Consolidation of long-term memory: evidence and alternatives.

Memory loss in retrograde amnesia has long been held to be larger for recent periods than for remote periods, a pattern usually referred to as the Ribot gradient. One explanation for this gradient is consolidation of long-term memories. Several computational models of such a process have shown how consolidation can explain characteristics of amnesia, but they have not elucidated how consolidation must be envisaged. Here findings are reviewed that shed light on how consolidation may be implemented in the brain. Moreover, consolidation is contrasted with alternative theories of the Ribot gradien

Memory consolidation, retrograde amnesia and the hippocampal complex.

Results from recent studies of retrograde amnesia following damage to the hippocampal complex of human and non-human subjects have shown that retrograde amnesia is extensive and can encompass much of a subject's lifetime; the degree of loss may depend upon the type of memory assessed. These and other findings suggest that the hippocampal formation and related structures are involved in certain forms of memory (e.g. autobiographical episodic and spatial memory) for as long as they exist and contribute to the transformation and stabilization of other forms of memory stored elsewhere in the brain

Retrograde amnesia and memory consolidation: a neurobiological perspective.

The fact that information acquired before the onset of amnesia can be lost (retrograde amnesia) has fascinated psychologists, biologists, and clinicians for over 100 years. Studies of retrograde amnesia have led to the concept of memory consolidation, whereby medial temporal lobe structures direct the gradual establishment of memory representations in neocortex. Recent theoretical accounts have inspired a simple neural network model that produces behavior consistent with experimental data and makes these ideas about memory consolidation more concrete. Recent physiological and anatomical findin

Selective attention in visual short-term memory consolidation.

Selection of visual short-term memory content takes place during perceptual and postperceptual processing. We studied postperceptual selection in a change detection task, in which spatially distributed cues preceding the memory array marked potential change targets. In the 550-730 ms interval after the memory array onset but not in the following retention interval, event-related potentials showed larger contralateral delay activity when the spatial cues were selective than when they were unselective. This finding implies that distributed precueing strengthens the visual short-term memory repre

Sleep consolidation of social memories and common ground.

Sleep consolidation effects are well established in the cognitive domains of memory and language learning, however very little is known regarding sleep effects in social contexts. Across three experiments, we investigated whether sleep enhances the accuracy of socially formed memories and communicative efficiency, including the use of partner-specific common ground between interlocutors in conversation. Participants completed a computerised (Experiment 1) or face-to-face (Experiment 2) matcher-director task with two other people before and after 12 h of sleep/wake. Both experiments showed more

Memory integration: An alternative to the consolidation/reconsolidation hypothesis.

The original concept of consolidation considers that memory requires time to be fixed. Since 2000, a comparable protein-dependent re-stabilization phase, called reconsolidation, has been assumed to take place after memory retrieval. This consolidation/reconsolidation hypothesis, has dominated the literature for more than 50 years, despite compelling evidence that is inconsistent with it. In this review, we present an historical overview and explain how, despite serious criticisms, this hypothesis has persisted for decades and become accepted as a dogma. Based on both older and more recent evid

Impairment of fear memory consolidation and expression by antihistamines.

Antihistamines are widely used to treat allergy symptoms. First-generation antihistamines have adverse effects on the central nervous system (CNS), such as hypnotic and amnesic effects, whereas second-generation antihistamines have poor brain penetration, and therefore, have fewer CNS-related adverse effects. Memory consists of several phases, including acquisition, consolidation, expression, and extinction. It remains unclear whether these phases are affected by antihistamines. We investigated the effects of diphenhydramine, a first-generation antihistamine, and levocetirizine and olopatadine

Acute exposure to blue wavelength light during memory consolidation improves verbal memory performance.

Acute exposure to light within the blue wavelengths has been shown to enhance alertness and vigilance, and lead to improved speed on reaction time tasks, possibly due to activation of the noradrenergic system. It remains unclear, however, whether the effects of blue light extend beyond simple alertness processes to also enhance other aspects of cognition, such as memory performance. The aim of this study was to investigate the effects of a thirty minute pulse of blue light versus placebo (amber light) exposure in healthy normally rested individuals in the morning during verbal memory consolida

The role of the ventromedial prefrontal cortex in memory consolidation.

"System-level memory consolidation theory" posits that the hippocampus an initially links the neocortical representations, followed by a shift to a hippocampus-independent neocortical network. With consolidation, an increase in activity in the human subgenual ventromedial prefrontal cortex (vmPFC) has repeatedly been shown. Previously we and others have proposed that this area might link the neocortical representational areas in remote memory, similarly as has been proposed for the rodent anterior cingulate cortex (ACC). Here, we review literature involving the human vmPFC to investigate if th

Procaine impairs learning and memory consolidation in the honeybee.

In vertebrates local anaesthetics lead to various learning deficits, depending on the site and time of injection. In this study we show the effects of the local anaesthetic procaine on learning and memory in an invertebrate, the honeybee. Reversible blocking of neuronal processes leads to different memory deficits depending on the time window during which the local anaesthesia is in effect.

