Optimization of NMR spectroscopy of encapsulated proteins dissolved in low viscosity fluids.

Comprehensive application of solution NMR spectroscopy to studies of macromolecules remains fundamentally limited by the molecular rotational correlation time. For proteins, molecules larger than 30 kDa require complex experimental methods, such as TROSY in conjunction with isotopic labeling schemes that are often expensive and generally reduce the potential information available. We have developed the reverse micelle encapsulation strategy as an alternative approach. Encapsulation of proteins within the protective nano-scale water pool of a reverse micelle dissolved in ultra-low viscosity non

Reconstitution of mammalian excision repair activity with mutant cell-free extracts and XPAC and ERCC1 proteins expressed in Escherichia coli.

Nucleotide excision repair in humans involves the coordinated actions of 8-10 proteins. To understand the roles of each of these proteins in excision it is necessary to develop an in vitro excision repair system reconstituted entirely from purified proteins. Towards this goal we have expressed in E. coli two of the 8 genes known to be essential for the excision reaction. XPAC and ERCC1 were expressed as fusion proteins with the Escherichia coli maltose binding protein (MBP) and purified to > 80% homogeneity by affinity chromatography. The purified proteins either as fusions or after cleavage f

Harnessing Affinity Tags for Recombinant Protein Purification: Concepts, Methods, and Troubleshooting.

Efficient purification of recombinant proteins is essential for both fundamental biochemical research as well as industrial biotechnology applications. The use of affinity tags has emerged as one of the most powerful and widely adopted method for protein purification. This paper contains recent advancements of design and functions of different affinity tags namely, polyhistidine (His-tag), glutathione S-transferase (GST), small ubiquitin-like modifier (SUMO) tag, maltose-binding protein (MBP), calmodulin-binding protein (CBP), intein-chitin-binding domain tag, N-utilization substance A tag, Ha

Effect of modification of sulfhydryl groups in soybean beta-amylase on the interaction with substrate and inhibitors.

Methyl 2,4-dinitrophenyl disulfide (MDPS) is shown to be an effective methanethiolating reagent for sulfhydryl groups in proteins via thiol-disulfide exchange reaction. It reacts with the two reactive sulfhydryl groups (SH1 and SH2) in soybean beta-amylase. A decrease of the enzymatic activity accompanies the methanethiolation of SH2. After complete methanethiolation of SH2, the modified enzyme still has 9% of the initial activity. Modification of SH2 with cyanide and iodoacetamide reduces the enzymatic activity to 65 and 2% of the initial activity, respectively. Apparently, the residual activ

Functional analysis and purification of a Pen-2 fusion protein for γ-secretase structural studies.

The 19-transmembrane, multisubunit γ-secretase complex generates the amyloid β-peptide (Aβ) of Alzheimer's disease (AD) by an unusual intramembrane proteolysis of the β-amyloid precursor protein. The complex, which similarly processes many other type 1 transmembrane substrates, is composed of presenilin, Aph1, nicastrin, and presenilin enhancer (Pen-2), all of which are necessary for proper complex maturation and enzymatic activity. Obtaining a high-resolution atomic structure of the intact complex would greatly aid the rational design of compounds to modulate activity but is a very difficult

The SARS coronavirus spike glycoprotein is selectively recognized by lung surfactant protein D and activates macrophages.

The severe acute respiratory syndrome coronavirus (SARS-CoV) infects host cells with its surface glycosylated spike-protein (S-protein). Here we expressed the SARS-CoV S-protein to investigate its interactions with innate immune mechanisms in the lung. The purified S-protein was detected as a 210 kDa glycosylated protein. It was not secreted in the presence of tunicamycin and was detected as a 130 kDa protein in the cell lysate. The purified S-protein bound to Vero but not 293T cells and was itself recognized by lung surfactant protein D (SP-D), a collectin found in the lung alveoli. The bindi

Expression and copper binding properties of the N-terminal domain of copper P-type ATPases of African trypanosomes.

