Apicomplexan parasites such as Toxoplasma gondii initiate motility through rapid, spatially confined cytoskeletal activation at their apical end. While calcium-, lipid-, and kinase-based signalling pathways have been partially elucidated, how these cues are translated into mechanical force remains unclear. Here, we uncover a dual methyltransferase mechanism that orchestrates this process. We characterise TgPCKMT, a PreConoidal ring-associated lysine (K) MethylTransferase, as an essential upstream regulator of motility. TgPCKMT anchors the actin nucleator Formin-1 (TgFRM1) at the conoid, enabli

Bacteria and fungi use nonribosomal peptide synthetases (NRPSs) to generate secondary metabolites called nonribosomal peptides (NRPs). A common feature of NRPs is the incorporation of D-stereocenter amino acids, which enhance chemical functionality and confer resistance to proteolytic degradation. Typically, these stereochemical inversions arise from dedicated epimerase domains, which are well-characterized. However, 2-hydroxyphenylthiazoline-containing natural products lack these conventional tailoring domains and are instead proposed to utilize noncanonical epimerase domains embedded, or "st

Hydrogen sulfide (H2S), methanethiol (MeSH), and dimethylsulfide (DMS) are abundant sulfur gases with crucial roles in global sulfur cycling, chemotaxis, and climate regulation. Microorganisms can S-methylate H2S and MeSH, which can be cytotoxic, to yield non-toxic DMS via MddA or MddH enzymes in largely terrestrial or marine environments, respectively. However, the potential of many important and abundant bacteria like Actinomycetota is underestimated due to unknown Mdd enzymes. Here, two novel S-adenosine-methionine-dependent H2S and MeSH S-methyltransferases, MddM1 and MddM2 are identified,

Although epigenetic abnormalities have been described in Huntington's disease (HD), the causal epigenetic mechanisms driving neurodegeneration in HD cortex and striatum remain undefined. Using an epigenetic pathway-targeted drug screen, we report that inhibitors of DNA methyltransferases (DNMTs), decitabine and FdCyd, block mutant huntingtin (Htt)-induced toxicity in primary cortical and striatal neurons. In addition, knockdown of DNMT3A or DNMT1 protected neurons against mutant Htt-induced toxicity, together demonstrating a requirement for DNMTs in mutant Htt-triggered neuronal death and sugg

Epigenetic regulation through protein posttranslational modifications is essential in development and disease. Among the key chemical modifications is protein methylation carried out by protein methyltransferases (PMTs). Quantitative and sensitive PMT activity assays can provide valuable tools to investigate PMT functions. Here we developed an enzyme-coupled luminescence assay for S-adenosyl-l-methionine (AdoMet/SAM)-based PMTs. In this assay, S-adenosyl-l-homocystine (AdoHcy/SAH), the by-product of PMT-involved methylation, is sequentially converted to adenine, adenosine monophosphate, and th

Metabolic syndrome (MetS) comprises a cluster of interconnected metabolic abnormalities that collectively elevate the risk of cardiovascular disease and mortality. With its global prevalence escalating, understanding the molecular underpinnings of MetS has become increasingly imperative. Protein arginine methyltransferases (PRMTs), classically studied for their epigenetic functions and oncogenic properties, are now recognized as pivotal regulators of metabolic homeostasis. Emerging research reveals that these enzymes coordinate crucial aspects of cellular metabolism through multiple mechanisms

BACKGROUND: The SET and MYND domain-containing (SMYD) protein family is a group of lysine methyltransferases with SET and MYND domains and plays a critical role in regulating gene expression through the methylation of histone and non-histone proteins.
SUMMARY: Studies have linked mutations or overexpression of SMYD2 and SMYD3 to various cancers, including renal carcinoma. Recent research also demonstrates that the expression levels and activity of SMYD2 and SMYD3 are increased in animal models of renal diseases such as autosomal dominant polycystic kidney disease, renal fibrosis, and diabetic

DNA methyltransferases (DNMTs) are important epigenetic modification during spermatogenesis. To further evaluate the pattern of DNMTs in horse testes during development, we investigated the expression and localization of DNMT1, DNMT3a and DNMT3b at different time points. The qRT-PCR results showed that DNMT1 expression was maintained in testes tissue from 6-month-old (0.5y) to 2-year-old (2y) of age and decreased after 3-year-old (3y) (P < 0.01). The expression levels of DNMT3a and DNMT3b peaked in testes tissue at 3y (P < 0.01). At 4-year-old (4y), the expression of DNMT3a and DNMT3b was decr

During the aging of the global population, the prevalence of neurodegenerative diseases will be continuously growing. Although each disorder is characterized by disease-specific protein accumulations, several common pathophysiological mechanisms encompassing both genetic and environmental factors have been detected. Among them, protein arginine methyltransferases (PRMTs), which catalyze the methylation of arginine of various substrates, have been revealed to regulate several cellular mechanisms, including neuronal cell survival and excitability, axonal transport, synaptic maturation, and myeli

