Catheter infection caused by Methylobacterium in immunocompromised hosts: report of three cases and review of the literature.

Three cases of catheter infection due to Methylobacterium extorquens are reported. Each patient had a history of acute leukemia and was immunocompromised; two had undergone bone marrow transplantation, and the third was receiving consolidation chemotherapy. All three patients survived after removal of the central venous catheter and antibiotic treatment. The clinical features of these cases are compared with those of the 12 previously reported cases of infection due to Methylobacterium species.

[Functionality of Metdi5511gene in Methylobacterium dichloromethanicum DM4].

A knockout mutant of Methylobacterium dichloromethanicum DM4 with an inactivated gene of a putative transcription regulator METDI5511 (ΔMETDI5511) has been obtained. The expression of this gene increases many times when the strain is grown on dichloromethane compared to methanol. The mutant had a low growth rate on dichloromethane as compared with the original strain and was found to be more sensitive to influences of various types of stress (oxidative, osmotic stress, heat, and drying). The cells were stained with Fluorescent Brightener 28 (Calcofluor white), and the intensity of their fluore

Adaptive laboratory evolution of Methylorubrum extorquens for lanthanide-dependent robust growth under high concentrations of methanol.

Methylorubrum extorquens AM1 has been extensively studied as a platform for biomanufacturing using a methanol feedstock. Given that lanthanide (Ln) ions are recognized as important metals related to methylotrophy of methylotrophic bacteria, this study applied adaptive laboratory evolution to M. extorquens under Ln-supplemented conditions to obtain strains with robust growth at high concentrations of methanol. The cultivation results indicated a possible Ln-dependent mechanism for methanol tolerance in this methylotroph. The parent strain was unable to grow at 5% (v/v) methanol under Ln-free co

Adaptively evolved Methylorubrum extorquens with enhanced formate tolerance and its application in 3-hydroxypropionic acid production.

UNLABELLED: Methylorubrum extorquens AM1, a native formate-utilizing bacterium, has exhibited limited capacity to tolerate formate. In this study, we employed an adaptive laboratory evolution (ALE) strategy to develop an evolved strain FT3 derived from M. extorquens AM1, with enhanced formate tolerance. When cultivated with a mixture of carbon sources containing 90 mM formate and 30 mM methanol, the FT3 strain exhibited 5.3 times higher optical density (OD600) compared to the parental strain. FT3 strain was shown to efficiently utilize both methanol and formate in experiments using 13C-labeled

Persistence preserves phenotypic heterogeneity in non-genetic formaldehyde resistance in Escherichia coli.

OBJECTIVES: Recently, it was discovered that Methylobacterium extorquens possesses a spectrum of phenotypic states conferring non-genetic resistance to formaldehyde. The non-genetic resistance allowed rapid growth on levels of formaldehyde which were lethal to the majority of the population. To investigate whether this is a common phenomenon, we tested the distantly related Escherichia coli for heterogeneity to formaldehyde stress during growth on glucose. RESULTS: Like M. extorquens, E. coli populations have a wide, reversible, continuous range of non-genetic resistance to formaldehyde. Howev

[Adaptation of aerobic methylobacteria to dichloromethane degradation].

A shortening of the lag phase in dichloromethane (DCM) consumption was observed in the methylobacteria Methylopila helvetica DM6 and Albibacter methylovorans DM10 after prior growth on methanol with the presence of 1.5% NaCI. Neither heat nor acid stress accelerated methylobacterium adaptation to DCM consumption. Sodium azide (1 mM) and potassium cyanide (1 mM) inhibited consumption of DCM by these degraders but not by transconjugants Methylobacterium extorquens AM1, expressing DCM dehalogenase but unable to grow on DCM. This indicates that the degrader strains possess energy-dependent systems

Phosphoribosylpyrophosphate synthetase as a metabolic valve advances Methylobacterium/Methylorubrum phyllosphere colonization and plant growth.

