The organic mercury compounds, methylmercury and ethylmercury, inhibited ciliary movement of ventricular ependymal cells in the mouse brain around the concentrations reported for human poisoning.

Functions of the nervous system are supported by the flow of cerebrospinal fluid (CSF), which is driven by the ciliary beating of ventricular ependymal cells. The aim of the present study was to examine whether methylmercury (MeHg), a substance with potent neurotoxicity in humans, affects the ciliary movement. The effects of another organic mercury compound, ethylmercury (EtHg), were also assessed for comparison. Toxicity of MeHg or EtHg was evaluated by measuring alterations in the ciliary beat frequency of ependymal cells lining the third ventricle of mouse brain slices. The obtained results

Sensitive determination of methylmercury δ13C compound specific stable isotopic analysis by purge and trap gas chromatography combustion isotope ratio mass spectrometry.

Despite several decades of mercury research, answering fundamental questions on where and how methylmercury (CH3Hg) toxin is naturally produced in aquatic ecosystems, is still highly challenging. Investigating complex and/or coupled processes in the context of global changes requires new high-resolution analytical tools. The purpose of the compound specific carbon stable isotopic analysis (δ13C-CSIA) of the methyl group of methylmercury (CH3Hg), is to explore how the carbon cycle contributes to CH3Hg sources and formation pathways. The main problem associated with recent CH3Hg δ13C-CSIA method

Modification of the toxic effects of methylmercury and thimerosal by testosterone and estradiol in SH-SY5Y neuroblastoma cell line.

Short-chained alkyl mercury compounds accumulate in particularly in the brain. Exposure to these compounds is associated with various neurotoxic effects. Gender-based differences are observed in neurodevelopmental disorders, and testosterone and estradiol may alter the toxic effect of the compounds. The present study aimed to investigate the toxic effects of methylmercury and thimerosal on SH-SY5Y cells in high testosterone/low estradiol and high estradiol/low testosterone containing cellular environment and estimate whether male and female brains react differently to the toxic effects of meth

Genetic variation in the susceptibility to mercury and other metal compounds in Drosophila melanogaster.

The tolerance of Drosophila melanogaster to heavy metal compounds was investigated with special emphasis on methylmercury. A pronounced variation in tolerance to CH3HgOH, HgCl2, (C2H5)3PbCl, (CH3)3SnCl, and CdCl2 was recorded between 12 wild-type strains. After ranking the tolerance of the strains with respect to the five compounds rank correlations for experiments within and between compounds were calculated. The results showed a high degree of correlation within compounds but no unequivocal indication of a correlation between compounds, indicating that different mechanisms of genetic control

Methylmercury targets photoreceptor outer segments.

Human populations experience widespread low level exposure to organometallic methylmercury compounds through consumption of fish and other seafood. At higher levels, methylmercury compounds specifically target nervous systems, and among the many effects of their exposure are visual disturbances, including blindness, which previously were thought to be due to methylmercury-induced damage to the visual cortex. Here, we employ high-resolution X-ray fluorescence imaging using beam sizes of 500 × 500 and 250 × 250 nm(2) to investigate the localization of mercury at unprecedented resolution in secti

Multiresponse biomarker evaluation of interactions between methylmercury and Arochlor 1260 in quail.

The single and combined effects of methylmercury and Arochlor 1260 were investigated in experimental quail treated chronically with the two compounds at low and high doses. A series of metabolic and biochemical biomarkers were evaluated together with mercury and PCB accumulation to pinpoint the effects of treatment with one or both chemicals. Methylmercury alone was associated with a decrease in serum cholesterol. Less PCBs were accumulated in tissues when Arochlor 1260 was combined with methylmercury than when the former was administered alone. Liver monooxygenase (MFO) activity was depressed

Mimicking receptor for methylmercury preconcentration based on ion-imprinting.

