Urinary excretion of N gamma-methylihistidine (3-methylihistidine): a tool to study metabolic responses in relation to nutrient and hormonal status in health and disease of man.

Some of the histidine residues of actin and myosin are methylated after synthesis of these contractile muscle proteins. During breakdown of muscle protein in the course of protein turnover, the product of methylation, N gamma-methylhistidine, is released and quantitatively excreted in the urine both of rat and of man. Since most of the N gamma-methylhistidine in the body occurs in muscle, the rate of its excretion becomes a convenient measure of muscle protein breakdown. Output per kilogram of body weight is highest in the infant, especially when related to creatinine, and is reduced in the el

Targeted liquid chromatography-tandem mass spectrometry profiling of arginine and histidine derivatives in chronic migraine: Discriminatory performance and changes after onabotulinumtoxinA therapy.

OBJECTIVE: This study was conducted to characterize plasma arginine, histidine, 1-methylhistidine, and 3-methylhistidine in chronic migraine and to evaluate their discriminatory ability and association with early clinical response to onabotulinumtoxinA (BoNT-A). BACKGROUND: Alterations in nitric oxide- and histamine-related metabolic pathways may contribute to chronic migraine biology and may provide circulating biomarkers for disease stratification and treatment monitoring. Targeted liquid chromatography-tandem mass spectrometry profiling enables precise quantification of candidate amino aci

Serum metabolomic signatures of fatty acid oxidation defects differentiate host-response subphenotypes of acute respiratory distress syndrome.

BACKGROUND: Fatty acid oxidation (FAO) defects have been implicated in experimental models of acute lung injury and associated with poor outcomes in critical illness. In this study, we examined acylcarnitine profiles and 3-methylhistidine as markers of FAO defects and skeletal muscle catabolism, respectively, in patients with acute respiratory failure. We determined whether these metabolites were associated with host-response ARDS subphenotypes, inflammatory biomarkers, and clinical outcomes in acute respiratory failure. METHODS: In a nested case-control cohort study, we performed targeted an

Nitrogen balance in extremely low birth weight infants with nonoliguric hyperkalemia.

We measured nitrogen balance and urinary 3-methylhistidine molar ratios in 33 extremely low birth weight infants (12 with hyperkalemia and 21 without) for the first 3 days of life. Although all infants were in negative nitrogen balance during the study, there was no difference in the degree of negative nitrogen balance between the two groups. There was also no difference in the 3-methylhistidine/creatinine molar ratios, indicating that muscle protein catabolism did not differ. We conclude that it is unlikely that catabolism contributes to the development of nonoliguric hyperkalemia in extremel

Application of metabolomic principles to disorders of nucleotide metabolism reveals new metabolic perturbations.

A metabolomic analysis of plasma amino acids and acylcarnitines was applied to four disorders of nucleotide metabolism. Multivariate analysis gave score plots that show segregation of hypoxanthine phosphoribosyltransferase and adenine phosphoribosyltransferase deficient plasma from controls with equivocal results for adenosine deaminase and dihydropyrimidine dehydrogenase deficiencies. Loadings plots revealed the principal metabolites responsible for the discrimination between these classes. There were increases for HPRT in C4-, C6-, and C3-DC (malonyl)-carnitines, and decreased serine. For AP

Growth hormone-induced changes in myofibrillar protein breakdown in hypopituitary children.

1. Creatinine and N tau-methylhistidine excretion rates have been measured in 13 hypopituitary children to calculate the body muscle contents and rates of myofibrillar protein breakdown. Analyses have been made during periods of growth hormone withdrawal and subsequent administration. 2. The creatinine excretion rate was lower in the hypopituitary children, indicating a lower muscle content per kg body weight. This difference persisted even in children who had received growth hormone for several years. 3. Excretion of N tau-methylhistidine was reduced by the administration of growth hormone. 4

Metabolomics approach using HR-MAS NMR spectroscopy for the assessment of metabolic profiles of uterine fibroids.

