Mannose-binding lectin: structure, function, genetics and disease associations.

Mannose-binding lectin (MBL), a serum protein characterised by both collagenous regions and lectin domains, plays an important role in innate immune defence. It binds to the repeating sugar arrays on many microbial surfaces through multiple lectin domains and, following binding, is able to activate the complement system via an associated serum protease, MASP-2. Serum levels of MBL are influenced by three mutations clustered in exon 1 of the gene and are further modulated by various promoter region polymorphisms. The exon 1 mutations lead to secondary structural abnormalities of the collagenous

Might there be a link between mannose binding lectin and vitiligo?

Mannose binding lectin (MBL) is a calcium dependent lectin that causes predisposition to infections and autoimmune diseases. This study aimed to examine the presence of any association between MBL2 gene variants and vitiligo. Codon 54 (allele B) and codon 57(allele C) polymorphisms in the exon 1 of the MBL2 gene were investigated in samples belonged to 50 healthy controls and 40 patients diagnosed as vitiligo. The PCR-RFLP method was used to investigate the polymorphisms in the MBL2 gene. Codon 57 polymorphism was not detected in any of the subjects from either group. The frequencies of low le

Impact of mannose-binding lectin on susceptibility to infectious diseases.

When the adaptive immune response is either immature or compromised, the innate immune system constitutes the principle defense against infection. Mannose-binding lectin (MBL) is a C-type serum lectin that plays a central role in the innate immune response. MBL binds microbial surface carbohydrates and mediates opsonophagocytosis directly and by activation of the lectin complement pathway. A wide variety of clinical isolates of bacteria, fungi, viruses, and parasites are bound by MBL. Three polymorphisms in the structural gene MBL2) and 2 promoter gene polymorphisms are commonly found that res

Mannose-binding lectin gene polymorphism in relation to periodontal infection.

BACKGROUND AND OBJECTIVE: Mannose-binding lectin (MBL) plays an important role in innate immunity. MBL deficiency is usually caused by mutations in exon 1 of the MBL structural gene (MBL2). Our aim was to investigate MBL2 polymorphisms and their relation to salivary levels of periodontal inflammatory/tissue destruction markers and two major periodontitis-associated bacteria. MATERIAL AND METHODS: Salivary samples from 222 subjects were available for genotyping by pyrosequencing. The subjects between 40 and 60 years of age and having a minimum of 20 teeth were divided into three periodontal gr

Mannose-binding Lectin Mediated Complement Pathway in Autoimmune Neurological Disorders.

Multiple sclerosis (MS) is a complex, demyelinating disease of the central nervous system (CNS) with variable phenotypic presentations, while Guillain-Barre Syndrome (GBS) is the prototypic acute inflammatory disorder that affects the peripheral nervous system. Myasthenia gravis (MG) is a T cell dependent and antibody mediated autoimmune disease. Although it has been shown that complement plays a critical role in the pathogenesis of MS, GBS, and MG, the role of mannose-binding lectin (MBL) as a biomarker of immunopathogensis of these diseases and also its association with the severity of them

Mannose Binding Lectin: A Potential Biomarker for Many Human Diseases.

The innate immune system plays a modulatory role in producing an inflammatory response during microbial infection and tissue regeneration. Mannose-binding lectin (MBL) is a predominant constituent of the innate immune system which initiates one of the complement activation, the lectin pathway. The activation of the complement system is also associated with many human diseases. We, therefore, try to summarize herewith the prognostic value of early detection of serum mannose-binding lectin (MBL) and measurement of its levels. The variant alleles and single nucleotide substitutions in MBL2 gene a

Complement and mannose-binding lectin 2 polymorphism in meningococcal disease.

BACKGROUND: Meningococcal disease is a leading cause of death due to infection in children. Differences in susceptibility and disease progression between children are unknown. The complement system plays a key role in the innate immunity against N. meningitides and could explain that differences in the response of individuals. The aim of this study was to evaluate the occurrence of complement system deficiencies and MBL2 polymorphism in patients with previous meningococcal disease. METHODS: We evaluated 40 children with confirmed previous diagnosis of any form of meningococcal disease and per

ROMP-based Glycopolymers with High Affinity for Mannose-Binding Lectins.

Well-defined, highly reactive poly(norbornenyl azlactone)s of controlled length (number-average degree of polymerization DPn¯ = 10 to 1,000) were made by ring-opening metathesis polymerization (ROMP) of pure exo-norbornenyl azlactone. These were converted into glycopolymers using a facile postpolymerization modification (PPM) strategy based on click aminolysis of azlactone side groups by amino-functionalized glycosides. Pegylated mannoside, heptyl-mannoside, and pegylated glucoside were used in the PPM. Binding inhibition of the resulting glycopolymers was evaluated against a lectin panel (Bc2

Mannose binding lectin gene polymorphism and the severity of chronic periodontitis.

OBJECTIVE: Mannose binding lectin (MBL) is a key molecule in first line defense against various microorganisms. Polymorphism of the MBL gene greatly affects serum MBL concentration, and is known to be associated with occurrence of infectious diseases. We aimed to determine whether there is a relationship between occurrence or severity of chronic periodontitis (CP) and polymorphism of the MBL gene. PATIENTS AND METHODS: Ninety-eight CP patients and 63 healthy subjects with no periodontitis were typed for the codon 54 polymorphism of the MBL gene by PCR-restriction fragment length polymorphism

Polymorphisms and serum level of mannose-binding lectin: an Iranian survey.

Mannose-binding lectin (MBL) is a Ca⁺² -dependent collagenous lectin, that is produced by liver and mediates innate immune responses by opsonization of pathogens. The serum level of MBL varies widely among healthy individuals, ranging from 0.05 µg/ml (or lower) to over 5 µg/ml, mainly depending on genetic variation. This study has examined promoter and exon 1 of mbl2 genotype among 117 Iranian healthy blood donors. MBL Single Nucleotide Polymorphisms (SNPs) were genotyped using polymerase chain reaction (PCR), and serum levels of MBL were quantified using a double-antibody enzyme linked immuno