Solid-phase extraction (SPE) based on molecularly imprinted polymers (MIPs) were used to develop selective separation and preconcentration for methylmercury ion from complex matrixes. In this study, an ion-imprinting polymer was prepared to make artificial organomercury lyase preorganizing three methacryloyl-(L)-cysteine methylester (MAC) monomers and one methylmercury ion in a three-coordinate form by template polymerization, with the goal preparing a solid-phase which has the high selectivity for methylmercury ions. Methylmercury-imprinted beads were produced by a dispersion polymerization t

Molecular composition of organic matter controls methylmercury formation in boreal lakes.

A detailed understanding of the formation of the potent neurotoxic methylmercury is needed to explain the large observed variability in methylmercury levels in aquatic systems. While it is known that organic matter interacts strongly with mercury, the role of organic matter composition in the formation of methylmercury in aquatic systems remains poorly understood. Here we show that phytoplankton-derived organic compounds enhance mercury methylation rates in boreal lake sediments through an overall increase of bacterial activity. Accordingly, in situ mercury methylation defines methylmercury le

Mercury compounds and the immune system: a review.

This article reviews the literature data concerning the immunologic monitoring of animals and cell cultures exposed to mercury compounds. Mercury is present in nature as metallic mercury, mono- and bivalent inorganic compounds, and organic alkyl, aryl and alloxy-alkyl compounds. Methylmercury is most important in terms of environmental exposure while metallic mercury is the most common form to which workers are exposed. The database on immune function disturbances in human induced by mercury compounds is limited. Immunotoxicity assessment in animals, mainly in rodents, with subsequent extrapol

Behavioral teratology of mercury and its compounds.

Mercury and its compounds have a wide spectrum of toxicities depending upon the chemical forms and modes of exposure. Among the various chemical forms, mercury vapor and methylmercury are well known and established as neurotoxic agents. Since the disasters in Minamata and Iraq, in which fetuses were more susceptible than adults to methylmercury exposure, much attention has been focused on prenatal exposure to mercury and its consequence. Recently postnatal effects of in utero exposure to methylmercury through fish (and marine mammals) consumption by mothers have been concerned and several epid

The toxicology of mercury.

The major physical forms of mercury to which humans are exposed are mercury vapor, Hg0, and methylmercury compounds, Ch3HgX. Mercury vapor emitted from both natural and anthropogenic sources is globally distributed in the atmosphere. It is returned as a water-soluble form in precipitation and finds its way into bodies of fresh and ocean water. Land run-off also accounts for further input into lakes and oceans. Inorganic mercury, present in water sediments, is subject to bacterial conversion to methylmercury compounds that are bioaccumulated in the aquatic food chain to reach the highest concen

Synergistic toxicity of the environmental neurotoxins methylmercury and β-N-methylamino-L-alanine.

Determination of the environmental factors involved in neurodegenerative diseases has been elusive. Methylmercury and β-N-methylamino-L-alanine (BMAA) have both been implicated in this role. Exposure of primary cortical cultures to these compounds independently induced concentration-dependent neurotoxicity. Importantly, concentrations of BMAA (10-100 μM) that caused no toxicity alone potentiated methylmercury (3 μM) toxicity. In addition, concentrations of BMAA and methylmercury that had no effect by themselves on the main cellular antioxidant glutathione together decreased glutathione levels.

Preparation of CH3 203HgCl of high specific activity.

In order to have a methylmercury compound of much higher specific activity than was obtainable from commercial suppliers of the compound, a procedure for the methylation of radioactive mercuric chloride with no addition of non-radioactive mercury has been developed. Methylation is accomplished by reacting the mercuric chloride with tetramethyl tin. Extractions into and out of benzene twice produces a methylmercury compound which is finally dissolved in a dilute sodium carbonate solution suitable for biological experiments. The entire procedure to produce a batch of the compound may be accompli

A laser Raman spectroscopic study on the interaction of alkylmercury with thiol and sulfur-containing compounds.

