2 years (2017 and 2019) in a deciduous forest and reedbeds. Collected engorged mosquitoes were identified morphologically. Total DNA was extracted from the abdomen and head-thorax parts of the vectors and was used to identify b

Vector-borne diseases in the United States have recently increased as a result of the changing nature of vectors, hosts, reservoirs, parasite/pathogens, and the ecological and environmental conditions. While most focus has been on mosquito-borne pathogens affecting humans, little is known regarding parasites of companion animal, livestock and wildlife and their potential mosquito hosts in the United States. This study assessed the prevalence of mature infections of Dirofilaria immitis and avian malaria parasites (Haemosporida) within urban mosquito (Diptera, Culicidae) communities in Oklahoma.

CASE HISTORY A little penguin (Eudyptula minor) of wild origin, in captivity at Wellington Zoo, became inappetent and lethargic in March 2013. Despite supportive care in the zoo's wildlife hospital, the bird died within 24 hours. CLINICAL FINDINGS Weight loss, dehydration, pale mucous membranes, weakness, increased respiratory effort and biliverdinuria were apparent on physical examination. Microscopic evaluation of blood smears revealed intra-erythrocytic stages of Plasmodium spp. and a regenerative reticulocytosis in the absence of anaemia. PATHOLOGICAL FINDINGS Post-mortem findings included

Plasmodium reticulum, the causative agent for avian malaria (a protozoan), and Dirofilaria immitis, the causative agent for canine heartworm (a filarial nematode), are 2 obligate parasites transmitted by mosquitoes. The objective of this project was to identify whether either parasite was present in Tennessee mosquitoes and to illustrate the need for collecting spatial and temporal vector-parasite data. During 2012, mosquitoes were collected from the East Tennessee Research and Education Center (ETREC) in eastern Tennessee and the Ames Plantation Research and Education Center (AMES) in western
