A practical learning and memory screening test in rats using a modified Biel water maze - A multi-site validation.

Developmental and adult neurotoxicity testing is an integral part of safety assessment guidelines. A key endpoint, typically performed in rodents, is the evaluation of learning and memory (L&M). Historically, a variety of equipment and testing paradigms have been developed and used across the industry. In recent years, regulatory guidance documents have become more prescriptive and stipulate specific types of tests to be used at different ages. The most common L&M tests for developmental neurotoxicity are performed using water mazes. Navigational L&M can be split into two principal types where

Assessment of verbal memory in Parkinson's disease utilizing a virtual reality-based Rey Auditory Verbal Learning Test.

The Rey Auditory Verbal Learning Test (RAVLT) is a commonly used tool for evaluating verbal learning and memory in neuropsychological assessments. In recent years, we developed a Virtual Reality (VR) adaptation of the RAVLT (VR-RVLT), aiming for increased ecological validity compared to the traditional pen and paper gold standard (GS-RAVLT). Following validation in healthy cohorts, the VR-RAVLT was validated with thirty individuals with Parkinson's Disease (PD) that completed both the GS-RAVLT and the VR- RAVLT. Validity of the VR-RAVLT was evaluated by assessing its construct and discriminant

Detecting simulated memory impairment with the Rey Auditory Verbal Learning Test: implications of base rates and study generalizability.

The accuracy of the Rey Auditory Verbal Learning Test (RAVLT) for detecting simulated neuropsychological impairment was investigated using discriminant function analyses and cross-validation procedures. In Study 1, a discriminant function analysis, using six RAVLT variables, correctly classified 80% of 50 simulating and 50 non-simulating participants. Base rates were changed and this discriminant function was tested in Study 2 with a 33% base rate of simulation. The discriminant function correctly classified 75% of 20 simulators and 90% of 40 non-simulators, yielding an overall classification

Comparative study on detectability of learning and memory disorder between two water maze tests commonly used in juvenile rat toxicity studies using isoflurane inhaled rat model.

Evaluation of learning and memory is crucial in juvenile animal toxicity studies (JAS) during the development of CNS active drugs, but there are no currently recommended test methods. We compared the ability of the Morris water maze (MWM) and the Biel water maze (BWM) to detect learning and memory disorder (LMD) using rats inhaled isoflurane (IFN). Rats were treated with 1% IFN using inhalation on postnatal day (PND) 7 for 6 h. All rats were subjected to the MWM on PND 33 and the BWM on PND 55/57 (Experiment 1), or the BWM on PND 32/33 and the MWM on PND 54/55 (Experiment 2). On PND 70, the br

Methods for assessment of Rey Auditory Verbal Learning Test performance in memory clinic patients and healthy adults - at the cross-roads of learning theory and clinical utility.

Background: Knowledge is still lacking regarding the preferred method for evaluation of learning in the Rey Auditory Verbal Learning Test (RAVLT). Validity of different methods was examined by the effect size in differentiating diagnostic stages in memory clinic patients versus healthy adults and the strength of association between RAVLT performance and brain atrophy. Method: The study included individuals with dementia (n = 247), Mild Cognitive Impairment (MCI, n = 709), Subjective Cognitive Impairment (SCI, n = 175) and cognitively unimpaired adults serving as healthy controls (HC, n = 102).

The Reliability and Clinical Validation of Automatically-Derived Verbal Memory Features of the Verbal Learning Test in Early Diagnostics of Cognitive Impairment.

BACKGROUND: Previous research has shown that verbal memory accurately measures cognitive decline in the early phases of neurocognitive impairment. Automatic speech recognition from the verbal learning task (VLT) can potentially be used to differentiate between people with and without cognitive impairment. OBJECTIVE: Investigate whether automatic speech recognition (ASR) of the VLT is reliable and able to differentiate between subjective cognitive decline (SCD) and mild cognitive impairment (MCI). METHODS: The VLT was recorded and processed via a mobile application. Following, verbal memory f

New Yes/No Recognition Memory Analysis on the California Verbal Learning Test-3: Clinical Utility in Alzheimer's and Huntington's Disease.

OBJECTIVES: The third edition of the California Verbal Learning Test (CVLT-3) includes a new index termed List A versus Novel/Unrelated recognition discriminability (RD) on the Yes/No Recognition trial. Whereas the Total RD index incorporates false positive (FP) errors associated with all distractors (including List B and semantically related items), the new List A versus Novel/Unrelated RD index incorporates only FP errors associated with novel, semantically unrelated distractors. Thus, in minimizing levels of source and semantic interference, the List A versus Novel/Unrelated RD index may yi

Odor preference learning and memory modify GluA1 phosphorylation and GluA1 distribution in the neonate rat olfactory bulb: testing the AMPA receptor hypothesis in an appetitive learning model.

An increase in synaptic AMPA receptors is hypothesized to mediate learning and memory. AMPA receptor increases have been reported in aversive learning models, although it is not clear if they are seen with memory maintenance. Here we examine AMPA receptor changes in a cAMP/PKA/CREB-dependent appetitive learning model: odor preference learning in the neonate rat. Rat pups were given a single pairing of peppermint and 2 mg/kg isoproterenol, which produces a 24-h, but not a 48-h, peppermint preference in the 7-d-old rat pup. GluA1 PKA-dependent phosphorylation peaked 10 min after the 10-min train

Wada test results contribute to the prediction of change in verbal learning and verbal memory function after temporal lobe epilepsy surgery.

