Cellular membrane potentials and contractile threshold in mammalian skeletal muscle susceptible to malignant hyperthermia.

An increase in external K+ can generate contractile force in muscle, and K+ contractures are an established means to determine the effect of an agent on the relation between membrane potential and mechanical activity [8]. We have used K+-contractures to further test our hypothesis [5] that abnormal cell membrane potential responses are intrinsic to skeletal muscle of Poland China pigs susceptible to malignant hyperthermia (MHS), and a nervous system is not required to initiate malignant hyperthermia (MH). Recently it has been shown that K+-induced contractures in certain skeletal and cardiac m

Effect of calcium antagonism or chelation on rabbit lacrimal gland secretion and membrane potentials.

1. ACh-induced secretion from the main excretory duct, as well as ACh-induced hyperpolarizations and resting membrane potentials of superficial, probably acinar, cells were recorded from the rabbit lacrimal gland perfused in vivo with either control or test solutions.2. During perfusion with test solution containing a Ca antagonist, verapamil (10(-5) to 10(-3)M), or a Ca chelator EGTA (10(-4) or 10(-3)M), ACh-induced secretion was only 30% of the control value, whereas the magnitude of the ACh-induced hyperpolarization was unchanged.3. With 10(-5)M-verapamil, but not with 10(-3)M-verapamil or

Membrane Microvesicles as Potential Vaccine Candidates.

The prevention and control of infectious diseases is crucial to the maintenance and protection of social and public healthcare. The global impact of SARS-CoV-2 has demonstrated how outbreaks of emerging and re-emerging infections can lead to pandemics of significant public health and socio-economic burden. Vaccination is one of the most effective approaches to protect against infectious diseases, and to date, multiple vaccines have been successfully used to protect against and eradicate both viral and bacterial pathogens. The main criterion of vaccine efficacy is the induction of specific humo

Membrane damaging potential of photosensitized riboflavin.

Riboflavin upon exposure to UV and visible radiations has been shown to produce active oxygen species. The present work deals with erythrocyte membrane as model system to study the damaging potential of photosensitized riboflavin. Membrane preparations (2.5 mg protein/ml) following exposure to sunlight in presence of riboflavin for different time intervals revealed significant inhibition of ATPases, p-nitrophenyl phosphatase and acetylcholinesterase. Considerable increase in lipid peroxidation was caused by the photosensitized riboflavin. Quenching studies using specific scavengers indicated r

Membrane surface potential of Spiroplasma floricola.

Anionic charges, cytochemically identified as lipid phosphate groups, cover the outer membrane surface of Spiroplasma floricola. They induce a negative membrane surface potential which affects the distribution of ions, including protons. Accordingly, the pH at the interface differs from the bulk pH. By using the fluorescent lipoid pH indicator 4-heptadecyl-7-hydroxycoumarin, the pH at the membrane surface was determined. From the difference of the bulk and the interfacial pH the membrane surface potential of S. floricola was calculated to be phi = -118 mV.

Muscarinic receptor activity has multiple effects on the resting membrane potentials of CA1 hippocampal interneurons.

Inhibitory interneurons appear to be an important target for the muscarinic actions of cholinergic inputs to the hippocampus. We investigated the effect of muscarinic receptor activity on the membrane potential (V(m)) and currents of rat hippocampal CA1 interneurons using whole-cell recording from visually identified CA1 interneurons. The predominant response observed was a muscarinic depolarization that was detected in interneurons from all layers of CA1. This depolarization was mediated by at least two mechanisms: a reduction in a potassium current and a mechanism that depended on extracellu

Exploring the Potentials of Membrane Gas Separation for CO Concentration After Plasma Catalytic CO2 Splitting.

Today, reducing carbon footprints requires the development of technologies to utilize CO2, particularly by converting it into valuable chemical products. One approach is plasma-catalytic CO2 splitting into CO and O2. The task of separating such a ternary mixture is nontrivial and requires the development of an efficient method. In this paper, we have developed a comprehensive scheme for the separation of a CO2/CO/O2 mixture using membrane technology. The novelty of this work lies in the development of a complete scheme for separating the products of plasma-chemical decomposition of CO2 to prod

Monitoring of compound resting membrane potentials of cell cultures with ratiometric genetically encoded voltage indicators.

The cellular resting membrane potential (Vm) not only determines electrical responsiveness of excitable cells but also plays pivotal roles in non-excitable cells, mediating membrane transport, cell-cycle progression, and tumorigenesis. Studying these processes requires estimation of Vm, ideally over long periods of time. Here, we introduce two ratiometric genetically encoded Vm indicators, rArc and rASAP, and imaging and analysis procedures for measuring differences in average resting Vm between cell groups. We investigated the influence of ectopic expression of K+ channels and their disease-c

Patterns of membrane potentials and distributions of the medullary respiratory neurons in the decerebrate rat.

