Receptor mechanisms in fish chromatophores--VI. Adenosine receptors mediate pigment dispersion in guppy and catfish melanophores.

Using the guppy, Lebistes reticulatus, and the siluroid catfish, Parasilurus asotus , the effects of purine and pyrimidine derivatives on the movement of melanophores were studied. All the substances tested did not aggregate pigment within melanophores. Adenosine and adenine nucleotides were very effective in dispersing melanosomes within the cell, although adenine itself lacked such action. Derivatives of other purines than adenine and of pyrimidines did not disperse melanosomes. The pigment dispersion induced by adenine derivatives was specifically antagonized by methylxanthines. It was conc…

A comparative ultrastructural and physiological study on melanophores of wild-type and periodic albino mutants of Xenopus laevis.

Pigment of tail-fin melanophores in periodic albino Xenopus laevis tadpoles is dispersed in response to darkness and to alpha-MSH in a manner similar to wild-type melanophores. However, periodic albino tadpoles lack the response to different background conditions and the melatonin-induced aggregation in darkness. The tyrosinase activity in cells of the latter type tadpoles is weak compared to the wild-type cells. Ultrastructural examination of melanophores from periodic albino mutants and cells from wild-type tadpoles shows similar organelles at corresponding sites. A morphological difference …

The regulation of skin pigmentation in response to environmental light by pineal Type II opsins and skin melanophore melatonin receptors.

Coupling skin colour with the light/dark cycle helps regulate body temperature in ectotherms. In X. laevis, nocturnal release of melatonin f … nvolvement of opsins with sensitivities therein. The effector-melatonin receptors expressed in skin melanophores are mtnr1a and mtnr1c. Our data point to candidate proteins required in the neuroendocrine circuit that underlies the circadian regulation of skin pigmentation, and suggest that multiple initiators and effectors likely participate.

Experimental evidence for autonomous action of the periodic albinism (ap) gene within developing retinal pigment cells and melanophores of Xenopus laevis.

Genes which affect pigment elaboration may do so by autonomous action within the developing pigment cells or by way of tissue interactions leading to pigment cell differentiation. The site of action of the periodic albinism (ap) gene was investigated by substituting presumptive neural ectoderm of gastrulae of one genotype with uncommitted ectoderm of different genotype. Retinal pigment cells and melanophores arising from such grafts were found to differentiate according to their own genotype in spite of having spent their entire developmental history in tissues of different genotype. This find…

Inhibition of adenylate cyclase activity in the goldfish melanophore is mediated by α₂-adrenoceptors and a pertussis toxin-sensitive GTP-binding protein.

The signal-transduction system that mediates the melanosome-aggregating response in melanophores of the black-moor goldfish, Carassius auratus, was investigated by examining the inhibition of adenylate cyclase activity mediated by α-adrenoceptors in cultured cells. When the melanophores were incubated with 1 mmol·1⁻¹ 3-isobutyl-1-methylxanthine for 5 min, the intracellular level of cyclic adenosine-3',5'-monophosphate increased two- to three-fold. Norepinephrine at 100 nmol· 1⁻¹ and naphazoline at 1 μmol· 1⁻¹ inhibited the 3-isobutyl-1-methylxanthine-induced accumulation of cyclic adenosine-3'…

Multiple splice variants within the bovine silver homologue (SILV) gene affecting coat color in cattle indicate a function additional to fibril formation in melanophores.

BACKGROUND: The silver homologue(SILV) gene plays a major role in melanosome development. SILV is a target for studies concerning melanoma diagnostics and therapy in humans as well as on skin and coat color pigmentation in many species ranging from zebra fish to mammals. However, the precise functional cellular mechanisms, in which SILV is involved, are still not completely understood. While there are many studies addressing SILV function upon a eumelaneic pigment background, there is a substantial lack of information regarding the further relevance of SILV, e.g. for phaeomelanosome developmen

AP-3 complex subunit delta gene, ap3d1, regulates melanogenesis and melanophore survival via autophagy in zebrafish (Danio rerio).

Zebrafish are an emerging model organism to study the syndromic albinism disorder, Hermansky-Pudlak syndrome (HPS), due to visible pigment development at 24 hours postfertilization, and conserved melanogenesis mechanisms. We describe crasher, a novel HPS type 10 (HPS10) zebrafish model, with a mutation in AP-3 complex subunit delta gene, ap3d1. Exon 14 of ap3d1 is overexpressed in crasher mutants, while the expression of ap3d1 as a whole is reduced. ap3d1 knockout in *AB zebrafish recapitulates the mutant crasher phenotype. We show ap3d1 loss-of-function mutations cause significant expression

Immunoglobulin superfamily receptor Junctional adhesion molecule 3 (Jam3) requirement for melanophore survival and patterning during formation of zebrafish stripes.

