Temperature dependent sensitivity of the harpacticoid copepod Nitokra spinipes to marine algal toxins.

Harmful algal blooms (HABs) - proliferated algae densities, often producing toxins - have increasingly been found in ocean and coastal areas. Recent studies show that rising temperatures contribute to HAB occurrence, but the broader influence of climate change on these outbreaks is less quantified. Of particular concern is the limited research on HAB toxin effects under varying temperatures, especially regarding primary consumers such as copepods, a crucial component of aquatic ecosystems. Therefore, we examined the impact of marine toxins on the harpacticoid copepod Nitokra spinipes, a model

First Report of Paralytic Shellfish Toxins in Marine Invertebrates and Fish in Spain.

A paralytic shellfish poisoning (PSP) episode developed in summer 2018 in the Rías Baixas (Galicia, NW Spain). The outbreak was associated with an unprecedentedly intense and long-lasting harmful algal bloom (HAB) (~one month) caused by the dinoflagellate Alexandrium minutum. Paralytic shellfish toxins (PSTs) were analyzed in extracts of 45 A. minutum strains isolated from the bloom by high-performance liquid chromatography with post-column oxidation and fluorescence detection (HPLC-PCOX-FLD). PSTs were also evaluated in tissues from marine fauna (invertebrates and fish) collected during the e

Distribution of Marine Lipophilic Toxins in Shellfish Products Collected from the Chinese Market.

To investigate the prevalence of lipophilic marine biotoxins in shellfish from the Chinese market, we used hydrophilic interaction liquid chromatography-tandem mass spectrometry (LC-MS/MS) to measure levels of okadaic acid (OA), azaspiracid (AZA1), pectenotoxin (PTX2), gymnodimine (GYM), and spirolide (SPX1). We collected and analyzed 291 shellfish samples from main production sites along a wide latitudinal transect along the Chinese coastline from December 2008 to December 2009. Results revealed a patchy distribution of the five toxins and highlighted the specific geographical distribution an

Anaerobic biotransformation mechanism of marine toxin domoic acid.

Domoic acid (DA) is a major marine neurotoxin, occurs frequently in most of the world's coastlines and seriously threatens ecosystem and public health. However, information on its biotransformation process in coastal anaerobic environments remains unclear. In this study, the underlying mechanism of anaerobic biotransformation of DA by marine consortium GLY was investigated using the combination of liquid chromatography-high-resolution Orbitrap mass spectrometry and comparative metatranscriptomics analysis. The results demonstrated that DA could be cometabolically biotransformed under anaerobic

Inhibition of actin polymerization by marine toxin pectenotoxin-2.

Pectenotoxin-2 (PCTX-2) is one of the polyether macrolide toxins isolated from scallops involved in diarrheic shellfish poisoning via actin depolymerization. In the present study, we examined the bioactive mechanism of PCTX-2 in smooth muscle cells and clarify mode of action of the PCTX-2-induced actin depolymerization using purified skeletal actin. PCTX-2 (300 nM-3 µM) non-selectively inhibited vascular smooth muscle contractions elicited by high K+ or phenylephrine in a dose-dependent manner. However, elevated cytosolic Ca2+ and myosin light chain phosphorylation stimulated by high K+ were o

Formation and detection of marine algal toxins and their distribution in Chinese coastal waters.

Marine algal toxins, a diverse group of bioactive secondary metabolites produced by harmful algal species, pose significant threats to human health, marine ecosystems, and aquaculture economies. Driven by human activities and climate change, the increasing frequency and scale of harmful algal blooms have expanded the contamination scope of these toxins. This trend underscores an urgent need to better understand their formation and distribution, and to develop more effective methods for their detection and monitoring. This review summarizes recent advances in the formation, detection and monito

Contamination status of lipophilic marine toxins in shellfish samples from the Bohai Sea, China.

Lipophilic marine toxins in shellfish pose significant threats to the health of seafood consumers. To assess the contamination status of shellfish by lipophilic marine toxins in the Bohai Sea, nine species of shellfish periodically collected from five representative aquaculture zones throughout a year were analyzed with a method of liquid chromatography-tandem mass spectrometry (LC-MS/MS). Lipophilic marine toxins, including okadaic acid (OA), dinophysistoxin-1 (DTX1), pectenotoxin-2 (PTX2), yessotoxin (YTX), homo-yessotoxin (homo-YTX), azaspiracids (AZA2 and AZA3), gymnodimine (GYM), and 13-d

Effects of marine toxins on the reproduction and early stages development of aquatic organisms.