Copper is an essential component of cuproproteins but can be toxic to cells, therefore copper metabolism is very carefully regulated within cells. To gain insight into trypanosome copper metabolism, Trypanosoma spp. genomic databases were screened for the presence of copper-containing and -transporting proteins. Among other genes encoding copper-binding proteins, a copper-transporting P-type ATPase (CuATPase) gene was identified. Sequence and phylogenetic analyses suggest that the gene codes for a Cu+ transporter belonging to the P1B-1 ATPase subfamily that has an N-terminal domain with copper

Disaccharide-Presenting Iron(II) Complexes as Synthetic Mimics of Natural Glycan Epitopes for Lectin Recognition.

Carbohydrates present on the surface of living cells mediate key biological processes, including viral infection, cell adhesion, and immune response. Mimicking the structural complexity of these naturally occurring sugars is therefore critical for gaining fundamental insights into protein-carbohydrate recognition and for guiding the design of glycomimetic architectures with therapeutic relevance. Covalently linking carbohydrates to molecular inorganic architectures provides a unique strategy for interrogating protein recognition that leverages the structural organization and precision of coord

Vaccination with recombinant filarial paramyosin induces partial immunity to Brugia malayi infection in jirds.

Vaccination with irradiated infective larvae induces partial protective immunity to infection with the filarial nematode Brugia malayi in jirds. Prior studies have shown that such immunization stimulates a much stronger antibody response to recombinant and native filarial paramyosin than that seen after normal infection. To determine whether vaccination with recombinant paramyosin could induce protective immunity to larval challenge, jirds were immunized with either B. malayi paramyosin and maltose binding protein (BM5-MBP) (fusion protein of B. malayi paramyosin and maltose-binding protein),

Visualizing conformational dynamics of proteins in solution and at the cell membrane.

Conformational dynamics underlie enzyme function, yet are generally inaccessible via traditional structural approaches. FRET has the potential to measure conformational dynamics in vitro and in intact cells, but technical barriers have thus far limited its accuracy, particularly in membrane proteins. Here, we combine amber codon suppression to introduce a donor fluorescent noncanonical amino acid with a new, biocompatible approach for labeling proteins with acceptor transition metals in a method called ACCuRET (Anap Cyclen-Cu2+ resonance energy transfer). We show that ACCuRET measures absolute

Potential usefulness of biotinylated neoglycoproteins as tumor markers.

We used several biotinylated neoglycoproteins as tumor markers to detect and localize endogenous carbohydrate-binding proteins in cultured hepatoblastoma, melanoma, and bladder carcinoma tumor cells. The neoglycoproteins used consisted of cellobiose, fucose, N-acetyl-galactosamine, N-acetyl-glucosamine, lactose, maltose, mannose, melibiose, and xylose. In addition, naturally occurring asialofetuin that was chemically disialylated was also used. Binding to the cultured tumor cells was made visible with the avidin-peroxidase technique. Depending on the type of neoglycoprotein used, markedly diff

Dual Protease-Mediated Proteolysis Coupled with Ultrafiltration for System-Level Profiling of Drug Targets and Conformational Dynamics.

Characterizing drug-binding proteins and the subsequent changes in protein complexes/interactions is pivotal to fully understanding drug mechanisms. Here, we developed a novel modification-free approach, Dual Protease-Mediated Proteolysis Coupled with Ultrafiltration (DPMU), which integrates limited proteolysis, ultrafiltration fractionation, and quantitative mass spectrometry to systematically map drug-induced structural alterations. DPMU employs thermolysin (broad specificity; prefers hydrophobic residues) and trypsin (specific for lysine and arginine residues) in a two-step workflow: thermo

Functional complementation of E. coli secD and secG mutants by Helicobacter pylori homologues.

The Sec machinery is one mechanism used by bacteria to translocate proteins across their cytoplasmic membrane. Most of the Sec components have been identified within the important gastric pathogen, Helicobacter pylori, however their functionality has not yet been demonstrated. Here we report the existence of putative homologues to the Sec components yajC (HP1450) and yidC (HP1551), and demonstrate the ability of the H. pylori secD (HP1550) and secG (HP1255) homologues to facilitate inner membrane translocation of the maltose-binding protein MalE, by complementation of the respective secretion-

Immunization of mice with recombinant L7/L12 ribosomal protein confers protection against Brucella abortus infection.