The aim of this study is to determine dysregulated metabolites and metabolic pathways in uterine fibroids and in the myometrial tissue from which uterine fibroids are derived. Fifteen (15) patients underwent hysterectomy because of uterine fibroids and 14 controls were included in this study. 1H HR-MAS NMR spectroscopy data were obtained from uterine fibroid tissue, the adjacent healthy myometrial tissue from cases, and myometrial tissue from controls. PCA and PLS-DA score plots from multivariate statistical analysis of pre-processed spectral data demonstrated a distinction between cases and c

Skeletal muscle and whole body protein turnover in thyroid disease.

The effects of disturbances of thyroid hormone secretion on leg and whole body amino acid and protein metabolism have been investigated in seven patients with untreated thyrotoxicosis and eight patients with untreated hypothyroidism; the results were compared to those obtained in 11 normal control subjects. After treatment, the patients were restudied. Arterio-venous exchanges of tyrosine and 3-methylhistidine across leg tissue in the post-absorptive state were used as indices of net protein balance and myofibrillar protein breakdown, respectively. Whole body protein turnover was measured usin

Altered body composition and muscle protein degradation in nutritionally growth-retarded children with cystic fibrosis.

To investigate nutritional growth retardation and the adaptive response to malnutrition in cystic fibrosis (CF), body composition and muscle protein catabolism were studied in nine malnourished CF children and eight healthy controls by anthropometry, measurement of whole body potassium, urinary creatinine excretion, creatinine height index, and urinary 3-methylhistidine excretion, an index of myofibrillar protein catabolism. CF children had a significant deficit of body mass (p less than 0.001), derived from both the body fat and the fat-free compartments, including a deficit in muscle mass (p

Histidine Nτ-methylation identified as a new posttranslational modification in histone H2A at His-82 and H3 at His-39.

Histone posttranslational modifications play critical roles in a variety of eukaryotic cellular processes. In particular, methylation at lysine and arginine residues is an epigenetic mark that determines the chromatin state. In addition, histone "histidine" methylation was initially reported over 50 years ago; however, further studies in this area were not conducted, leaving a gap in our understanding. Here, we aimed to investigate the occurrence of histidine methylation in histone proteins using highly sensitive mass spectrometry. We found that acid hydrolysates of whole histone fraction from

Activity of homocarnosine and other compounds against staphylococcal infections in mice.

Homocarnosine and carnosine have been identified in bovine brain extracts which are effective in protecting mice against infections by Staphylococcus aureus. These peptides, as well as l-1-methylhistidine, beta-alanine, gamma-aminobutyric acid, delta-aminovaleric acid, epsilon-aminocaproic acid, 1-aminomethylcyclohexane-4-carboxylic acid, and anserine, were tested as prophylactic agents against S. aureus infections in C3H and Swiss mice. Histidine and methylhistidine were ineffective in preventing mortality in both mouse strains. Carnosine, anserine, and epsilon-aminocaproic acid were effectiv

Pharmacology of alpha-fluoromethylhistidine, a specific inhibitor of histidine decarboxylase.

alpha-Fluoromethyl-[S]-histidine (FMH) is a specific and potent inhibitor of histidine decarboxylase, which forms histamine from histidine. It acts selectively and irreversibly by formation of a covalent linkage, possibly with the serine residue in the active site of the enzyme. A single administration of FMH decreases the histamine content only of non-mast cells in the brain and stomach of rodents, but repeated administration gradually decreases the histamine content of mast cells in all tissues. Thus, FMH can be used to deplete histamine in pharmacological studies. As no marked side-effects

Amino Acid Dysregulation in the Mother-Fetus Unit: Multi-Compartment Metabolomic Signatures of Gestational Diabetes Mellitus and Fetal Macrosomia.

The role of amino acid disturbances in the mother-fetus system remains poorly understood, despite their critical involvement in gestational diabetes mellitus (GDM), fetal macrosomia (FM) and offspring metabolic programming. This study included 62 mother-newborn dyads stratified by GDM and FM status. An analysis of the association of amino acids with clinical parameters was performed using the Spearman test. Amino acid markers of GDM were identified using the mutual information index and the Mann-Whitney test. A random forest method was used to identify amino acid markers, with the SHAP value u

Branched-chain ketoacids reduce muscle protein degradation in Duchenne muscular dystrophy.

