Distinguishing Mast Cell Progenitors from Mature Mast Cells in Mice.

Mast cells originate from the bone marrow and develop into c-kit(+) FcɛRI(+) cells. Both mast cell progenitors (MCp) and mature mast cells express these cell surface markers, and ways validated to distinguish between the two maturation forms with flow cytometry have been lacking. Here, we show that primary peritoneal MCp from naïve mice expressed high levels of integrin β7 and had a low side scatter (SSC) light profile; whereas mature mast cells expressed lower levels of integrin β7 and had a high SSC light profile. The maturation statuses of the cells were confirmed using three main strategie

Interaction between conjunctival epithelial cells and mast cells induces CCL2 expression and piecemeal degranulation in mast cells.

PURPOSE: Intraepithelial mast cells are observed in giant papillae tissue samples obtained from patients with atopic keratoconjunctivitis (AKC)/vernal keratoconjunctivitis (VKC). We examined the roles of interaction between the conjunctival epithelial cells and mast cells. METHODS: The interaction between human mast cells and conjunctival epithelial cells (HCjE) was investigated using a coculture model. Protein array analysis, ELISA, and real-time PCR were performed to test the interaction. Tissue samples (n = 6) from giant papillae were resected for therapeutic purposes, and subjected to imm

Expression of cell surface antigens on mast cells: mast cell phenotyping.

During the past few decades, a number of functionally important cell surface antigens have been detected on human mast cells (MCs). These antigens include the stem cell factor receptor (SCFR/CD117), the high-affinity immunoglobulin E receptor, adhesion molecules, and activation-linked membrane determinants. Several of these antigens (CD2, CD25, CD35, CD88, CD203c) appear to be upregulated on MCs in patients with systemic mastocytosis and therefore are used as diagnostic markers. Quantitative measurement of these markers on MCs is thus of diagnostic value and is usually performed by multicolor-

[Morphological characteristics of human mast cells in normal and pathological thyroid glands. Functional aspect of human mast cells in comparison with rat mast cells].

We examined the distribution and population density of human mast cells in thyroid glands. The results were compared with those of Sprague-Dawley (SD) rats because they thyroid function of SD rats is known to be under the control of bioactive amines discharged from mast cells. Normal thyroid tissues were obtained either form autopsy or from a normal portion of the tissue distant from nodular lesions. Thyroid tissues were surgically removed from cases of Graves' disease and other tumorous lesions such as follicular adenoma, follicular carcinoma, papillary carcinoma and medullary carcinoma. The

Mast Cells in Neurodegenerative Disease.

Neurodegenerative diseases affect millions of people worldwide, yet there are currently no effective treatments. Because risk of neurodegenerative disease substantially increases with age, greater life expectancy with a concomitant aging population means more individuals will be affected in the coming decades. Thus, there is an urgent need for understanding the mechanisms driving neurodegenerative diseases in order to develop improved treatment strategies. Inflammation in the nervous system, termed "neuroinflammation," has become increasingly recognized as being associated with neurodegenerati

Skin microbiome and mast cells.

Microbiotas in the skin have high levels of diversity at the species level, but low phylum-level diversity. The human skin microbiota is composed predominantly of Gram-positive bacteria especially Actinobacteria, which are the dominant bacterial phylum on the skin. Lipoteichoic acid (LTA) is a major constituent of the cell wall of Gram-positive bacteria and is therefore abundant in the skin microbiome. Recent studies have shown that LTA, and other bacterial products, permeates the whole skin and comes into contact with epidermal and dermal cells, including mast cells (MCs), with the potential

Toxoplasma gondii and mast cells.

Mast cells, discovered by Paul Burnet over one century ago, have been long recognized only as inductors of IgE-dependent allergic diseases (allergy of type I, Th2 lymphocytes dependent). However, numerous recent studies have indicated that they play an essential role in many other immunological and non-immunological processes. Infection with Toxoplasma gondii elicits the induction of a strong cell-mediated immunity characterized by a highly-polarized Th1 response, which can protect against allergy. Knowledge of the contribution of mast cells to Toxoplasma invasion is still limited, and the pre

Mast cells in gastrointestinal disease.

The function of mast cells in allergic inflammatory reactions is well documented in the literature. Mast cells also play an important role in the regulation of gastrointestinal visceral sensitivity and vascular permeability. Several studies have noted an increased number of mast cells in the mucosa of patients with gastrointestinal diseases such as irritable bowel syndrome, mastocytic enterocolitis, and systemic mastocytosis. The role of mast cells in the symptomatology of these and other diseases has only recently been fully appreciated and could provide avenues for new therapeutic opportunit

Vagal influences over mast cells.

