Thermosensitive hydrogel of hydrophobically-modified methylcellulose for intravaginal drug delivery.

Hydrogels with the advantages of prolonging drug release and administration convenience are necessary for intravaginal drug delivery to prevent sexual transmission of human immunodeficiency virus and other vaginal infections. In this study, the thermosensitive hydrogel of methylcellulose modified by stearic acid (MCS) were evaluated in the presence of NaCl and phosphates, which exhibited sol-to-gel transition performance at body temperature or even lower. The in vitro cytotoxicity and in vivo mucosal irritation were investigated and the results showed that MCS hydrogel possessed good biocompat

Involvement of the spleen in the anti-prion activity of hydroxypropyl methylcellulose in mice.

Prion diseases are a unique group of fatal infectious neurodegenerative disorders. Previously, we reported that hydroxypropyl methylcellulose (HPMC) exerts long-term anti-prion activity in prion-infected rodents after a single peripheral administration, but the mechanism of action is not well understood. Interestingly, the distribution of HPMC in mice partially resembles that of peripheral pathogenic prions, since peripherally administered HPMC remains in the spleen and lymphoid tissues for a long period. Here, we characterized the anti-prion activity of HPMC in relation to the spleen in mice.

The Impact of Plasticizer Polarity on the Permeability of Hydroxypropyl Methylcellulose Phthalate Films.

The aim of this study was to gain detailed insights into how different plasticizers influence the permeability of isolated hydroxypropyl methyl cellulose phthalate (HPMC-P) films. Since the migration of plasticizers from film coatings may critically affect their functionality, it is essential to monitor the impact of their leaching on the associated film properties. In this work, the leaching behavior of plasticizers from isolated HPMC-P films was estimated using their thermal properties by modulated temperature differential scanning calorimetry (MT-DSC) at varying plasticizer concentrations.

Evaluation of a viscoelastic solution of hydroxypropyl methylcellulose as a potential vitreous substitute.

Hydroxypropyl methylcellulose (HPMC), an inert polymer that forms a viscoelastic solution in aqueous media, is useful for anterior segment surgery. A 2.2% solution of HPMC (MW 86,000 daltons) of 6,000 centistokes viscosity was evaluated as a vitreous substitute in rabbit eyes. Four days after perfluoropropane gas compression of the vitreous, the gas was exchanged with 0.5 ml HPMC. The aqueous was removed 1 and 2 weeks postinjection and at sacrifice, the vitreous was removed at sacrifice. HPMC concentrations in aqueous humor were: 0 hr, 0%; 48 hr, 0.002%; 1 week, 0.004%; 2 weeks, 0.001%; 4 week

Colony growth of normal and neoplastic cells in various concentrations of methylcellulose.

To assess the semisolid character of methylcellulose (MC) and its ability to prevent cell migration and aggregation in clonogenic assays, we studied the influence of various concentrations of MC (0.7%-1.26%) on colony growth of neoplastic cell lines, normal bone marrow cells, and hairy cell leukemia (HCL). All cell lines (K562, HL-60, JOK-1, Daudi, and BB3, an IgM-kappa B-cell line) showed a prominent decrease in colony numbers and remarkable changes in colony morphology at rising MC concentrations, whereas no such influence could be demonstrated for HCL, mixed lineage colony-forming units (CF

Methylcellulose instead of Healon in extracapsular surgery with intraocular lens implantation.

Methylcellulose 1% was used in the anterior chamber to facilitate posterior chamber lens implantation at the time of extracapsular cataract extraction. In parallel series with similar preoperative endothelial cell counts, 70 eyes implanted under methylcellulose had a mean central endothelial cell loss of 8 +/- 5.3% at 8 to 12 weeks postoperative, whereas 63 eyes implanted under an air bubble lost a mean 25.3 +/- 14.6% (P less than 0.001). No differences in postoperative visual acuity, intraocular pressure, or inflammation were noted. A YAG laser preoperative anterior capsulotomy was used in al

Physicochemical Characterization of Hyaluronic Acid-Methylcellulose Semi-Gels for Mitochondria Transplantation.

Traumatic spinal cord injury (SCI) presents a significant medical challenge due to its intricate nature and treatment complexities. SCI can cause physical impairments by affecting neural and motor functions as well as initiating a series of pathophysiological events exacerbating the initial trauma. Leakage from ruptured neurons and vessels disrupt ionic balance and induces excitotoxicity, leading to progressive cellular degeneration. Introducing mitochondria to the SCI lesion has shown potential in attenuating secondary injury. Mitochondria transplantation improves cellular bioenergetics and r

Injectable in situ gelling methylcellulose-based hydrogels for bone tissue regeneration.

Injectable bone substitutes (IBSs) represent a compelling choice for bone tissue regeneration, as they can be exploited to optimally fill complex bone defects in a minimally invasive manner. In this context, in situ gelling methylcellulose (MC) hydrogels may be engineered to be free-flowing injectable solutions at room temperature and gels upon exposure to body temperature. Moreover, incorporating a suitable inorganic phase can further enhance the mechanical properties of MC hydrogels and promote mineralization, thus assisting early cell adhesion to the hydrogel and effectively guiding bone ti

Methylcellulose enhances resolution in gravitational field-flow fractionation: Going beyond viscosity.