We analyzed the membrane potential of 161 respiratory neurons in the medulla of decerebrate rats which were paralyzed and ventilated. Three types of inspiratory (I) neurons were observed: those displaying progressive depolarization in inspiration (augmenting I neurons), those which gradually repolarized after maximal depolarization at the onset of inspiration (decrementing I neurons) and those exhibiting a plateau or bell-shaped membrane potential trajectory throughout inspiration (I-all neurons). Three types of expiratory (E) neurons were also encountered: those in which the membrane potentia

Mechanism of the effect of cyanide on cell membrane potentials in Necturus gall-bladder epithelium.

1. Addition of sodium cyanide to the mucosal or the serosal medium bathing the isolated gall-bladder of Necturus maculosus causes hyperpolarization of both apical and basolateral membrane of the epithelial cells. The effect of cyanide is practically immediate, reversible (if exposure is brief), and long-lasting (greater than 30 min). 2. The hyperpolarization is accompanied by: (a) reduction of the equivalent resistance of the cell membranes, as shown by cable analysis and input resistance measurements, and (b) increase of the potassium selectivity of both cell membranes, as evidenced by the ef

Effect of neural stimulation on acinar cell membrane potentials in isolated pancreas and salivary gland segments.

The effect of cholinergic neural excitation by field stimulation on the acinar cell membrane potential was investigated in superfused segments of mouse pancreas and salivary glands (sublingual, submaxillary, and parotid glands). Responses of acinar cells in both exocrine pancreas and salivary glands to the neural excitation obtained by field stimulation were similar to responses previously described in each gland to the externally applied acetylcholine. In the pancreatic acinar cell, electrical field simulation induced depolarization with a latency of 0.3 to 1.2 sec. This depolarization was ac

Comparative action of methylmercury and divalent inorganic mercury on nerve terminal and intraterminal mitochondrial membrane potentials.

Both methylmercury (MeHg) and inorganic divalent mercury (Hg++) alter the flux of ions and small molecules across nerve terminal membranes by mechanisms that may involve membrane depolarization. We compared the effects of MeHg and Hg++ on plasma (psi p) and mitochondrial membrane potentials (psi m) in synaptosomes using the potentiometric carbocyanine dye 3,3'-diethylthiadicarbocyanine iodide [diS-C2(5)]. Both mercurials (1-20 microM) produced concentration-dependent increases in dye fluorescence after 5 min of exposure which were not altered by removal of Ca++ from the medium. To determine di

Development of resting membrane potentials of embryonic murine spinal cord cells evaluated by flow cytometric analysis.

The membrane electrical properties of embryonic murine spinal cord cells of embryonic day-10 (E10) to E14 were studied using a voltage-sensitive oxonol dye combined with fluorescence-activated cell sorting techniques. This period of development corresponds to the time when neurons differentiate from their neuroblast precursors. Bovine oligodendrocytes were also investigated in these studies as they have an exclusive K+ dependence on resting membrane potential (RMP). Fluorescence emission histograms of spinal cord cells on E11-E14 exhibited little difference, suggesting that the RMP of cells of

Hydrogen peroxide induced changes in membrane potentials in guinea pig ventricular muscle: permissive role of iron.

STUDY OBJECTIVE: It has been proposed that oxygen free radicals trigger reperfusion arrhythmias. The mechanism of these arrhythmias is not clear. Thus, the effect of H2O2 on cellular action potentials was examined. DESIGN: Trabecular muscles were superfused either with H2O2 alone or with H2O2 in combination either with H2O2 alone or with H2O2 in combination with iron ions or an iron chelating agent or various scavengers of oxygen free radicals. The effect of reduction of the superfusate calcium from 1.8 to 0.2 mmol.litre-1 on H2O2 induced changes was also studied. EXPERIMENTAL MATERIAL: Thin

Atherosclerosis: From the Disruption of Mitochondrial Membrane Potential to the Potential Interventional Strategies.

Atherosclerosis (AS) is the major factor of cardiovascular disease (CVD) and is characterized by a progressive and chronic inflammatory process in the arterial wall. Recent studies have shown that disruption of the mitochondrial membrane potential (deltapsi (m)) directly affects the electron transport chain (ETC), which in turn leads to oxidative stress, and furthermore, its alteration leads to apoptosis and activation of the NLRP3 inflammasome, thereby promoting the development of AS. Here, this review describes how deltapsi (m) contributes to the development of AS by mediating oxidative stre

The ionic mechanism of membrane potential oscillations and membrane resonance in striatal LTS interneurons.

Striatal low-threshold spiking (LTS) interneurons spontaneously transition to a depolarized, oscillating state similar to that seen after sodium channels are blocked. In the depolarized state, whether spontaneous or induced by sodium channel blockade, the neurons express a 3- to 7-Hz oscillation and membrane impedance resonance in the same frequency range. The membrane potential oscillation and membrane resonance are expressed in the same voltage range (greater than -40 mV). We identified and recorded from LTS interneurons in striatal slices from a mouse that expressed green fluorescent protei
