Medicarpin confers powdery mildew resistance in Medicago truncatula and activates the salicylic acid signalling pathway.

Powdery mildew (PM) caused by the obligate biotrophic fungal pathogen Erysiphe pisi is an economically important disease of legumes. Legumes are rich in isoflavonoids, a class of secondary metabolites whose role in PM resistance is ambiguous. Here we show that the pterocarpan medicarpin accumulates at fungal infection sites, as analysed by fluorescein-tagged medicarpin, and provides penetration and post-penetration resistance against E. pisi in Medicago truncatula in part through the activation of the salicylic acid (SA) signalling pathway. Comparative gene expression and metabolite analyses r

Dwarf and Increased Branching 1 controls plant height and axillary bud outgrowth in Medicago truncatula.

Optimizing plant architecture is an efficient approach for breeders to increase crop yields, and phytohormones such as gibberellins (GAs) play an important role in controlling growth. Medicago truncatula is a model legume species, but the molecular mechanisms underlying its architecture are largely unknown. In this study, we examined a tobacco retrotransposon Tnt1-tagged mutant collection of M. truncatula and identified dwarf and increased branching 1 (dib1), which exhibited extreme dwarfism and increased numbers of lateral branches. By analysis of the flanking sequences of Tnt1 insertions in

Regulatory divergence of duplicated VIH genes links phosphate signaling to nodule development in Medicago truncatula.

Primary metabolites and their derivatives often serve as intracellular signals. Inositol pyrophosphates are central regulators of phosphate signaling, but their roles in legume nodulation remain poorly understood. Here, we identify two conserved Vip1 Homolog/Diphosphoinositol Pentakisphosphate Kinase genes, MtVIH1 and MtVIH2, in Medicago truncatula. Biochemical analyses show that the kinase domains of both MtVIH1 and MtVIH2 retain PP-InsP kinase activity. However, transcriptomic and expression analyses reveal regulatory divergence between the duplicated genes, with MtVIH2 being preferentially

Identification, characterization, and evolutionary analysis of aldehyde dehydrogenase (ALDH) genes superfamily in Medicago truncatula L.

Aldehydes are reactive compounds that play crucial roles in various metabolic processes within plants. However, their accumulation can lead to toxic effects, Aldehyde dehydrogenases (ALDHs) represent a diverse family of enzymes that catalyze the oxidation of aldehydes to carboxylic acids. ALDHs help mitigate the toxic effects of these compounds and maintain cellular homeostasis in plants. In this study, a bioinformatics analysis of the Medicago truncatula genome identified 27 MtALDHs, which were classified into ten distinct groups based on their phylogenetic relationships. The distribution of

Indole-3-acetaldoxime delays root iron-deficiency responses and modify auxin homeostasis in Medicago truncatula.

Iron (Fe) is an essential plant micronutrient, being a major limiting growth factor in calcareous soils. To increase Fe uptake, plants induce lateral roots growth, the expression of a Fe(III)-chelate reductase (FCR), a Fe(II)-transporter and a H+-ATPase and the secretion of flavins. Furthermore, auxin hormone family is involved in the Fe-deficiency responses but the action mechanism remains elusive. In this work, we evaluated the effect of the auxin-precursor indole-3-acetaldoxime (IAOx) on hydroponically grown Medicago truncatula plants under different Fe conditions. Upon 4-days of Fe starvat

Physiological and transcriptomic response of Medicago truncatula to colonization by high- or low-benefit arbuscular mycorrhizal fungi.

Arbuscular mycorrhizal (AM) fungi form a root endosymbiosis with many agronomically important crop species. They enhance the ability of their host to obtain nutrients from the soil and increase the tolerance to biotic and abiotic stressors. However, AM fungal species can differ in the benefits they provide to their host plants. Here, we examined the putative molecular mechanisms involved in the regulation of the physiological response of Medicago truncatula to colonization by Rhizophagus irregularis or Glomus aggregatum, which have previously been characterized as high- and low-benefit AM fung

Transcriptional snapshots provide insights into the molecular basis of arbuscular mycorrhiza in the model legume Medicago truncatula.