The established microanatomical association of rat intestinal mucosal mast cells (IMMC) and mucosal nerves raises the possibility that there is crosstalk between mast cells and extrinsic nerves that connect to the CNS. The idea of mast cell-CNS interactions is supported by the demonstration that rat mast cell protease II (RMCPII), found predominantly in IMMC, can be conditionally released by pairing an audio-visual cue with antigen challenge. That the vagus nerve is involved in the IMMC-nerve axis was further demonstrated in a series of our studies showing that: (a) vagal afferents penetrate t

Mast cells and oral inflammation.

Mast cells are mobile granule-containing secretory cells that are distributed preferentially about the microvascular endothelium in oral mucosa and dental pulp. The enzyme profile of mast cells in oral tissues resembles that of skin, with most mast cells expressing the serine proteases tryptase and chymase. Mast cells in oral tissues contain the pro-inflammatory cytokine tumour necrosis factor-alpha in their granules, and release of this promotes leukocyte infiltration during evolving inflammation in several conditions, including lichen planus, gingivitis, pulpitis, and periapical inflammation

Mast cells in clinical dermatology.

Recent advances in cutaneous mast cell biology are briefly reviewed with special reference to our own studies on cultured human mast cells. Of note are the heterogeneity of mastocytosis, the important participation of mast cells in allergic inflammation by releasing cytokines and the inhibitory effect of histamine release from mast cells by phototherapy. It is also stressed that mast cells play a major role in tissue remodelling. These novel findings suggest that mast cells can no longer be regarded simply as cells that initiate immediate allergic reactions, but that they are responsible for v

Mast cells and their mediators.

Mast cells (MC) are the most important cells in hypersensitivity reactions. Their functions in other pathological conditions are, however, not completely understood. At least two different MC subtypes have been isolated and it seems that due to environmental changes further differences in the morphology and functions of MC:s exist. The activation of MC:s can be both due to immunological and non-immunological factors and results in a release of different mediators and production of so called newly generated MC mediators. The functions following mediator release are complicated and several other

[Mast cells in bone pathology].

The authors have endeavoured to determine the place occupied by mast cells in bone tissue from the mode of action on the bone of their secretion products (metachromatic granules, heparin, prostaglandins), from their proliferation in reaction to some bone diseases (secondary mastocytosis) and from the bone lesions associated with systemic mastocytosis. Metachromatic granules are capable of chelating calcium, and heparin and prostaglandins stimulate bone resorption. However, the role of mast cells in bone remodeling through these mechanisms has not yet been demonstrated. Similarly, the significa

Mast Cells and Cardiovascular Disorders.

Mastocytosis is a neoplasm characterised by clonal proliferation of mast cells (MCs) in the skin and other organs, including the heart, and symptoms produced by MC-derived mediators. MC activation syndrome (MCAS) relates to increased and inappropriate activation of MCs without clonal proliferation. Mast cells are tissue-resident immune cells strategically located in different cardiac sites, e.g., myocardium, pericardium, aortic valve, and close to nerves, and in atherosclerotic plaques. The mediators (e.g., histamine, tryptase, chymase, cytokines, and chemokines) have roles in inflammation, an

Targeting Mast Cells with Biologics.

Mast cells are tissue-resident allergic effector cells that cause many symptoms associated with IgE-mediated immediate hypersensitivity reactions. Beginning with allergen-specific therapy over a century ago, biologics have been used to target mast cells in patients in order to reduce allergic symptoms and reactions. This review discusses the history and current outlook of the use of biologics in mast cell-associated diseases and reactions.

Coeliac Disease and Mast Cells.

Over the last decades, there has been an impressive progress in our understanding of coeliac disease pathogenesis and it has become clear that the disorder is the final result of complex interactions of environmental, genetic, and immunological factors. Coeliac disease is now considered a prototype of T-cell-mediated disease characterized by loss of tolerance to dietary gluten and the targeted killing of enterocytes by T-cell receptor αβ intraepithelial lymphocytes. Accumulating evidence, however, indicates that the induction of a gluten-specific T helper-1 response must be preceded by the act

Mast cells in gastrointestinal disorders.

Mast cells are constitutively found in the gastrointestinal (GI) tract. The three major physiological functions of GI mast cells comprise of - as far as we know - regulation of GI functions, namely epithelial and endothelial functions, crosstalk with the enteric nervous system, and contribution to the host defense against bacterial, viral and parasitic agents. A number of chronic GI diseases, including inflammatory bowel disease (Crohn's disease, ulcerative colitis), celiac disease, irritable bowel syndrome, and food allergies, are thought to be associated with mast cell hyperplasia and humora
