Sensory Innervation of the Larynx and the Search for Mucosal Mechanoreceptors.

Purpose The larynx is a uniquely situated organ, juxtaposed between the gastrointestinal and respiratory tracts, and endures considerable immunological challenges while providing reflexogenic responses via putative mucosal mechanoreceptor afferents. Laryngeal afferents mediate precise monitoring of sensory events by relay to the internal branch of the superior laryngeal nerve (iSLN). Exposure to a variety of stimuli (e.g., mechanical, chemical, thermal) at the mucosa-airway interface has likely evolved a diverse array of specialized sensory afferents for rapid laryngeal control. Accordingly, m

Mechanoreceptors sensitive to flow at the gastroduodenal junction of the cat.

Vagal afferent activity was recorded from the cervical vagus simultaneously with intraluminal pressure and flow rate at the proximal duodenum, in cats anesthetized with pentobarbital sodium. Thirty-seven mechanoreceptors that adapted slowly to maintained pressure changes were found to be sensitive to the flow of saline through the pylorus. The passage of liquid was associated with an increase of pressure equal to one-tenth of that necessary to trigger a response of the receptors during the distension of a small balloon. The frequency of discharge was increased by the passage of liquid for posi

Quantification of myelinated endings and mechanoreceptors in human digital skin.

We used immunohistochemistry and confocal microscopy applied to fingertip punch biopsy to study glabrous skin innervation in 14 healthy subjects. In addition to epidermal nerve fibers, we quantified mechanoreceptors and their myelinated afferents. Using digital images and dedicated software, we calculated caliber, internodal and nodal length, and G-ratio of the last four internodes of the myelinated endings. In our skin samples, we found a mean density of 59.0 +/- 29.3 myelinated endings per square millimeter with a mean diameter of 3.3 +/- 0.5 microm and an internodal length of 79.1 +/- 13.8

Anterior cruciate ligament mechanoreceptors. Histologic studies on lesions and reconstruction.

The fate of mechanoreceptors was studied in torn and reconstructed anterior cruciate ligaments in animals and in humans. In untreated anterior cruciate ligament lesions in humans, morphologically normal mechanoreceptors remained in the ligament for 3 months after the injury. After that time, their number gradually decreased. By the 9th postinjury month, only a few free nerve endings were present. Even free nerve endings were totally absent in the biopsy specimens from 1-year-old lesions. In experimental animals, anterior cruciate ligament reconstruction with autologous patellar tendon resulted

Ventilatory response to mechanoreceptor activation not altered in human hypertension.

The purpose of this study was to determine if the ventilatory responses during mechanically sensitive afferent stimulation were augmented in hypertension (HTN). Participants with HTN (n = 11; 52 ± 12 yrs; 27 ± 3 kg/m2) and controls (CTL, n = 11; 56 ± 12 yrs; 27 ± 4 kg/m2) were recruited. Following a resting period, the muscle mechanically sensitive afferents were stimulated with passive leg movement for 30-seconds. Ventilatory variables were measured (breath-by-breath) at rest and during passive leg movement protocols. All data was analyzed with a two-way repeated measures ANOVA. Significance

Encoding of Tactile Stimuli by Mechanoreceptors and Interneurons of the Medicinal Leech.

For many animals processing of tactile information is a crucial task in behavioral contexts like exploration, foraging, and stimulus avoidance. The leech, having infrequent access to food, developed an energy efficient reaction to tactile stimuli, avoiding unnecessary muscle movements: The local bend behavior moves only a small part of the body wall away from an object touching the skin, while the rest of the animal remains stationary. Amazingly, the precision of this localized behavioral response is similar to the spatial discrimination threshold of the human fingertip, although the leech ski

The effect of mechanoreceptor stimulation of the laryngopharynx on the oesophago-gastric junction.

Stimulation of the pharynx by injection of minute amounts of water induces prolonged period of lower oesophageal sphincter (LOS) relaxation and augmentation of the crural diaphragm (CD) contraction. The purpose of this study was to determine the effects of stimulating mechano-receptors at the laryngopharynx on the LOS and CD by using an air pulse stimulation device. Air pulses were delivered via the internal channel of a flexible endoscope. Oesophageal, LOS, and CD pressures; CD electromyogram; and oesophageal pH 5 cm above the LOS were recorded in nine healthy subjects. Stimulation of the lar

Studies of mechanoreceptors in skin of the snout of the echidna Tachyglossus aculeatus.

The echidna Tachyglossus aculeatus, together with the platypus, belongs to the monotremes, a group of mammals with a number of reptilian characteristics. A structure unique to the skin of monotremes is the push rod-a compacted column of epidermal cells that is 20 microns wide and 100 microns long with its tip at the skin surface, and that is able to move relatively independently of adjacent tissue. At the base of each push rod is a cluster of encapsulated nerve endings. Push rods are common in skin of the snout and have been postulated to have a mechanosensory function. Experiments were carrie

Rapidly and slowly adapting mechanoreceptors in the glans penis of the cat.

Sixty-one single mechanoreceptive fibers were surgically isolated from the dorsal nerve of the penis in anesthetized, mature, and sexually intact male cats. Impulse activity was recorded extracellularly. Receptive fields on the glans penis were stimulated with an accurate computer controlled mechanostimulator. Thirty-four units were categorized as rapidly adapting (RA) based on the absence of a response to sustained skin displacement. The remaining were slowly adapting (SA) units responding to sustained displacement. Most of the SA units were located in the distal smooth glans whereas RA units

Cholecystokinin octapeptide and gastric mechanoreceptor activity in rat brain.