The interaction of the methylmercury cation with sulfur compounds in aqueous solution at physiological pH was studied by laser Raman spectroscopy. Metal binding is shown to occur preferentially at the sulfhydryl group of sulfur compounds. Raman frequencies of S-Hg stretching of the one-to-one methylmercury-sulfhydryl or sulfur-containing complexes were observed at approximately 330 cm-1. There was no frequency shift when ligands were exchanged. However, the relative intensity (I S-Hg/I C-Hg) was different. The relative intensities of MeHg-thioglycerol, MeHg-cysteine and MeHg-2-mercaptobenzothi

Overexpression of the ubiquitin-conjugating enzyme Cdc34 confers resistance to methylmercury in Saccharomyces cerevisiae.

A search was made for genes that confer resistance to methylmercury in yeast using a genomic DNA library derived from Saccharomyces cerevisiae. The genomic library was introduced into yeast and transformants that grew in the presence of a normally toxic concentration of methylmercury were selected. We sequenced the genomic DNA fragment in the plasmid from the clone with the highest resistance to methylmercury and analyzed the sequence for presence of an open reading frame that might confer resistance to methylmercury. We identified a gene, CDC34 (also known as UBC3), that increased resistance

Lymphocyte proliferative response and tissue distribution of methylmercury sulfide and chloride in exposed rats.

The immunotoxic effects and tissue distribution of different forms of methylmercury compounds were studied in rats. Methylmercury sulfide or methylmercury chloride was fed to rats at concentrations of 5 or 500 microg/L in drinking water for 8 wk. T-cell lymphocyte proliferative response to phytohemagglutinin (PHA) and determination of tissue distribution of mercury by gas chromatography using electron capture were assayed. Four different forms of mercury compounds were employed: MeHgS-, (MeHg)2S, (MeHg)3S+, and MeHgCl. Results indicated that exposure to methylmercury significantly enhanced lym

Impaired biliary excretion and whole body elimination of methylmercury in rats with congenital defect in biliary glutathione excretion.

Biliary excretion of methylmercury, a major route of elimination of this toxic compound, was less than 2% of control in Eisai hyperbilirubinemic (EHBR) rats, a mutant Sprague-Dawley strain with a defect in biliary excretion of a variety of organic anions, including glutathione S-conjugates and reduced glutathione (GSH). Biliary GSH excretion in EHBR rats was also < 2% of controls, confirming previous findings. Impaired biliary methylmercury and GSH excretion was not explained by decreased hepatic content of these compounds. Indeed, hepatic methylmercury and GSH concentrations in EHBR rats were

Development of a quadruple isotope spiking methodology to study mercury methylation and methylmercury demethylation in environmental and biological media.

A quadruple isotope spiking methodology has been developed and applied for the study of Hg-species-specific transformations in incubation experiments. The method enables for the first time discrimination between interconversions reactions occurring during the incubation step from those occurring during the analytical procedure. Two different mathematical models for quadruple spiking were developed and applied here in model incubation experiments to track inorganic Hg(II) methylation and methylmercury demethylation. The results showed identical Hg compounds concentrations and interconversion re

Glutathione reductase plays a role in the metabolism of methylmercury degradation in Rhodotorula mucilaginosa.

UNLABELLED: Mercury pollution is a kind of heavy metal pollution with great harm and strong toxicity which exists worldwide. Some microorganisms can convert highly toxic methylmercury into inorganic mercury compounds with significantly reduced toxicity. This is an effective means of methylmercury pollution remediation. As a safe microorganism with great potential in the remediation of heavy metal pollution, Rhodotorula mucilaginosa has not been studied in the remediation of methylmercury. Here, a R. mucilaginosa strain Rm4 with high methylmercury resistance was obtained by wild-strain screenin

The blood-cerebrospinal fluid barrier--first evidence for an active transport of organic mercury compounds out of the brain.

Exposure to organic mercury compounds promotes primarily neurological effects. Although methylmercury is recognized as a potent neurotoxicant, its transfer into the central nervous system (CNS) is not fully evaluated. While methylmercury and thiomersal pass the blood-brain barrier, limited data are available regarding the second brain regulating interface, the blood-cerebrospinal fluid (CSF) barrier. This novel study was designed to investigate the effects of organic as well as inorganic mercury compounds on, and their transfer across, a porcine in vitro model of the blood-CSF barrier for the