The proficiency of phyllosphere microbiomes in efficiently utilizing plant-provided nutrients is pivotal for their successful colonization of plants. The methylotrophic capabilities of Methylobacterium/Methylorubrum play a crucial role in this process. However, the precise mechanisms facilitating efficient colonization remain elusive. In the present study, we investigate the significance of methanol assimilation in shaping the success of mutualistic relationships between methylotrophs and plants. A set of strains originating from Methylorubrum extorquens AM1 are subjected to evolutionary press

[Effect of DNA-damaging agents on the aerobic methylobacteria capable and incapable of utilizing dichloromethane].

Methylobacterium dichloromethanicum DM4, a degrader of dichloromethane (DCM), was more tolerant to the effect of H2O2 and UV irradiation than Methylobacterium extorquens AM1, which does not consume DCM. Addition of CH2Cl2 to methylobacteria with active serine, ribulose monophosphate, and ribulose bisphosphate pathways of C1 metabolism, grown on methanol, resulted in a 1.1- to 2.5-fold increase in the incorporation of [alpha-32P]dATP into DNA Klenow fragment (exo-). As DCM dehalogenase was not induced in this process, the increase in total lengths of DNA gaps resulted from the action of DCM rat

Diversity of Methylobacterium species associated with New Zealand native plants.

Methylobacterium species are abundant colonizers of the phyllosphere due to the availability of methanol, a waste product of pectin metabolism during plant cell division. The phyllosphere is an extreme environment, with a landscape that is heterogeneous and continuously changing as the plant grows and is exposed to high levels of ultraviolet irradiation. Geographically, New Zealand (NZ) has been isolated for over a million years, has a biologically diverse flora, and is considered a biodiversity hotspot, with most native plants being endemic. We therefore hypothesize that the phyllosphere of N

Phylogeny poorly predicts the utility of a challenging horizontally transferred gene in Methylobacterium strains.

Horizontal gene transfer plays a crucial role in microbial evolution. While much is known about the mechanisms that determine whether physical DNA can be transferred into a new host, the factors determining the utility of the transferred genes are less clear. We have explored this issue using dichloromethane consumption in Methylobacterium strains. Methylobacterium extorquens DM4 expresses a dichloromethane dehalogenase (DcmA) that has been acquired through horizontal gene transfer and allows the strain to grow on dichloromethane as the sole carbon and energy source. We transferred the dcmA ge

[Aerobic methylobacteria as the basis for a biosensor for dichloromethane detection].

Cells of dichloromethane (DChM) bacteria-destructors were immobilized by sorption on different types of membranes, which were fixed on the measuring surface of a pH-sensitive field transistor. The presence of DChM in the medium (0.6-8.8 mM) led to a change in the transistor's output signal, which was determined by the appearance of H+ ions in the medium due to DChM utilization by methylobateria. Lanthanide-dependent isolation of phyllosphere methylotrophs selects for a phylogenetically conserved but metabolically diverse community.

Lanthanides have emerged as important metal cofactors for biological processes. Lanthanide-associated metabolisms are well-studied in leaf symbiont methylotrophic bacteria, which utilize reduced one-carbon compounds such as methanol for growth. Yet, the importance of lanthanides in plant-microbe interactions and on microbial physiology and colonization in plants remains poorly understood. To investigate this, 344 pink-pigmented facultative methylotrophs were isolated from soybean leaves by selecting for bacteria capable of methanol oxidation with lanthanide cofactors, but none were obligately

A tungsten-specific maturation pathway governs cofactor assembly of a CO2-reducing formate dehydrogenase in Methylorubrum extorquens.

Tungsten-dependent formate dehydrogenases catalyze the reversible interconversion of carbon dioxide and formate and play key roles in microbial redox metabolism, yet the molecular basis for selective tungsten cofactor assembly in aerobic bacteria remains poorly understood. Here, we define a dedicated tungsten-specific maturation pathway that governs cofactor assembly and delivery to the carbon dioxide-reducing formate dehydrogenase MeFDH1 in Methylorubrum extorquens AM1. Targeted genetic deletions identified moeA1, mobB, and fdhD as essential for MeFDH1 activity and tungsten incorporation. Pro

Engineering photo-methylotrophic Methylobacterium for enhanced 3-hydroxypropionic acid production during non-growth stage fermentation.

