NaCl preference and water intake effects of food availability in median eminence polydipsia.

Lesions in the median eminence (ME) produce a well-known neurological model of polydipsia and polyuria in rats. The effect of food availability (ad lib/deprivation) on the polydipsic/polyuric behaviour of animals was tested. As expected, all lesioned rats developed strong polyuria and polydipsia during the first postoperative 24h. This effect was maintained during day 2 when food was available ad lib (experiment A), but both polyuria and polydipsia were abolished when animals were deprived of food during the next 24h (day 3). Animals deprived of food from the first post-operative day (experime

The fetal baboon median eminence as a circumventricular organ: I. Transmission electron microscopy.

This investigation has focused upon a set of neuroanatomical correlates that underscore functional changes in the median eminence of the fetal baboon Papio anubis. Eight fetal primate brains were harvested at mid-gestation (100 days post-coitus) and prepared for routine light and transmission election microscopic examination following ventriculo-cisternal perfusion with high osmolarity aldehyde fixatives. The median eminence and other adjacent circumventricular organs (CVO) were blocked and embedded in epon. Routine transmission electron microscopic examination revealed discreet regional diffe

Blood supply of the rat hypothalamus. IV. Retrochiasmatic area, median eminence, arcuate nucleus.

The vascular supply of the rat hypothalamo-hypophyseal complex was studied by aid of a double injection technique (two dyes different in colour injected successively). The vascular bed of the median eminence, the arcuate nucleus, and the retrochiasmatic area are intimately interconnected among themselves, but isolated from the vascular system of all other parts of the hypothalamus. The arterial supply of this ensemble of the medial basal hypothalamus is common from the hypophyseal arteries, via the primary plexus and the specific vascular loops of the median eminence. From here a separation of

SEMA6A drives GnRH neuron-dependent puberty onset by tuning median eminence vascular permeability.

Innervation of the hypothalamic median eminence by Gonadotropin-Releasing Hormone (GnRH) neurons is vital to ensure puberty onset and successful reproduction. However, the molecular and cellular mechanisms underlying median eminence development and pubertal timing are incompletely understood. Here we show that Semaphorin-6A is strongly expressed by median eminence-resident oligodendrocytes positioned adjacent to GnRH neuron projections and fenestrated capillaries, and that Semaphorin-6A is required for GnRH neuron innervation and puberty onset. In vitro and in vivo experiments reveal an unexpe

Thenar eminence quantitative sensory testing in the diagnosis of proximal median nerve compression.

Diagnosis of proximal median nerve compression (PMNC) remains a clinical challenge. The authors hypothesized that measurement of the sensibility of the thenar eminence might identify PMNC by demonstrating abnormal function in the palmar cutaneous branch of the median nerve. This hypothesis was evaluated by means of quantitative sensory testing of the thenar eminence in 33 healthy volunteers, 14 patients with carpal tunnel syndrome, and 35 patients with PMNC. The cutaneous pressure thresholds for one-point static touch (1PS) and two-point static touch (2PS) were measured with the Pressure-speci

Effect of oestrogen-dependent vasopressin on HPA axis in the median eminence of female rats.

The median eminence (ME) anatomically consists of external (eME) and internal (iME) layers. The hypothalamic neurosecretory cells terminate their axons in the eME and secrete their neurohormones regulating anterior pituitary hormone secretion involved in stress responses into the portal vein located in the eME. Magnocellular neurosecretory cells (MNCs) which produce arginine vasopressin (AVP) and oxytocin in the paraventricular (PVN) and supraoptic nuclei (SON) terminate their axons in the posterior pituitary gland (PP) through the iME. Here, we provide the first evidence that oestrogen modula

Modulation of LH secretion by progesterone and estradiol after naloxone injection in the median eminence.

To evaluate whether the median eminence (ME) is a site of action of opioids on gonadotropin secretion, and to determinate the role of estradiol (E) and progesterone (P) in modifying these secretion in response to naloxone, we injected naloxone (1, 2.5 and 5 micrograms) into the ME in ovariectomized rats treated with estradiol benzoate (OVX+E) or ovariectomized rats treated with estradiol benzoate plus progesterone (OVX+E+P), and measured LH and FSH serum levels 10 and 30 min postinjection. Naloxone injection at doses of 2.5 and 5 micrograms significantly increased serum LH levels in OVX+E anim

Evidence for the existence of a dopamine receptor of the D-1 type in the rat median eminence.

By means of receptor autoradiography using the dopamine (DA) receptor radioligands [3H]cis(z)-flupenthixol ([3H]FLU), [3H]spiperone, [3H]N-propyl-norapomorphine and amino-6,7-dihydroxy-1,2,3,4-tetrahydronaphtalene-2-(5,8[3H]) in combination with a microdensitometrical analysis indications have been obtained for the existence of a DA receptor of the D-1 type in the median eminence of the male rat. Thus, only [3H]FLU (10-20 nM) strongly labeled both the nuc. caudatus putamen and the median eminence and the labeling was markedly prevented by (+)-butaclamol in both regions. Furthermore, a DA recep

The hypothalamo-hypophysial system of hypophysectomized rats. II. Structure and ultrastructure of the median eminence.

The median eminence (ME) of hypophysectomized rats was studied by means of light and electron microscopy. Paraldehyde-fuchsin (PAF)-positive material is seen in the external zone (EZ) of the ME 2--5 days after the operation. Its amount gradually increases especially in the caudal part of the ME during the following few days. Some PAF-positive fibers make contact with the subependymally located blood capillaries. In the most caudal region of the recessus infundibuli they penetrate into the third ventricle. PAF-positive material decreases markedly from the ME of rats two months after hypophysect

Immunocytochemical delineation of thyrotrophic area: origin of thyrotropin-releasing hormone in the median eminence.