The arbuscular mycorrhizal (AM) association between terrestrial plants and soil fungi of the phylum Glomeromycota is the most widespread beneficial plant-microbe interaction on earth. In the course of the symbiosis, fungal hyphae colonise plant roots and supply limiting nutrients, in particular phosphorus, in exchange for carbon compounds. Owing to the obligate biotrophy of mycorrhizal fungi and the lack of genetic systems to study them, targeted molecular studies on AM symbioses proved to be difficult. With the emergence of plant genomics and the selection of suitable models, an application o

Metabolic profiling of Medicago truncatula cell cultures reveals the effects of biotic and abiotic elicitors on metabolism.

GC-MS-based metabolite profiling was used to analyse the response of Medicago truncatula cell cultures to elicitation with methyl jasmonate (MeJa), yeast elicitor (YE), or ultraviolet light (UV). Marked changes in the levels of primary metabolites, including several amino acids, organic acids, and carbohydrates, were observed following elicitation with MeJa. A similar, but attenuated response was observed following YE elicitation, whereas little response was observed following UV elicitation. MeJa induced the accumulation of the triterpene beta-amyrin, a precursor to the triterpene saponins, a

Genome-wide identification of MtTCTPs and functional analysis of MtTCTP3 as a positive regulator of nodulation in Medicago truncatula.

The Translationally Controlled Tumor Protein (TCTP) is a conserved eukaryotic regulator of cell proliferation and stress responses, but its function in legume-rhizobium symbiosis remains unclear. Here, we identified five MtTCTP genes in the model legume Medicago truncatula and characterized the role of MtTCTP3 in symbiotic nodulation. MtTCTP3 is highly expressed in root nodules, particularly in the nodule apical meristem. Overexpression of MtTCTP3 significantly enhanced nodule formation, nitrogenase activity, and biomass accumulation, while silencing of MtTCTP3 caused reduced nodule number and

The effect of Sinorhizobium meliloti volatilomes and synthetic long-chain methylketones on soil and Medicago truncatula microbiomes.

Bacterial volatile compounds play important roles in intra- and interkingdom interactions but little is known about their effects on soil and plant microbiomes. The legume symbiont Sinorhizobium meliloti (Sm) releases volatile methylketones (MKs), one of which acts as an infochemical among bacteria and hampers plant-bacteria interactions. Inactivation of the fatty acyl-CoA ligase FadD in Sm moderately enhances MK production. To further explore the ecological role of MKs on soil and plant bacterial communities, we aimed at obtaining an MK-overproducing Sm strain by deleting the 3-oxoacyl-CoA th

Plant-Compatible Xenium In Situ Sequencing: Protocol for Spatial Transcriptomics in Medicago truncatula Roots and Nodules.

Elucidating the spatial and temporal regulation of gene expression during plant organogenesis is crucial for enabling precise crop improvement strategies that incorporate beneficial traits into crops while avoiding adverse effects. Root nodules, specialized organs formed in symbiosis with nitrogen-fixing bacteria, provide a valuable system to study cell-type-specific gene networks in a symbiosis-induced developmental context. However, capturing these dynamics at cellular resolution in intact plant tissues remains technically challenging. Spatial transcriptomics technologies developed for anima

Differential regulation of the "phytoglobin-nitric oxide respiration" in Medicago truncatula roots and nodules submitted to flooding.

Flooding induces hypoxia in plant tissues, impacting various physiological and biochemical processes. This study investigates the adaptive response of the roots and nitrogen-fixing nodules of Medicago truncatula in symbiosis with Sinorhizobium meliloti under short-term hypoxia caused by flooding. Four-week-old plants were subjected to flooding for 1-4 days. Physiological parameters as well as the expression of the senescence marker gene MtCP6 remained unchanged after 4 days of flooding, indicating no senescence onset. Hypoxia was evident from the first day, as indicated by the upregulation of

The importance of the urea cycle and its relationships to polyamine metabolism during ammonium stress in Medicago truncatula.