Radish, a Drosophila mutant deficient in consolidated memory.

We have characterized the behavior and genetics of the Drosophila mutant radish (rsh gene). Initial learning of radish flies in two olfactory discrimination tests is high, but subsequent memory decays rapidly at both early and late times after training. Anesthesia-resistant memory (consolidated memory) is undetectable in radish flies 3 hr after training. The mutant shows normal locomotor activity and normal sensitivity to the odor cues and electric-shock reinforcement used in the learning tests. The radish gene maps within a 180-kb interval in the 11D-E region of the X chromosome.

Rehearsal initiates systems memory consolidation, sleep makes it last.

After encoding, memories undergo a transitional process termed systems memory consolidation. It allows fast acquisition of new information by the hippocampus, as well as stable storage in neocortical long-term networks, where memory is protected from interference. Whereas this process is generally thought to occur slowly over time and sleep, we recently found a rapid memory systems transition from hippocampus to posterior parietal cortex (PPC) that occurs over repeated rehearsal within one study session. Here, we use fMRI to demonstrate that this transition is stabilized over sleep, whereas wa

Visual Selection Is Spatially Constrained During Working Memory Consolidation.

Working memory consolidation (WMC) is known to consume central attentional resources, impairing performance on subsequent attention-demanding tasks. However, the specific nature of this impairment, particularly its effect on the initial stage of visual selection, remains unclear. The current study tested three competing hypotheses: No-suppression, Pure-temporal-suppression, and Spatial-suppression hypotheses. We used a sequential target-matching task where the inter-stimulus interval (ISI) between a target item and a subsequent probe item was varied to modulate the state of consolidation. Expe

Latent persistence in fMRI dynamics during human memory consolidation.

Human memory consolidation involves the gradual stabilization and reorganization of memory traces over time. Despite numerous empirical and computational accounts emphasizing different aspects of this process, an integrated framework for evaluating consolidation theories against brain data remains limited. We propose a biologically informed, data-driven framework based on Koopman operator analysis to examine latent dynamical structure in fMRI signals associated with memory consolidation. The Koopman framework lifts nonlinear brain dynamics into a linear function space, enabling spectral charac

Effects of theta tACS on tactile texture memory consolidation.

Tactile texture memory has been underexplored compared to visual and auditory memory, despite its importance in daily functioning. Prior research indicates that theta oscillations support memory consolidation across modalities, and frontal midline theta transcranial alternating current stimulation (tACS) has been shown to enhance long-term visual and auditory memory. This study examined whether theta-frequency tACS similarly influences tactile texture memory. 59 participants were randomly assigned to theta tACS (3 Hz), beta tACS (16 Hz), or sham stimulation administered immediately after encod

Sleep-dependent memory consolidation in patients with sleep disorders.

Sleep can improve the off-line memory consolidation of new items of declarative and non-declarative information in healthy subjects, whereas acute sleep loss, as well as sleep restriction and fragmentation, impair consolidation. This suggests that, by modifying the amount and/or architecture of sleep, chronic sleep disorders may also lead to a lower gain in off-line consolidation, which in turn may be responsible for the varying levels of impaired performance at memory tasks usually observed in sleep-disordered patients. The experimental studies conducted to date have shown specific impairment

Sleep-dependency of episodic-like memory consolidation in rats.

Episodic memory refers to the recollection of a representation that binds together into a unique past experience "what" happened, "where" and "when". Sleep has been identified as a state that optimizes the consolidation of newly acquired memory. To determine if sleep is important for the consolidation of episodic-like memory, we tested rats on an episodic-like memory task requiring the binding of an object memory into a spatio-temporal context, as well as retention of its individual components, using separate tests of novel-object recognition ("what"), object-place recognition ("where") and te

Hippocampal dysfunction during aging I: deficits in memory consolidation.

Numerous ablation studies indicate a critical role for the hippocampal system in establishing or consolidating certain types of memory. Normal aging manifests by selective neurobiological changes in the hippocampal formation and on performance of tasks that require a functional hippocampus, including retention of contextual fear conditioning. To determine if impairments in the consolidation process contribute to memory dysfunction in aging, middle-aged and aged rats were fear conditioned and subsequently received dorsal hippocampal lesions or sham surgery after a 1, 7, 14, or 28-day interval.

Kinetics of memory consolidation: role of amnesic treatment parameters.

The consolidation theory states that with the passage of time the engram of a recent learning experience grows increasingly resistant to disruption by amnesic treatment. The time required to reach complete resistance ("consolidation time") is a controversial issue; current estimates range from 10(1) to 10(5) seconds. The present study suggests a parsimonious interpretation of the divergence, namely, that weak amnesic treatments fail to block memory consolidation but do slow its rate, so that post-treatment consolidation inflates the retention scores measured 24 hours later and leads to variabl