BALB/c mice were immunized with the recombinant Brucella abortus L7/L12 ribosomal protein fused to maltose binding protein (MBP). Vaccinated animals mounted a specific immune response to the recombinant fusion protein as demonstrated by immunoblot analyses. Additionally, B. abortus L7/L12 ribosomal protein conferred a significant degree of protection when compared to mice vaccinated with adjuvant alone, adjuvant plus MBP or B. abortus. These results indicate that a recombinant B. abortus protein, previously identified as T-cell-reactive, engendered protective immunity to mice against brucellos

Immunization of Mice with Recombinant Brucella abortus Organic Hydroperoxide Resistance (Ohr) Protein Protects Against a Virulent Brucella abortus 544 Infection.

In this study, the Brucella abortus ohr gene coding for an organic hydroperoxide resistance protein (Ohr) was cloned into a maltose fusion protein expression system (pMAL), inserted into Escherichia coli, and purified, and its immunogenicity was evaluated by western blot analysis using Brucella-positive mouse sera. The purified recombinant Ohr (rOhr) was treated with adjuvant and injected intraperitoneally into BALB/c mice. A protective immune response analysis revealed that rOhr induced a significant increase in both the IgG1 and IgG2a titers, and IgG2a reached a higher level than IgG1 after

Expression of in vivo biotinylated recombinant antigens SAG1 and SAG2A from Toxoplasma gondii for improved seroepidemiological bead-based multiplex assays.

BACKGROUND: Few bead-based multiplex assays have been described that detect antibodies against the protozoan parasite Toxoplasma gondii in large-scale seroepidemiological surveys. Moreover, each multiplex assay has specific variations or limitations, such as the use of truncated or fusion proteins as antigens, potentially masking important epitopes. Consequently, such an assay must be developed by interested groups as none is commercially available. RESULTS: We report the bacterial expression and use of N-terminal fusion-free, soluble, in vivo biotinylated recombinant surface antigens SAG1 an

Cloning and expression of mamba toxins.

Mamba venoms contain pharmacologically active proteins that interfere with neuromuscular transmission by binding to and altering the normal functioning of neuronal proteins involved, directly or indirectly, with regulating nerve transmission. Of the mamba toxins studied to date, many act on voltage-sensitive K+ channels, nicotinic or muscarinic acetylcholine receptors, or acetylcholinesterase. In an attempt to clone, characterize, and express the genes encoding these toxins, as well as other genes specifying activities not completely elucidated as yet, a cDNA library was constructed from mRNA

Glycogen-Degrading Activities of Catalytic Domains of α-Amylase and α-Amylase-Pullulanase Enzymes Conserved in Gardnerella spp. from the Vaginal Microbiome.

Gardnerella spp. are associated with bacterial vaginosis in which normally dominant lactobacilli are replaced with facultative and anaerobic bacteria, including Gardnerella spp. Co-occurrence of multiple species of Gardnerella is common in the vagina, and competition for nutrients such as glycogen likely contributes to the differential abundances of Gardnerella spp. Glycogen must be digested into smaller components for uptake, a process that depends on the combined action of glycogen-degrading enzymes. In this study, the ability of culture supernatants of 15 isolates of Gardnerella spp. to pro

Successful recombinant production of Allochromatium vinosum cytochrome c' requires coexpression of cmm genes in heme-rich Escherichia coli JCB712.

Cytochrome c' from the purple photosynthetic bacterium Allochromatium vinosum (CCP) displays a unique, reversible dimer-to-monomer transition upon binding of NO, CO, and CN(-). This small, four helix bundle protein represents an attractive model for the study of other heme protein biosensors, provided a recombinant expression system is available. Here we report the development of an efficient expression system for CCP that makes use of a maltose binding protein fusion strategy to enhance periplasmic expression and allow easy purification by affinity chromatography. Coexpression of cytochrome c

Engineering Coil-Coiled Domains for the Design of Modular Theranostic Agents: Galectin-3 as a Targeting Moiety.

To generate drug-targeting agents with a modular and efficient design for disease treatments, including cancer, we used protein coil-coiled dimerization motifs to eliminate the need for chemical conjugation techniques, generating a versatile theranostic system: a targeting moiety genetically fused to an E3 coil and a fusion protein of maltose-binding protein and a monomeric streptavidin that includes a K3 coil. The system can be directed to other targets by changing the targeting moiety. The second component enables simultaneous binding to two different ligands (maltosylated or biotinylated).