This study explored the potential of methanol as a sustainable feedstock for biomanufacturing, focusing on Methylobacterium extorquens, a well-established representative of methylotrophic cell factories. Despite this bacterium's long history, its untapped photosynthetic capabilities for production enhancement have remained unreported. Using genome-scale flux balance analysis, it was hypothesized that introducing photon fluxes could boost the yield of 3-hydroxypropionic acid (3-HP), an energy- and reducing equivalent-consuming chemicals. To realize this, M. extorquens was genetically modified b

A novel engineered strain of Methylorubrum extorquens for methylotrophic production of glycolic acid.

The conversion of CO2 into methanol depicts one of the most promising emerging renewable routes for the chemical and biotech industry. Under this regard, native methylotrophs have a large potential for converting methanol into value-added products but require targeted engineering approaches to enhance their performances and to widen their product spectrum. Here we use a systems-based approach to analyze and engineer M. extorquens TK 0001 for production of glycolic acid. Application of constraint-based metabolic modeling reveals the great potential of M. extorquens for that purpose, which is no

Transposon mutagenesis for methylotrophic bacteria using Methylorubrum extorquens AM1 as a model system.

Transposon mutagenesis utilizes transposable genetic elements that integrate into a recipient genome to generate random insertion mutations which are easily identified. This forward genetic approach has proven powerful in elucidating complex processes, such as various pathways in methylotrophy. In the past decade, many methylotrophic bacteria have been shown to possess alcohol dehydrogenase enzymes that use lanthanides (Lns) as cofactors. Using Methylorubrum extorquens AM1 as a model organism, we discuss the experimental designs, protocols, and results of three transposon mutagenesis studies t

Methanol metabolism in synthetic methylotrophic microorganisms.

Methanol has garnered increasing attention as an ideal carbon source for chemical biomanufacturing due to its abundance and easy availability. Microbial methylotrophy mainly refers to the ability to utilize reduced one-carbon compounds such as methanol, methane or formaldehyde as sole or primary sources of carbon and energy to sustain growth. Besides native methylotrophic microbes, such as Komagataella phaffii (K. phaffii), Methylobacterium extorquens (M. extorquens), Bacillus methanolicus (B. methanolicus), many other yeasts or bacteria have been engineered to exhibit methylotrophy or by reco

Functional verification of bacteriochlorophyll biosynthetic genes distributed in pink-pigmented facultative methylotrophs.

Pink-pigmented facultative methylotrophs (PPFMs), represented by the genera Methylobacterium and Methylorubrum, are ubiquitous bacteria commonly found in nutrient-poor environments such as plant leaf surfaces. Comparative genomics has revealed that many PPFMs possess genes for bacteriochlorophyll (BChl) biosynthesis; however, whether these genes support functional phototrophy has remained largely unknown. Here, we systematically investigated BChl production and its physiological function in PPFMs. Among 43 Methylobacterium/Methylorubrum type strains examined, 36 produced BChl, demonstrating th

Comparative Binding Studies of the Chelators Methylolanthanin and Rhodopetrobactin B to Lanthanides and Ferric Iron.

The question of lanthanide (Ln) uptake in Ln-using bacteria has gained a lot of attention in recent years, and the existence of specific Ln-binding metallophores, termed lanthanophores, has been postulated. Here, the recently isolated metallophore methylolanthanin (MLL), which is shown to be involved in Ln metabolism of Methylobacterium extorquens AM1 along the structurally related siderophore rhodopetrobactin B (RPB B), is investigated. The total synthesis of both chelators as well as Ln-binding investigations employing a multitude of spectroscopic methods is reported. Compared to MLL, RPB B