Marine organisms, and specially phytoplankton species, are able to produce a diverse array of toxic compounds that are not yet fully understood in terms of their main targets and biological function. Toxins such as saxitoxins, tetrodotoxin, palytoxin, nodularin, okadaic acid, domoic acid, may be produced in large amounts by dinoflagellates, cyanobacteria, bacteria and diatoms and accumulate in vectors that transfer the toxin along food chains. These may affect top predator organisms, including human populations, leading in some cases to death. Nevertheless, these toxins may also affect the rep

Evidence of freshwater algal toxins in marine shellfish: Implications for human and aquatic health.

The occurrence of freshwater harmful algal bloom toxins impacting the coastal ocean is an emerging threat, and the potential for invertebrate prey items to concentrate toxin and cause harm to human and wildlife consumers is not yet fully recognized. We examined toxin uptake and release in marine mussels for both particulate and dissolved phases of the hepatotoxin microcystin, produced by the freshwater cyanobacterial genus Microcystis. We also extended our experimental investigation of particulate toxin to include oysters (Crassostrea sp.) grown commercially for aquaculture. California mussels

Natural attenuation and engineering-based removal of marine algal toxins: current status and future perspectives.

The global surge in harmful algal blooms (HABs), amplified by ocean warming and eutrophication, has intensified ecological and public health risks through the production of potent marine algal toxins. Given their ubiquitous occurrence, environmental persistence, and adverse effects on marine ecosystems and coastal economies, increasing efforts have been devoted to mitigating toxic HABs and associated toxin pollution. This review presents a systematic, integrated analysis of natural attenuation processes and engineering-based removal technologies for marine algal toxins, addressing a critical g

Marine toxin domoic acid alters nitrogen cycling in sediments.

As a red tide algal toxin with intense neurotoxicity distributed worldwide, domoic acid (DA) has attracted increasing concerns. In this work, the integrative analysis of metagenome and metabolome are applied to investigate the impact of DA on nitrogen cycling in coastal sediments. Here we show that DA can act as a stressor to induce the variation of nitrogen (N) cycling by altering the abundance of functional genes and electron supply. Moreover, microecology theory revealed that DA can increase the role of deterministic assembly in microbial dynamic succession, resulting in the shift of niches

A new pathway for anaerobic biotransformation of marine toxin domoic acid.

Domoic acid (DA) is a harmful algal toxin produced by marine diatom Pseudo-nitzschia and seriously threatens ecosystem and human health. However, the current knowledge on its biotransformation behavior in coastal anaerobic environment is lacking. This study investigated the anaerobic biotransformation of DA by a new marine consortium GH1. The results demonstrated that 90% of DA (1 mg L-1) was cometabolically biotransformed under sulfate-reducing condition. A new anaerobic biotransformation pathway involving DA hydration, dehydrogenation, and C-C bond cleavage was proposed, where the conjugated

Foodborne Disease Outbreaks Associated with Marine Toxins - Foodborne Disease Outbreak Surveillance System, United States, 2011-2023.

PROBLEM/CONDITION: Marine toxins cause most of the noninfectious outbreaks reported to CDC's Foodborne Disease Outbreak Surveillance System (FDOSS) each year. Certain marine toxins are produced by algae that accumulate in aquatic animals through the food chain, whereas others appear as a result of improper food storage and are therefore preventable. Overgrowths of toxin-producing algae (harmful algal blooms) have occurred on both U.S. coasts; the historical geographic range for some species (e.g., Gambierdiscus in tropical and subtropical regions) have expanded. Marine toxins that cause foodbo

Light and scanning electron microscopic studies on effects of marine algal toxins toward freshly prepared hepatocytes.

