Pink powerhouses: insights into the multifaceted role of Methylobacterium in climate-resilient farming.

The plant microbiomes consist of a myriad of microorganisms that inhabit and interact with plant tissues and play pivotal roles in improving crop productivity and sustainability. These microbiomes constitute bacteria, fungi, archaea and viruses that have coevolved and supported plants inhabiting the Earth for millions of years. Among these, bacterial members play major functional roles in fostering plant growth and are regarded as plant growth-promoting bacteria (PGPB). One of the major bacterial genera of the plant microbiome that colonizes the entire plant system is the genus Methylobacteriu

Genomic and Transcriptomic Analysis of Growth-Supporting Dehalogenation of Chlorinated Methanes in Methylobacterium.

Bacterial adaptation to growth with toxic halogenated chemicals was explored in the context of methylotrophic metabolism of Methylobacterium extorquens, by comparing strains CM4 and DM4, which show robust growth with chloromethane and dichloromethane, respectively. Dehalogenation of chlorinated methanes initiates growth-supporting degradation, with intracellular release of protons and chloride ions in both cases. The core, variable and strain-specific genomes of strains CM4 and DM4 were defined by comparison with genomes of non-dechlorinating strains. In terms of gene content, adaptation towar

Engineering phosphoribosylpyrophosphate synthetase in Methylobacterium radiotolerans for lettuce growth and quality enhancement.

A bacterial strain designated DS15 was isolated from the leaves of Salvia miltiorrhiza and characterized to assess its plant growth-promoting (PGP) traits. Based on 16 S rRNA gene and genome alignment, DS15 strain was identified as Methylobacterium radiotolerans. Its PGP traits, including indole acetic acid (IAA) production, phosphate solubilization, and siderophore secretion were determined. Foliar application of DS15 strain significantly increased 35.0% of fresh weight of lettuce seedlings grown in a standard growth chamber compared to the control group. Genomic analysis revealed the presenc

Effective utilization of dichloromethane by a newly isolated strain Methylobacterium rhodesianum H13.

An effective dichloromethane (DCM) utilizer Methylobacterium rhodesianum H13 was isolated from activated sludge. A response surface methodology was conducted, and the optimal conditions were found to be 4.5 g/L Na2HPO4·12H2O, 0.5 g/L (NH4)2SO4, an initial pH of 7.55, and a temperature of 33.7 °C. The specific growth rate of 0.25 h(-1) on 10 mM DCM was achieved, demonstrating that M. rhodesianum H13 was superior to the other microorganisms in previous investigations of DCM utilization. DCM mineralization paralleled the production of cells, CO2, and water-soluble metabolites, as well as the rele

Aromatic acid metabolism in Methylobacterium extorquens reveals interplay between methylotrophic and heterotrophic pathways.

Efforts towards microbial conversion of lignin to value-added products face many challenges because lignin's methoxylated aromatic monomers release toxic C1 byproducts such as formaldehyde. The ability to grow on methoxylated aromatic acids (e.g., vanillic acid) has recently been identified in certain clades of methylotrophs, bacteria characterized by their unique ability to tolerate and metabolize high concentrations of formaldehyde. Here, we use a phyllosphere methylotroph isolate, Methylobacterium extorquens SLI 505, as a model to identify the fate of formaldehyde during methylotrophic grow

Methylobacterium extorquens PA1 utilizes multiple strategies to maintain formaldehyde homeostasis during methylotrophic growth.

Metabolic homeostasis is a central organizing principle of physiology whereby dynamic processes work to maintain a balanced internal state. Highly reactive essential metabolites are ideally maintained at equilibrium to prevent cellular damage. In the facultative methylotrophic bacterium Methylobacterium extorquens PA1, the utilization of one-carbon growth substrates, including methanol, generates formaldehyde as an obligate intermediate. Formaldehyde is highly chemically reactive and capable of damaging various biomolecules, making formaldehyde homeostasis critical during methylotrophic growth

Rare earth element-dependent microbial growth and biomining potential in Methylobacterium fujisawaense LAC1.

Rare earth elements (REEs) are essential for modern technology and have recently been recognized for their role in microbial methanol oxidation, particularly in methanotrophs and methylotrophs. We report a novel acidophilic methylotroph, Methylobacterium fujisawaense LAC1 that exhibits a remarkable ability to utilize and accumulate both light and heavy REEs. This is the first methylotroph reported to utilize and accumulate heavy REEs. Strain LAC1 can grow on acetate, glycerol, succinate, 1-butanol, glucose, and dodecane with calcium. Specifically, with certain REEs, LAC1 can also metabolize et

[Isolation and degradation characteristics of dichloromethane-degradation bacterial strain by Methylobacterium rhodesianum H13].