To identify the specific thyrotropin-releasing hormone (TRH)-containing neurons projecting to the median eminence (ME), a retrograde tracing method with horseradish peroxidase (HRP) was combined with immunocytochemical staining for TRH. Three days after HRP injection restricted to the ME, several TRH-positive neuronal perikarya were found to contain HRP. Such double-stained cells were exclusively distributed in the anterior parts of the periventricular nucleus and the most medial parts of the paraventricular nucleus. Few double-stained cells were observed in other parts of the brain examined.

Translingual approach of the basal surface of the diencephalon of the rat and retrograde labeling from the median eminence.

A unique surgical procedure is described by which the basal surface of the diencephalon can be exposed without cannulation of the trachea. The basal surface of the diencephalon is exposed through a midsagittal incision by splitting the oral diaphragm, tongue, and soft palate respectively. Then a small hole is drilled in the base of the skull. After manipulation on the base of the brain is completed, the hole in the skull is plugged with Histoacryl and the soft palate and tongue are sutured. Rats having such interventions survive in excellent condition for days without the need of intensive med

Effects of p-chloroamphetamine, a serotonin-depleting drug, on the median eminence and pituitary pars intermedia.

p-Chloroamphetamine (PCA), an agent known to cause depletion of levels of brain serotonin in rodents, was administered to rats in three sequential injections (10mg/kg) to study effects on the hypothalamic median eminence and pituitary gland. One week following the initial sequence of injections of PCA, light and electron micrographs revealed degenerate fibers in the outer zone of the median eminence. Lower drug doses or single 10-mg/kg doses did not lead to morphologic changes. Neuronal processes located in the pituitary intermediate lobe appeared normal although there was a significant increa

Neuroestradiol in the Stalk Median Eminence of Female Rhesus Macaques Decreases in Association With Puberty Onset.

In primates, despite the fact that GnRH neurons are mature at birth, a gonadal steroid independent central inhibition restrains the initiation of puberty. The neural substrates responsible for this central inhibition, however, are unclear. In this study, we tested the hypothesis that neuroestradiol release in the hypothalamus decreases prior to the pubertal increase in GnRH release. We found that in female monkeys at the prepubertal stage, when GnRH release was low, estradiol (E2) levels in the stalk-median eminence of the hypothalamus were higher than those in older, early pubertal females in

Projections to the median eminence and the arcuate nucleus with special reference to monoamine systems: effects of lesions.

The external layer of the median eminence (ELME) and the arcuate nucleus of male rats were studied with the Falck-Hillarp technique and electron microscopy of aldehyde-OsO4 or KMnO4 fixed material after various types of hypothalamic deafferentation experiments with the Haláz knife. Special reference was paid to the monoamine systems and the results can be summarized as follows. 1. The main monoaminergic input to the ELME comes from the arcuate nucleus-periventricular area via a dorsal approach. A horizontal transection through the arcuate nucleus decreases the percentage of monoamine boutons i

Adult NG2-Glia Are Required for Median Eminence-Mediated Leptin Sensing and Body Weight Control.

While leptin is a well-known regulator of body fat mass, it remains unclear how circulating leptin is sensed centrally to maintain energy homeostasis. Here we show that genetic and pharmacological ablation of adult NG2-glia (also known as oligodendrocyte precursors), but not microglia, leads to primary leptin resistance and obesity in mice. We reveal that NG2-glia contact the dendritic processes of arcuate nucleus leptin receptor (LepR) neurons in the median eminence (ME) and that these processes degenerate upon NG2-glia elimination, which explains the consequential attenuation of these neuron

Gene expression in arcuate nucleus-median eminence of rats treated with leptin or ciliary neurotrophic factor.

Ciliary neurotrophic factor (CNTF) and leptin are cytokine-like% hormones and act on their corresponding receptors in the hypothalamic arcuate nucleus (ARC). The present study was designed to assess effects of intracerebroventricular (ICV) injection of leptin and CNTF on gene expression in micropunched hypothalamic arcuate nucleus-median eminence (ARC-ME) complex samples from rats. Male Sprague Dawley rats were implanted with lateral cerebroventricular cannulas for administration of control, 10 microg/d leptin or 5 microg/d CNTF for four days. Real-time Taqmantrade mark RT-PCR was used to quan

Blockade of volume expansion-induced release of atrial natriuretic peptide by median eminence lesions in the rat.

Since stimulation of the anteroventral third ventricle region (AV3V) induced a rapid elevation of plasma atrial natriuretic peptide (ANP) associated with rapid changes in brain and pituitary content of ANP, whereas lesions of the AV3V were followed by a marked decline in plasma, brain and pituitary content of the peptide, we hypothesized that release of ANP from the median eminence (ME) might be an important pathway to control plasma ANP. Consequently, electrolytic lesions were placed in the ME and the response to hypertonic-volume expansion was determined in conscious rats. In sham-operated c

The location of LH-RH neurons in the rat hypothalamus and their pathways to the median eminence. Experimental immunohistochemistry and radioimmunoassay.

The location of the perikarya of LH-RH neurons in the rat hypothalamus and their pathways to the median eminence were studied by immunohistochemistry and radioimmunoassay after placing stereotaxic electrolytic lesions in several parts of the hypothalamus. The principal location of the cell somata was found to be in the ventral part of the medial preoptic area; their pathways were classified into a main baso-lateral pathway and an accessory descending pathway branching off from the former. The main pathway was found to cross in the vicinity of the corresponding neuronal perikarya. The central m