Mussels exposed to dinoflagellates may represent a human health risk due to accumulation of a variety of algal toxins. In several parts of the world, algal toxins leading to diarrhea (diarrhetic shellfish poisons, DSP) are found in mussels for extended periods of the year. Routine monitoring of these toxins involves ip injections in mice. Chemical analytical methods have been developed for only some of the toxins in question, namely, those giving diarrhea. Other toxins in the DSP complex are not easily detected by analytical methods. In this report we show that freshly prepared hepatocytes fro

Occurrence of lipophilic marine toxins in shellfish from Galicia (NW of Spain) and synergies among them.

Lipophilic marine toxins pose a serious threat for consumers and an enormous economic problem for shellfish producers. Synergistic interaction among toxins may play an important role in the toxicity of shellfish and consequently in human intoxications. In order to study the toxic profile of molluscs, sampled during toxic episodes occurring in different locations in Galicia in 2014, shellfish were analyzed by liquid chromatography tandem mass spectrometry (LC-MS/MS), the official method for the detection of lipophilic toxins. The performance of this procedure was demonstrated to be fit for purp

Sublethal oral exposure to the marine toxin PbTx3: Neurotoxicity assessment in mice.

Brevetoxins (PbTxs) are marine polyether compounds produced by the alga Karenia brevis, which accumulate in shellfish and fish, and transferred through the marine food web. These toxins impact both marine ecosystems and human health. Consumption of contaminated shellfish is the primary cause of Neurotoxic Shellfish Poisoning (NSP), which presents with neurological, gastrointestinal, and/or cardiovascular symptoms that appear in a few hours and typically resolves within 1-3 days, although some disturbances may persist for weeks or months. The acute toxicity of PbTxs involves activation of volta

Biogenic ROS mediated degradation mechanism of marine toxin domoic acid.

Domoic acid (DA) is a compound generated as a secondary metabolite during harmful algal blooms, has historically received attention as the potent neurotoxicity in marine environment. However, the aerobic degradation mechanism of DA and the DA-degrader remain largely unknown. Here, we revealed the mechanism of aerobic degradation of DA by a ubiquitous marine Pseudoalteromonas sp., and more importantly, we confirmed that the degradation of DA is mediated by biogenic reactive oxygen species (ROS), rather than direct enzyme-mediated as traditionally conceived. Results indicated that DA degradation

Fundamental Studies of the Singlet Oxygen Reactions with the Potent Marine Toxin Domoic Acid.

Domoic acid (DA), a potent marine toxin, is readily oxidized upon reaction with singlet oxygen (1O2). Detailed product studies revealed that the major singlet oxygenation reaction pathways were the [2 + 2] cycloaddition (60.2%) and ene reactions (39.8%) occurring at the Z double bond. Diene isomerization and [4 + 2] cycloaddition, common for conjugated diene systems, were not observed during the singlet oxygenation of DA. The bimolecular rate constant for the DA reaction with 1O2 determined by competition kinetics was 5.1 × 105 M-1 s-1. Based on the rate constant and steady-state concentration

Antillatoxin is a marine cyanobacterial toxin that potently activates voltage-gated sodium channels.

Antillatoxin (ATX) is a lipopeptide derived from the pantropical marine cyanobacterium Lyngbya majuscula. ATX is neurotoxic in primary cultures of rat cerebellar granule cells, and this neuronal death is prevented by either N-methyl-d-aspartate (NMDA) receptor antagonists or tetrodotoxin. To further explore the potential interaction of ATX with voltage-gated sodium channels, we assessed the influence of tetrodotoxin on ATX-induced Ca2+ influx in cerebellar granule cells. The rapid increase in intracellular Ca2+ produced by ATX (100 nM) was antagonized in a concentration-dependent manner by tet

Blurred lines: Multiple freshwater and marine algal toxins at the land-sea interface of San Francisco Bay, California.

San Francisco Bay (SFB) is a eutrophic estuary that harbors both freshwater and marine toxigenic organisms that are responsible for harmful algal blooms. While there are few commercial fishery harvests within SFB, recreational and subsistence harvesting for shellfish is common. Coastal shellfish are monitored for domoic acid and paralytic shellfish toxins (PSTs), but within SFB there is no routine monitoring for either toxin. Dinophysis shellfish toxins (DSTs) and freshwater microcystins are also present within SFB, but not routinely monitored. Acute exposure to any of these toxin groups has s