A dichloromethane-degrading bacterium Methylobacterium rhodesianum H13 which utilized the DCM as the sole carbon and energy source was isolated. According to the research, M. rhodesianum H13 could completely degrade 5 mmol x L(-1) DCM in 23 h with the initial cell concentration of 0.82 mg x L(-1), pH 7.0, 30 degrees C, and the cell yield rate was about 0.136 g x g(-1) DCM. With the degradation of DCM, Cl- concentration gradually raised (the release of Cl- concentration was about 2 times higher as the DCM), pH value dropped to 6.75, and the solution was weakly acidic. Temperature, pH, DCM conce

Phylogeny poorly predicts the utility of a challenging horizontally transferred gene in Methylobacterium strains.

Horizontal gene transfer plays a crucial role in microbial evolution. While much is known about the mechanisms that determine whether physical DNA can be transferred into a new host, the factors determining the utility of the transferred genes are less clear. We have explored this issue using dichloromethane consumption in Methylobacterium strains. Methylobacterium extorquens DM4 expresses a dichloromethane dehalogenase (DcmA) that has been acquired through horizontal gene transfer and allows the strain to grow on dichloromethane as the sole carbon and energy source. We transferred the dcmA ge

Metabolism-linked methylotaxis sensors responsible for plant colonization in Methylobacterium aquaticum strain 22A.

Motile bacteria take a competitive advantage in colonization of plant surfaces to establish beneficial associations that eventually support plant health. Plant exudates serve not only as primary growth substrates for bacteria but also as bacterial chemotaxis attractants. A number of plant-derived compounds and corresponding chemotaxis sensors have been documented, however, the sensors for methanol, one of the major volatile compounds released by plants, have not been identified. Methylobacterium species are ubiquitous plant surface-symbiotic, methylotrophic bacteria. A plant-growth promoting b

Methylobacterium as a key symbiont in plant-microbe interactions: Its ecological and agricultural significance.

Pink-pigmented facultative methylotrophs (PPFMs), encompassing the genera Methylobacterium and Methylorubrum, can utilize reduced one-carbon compounds such as methanol, methylamine, formaldehyde, and formate as carbon and energy sources. They are commonly associated with plants, particularly on leaf surfaces (phyllosphere), where their methylotrophic metabolism offers a significant adaptive advantage over other bacterial species. These genera hold quite diverse species with unique phenotypes. Many studies report plant growth-promoting activity of the genera due to their ability to produce plan

Decreasing peptide deformylase activity is a beneficial strategy for increasing formaldehyde resistance in Methylobacterium extorquens.

Formaldehyde is a highly toxic metabolite that can cause extensive damage to DNA and proteins, and strategies to mitigate formaldehyde toxicity are poorly understood. Methylotrophic bacteria, such as Methylobacterium extorquens, thrive on one-carbon compounds as sole sources of carbon and energy. These organisms are excellent models for discovering formaldehyde stress response systems because formaldehyde is an obligate intermediate in their central carbon metabolism. Here, we characterize an evolved def allele (def evo ) that increases formaldehyde resistance in M. extorquens. The def gene en

N-terminome and proteogenomic analysis of the Methylobacterium extorquens DM4 reference strain for dichloromethane utilization.

UNLABELLED: Methylobacterium strains can use one-carbon compounds, such as methanol, for methylotrophic growth. In addition to methanol, a few strains also utilize dichloromethane, a major industrial chlorinated solvent pollutant. With a fully assembled and annotated genome, M. extorquens DM4 is the reference bacterium for aerobic dichloromethane degradation. The doublet N-terminal oriented proteomics (dN-TOP) strategy was applied to further improve its genome annotation and a differential proteomics approach was performed to compare M. extorquens DM4 grown either with methanol or dichlorometh

Catheter infection caused by Methylobacterium in immunocompromised hosts: report of three cases and review of the literature.

Three cases of catheter infection due to Methylobacterium extorquens are reported. Each patient had a history of acute leukemia and was immunocompromised; two had undergone bone marrow transplantation, and the third was receiving consolidation chemotherapy. All three patients survived after removal of the central venous catheter and antibiotic treatment. The clinical features of these cases are compared with those of the 12 previously reported cases of infection due to Methylobacterium species.

Phosphoribosylpyrophosphate synthetase as a metabolic valve advances Methylobacterium/Methylorubrum phyllosphere colonization and plant growth.

The proficiency of phyllosphere microbiomes in efficiently utilizing plant-provided nutrients is pivotal for their successful colonization of plants. The methylotrophic capabilities of Methylobacterium/Methylorubrum play a crucial role in this process. However, the precise mechanisms facilitating efficient colonization remain elusive. In the present study, we investigate the significance of methanol assimilation in shaping the success of mutualistic relationships between methylotrophs and plants. A set of strains originating from Methylorubrum extorquens AM1 are subjected to evolutionary press