In order to determine whether branched-chain ketoacids can reduce the excessive rate of muscle protein degradation that characterizes Duchenne muscular dystrophy, nine boys affected with the disease were studied in a metabolic ward while receiving meat-free diets. After a three-day equilibration period, excretion rates of 3-methylhistidine and creatinine were measured in two consecutive four-day periods. In the second period, a supplement containing a mixture of ornithine alpha-ketoisocaproate, alpha-ketoisovalerate, and alpha-keto-beta-methylvalerate in a proportion of 4:1:1 was administered

Susceptibility to mild and severe acute mountain sickness is associated with distinct urine metabolite profiles.

Rapid ascent to high altitude (HA) in unacclimatized lowlanders elicits a series of hematologic, ventilatory, metabolic, and cardiovascular adaptations to counteract the lower partial pressure of oxygen. When ascent occurs faster than the body can acclimatize, high-altitude illnesses (HAIs) can occur. The most prevalent of the HAIs is acute mountain sickness (AMS), which can range in severity from mild (minor inconvenience) to severe (total incapacitation). Symptoms of AMS include headache, fatigue, gastrointestinal distress, dizziness, and in some cases sleep disturbances. Identifying individ

Magnolin and Lanosterol in WenYangTongbi pill-mediated Foxs1-Nlrp3 axis suppression alleviates chronic sleep deprivation-induced cognitive impairment.

BACKGROUND: Chronic sleep deprivation (CSD) impairs cognitive function, yet current therapeutic options remain limited. Evidence suggests CSD induces cognitive deficits through both central and peripheral mechanisms. The WenYang Tongbi (WYT) pill, composed of five herbs and Hirudo nipponica Whitman, has the potential on both improving nerve injury and peripheral homeostasis. Here, we hypothesized that the WYT pill can ameliorate related cognitive impairment and investigate its mechanism through a CSD mouse model. PURPOSE: This work was designed to evaluate the therapeutic potential of WYT aga

Investigation of an active site histidine-aspartate couple in Trimeresurus flavoviridis phospholipase A2.

When Trimeresurus flavoviridis phospholipase A2 was reacted with methyl p-nitrobenzenesulfonate, its activity decreased following first-order kinetics. The pH dependence of the rate constants of inactivation showed that His-48 with an apparent pKa of 6.5 controls the reaction. In the pH region below 6.5, N1-methylhistidine was predominantly formed. On the other hand, N1,N3-dimethylhistidine was almost exclusively produced in the pH region above 6.5. No N3-methylhistidine was detected at any pH tested. Such observations suggested that the first methylation occurred at the N1-position of the imi

Reversal of cancer cachexia in rats by cimaterol and supplemental nutrition.

The anabolic beta 2-agonist cimaterol was used in conjunction with supplemental nutrition to reverse cancer-induced cachexia and malnutrition in tumor-bearing rats. Cimaterol was administered to tumor-bearing rats receiving total parenteral nutrition or enteral nutrition for 10 days, beginning 2 weeks after subcutaneous transplantation of methylcholanthrene sarcoma. A significant increase occurred in both muscle weight and muscle protein in animals receiving cimaterol in conjunction with either enteral or parenteral feeding, compared to food fed tumor-bearing animals. NAT8 Variants, N-Acetylated Amino Acids, and Progression of CKD.

BACKGROUND AND OBJECTIVES: Genetic variants in NAT8, a liver- and kidney-specific acetyltransferase encoding gene, have been associated with eGFR and CKD in European populations. Higher circulating levels of two NAT8-associated metabolites, N-δ-acetylornithine and N-acetyl-1-methylhistidine, have been linked to lower eGFR and higher risk of incident CKD in the Black population. We aimed to expand upon prior studies to investigate associations between rs13538, a missense variant in NAT8, N-acetylated amino acids, and kidney failure in multiple, well-characterized cohorts.