Gravitational Field-Flow Fractionation (GrFFF) is an elution-based method designed for the separation of particles ranging from a few micrometers up to approximately 100 μm in diameter. Separation occurs over time, with particles being fractionated based on size and other physico-chemical properties. GrFFF takes advantage of gravitational forces acting perpendicularly to a laminar flow in a thin channel. The fluid exhibits a parabolic velocity profile, with the maximum velocity at the center of the channel and zero velocity at the walls. The exit time of particles depends on their equilibrium

Air, methylcellulose, sodium hyaluronate and the corneal endothelium. Endothelial protective agents.

In a randomised trial the endothelial protective agent used during extracapsular cataract extraction and intraocular lens insertion was air in 19 eyes (group 1), methylcellulose in 25 eyes (group 2) and sodium hyaluronate in 22 (group 3). The cell population densities of each eye were estimated immediately before and three months after the operations to determine the degree of cell loss. Eyes showing mechanical (touch) damage on the second postoperative day were eliminated. The numbers of eyes in each group which showed a statistically significant cell loss were compared, and the mean cell los

Thermo-Responsive Methylcellulose Hydrogels: From Design to Applications as Smart Biomaterials.

Methylcellulose (MC) is an attractive material used to produce thermo-responsive hydrogels. They undergo sol-gel transition when a critical temperature is reached, thus modifying their properties (e.g., physicochemical and mechanical) in response to temperature changes. This behavior is particularly attractive when the body temperature acts as a trigger to modulate the thermo-responsive behavior of MC hydrogels. In this regard, exciting advances have been achieved in the field of cell and drug delivery, tissue engineering, and regenerative medicine, making MC a very attractive and versatile bi

Hydroxypropyl methylcellulose: A better submucosal fluid cushion for endoscopic mucosal resection.

BACKGROUND: Formation of a submucosal fluid cushion has become integral to endoscopic mucosal and polyp resection. Solutions available for injection into the submucosa of the GI tract create short-lasting submucosal fluid cushions or are costly. The feasibility, durability, and histologic response to submucosal fluid cushions created by injection of hydroxypropyl methylcellulose were studied in the esophagus of pigs. METHODS: Thirty-six esophageal submucosal fluid cushions created with hydroxypropyl methylcellulose were studied in 12 animals divided in 2 groups of 6 animals each. In Group I,

Method to recover a lipophilic drug from hydroxypropyl methylcellulose matrix tablets.

A reverse-phase high-performance liquid chromatographic (HPLC) method for recovery of the lipophilic drug, alprazolam, from matrix tablets containing the hydrophilic polymer hydroxypropyl methylcellulose (HPMC) was developed. Lipophilic drugs, such as alprazolam, are difficult to completely extract and quantitate from tablets containing HPMC polymer. The percentage of recoveries of alprazolam from placebo powder spiked with alprazolam stock solution and from placebo powder mixed with alprazolam powder were about 100% and 85% to 95%, respectively. The validated method using water to completely

Evaluation of a Methylcellulose and Hyaluronic Acid Hydrogel as a Vehicle for Rectal Delivery of Biologics.

Biologics have changed the management of Inflammatory Bowel Disease (IBD), but there are concerns regarding unexpected systemic toxicity and loss of therapeutic response following administration by injection. Local delivery of biologics directly to the inflamed mucosa via rectal enema administration addresses the problems associated with systemic administration. Hydrogels are potentially useful delivery vehicles enabling rectal administration of biologics. Here, we prepared a hydrogel system based on methylcellulose (MC) and hyaluronic acid (HA), which possesses mucosal healing properties, inc

The use of hydroxypropyl methylcellulose in extracapsular cataract extraction with intraocular lens implantation.

We conducted a prospective randomized clinical trial on the use of 2% hydroxypropyl methylcellulose and 1% sodium hyaluronate in the anterior chamber during anterior capsulotomy and nuclear expression in extracapsular cataract extraction with posterior chamber lens implantation in 70 patients. Both viscoelastic substances helped maintain the anterior chamber and facilitated the operation, although sodium hyaluronate was preferred. There was no excessive intraocular inflammation or toxicity noted with either agent. Patients given sodium hyaluronate had a larger increase in intraocular pressure

Safety and efficacy of 2% methylcellulose in cat and monkey cataract-implant surgery.

We evaluated the safety and efficacy of 2% methycellulose as an adjunct for cataract extraction with implantation in cat and monkey models. When used intraoperatively, methylcellulose reduced the iridovitreal bulge during surgery. No significant increase in clinical inflammation occurred nor was there statistically significant intraocular pressure elevation at 24 hours, 7 days, or 90 days. In the cat model, the central corneal thickness increased at day seven in both control and methylcellulose eyes; this thickness persisted to 90 days. The endothelial cell loss decreased significantly at day
