UL24 deletion attenuates Marek's disease virus replication and pathogenicity.

Marek's disease virus (MDV), a highly oncogenic alphaherpesvirus of chickens, causes severe immunosuppression and fatal T-cell lymphomas. The UL24 gene is conserved among alphaherpesviruses and contributes to viral replication and pathogenesis in several members of this subfamily; however, its role in MDV remains undefined. Here, we constructed a UL24-deletion mutant (Md5BACΔUL24) and its repaired revertant (Md5BACΔUL24-Re) in the virulent Md5 strain using Red-mediated recombination. In vitro, UL24 deletion significantly impaired viral replication, as evidenced by smaller plaque size and reduc

Effects of Ikaros (IKZF1) gene in the virulence of Marek's disease virus.

Marek's disease (MD) caused by the oncogenic avian herpesvirus, Marek's disease virus (MDV) has significant economic impacts on the poultry industry because MDV is ubiquitous in the environment and most chickens are exposed to the threat by MDV from the first day of age. Meq, a bZIP transactivator, is required for tumor formation by MDV, mostly T cell lymphomas. Additionally, Ikaros (IKZF1) has been identified as a cancer driver gene for MDV tumorigenesis. The safety of G2M-WT-Ikaros, which contains wildtype IKZF1 gene in the virulent MDV genome as a potential vaccine candidate, was compared w

Exophthalmos and Buphthalmos in Broiler Breeders Associated with Marek's Disease Virus.

This study investigated blindness accompanied by unilateral or bilateral exophthalmos in four broiler breeder flocks. Histopathologic examination revealed keratitis, corneal edema, cataracts, uveitis, and pectenitis. In addition, retinal degeneration and occasional intranuclear inclusion bodies, consistent with herpesvirus, were observed in the ganglion layer. Immunohistochemical analysis of the ocular sections revealed a mixture of T cells (CD3+) and B cells (Pax5). Marek's disease virus type 1 (MDV-1) DNA was detected in all samples of optic nerve, vitreous fluid, retina, cornea, iris, and l

Pathology and Molecular Diagnosis of Marek's Disease Virus in Chickens in Nigeria.

Marek's disease caused by Marek's disease virus (MDV) serotype 1 is an economically important neoplastic disease of poultry. Diagnosis of this disease is usually based on clinical signs, postmortem lesions, and diagnostic tests like cytology, histopathology, and molecular-based methods. However, there might be the problem of inaccurate diagnosis in Nigeria. Employment of gross pathology, histopathology, immunohistochemistry, and polymerase chain reaction (PCR) coupled with sequence analysis provides a reliable approach to arrive at precise confirmatory diagnosis of the disease. Therefore, visc

Marek's Disease Virus Virulence Genes Encode Circular RNAs.

Circular RNAs (circRNAs) are a recently rediscovered class of functional noncoding RNAs that are involved in gene regulation and cancer development. Next-generation sequencing approaches identified circRNA fragments and sequences underlying circularization events in virus-induced cancers. In the present study, we performed viral circRNA expression analysis and full-length sequencing in infections with Marek's disease virus (MDV), which serves as a model for herpesvirus-induced tumorigenesis. We established inverse PCRs to identify and characterize circRNA expression from the repeat regions of

Genetic characterization of a Marek's disease virus strain isolated in Japan.

BACKGROUND: Marek's disease virus (MDV) causes malignant lymphomas in chickens (Marek's disease, MD). MD is currently controlled by vaccination; however, MDV strains have a tendency to develop increased virulence. Distinct diversity and point mutations are present in the Meq proteins, the oncoproteins of MDV, suggesting that changes in protein function induced by amino acid substitutions might affect MDV virulence. We previously reported that recent MDV isolates in Japan display distinct mutations in Meq proteins from those observed in traditional MDV isolates in Japan, but similar to those in

Marek's disease virus-induced transient cecal tonsil atrophy.

Marek's disease (MD) is a lymphoproliferative disease of domestic chickens that is caused by a highly cell-associated oncogenic alpha-herpesvirus, Marek's disease virus (MDV). MDV replicates in chicken lymphocytes and establishes a latent infection within CD4+ T cells. MD is characterized by bursal and thymic atrophy and rapid onset of T cell lymphomas that infiltrate lymphoid tissues, visceral organs, and peripheral nerves with severe clinical symptoms that include transient paralysis, anemia, weight loss, and neurologic disorders. The cecal tonsils (CT) are considered the largest lymphoid ag

Detection and characterization of avian leukosis virus in Marek's disease vaccines.

