The efficacy of treatment with intralesional meglumine antimoniate alone, compared with that of cryotherapy combined with the meglumine antimoniate or intralesional sodium stibogluconate, in the treatment of cutaneous leishmaniasis.

It would be very useful to have a more effective and more rapid method available for the treatment of cutaneous leishmaniasis (CL). The main aim of the present, Iranian study, was to see if the combination of cryotherapy and intralesional injections with meglumine antimoniate (C + MA) would be more effective than the injections given alone (MA) or the combination of cryotherapy plus intralesional sodium stibogluconate (C + SS). Forty patients (with 67 lesions) were treated with C + MA, another 40 (with 65 lesions) were treated with C + SS and 100 patients (with 180 lesions) were treated with M

Effectiveness of harmaline along with meglumine antimoniate on Leishmania major.

Leishmaniasis is a disease caused by Leishmania species and transmitted via sandflies. Current control strategies against reservoir hosts and vectors are not eco-friendly. Using harmaline (HA) from Peganum harmala, and meglumine antimoniate (MA) could be a promising therapy. The study aimed to explore the potential treatment outcomes and action mechanisms of HA and MA against Leishmania major stages by investigating their effectiveness through molecular docking, anti-leishmanial effects, safety assessment, and apoptotic profile evaluations. According to the molecular docking results, the prote

Comparison of meglumine antimoniate and pentamidine for peruvian cutaneous leishmaniasis.

Pentamidine was compared with meglumine antimoniate (Glucantime) for 80 patients with cutaneous leishmaniasis due to Leishmania braziliensis in Peru. Of the 40 patients administered Glucantime (20 mg of antimony [Sb]/kg/day intravenously for 20 days), 31 cured (78%), 6 failed (15%), of which 5 were due to relapse, and 3 were lost to follow-up (7%). Of the 40 patients administered pentamidine (2 mg/kg every other day for seven injections), 14 were cured (35%), 23 failed (58%), and 3 were lost to follow-up (7%). Five pentamidine failures were due to relapse, and 14 failures were due to the prese

[Miltefosine versus meglumine antimoniate in the treatment of mucosal leishmaniasis].

The conventional treatment for tegumentary leishmaniasis is meglumine antimoniate, which needs parenteral administration, has increased therapeutic failure, and produces serious adverse effects, justifying the search for therapeutic alternatives. We report here the preliminary results of a phase II clinical trial in patients with mucosal leishmaniasis, in which the efficacy of oral miltefosine versus the antimonial compound was assessed. The evaluation of response to the treatment was performed by monitoring with nasopharyngeal video-fibroscopy, using a score of mucosal injury severity for pat

Generation of formaldehyde by pharmaceutical excipients and its absorption by meglumine.

Formaldehyde is a well-known air impurity. The possibility was investigated in this study that pharmaceutical excipients commonly used in oral solid dosage forms might also be sources of formaldehyde. The results showed that formaldehyde is generated by the excipients lactose, D-mannitol, microcrystalline cellulose, low-substituted hydroxypropylcellulose, magnesium stearate and light anhydrous silicic acid. Since the quality and safety of pharmaceutical products can be significantly affected by the presence of formaldehyde, various amines were then investigated for their ability to decrease le

Pharmacokinetics of antimony in children treated for leishmaniasis with meglumine antimoniate.

BACKGROUND: In some settings, the response to pentavalent antimonial therapy for leishmaniasis may be lower in children than in adults. We hypothesized that there are age-dependent pharmacokinetic differences of potential clinical relevance. METHODS: We compared the pharmacokinetics of antimony (Sb) in adults and 2 groups of children 3-6 years old who had cutaneous leishmaniasis treated with intramuscular meglumine antimoniate. Adults (n=9) and the first group of children (n=9) received 20 mg Sb/kg/day for 20 days; the second group of children (n=6) received 20 mg Sb/kg for 19 days and 30 mg

Absence of complement activation in vitro by sodium meglumine ioxitalamate.

The effect of radiographic contrast media on proteins of the complement system in vitro was investigated using protein level measurement and crossed immunoelectrophoresis. Despite the 20% loss of total serum hemolytic activity (CH50) induced by sodium meglumine ioxitalamate (0.5 M), no significant changes in C3c, C3N or B were detected by radial immunodiffusion. These contrast media did not cause electrophoretic conversion of C3, as assessed by the absence of split product, and the small degree of spontaneous C3 conversion (loss of 20% C3N antigen and the presence of two peaks with C3c antigen

Safety profile of meglumine antimoniate intralesional infiltration for cutaneous leishmaniasis.

Objectives: Cutaneous leishmaniasis is a neglected disease, associated with high morbidity, which is partially due to the toxicity of available therapies. The pentavalent antimonial derivatives intralesional infiltration has proven to be as effective as the intravenous drug-based therapy, however, there is a lack of robust safety data. Methods: Phase II, uncontrolled, unicenter clinical trial to assess the safety profile of a standardized meglumine antimionate intralesional therapy, based on weekly infiltrations. Results: Fifty-three patients were studied, predominantly men (60%) and young adult

Systemic meglumine antimoniate in acute cutaneous leishmaniasis: children versus adults.

Some studies showed that children have a lower response to systemic use of pentavalent antimoniate than adults. We aimed to evaluate the response rate to Glucantime therapy in children and compare it with adults. One hundred and twelve patients with acute cutaneous leishmaniasis: (ACL) were divided into two equal groups of adults (> 15 yrs) and children (≤ 15 yrs). They received meglumine antimoniate; 20 mg/kg/day for 20 days, their improvement rate was evaluated 20 and 45 days after treatment. Per-protocol analysis showed a significantly lower response in the children group 20 and 45 days aft

Gastroduodenal ulceration associated with flunixin meglumine administration in three dogs .

In 3 clinically ill dogs, signs of gastroduodenal ulceration were first noticed within 7 days of beginning flunixin meglumine administration and included pyrexia, anorexia, weight loss, vomiting, melena, pain on abdominal palpation, and abdominal distention. One dog was euthanatized and 2 dogs recovered after surgical repair of the perforated ulcers and treatment of peritonitis. Prolonged administration of flunixin meglumine should be avoided, especially in debilitated dogs or when concurrently administering other nonsteroidal anti-inflammatory drugs or corticosteroids.

Noninvasive CT determination of arterial blood concentration of meglumine iothalamate.

A quantity that often must be determined in physiological imaging studies is the blood concentration of the tracer over time. This is usually performed by direct arterial or venous blood sampling. We studied the relationship between the concentration of meglumine iothalamate in arterial blood and values determined from voxels containing large blood vessels in a series of CT images at the same location over time. After correction for volume averaging based on a single venous blood sample, there was an excellent correlation between the two blood curves. Differences between the curves were shown

Intussusception: hydrostatic pressure equivalents for barium and meglumine sodium diatrizoate.

The pressures generated by a barium suspension and various solutions of meglumine sodium diatrizoate in water were measured with a manometer. A pressure of 120 mm Hg was produced by a 3.5-foot (105-cm) column of 60% wt/vol barium and a 5-foot (150-cm) column of either a 1:3 or 1:4 solution of meglumine sodium diatrizoate and water. This is the pressure used to reduce an intussusception with air.