Adhesive interactions are essential for tissue patterning and morphogenesis yet difficult to study owing to functional redundancies across genes and gene families. A useful system in which to dissect roles for cell adhesion and adhesion-dependent signaling is the pattern formed by pigment cells in skin of adult zebrafish, in which stripes represent the arrangement of neural crest derived melanophores, cells homologous to melanocytes. In a forward genetic screen for adult pattern defects, we isolated the pissarro (psr) mutant, having a variegated phenotype of spots, as well as defects in adult …

Alpha-melanophore-stimulating hormone in the brain, cranial placode derivatives, and retina of Xenopus laevis during development in relation to background adaptation.

The amphibian Xenopus laevis can adapt the color of its skin to the light intensity of the background. A key peptide in this adaptation process is alpha-melanophore-stimulating hormone (alpha-MSH), which is derived from proopiomelanocortin (POMC) and released by the endocrine melanotrope cells in the pituitary pars intermedia. In this study, the presence of alpha-MSH in the brain, cranial placode derivatives, and retina of developing Xenopus laevis was investigated using immunocytochemistry, to test the hypothesis that POMC peptide-producing neurons and endocrine cells have a common embryonic

RPE65 knockout Xenopus laevis have a compromised but detectable electroretinogram and altered visual responses, without retinal degeneration or altered melanophore dispersion.

The isomerohydrolase RPE65 is a critical element of the visual cycle, the series of enzymatic reactions by which the chromophore of the visual pigments is regenerated following light exposure. In humans, mutations in the RPE65 gene cause a severe form of blindness called Leber's congenital amaurosis. Studies of Rpe65-/- mice have shown dramatic depletion of 11-cis-retinal in the retina, resulting in a slow retinal degeneration. However, a number of studies suggest that RPE65 may not be necessary for the regeneration of photopigment in all photoreceptor types. Using CRISPR/Cas9 technology, we p

oca2 Regulation of chromatophore differentiation and number is cell type specific in zebrafish.

We characterized a zebrafish mutant that displays defects in melanin synthesis and in the differentiation of melanophores and iridophores of the skin and retinal pigment epithelium. Positional cloning and candidate gene sequencing link this mutation to a 410-kb region on chromosome 6, containing the oculocutaneous albinism 2 (oca2) gene. Quantification of oca2 mutant melanophores shows a reduction in the number of differentiated melanophores compared with wildtype siblings. Consistent with the analysis of mouse Oca2-deficient melanocytes, zebrafish mutant melanophores have immature melanosomes…

Anemonefish Chromatophore Distribution and Organization Revealed by TEM Studies.

Anemonefish have a characteristic vertical white barred color pattern on an orange background made by a specific distribution of three types of pigment cells: melanophores, xanthophores, and iridophores. This color pattern is an interesting alternative model to zebrafish to understand the cellular and molecular basis of complex color pattern formation. Using transmission electron microscopic observations, we have investigated the pigment cell composition in the skin of the anemonefish Amphiprion ocellaris and found that: 1) white skin comprises iridophores and isolated melanophores; 2) orange …

Light-induced body color change in developing zebrafish.

In response to ambient light levels, many lower vertebrates darken or lighten their body colors by regulating dispersion or aggregation, respectively, of melanin granules (melanosomes) in the melanophore. This physiological reaction is mediated by photoreception in the eyes, the pineal gland, the deep brain and the melanophores themselves, depending on species and their developmental stages. In this study, we established a method for quantitative measurement of the light-induced body color change in zebrafish larvae. From 2 days post-fertilization (dpf), the dermal melanophores responded to li…

Chromatophore distribution and inferior performance of albino Japanese flounder Paralichthys olivaceus with special reference to different chromatophore expression between albinism and pseudo-albinism.

Albinism with a large variation in body color was found in a hatchery population of Japanese flounder. In addition to albinism, ambicoloration and pseudo-albinism were simultaneously observed in some individuals. Albinos had a remarkably lower number of melanophores on the scales of ocular side than wild-type individuals did, although no significant difference was observed in the numbers of xanthophores and iridophores. The intensity of body color significantly correlated with the number of melanophores among the albinos. No significant differences were observed in the intensity of body color …