Extracellular unitary recordings were made from neurons in the rat dorsal vagal nucleus, and the response of these neurons to iontophoretically applied cholecystokinin octapeptide (CCK-OP) and proglumide (a novel CCK antagonist) was studied. The effect of CCK-OP and proglumide on the response of the neurons to gastric distension was also studied. Seventy-four percent (n = 39) of the neurons studied were excited by CCK-OP. It was not possible to antagonize the response to CCK-OP with proglumide. CCK-OP was also found to be an effective modulator of the response to gastric distension; the effect

Aβ low threshold mechanoreceptors contribute to sensory abnormalities in fibromyalgia.

Fibromyalgia syndrome (FM) is characterized by widespread pain and fatigue. People living with FM also experience tactile allodynia, cold-evoked pain, paraesthesia and dysaesthesia. There is evidence of small fibre neuropathy and hyperexcitability of nociceptors in FM; however, the presence of other sensory abnormalities suggests involvement of large diameter sensory fibres. The passive transfer of FM IgG to mice causes cold and mechanical hyperalgesia associated with changes in A- and C-nociceptor function. However, whether FM IgG also confers sensitivity to light touch and whether large diam

Evidence of extraganglionic vagal mechanoreceptors in the mouse vagus nerve.

Vagal afferent neuronal somas are in the nodose and jugular ganglia. In this study, we identified extraganglionic neurons in whole-mount preparations of the vagus nerves from Phox2b-Cre-ZsGreen transgenic mice. These neurons are typically arranged in small clusters and monolayers along the cervical vagus nerve. Although infrequent, these neurons were sometimes observed along the thoracic and esophageal vagus. We performed RNAscope in situ hybridization and confirmed that the extraganglionic neurons detected in this transgenic mouse strain expressed vagal afferent markers (i.e., Phox2b and Slc1

Contribution of mechanoreceptors to spinal cord injury-induced mechanical allodynia.

Evidence from previous studies supports the concept that spinal cord injury (SCI)-induced neuropathic pain (NP) has its neural roots in the peripheral nervous system. There is uncertainty about how and to which degree mechanoreceptors contribute. Sensorimotor activation-based interventions (eg, treadmill training) have been shown to reduce NP after experimental SCI, suggesting transmission of pain-alleviating signals through mechanoreceptors. The aim of the present study was to understand the contribution of mechanoreceptors with respect to mechanical allodynia in a moderate mouse contusion SC

Changes in mechanoreceptors in rabbits' anterior cruciate ligaments with age.

At present, a few studies have been done on the changes in the distribution, morphology and quantity of mechanoreceptors in anterior cruciate ligament (ACL) with age. In this study, we observed the changes in mechanoreceptors of healthy rabbits' ACL with age. We found that rabbits' ACLs contained 5 kinds of mechanoreceptors including Ruffini corpuscles, Pacinian corpuscles, Golgitendon bodies, free nerve endings and atypical mechanoreceptors. In each ACL, free nerve endings were the most followed by Ruffini corpuscles, Pacinian corpuscles, Golgitendon bodies and atypical mechanoreceptors in th

Characteristics of periodontal mechanoreceptors supplying reimplanted canine teeth in cats.

These characteristics were investigated 12 weeks and one year after tooth reimplantation. Electrophysiological recordings were made from single mechanosensitive units dissected from the inferior alveolar nerve and each unit was characterized by applying forces to the crown of the tooth. The characteristics were compared with those of receptors innervating normal teeth. The periodontal mechanoreceptors supplying teeth reimplanted 12 weeks earlier responded to applied forces over a narrower range of directions, had higher force thresholds, lower discharge frequencies and adapted more rapidly tha

Mechanoreceptor synapses in the brainstem shape the central representation of touch.

Mammals use glabrous (hairless) skin of their hands and feet to navigate and manipulate their environment. Cortical maps of the body surface across species contain disproportionately large numbers of neurons dedicated to glabrous skin sensation, in part reflecting a higher density of mechanoreceptors that innervate these skin regions. Here, we find that disproportionate representation of glabrous skin emerges over postnatal development at the first synapse between peripheral mechanoreceptors and their central targets in the brainstem. Mechanoreceptor synapses undergo developmental refinement t

A study of mechanoreceptors in fibrocartilage masses in the defect of pars interarticularis.

We investigated the origin of low back pain associated with lumbar spondylolysis and spondylolytic spondylolisthesis by removing fibrocartilage masses from the lytic sites in symptomatic patients and staining the masses by the Gairns gold chloride method to examine mechanoreceptors. The fibrocartilage masses were found to contain four types of mechanoreceptors: Pacinian corpuscles, Ruffini receptors, Golgi tendon organ-like receptors, and free nerve endings. All of these mechanoreceptors were present at the periphery of the specimens, and Ruffini receptors and free nerve endings were abundant.

The interaction of chemo- and mechanoreceptor signals in the control of airway calibre.

The volume of an isolated tracheal segment and of the intrathoracic airways (intrathoracic portion of anatomical dead space) were measured during independent changes in ventilation minute volume and alveolar PCO2 and PO2, in order to assess the separate effects of chemoreceptor and mechanoreceptor signals upon airway calibre. The results were highly dependent upon initial airway tone, initial airway constriction favouring dilator responses and vice versa. Increasing alveolar PCO2 caused reflex airway constriction if ventilation was held constant. The magnitude of this response was reduced if v
