Odontoblasts enhance the maturation of enamel crystals by secreting EMSP1 at the enamel-dentin junction.

The temporal expression patterns and activity distributions of enamelysin and EMSP1, which are the major proteinases in immature enamel, were characterized. Extracellular matrix fractions from developing porcine incisors, individually comprised of predentin, dentin, and four secretory-stage enamel samples, including the highly mineralized enamel (HME) at the enamel-dentin junction (EDJ), were isolated, and their resident proteinases were identified by zymography. Soft-tissue fractions, which included cells from the extension site of enamel formation (ESEF), secretory- and maturation-stage amel

Activation of extracellular signal-related kinase in abdominal aortic aneurysm.

BACKGROUND: Extracellular matrix degeneration, caused by matrix metalloproteinase-2, facilitates smooth muscle cell migration leading to medial layer decline and, ultimately, abdominal aortic aneurysm. It remains unclear what exactly causes aneurysms to rupture, which leads to death in most patients. The extracellular signal-related kinase may be linked to the latter process. We aimed to clarify the role of extracellular signal-related kinase in aortic aneurysm development and rupture in patients. DESIGN: Aortic fragments were harvested during open repair of nonruptured (n = 20) and ruptured

Prenatal plasma matrix metalloproteinase-9 levels to predict spontaneous preterm birth.

OBJECTIVE: Matrix metalloproteinase-9, a zinc-dependent proteinase, may be important in initiating labor or rupture of membranes. We determined plasma levels of this enzyme in nonpregnant and pregnant women and evaluated whether they predict spontaneous preterm delivery. METHODS: A sandwich enzyme-linked immunosorbent assay (ELISA) was used to measure matrix metalloproteinase-9 levels in plasma samples from 25 nonpregnant women (mean age 39+/-9 years) and in stored plasma samples obtained during a randomized trial of zinc supplementation in pregnancy. Women were selected who delivered followi

Posteruptive Loss of Proteins in Porcine Enamel.

Tooth enamel maturation requires the removal of proteins from the mineralizing enamel matrix to allow for crystallite growth until full hardness is reached to meet the mechanical needs of mastication. While this process takes up to several years in humans before the tooth erupts, it is greatly accelerated in the faster-developing pigs. Pig teeth erupt with softer, protein-rich enamel that is similar to hypomineralized human enamel but continues to harden quickly after eruption. Proteins that bind to enamel crystals and prevent crystal growth and enamel hardening (e.g., albumin) have been sugge

Analysis of MMP-8, MMP-20, and TGF-β1 in molar-incisor hypomineralization and assessment of periodontal health.

BACKGROUND: Gingival crevicular fluid (GCF) volume can be used to evaluate periodontal diseases, along with matrix metalloproteinase-8 (MMP-8) and transforming growth factor-beta 1 (TGF-β1) found in GCF. Conversely, matrix metalloproteinase-20 (MMP-20) is implicated in amelogenesis and serves as a potential biomarker for assessing enamel mineralization. In this context, the present study aims to investigate the effect of molar incisor hypomineralization (MIH) on periodontal health and to evaluate the relationship between MIH and GCF volume, as well as GCF biomarkers such as MMP-8, MMP-20, and

[Expression of the small molecular weight matrix metalloproteinase in adenomyosis of the mouse uterus].

The etiology and the pathogenesis of adenomyosis, which is a benign disease featuring ectopic proliferation and invasion of the endometrial stromas and glands into the myometrium, as seen with malignant tumor cells, are still unknown. Adenomyosis induced in mice by intrauterine pituitary isografts was analyzed to study the relationship between adenomyosis and matrix metalloproteinase. Under zymography, adenomyosis showed a gelatiolytic band of 20-30kDa, which was inhibited by EDTA. We then compared matrix metalloproteinase, which is a small protein, with the known pump-1 (MMP7) utilizing RT-PC

Temporal changes in myocardial collagen, matrix metalloproteinases, and their tissue inhibitors in the left ventricular myocardium in experimental chronic mitral regurgitation in rodents.

Mitral regurgitation (MR) imposes left ventricular volume overload, triggering rapid ventricular dilatation, increased myocardial compliance, and, ultimately, cardiac dysfunction. Breakdown of the extracellular matrix has been hypothesized to drive these rapid changes, partially from an imbalance in the matrix metalloproteinases (MMPs) and their tissue inhibitors [tissue inhibitors of metalloproteinase (TIMPs)]. In the present study, we developed a rat model of severe MR that mimics the human condition and investigated the temporal changes in extracellular matrix-related genes, collagen biosyn

[Matrix metalloproteinases and atherosclerosis. Therapeutic aspects].

Matrix metalloproteinases (MMPs) play a key role in the physiology of connective tissue development, morphogenesis and wound healing, but their unregulated activity has been implicated in numerous disease processes including arthritis, tumor cell metastasis and atherosclerosis. MMP family consists of at least 20 members; MMPs are produced by the different cell types (vascular smooth muscle cells, monocytes, endothelial cells) involved in the atheromatous plaque formation and participate to extracellular matrix remodelling and cell infiltration or migration. Since excessive tissue remodelling a

Anti-IL-20 monoclonal antibody suppresses breast cancer progression and bone osteolysis in murine models.

IL-20 is a proinflammatory cytokine involved in rheumatoid arthritis, atherosclerosis, and stroke. However, little is known about its role in breast cancer. We explored the function of IL-20 in tumor growth and metastasis, as well as in clinical outcome. Tumor expression of IL-20 was assessed by immunohistochemical staining among 198 patients with invasive ductal carcinoma of the breast, using available clinical and survival data. IL-20 expression was associated with advanced tumor stage, greater tumor metastasis, and worse survival. Reverse transcription quantitative polymerase chain reaction

Multi-omics integration and functional validation identify thrombospondin-1 as a functionally relevant mediator of doramectin-associated inhibition of glioblastoma migration and invasion.

Glioblastoma (GBM) exhibits highly infiltrative growth that limits therapeutic efficacy and promotes recurrence. Although doramectin (DRM) has been shown to induce apoptosis and autophagy in GBM cells, its effects on tumor migration and invasion remain unclear. We performed integrative transcriptomic and functional analyses to identify mediators associated with DRM anti-invasive activity. RNA sequencing of DRM-treated versus control rat C6 glioma cells identified 3747 differentially expressed genes, including 1660 upregulated and 2087 downregulated genes, with thrombospondin-1 (THBS1) markedly

Nobiletin Inhibits IL-1β-Induced Inflammation in Chondrocytes via Suppression of NF-κB Signaling and Attenuates Osteoarthritis in Mice.

Osteoarthritis (OA), a common degenerative joint disease, is principally characterized by inflammation and destruction of cartilage. Nobiletin, an extract of the peel of citrus fruits, is known to have anti-inflammatory properties. However, the mechanisms by which nobiletin plays a protective role in osteoarthritis (OA) are not completely understood. In the present study, we investigated the anti-inflammatory effects of nobiletin in the progression of OA in both in vitro and in vivo experiments. Mouse chondrocytes were pretreated with nobiletin (0, 10, 20, 40 μM) for 24 h and then incubated wi