Uterine leiomyosarcoma (LMS) is an aggressive tumor that presents poor prognosis, high rates of recurrence and metastasis. Because of its rarity, there is no information available concerning LMS molecular mechanisms of origin and development. Here, we assessed the expression profile of Hedgehog (HH) signaling pathway markers and the effects of their pharmacological inhibition on uterine smooth muscle (UTSM), leiomyoma and LMS cells. Additionally, we also evaluated the effects of DNMTs inhibition on LMS cells behavior. Cell proliferation, migration and apoptosis rates were evaluated by MTT, Scr

BACKGROUND: Hepatocellular carcinoma (HCC) is a major health concern in many countries including Egypt. The alteration in DNA methylation that was observed in HCC patients suggests a possible role of DNA methyltransferases (DNMTs) in the disease pathogenesis in addition to potential role as a disease biomarker.
AIM: To study the change in DNMTs expression in chronic HCV infected patients as potential non- invasive biomarker for diagnosis of hepatocellular carcinoma.
METHODS: 26 patients with HCC, 45 patients with liver cirrhosis, 20 chronic HCV patients and 20 apparently healthy individuals

Successful human pregnancy depends on the precise spatio-temporal orchestration of trophoblast lineages to establish a functional feto-maternal interface. While DNA methyltransferases (DNMTs) are recognized as core drivers of epigenetic programming, emerging evidence reveals that they are not merely passive transcriptional silencers, but also dynamic architectural coordinators of placental morphogenesis. This work presents the roles of maintenance (DNMT1) and de novo (DNMT3A, DNMT3B) methyltransferases in dictating the precise timing of trophoblast fate transitions. During trophoblast fusion,

Methylation, catalyzed by a diverse family of S-adenosylmethionine (SAM)-dependent methyltransferases, is a fundamental biochemical process that regulates the function of proteins, DNA, and RNA. Among these modifications, protein arginine methylation, catalyzed by protein arginine methyltransferases (PRMTs), represents a critical post-translational mechanism regulating transcription, DNA repair, RNA splicing, and signal transduction. Emerging evidence highlights important roles of PRMTs in cardiovascular diseases (CVDs); however, current research is limited by several challenges, including the

Aminoglycoside-producing Actinobacteria are known to protect themselves from their own aminoglycoside metabolites by producing 16S ribosomal RNA methyltransferase (16S-RMTase), which prevents them from binding to the 16S rRNA targets. Ten acquired 16S-RMTases have been reported from gram-negative pathogens. Most of them posttranscriptionally methylate residue G1405 of 16S rRNA resulting in high-level resistance to gentamicin, tobramycin, amikacin, and plazomicin. Strains that produce 16S-RMTase are frequently multidrug-resistant or even extensively drug-resistant. Although the direct clinical

16S rRNA methyltransferases confer resistance to most aminoglycosides, but discriminating their activity from that of aminoglycoside-modifying enzymes (AMEs) is challenging using phenotypic methods. We demonstrate that arbekacin, an aminoglycoside refractory to most AMEs, can rapidly detect 16S methyltransferase activity in Enterobacteriaceae with high specificity using the standard disk susceptibility test.

Methylation of histones is one of the important "epigenetic" mechanisms associated with the transcriptional silencing and/or activating of tumor suppressor genes. To assess whether epigenetic phenomena could be involved in salivary gland carcinogenesis, the expression levels of four histone lysine-methyltransferases (HMT) were investigated, in both pleomorphic adenoma and the adjacent normal tissue of the parotid glands. The expression levels of three HMTs, SETB1, Eu-HMTase and SET08, were higher in tumor tissues. On the contrary, DOTL1 presented a lower expression level in the tumor tissues t

DNA methylation is important in cellular, developmental and disease processes, as well as in bacterial restriction-modification systems. Methylation of DNA at the amino groups of cytosine and adenine is a common mode of protection against restriction endonucleases afforded by the bacterial methyltransferases. The first structure of an N:6-adenine methyltransferase belonging to the beta class of bacterial methyltransferases is described here. The structure of M. RSR:I from Rhodobacter sphaeroides, which methylates the second adenine of the GAATTC sequence, was determined to 1.75 A resolution us

SETD2 is a critical tumor suppressor in lung cancer that catalyzes histone H3 lysine 36 (H3K36) trimethylation. Oncogenic histone H3 alleles, including H3K36M, causally drive cancer formation by dominantly inhibiting SETD2 and other H3K36 methyltransferases. Here we show that expression of the H3K36M oncogene paradoxically suppresses tumor growth in KRAS-driven lung cancer by promoting transcription of repetitive elements and accumulation of cytoplasmic dsRNA, activating antiviral-like immune clearance. Unexpectedly, all attendant effects of H3K36M expression require SETD2 activity despite wid