The ornithine-urea cycle (urea cycle) makes a significant contribution to the metabolic responses of lower photosynthetic eukaryotes to episodes of high nitrogen availability. In this study, we compared the role of the plant urea cycle and its relationships to polyamine metabolism in ammonium-fed and nitrate-fed Medicago truncatula plants. High ammonium resulted in the accumulation of ammonium and pathway intermediates, particularly glutamine, arginine, ornithine, and putrescine. Arginine decarboxylase activity was decreased in roots, suggesting that the ornithine decarboxylase-dependent produ

Legume adaptation to sulfur deficiency revealed by comparing nutrient allocation and seed traits in Medicago truncatula.

Reductions in sulfur dioxide emissions and the use of sulfur-free mineral fertilizers are decreasing soil sulfur levels and threaten the adequate fertilization of most crops. To provide knowledge regarding legume adaptation to sulfur restriction, we subjected Medicago truncatula, a model legume species, to sulfur deficiency at various developmental stages, and compared the yield, nutrient allocation and seed traits. This comparative analysis revealed that sulfur deficiency at the mid-vegetative stage decreased yield and altered the allocation of nitrogen and carbon to seeds, leading to reduced

Biochemical analysis of the TPS-b subfamily reveals a cineole-centered monoterpene biosynthetic module in Medicago truncatula.

Terpenoids constitute one of the largest and most structurally diverse classes of plant specialized metabolites, with diversity generated by terpene synthases (TPSs) and downstream tailoring enzymes. In Medicago truncatula, the TPS-b subfamily comprises five putative synthases, two of which are embedded within a previously uncharacterized genomic locus containing a cytochrome P450 (CYP) and a BAHD-type acyltransferase. Here, we present a comprehensive biochemical analysis of the M. truncatula TPS-b subfamily and define a cineole-centered monoterpene biosynthetic module. Heterologous expression

Duplicated CYP71D enzymes promote metabolic innovation through regiospecific oxidation at a sesquiterpenoid biosynthetic locus in Medicago truncatula.

Sesquiterpenoid chemical diversity is frequently generated through cytochrome P450 (CYP)-mediated oxidation of terpene scaffolds. In Medicago truncatula, the sesquiterpene alcohol himachalol, produced by the terpene synthase MtTPS10, has been implicated in pathogen defense, yet the enzymes responsible for its downstream modification remain undefined. Here, we characterize a compact locus containing two sesquiterpene synthases and three CYP71D genes. Using heterologous expression, we show that two duplicated CYP71D enzymes catalyze regiospecific oxidations of himachalol. CYP71D81 hydroxylates t

NO loading: Efficiency assessment of five commonly used application methods of sodium nitroprusside in Medicago truncatula plants.

Nitric oxide (NO) is a bioactive, diffusible molecule involved in a multitude of physiological and developmental processes in plants, which has been reported to display both antioxidant and pro-oxidant properties in plants. Several reports exist highlighting the protective action of sodium nitroprusside (SNP), an NO donor, which demonstrate its important role as a signal molecule in plants responsible for the expression regulation of antioxidant and other defense enzymes. However, the mode of application of this compound varies greatly between studies. The present study provides a comprehensiv

Elevated CO2 alters the feeding behaviour of the pea aphid by modifying the physical and chemical resistance of Medicago truncatula.

Elevated CO(2) compromises the resistance of leguminous plants against chewing insects, but little is known about whether elevated CO(2) modifies the resistance against phloem-sucking insects or whether it has contrasting effects on the resistance of legumes that differ in biological nitrogen fixation. We tested the hypothesis that the physical and chemical resistance against aphids would be increased in Jemalong (a wild type of Medicago truncatula) but would be decreased in dnf1 (a mutant without biological nitrogen fixation) by elevated CO(2). The non-glandular and glandular trichome density

Genome-Wide Characterization of the PLATZ Gene Family in Medicago truncatula and Their Expression Profiles in Response to Abiotic Stress.

Plant A/T-rich sequence and zinc-binding protein (PLATZ) transcription factors are pivotal regulators of plant growth, development, and abiotic stress responses. Although PLATZ genes have been extensively studied and functionally characterized in various plant species, little information is available regarding these genes in the legume model plant Medicago truncatula. In this study, a total of 16 PLATZ genes (designated MtPLATZ1 to MtPLATZ16) were identified from the M. truncatula genome and distributed across six of the eight chromosomes. Subcellular localization prediction analysis indicated