In recent years, the clinical usefulness of the Wada test (WT) has been debated among researchers in the field. Therefore, we aimed to assess its contribution to the prediction of change in verbal learning and verbal memory function after epilepsy surgery. Data from 56 patients with temporal lobe epilepsy who underwent WT and subsequent surgery were analyzed retrospectively. Additionally, a standard neuropsychological assessment evaluating attentional, learning and memory, visuospatial, language, and executive function was performed both before and 12 months after surgery. Hierarchical linear

Investigation of the component processes involved in verbal declarative memory function in bipolar disorder: utility of the Hopkins Verbal Learning Test-Revised.

Evidence suggests that standard learning and recall indexes are sensitive markers of verbal declarative memory ability in bipolar disorder (BD), but no study has examined performance across the full range of component process measures on the Hopkins Verbal Learning Test (HVLT-R) in a BD cohort. As the HVLT-R is part of a widely used battery of cognitive functioning backed by the U.S. Federal Drug Administration as the accepted battery for use in pro-cognitive trials assessing cognitive-enhancing drugs in the related disorder schizophrenia, estimating the utility of its measures in BD is import

An interpretable deep-learning approach to detect biomarkers in anxious-depressed symptoms from prefrontal fNIRS signals during an autobiographical memory test.

BACKGROUND: Individuals with anxious-depressed (AD) symptoms have more severe mood disorders and cognitive impairment than those with non-anxious depression (NAD) symptoms. Therefore, it is important to clarify the underlying neurophysiology of these two symptom groups to optimize treatment. METHODS: We developed an interpretable deep-learning framework based on two convolutional neural networks (CNN) to diagnose depression from functional near-infrared spectroscopy (fNIRS) neuroimaging data recorded during an autobiographical memory test (AMT) from 824 participants. This system was designed

Normative Data for Learning and Memory Test (TAMV-I) in Latin American and Spanish Children: An item response theory and linear mixed models approach.

Robust normative data for pediatric learning and memory tests in Spanish-speaking populations are scarce, and existing approaches often rely on univariate methods that overlook item-level properties and inter-trial dependencies. The aim was to evaluate the item parameters of the TAMV-I using Item Response Theory (IRT) and to generate covariate-adjusted normative data through Linear Mixed Models (LMM). We hypothesized that the 2-parameter logistic (2PL) model would outperform the Rasch model and that demographic and contextual factors would show significant interactions influencing test perform

The effects of selective antagonists of serotonin 5-HT7 and 5-HT1A receptors on MK-801-induced impairment of learning and memory in the passive avoidance and Morris water maze tests in rats: mechanistic implications for the beneficial effects of the novel atypical antipsychotic lurasidone.

We have previously reported that lurasidone, a novel atypical antipsychotic with potent serotonin 5-HT(7) antagonist and 5-HT(1A) partial agonist activities, is superior to other antipsychotics in improving the N-methyl-D-aspartate (NMDA) receptor antagonist MK-801-induced learning and memory impairment in the passive avoidance (PA) and Morris water maze (MWM) tests in rats. In this study, we investigated the effects of selective antagonists of 5-HT(7) and 5-HT(1A) receptors (SB-656104-A and WAY-100635, respectively) on MK-801-induced learning and memory impairment in the same tests. Refinement of the Barnes and Morris water maze protocols improves characterization of spatial cognitive deficits in the lithium-pilocarpine rat model of epilepsy.

Temporal lobe epilepsy (TLE) often causes cognitive impairment, especially a decline in spatial memory. Reductions in spatial memory and learning are also common in rodent models of TLE. The Morris water maze and the Barnes maze are the standard methods for evaluating spatial learning and memory in rodents. However, animals with TLE may exhibit agitation, distress, and fail to follow the paradigmatic context of these tests, making the interpretation of experimental data difficult. This study optimized the procedure of the Morris water maze and the Barnes maze to evaluate spatial learning and m

The mechanisms of learning and memory impairment caused by nonylphenol: a narrative review based on in vivo and in vitro studies.

Learning and memory play a fundamental role on brain cognitive functions which are crucial for human life. Nonylphenol (NP), a serious environmental pollutant over the world, is proven to be harmful for learning and memory mainly via diet exposure. Currently, besides the administrative restrictions for the use of NP, there are rarely other effective approaches against learning and memory impairment caused by NP. This review summarized the mechanisms underlying NP-induced learning and memory impairment according to in vivo and in vitro experiments. Based on the studies involved in behavior test

A review of behavioral methods for the evaluation of cognitive performance in animal models: Current techniques and links to human cognition.

INTRODUCTION: Memory is defined as the ability to store, maintain and retrieve information. Learning is the acquisition of information that changes behavior and memory. Stress, dementia, head trauma, amnesia, Alzheimer's, Huntington, Parkinson's, Wernicke-Korsakoff syndrome (WKS) may be mentioned among the diseases in which memory and learning are affected. The task of understanding deficits in memory and learning in humans is daunting due to the complexity of neural and cognitive mechanisms in the nervous system. This job is made more difficult for clinicians and researchers by the fact that