Avian leukosis virus (ALV) infection in chickens is known to induce increased mortality, tumors, delayed growth, and suboptimal egg production. Countries importing specified pathogen-free eggs, vaccines, and poultry breeding stock require freedom of infection or contamination with ALV in such products among other avian pathogens. Recently, ALV was found as a contaminant in a limited number of commercial poultry vaccines, even after routine quality assurance procedures cleared the vaccines for commercialization. The contaminated vaccines were promptly withdrawn from the market, and no direct de

Marek's disease in Japanese quail -- a pathological, virological and serological study.

Experimental studies were performed to see whether Japanese quail (Coturnix coturnix japonica) are susceptible to JM strain of Marek's disease virus (MDV). In three identical trials, a total of 120, one-day-old quail were inoculated intra-abdominally with 0.2 ml. of chicken blood infected with MDV (JM strain) and raised in FAPP isolators. Uninfected controls were inoculated with normal saline only and raised separately. During the 18-week post infection observation period, 65-80% of the infected quail died within six weeks, while the highest mortality in control groups was only 0-5%. In each t

Transgenerational epigenetic inheritance and immunity in chickens that vary in Marek's disease resistance.

Marek's disease virus (MDV), a naturally oncogenic, highly contagious alpha herpesvirus, induces a T cell lymphoma in chickens that causes severe economic loss. Marek's disease (MD) outcome in an individual is attributed to genetic and environmental factors. Further investigation of the host-virus interaction mechanisms that impact MD resistance is needed to achieve greater MD control. This study analyzed genome-wide DNA methylation patterns in 2 highly inbred parental lines 63 and 72 and 5 recombinant congenic strains (RCS) C, L, M, N, and X strains from those parents. Lines 63 and 72, are MD

Marek's Disease Virus Disables the ATR-Chk1 Pathway by Activating STAT3.

Oncogenic virus replication often leads to genomic instability, causing DNA damage and inducing the DNA damage response (DDR) pathway. The DDR pathway is a cellular pathway that senses DNA damage and regulates the cell cycle to maintain genomic stability. Therefore, the DDR pathway is critical for the viral lifecycle and tumorigenesis. Marek's disease virus (MDV), an alphaherpesvirus that causes lymphoma in chickens, has been shown to induce DNA damage in infected cells. However, the interaction between MDV and the host DDR is unclear. In this study, we observed that MDV infection causes DNA s

Two sublines of a lymphoblastoid cell line associated with Marek's disease virus.

From MDCC-MSB1, the original Marek's disease virus-associated lymphoblastoid cell line, two sublines named MDCC-MSB1-33C and MDCC-MSB1-41C were differentiated. The MDCC-MSB1-33C cell line grew well at 33 degrees, but grew poorly and finally stopped growing at 41 degrees. On the other hand, the MDCC-MSB1-41C cell line grew actively at 41 degrees, but at 33 degrees, continuous cultivation was difficult to achieve. MDCC-MSB1-33C grew suspended in the liquid medium, as did the original producer MDCC-MSB1 cell line, while MDCC-MSB1-41C grew attached to the substrate. Moreover, in soft agar, MDCC-MS

Marek's Disease in France: Ten Cases of Central Nervous System Syndromes.

Between 2017 and 2019, 10 broiler flocks, with birds exhibiting neck paralysis, nervous tics, and torticollis, were submitted for necropsy and additional tests. These neurologic signs were mostly observed in certified, nonvaccinated broilers (8 of 10), ranging between 45 and 56 days of age. No specific lesions were described at macroscopic examination. Histopathology revealed moderate to severe lymphocytic infiltration of the central nervous system (CNS). In addition, peripheral nerves and visceral organs were commonly affected, although less severely. Immunohistochemistry identified the infil

Virulence of Marek's disease virus in Japan is linked to polymorphisms in the meq oncogene.

Marek's disease virus (MDV) causes lymphomas and neurological disorders in chickens. Although vaccination largely controls outbreaks, highly virulent strains continue to emerge. The major MDV-encoded oncoprotein is Meq, functions as a transcription factor. Amino acid polymorphisms in Meq have been reported to influence virulence. Despite routine vaccination, MD still occurs sporadically in Japan. Japanese isolates harbour characteristic Meq polymorphisms, but their impact on MDV virulence remains unclear. We investigated the transcriptional regulation by Meq from Japanese isolates and evaluate

Role of T Cells in Vaccine-Mediated Immunity against Marek's Disease.

Marek's disease virus (MDV), a highly cell-associated oncogenic α-herpesvirus, is the etiological agent of T cell lymphomas and neuropathic disease in chickens known as Marek's disease (MD). Clinical signs of MD include neurological disorders, immunosuppression, and lymphoproliferative lymphomas in viscera, peripheral nerves, and skin. Although vaccination has greatly reduced the economic losses from MD, the molecular mechanism of vaccine-induced protection is largely unknown. To shed light on the possible role of T cells in immunity induced by vaccination, we vaccinated birds after the deplet
