Different alpha-adrenoreceptors in the central nervous system mediating biochemical and functional effects of clonidine and receptor blocking agents.

The influence of clonidine on alpha-adrenoreceptors in the central nervous system of rats and mice has been investigated. Both functional events due to postsynaptic receptor stimulation (flexor reflex activity, motor activity) and biochemical changes have been considered. 1. Clonidine was less potent in stimulating the hindlimb flexor reflex activity of spinal rats than in inhibiting the alpha-methyltyrosine-induced disappearance of noradrenaline in the spinal cord and in the whole brain of rats. 2. The increase in flexor reflex activity due to clonidine (0.4 mg/kg) was virtually completely in

The effects of phenylpropanolamine and other sympathomimetics on food consumption and motor activity in mice.

The effects of phenylpropanolamine on motor activity and on food intake were compared with those of S-amphetamine, ephedrine, 2-aminoindane and fenfluramine in groups of mice. Motor activity was additionally measured in mice pretreated with levodopa and benserazide, and food intake in mice pretreated with alpha-methyl-p-tyrosine. Amphetamine (2.5 mg kg-1) increased motor activity, phenylpropanolamine (10-40 mg kg-1) and 2-aminoindane (2.5-10 mg kg-1) decreased activity whilst ephedrine (2.5-40 mg kg-1) had a biphasic effect. Fenfluramine (10-40 mg kg-1) had negligible effect on activity. In mi

On the prolactin-inhibiting effect of neurotensin. The role of dopamine.

Neurotensin (NT) when injected in a dose of 5 micrograms into the third cerebral ventricle of conscious, unrestrained male rats, outfitted with chronic third ventricle and jugular cannulae, decreased the resting prolactin (PRL) levels. NT (5 micrograms) also markedly inhibited the PRL-releasing effect of activation of central serotonin receptors by 5-hydroxytryptophan (5HTP, 15 mg/kg) in animals pretreated with fluoxetine (4 mg/kg). however, interruption of dopamine (DA) neurotransmission by either alpha-methyl-p-tyrosine (alpha MT, 250 mg/kg) or spiroperidol (0.3 mg/kg) led to a blockade of t

Increased 5-HT2 receptor number in brain as a probable explanation for the enhanced 5-hydroxytryptamine-mediated behaviour following repeated electroconvulsive shock administration to rats.

Following electroconvulsive shock (ECS) administration daily for 10 days there was an increase (35%) in 5-hydroxytryptamine2 (5-HT2) receptor number in rat frontal cortex 24 h after the last ECS, compared with handled controls. A similar increase was seen after intermittent ECS administration (5 ECS over 10 days) given during halothane anaesthesia, compared with halothane-treated controls. The dissociation constant was also increased at this time. A single ECS had no effect. Treatment of rats with pentylenetetrazol, p-chlorophenylalanine or alpha-methyl p-tyrosine during the intermittent ECS a

Compulsive gnawing in rats after implantation of drugs into the ventral thalamus. A contribution to the mechanism of morphine action.

1 Implantation of morphine into various parts of the corpus striatum of rats evokes only weak gnawing responses.2 Deposition of apomorphine, morphine or methadone in the region of the nucleus ventralis thalami produces a biphasic response, i.e. general excitation, followed by a period of intense gnawing.3 The effect of both apomorphine and morphine is blocked by chlorpromazine, haloperidol and pimozide. However, pretreatment with alpha-methyltyrosine methyl ester or alpha-methyldopa prevents only the gnawing response to morphine, but not to apomorphine.4 Systemic nalorphine, morphine or pethid

Gastric ulcer formation in cold-stressed mice related to a central calcium-dependent-dopamine synthesizing system.

Stress ulceration of the stomach in mice was investigated from the aspect of the calcium/calmodulin-dependent dopamine synthesizing system in the brain. Cold stress was induced in mice by restraining them at 4 degrees C. Serum and brain calcium levels were increased by cold stress, and an increased brain calcium level was found to enhance dopamine synthesis and a successively increased brain dopamine level induced gastric ulcer formation. Development of gastric ulcers elicited by cold stress was significantly decreased by i.p. pretreatment with EDTA (1 micromol/mouse, 1 h before restraint) or

Effect of dexamethasone on plasma free dopamine: dopaminergic modulation in hypertensive patients.

To investigate the peripheral dopaminergic modulation in the pathogenesis of human hypertension, we examined the responses of plasma free dopamine (DA) to dexamethasone (Dx) administration, which is suggested to activate dopaminergic activity. We administered Dx 2 mg intravenously to patients with primary aldosteronism (PA), essential hypertension (EH), and normotensive controls (NT). Plasma free DA was increased significantly in all groups and the responses were more remarkable in PA than in EH and NT. Plasma epinephrine (E) showed a gradual increase while plasma norepinephrine (NE) tended to

Noradrenergic involvement in pinealectomy induced convulsions in gerbils.

Gerbils treated with various noradrenergic compounds were pinealectomized (PINX) or sham-operated (SHAM) and observed for convulsive activity. Although declines in cortical and hypothalamic norepinephrine content were found after alpha-methylparatyrosine (alpha MPT) or PINX, treatment with alpha MPT did not produce convulsions in SHAM gerbils or in gerbils which were PINX one week earlier. Likewise, alpha MPT did not increase the number or severity of convulsions observed in acutely PINX gerbils. Treatment with propranolol or isoproterenol did not have any effect on convulsive activity of PINX

Catecholamine-secreting infratemporal fossa paraganglioma.

Extraadrenal paragangliomas are tumors of the paraganglion system, usually arising from the carotid bodies, the glomus jugulare, or the glomus tympanicum. These tumors are capable of secreting catecholamines which can cause severe hypertensive crises. This paper reports a case of a patient who suffered a cerebral vascular accident due to hypertension resulting from a catecholamine-secreting infratemporal fossa paraganglioma. Any patient with a history of paroxysmal hypertension, headaches, and palpitations should be evaluated for a catecholamine-secreting tumor. Diagnosis and management of the

Prolactin and TSH responses to both domperidone and TRH in normal and hyperprolactinaemic women after dopamine synthesis blockade.

A study of the effect of alpha-methyl-1-tryosine (metyrosine) blockade (2 g/d for 2 d) of dopamine (DA) synthesis on the PRL and TSH response to domperidone (DOM) and TRH in normal women and subjects with pathological hyperprolactinaemia is reported. In the normal subjects, there was a marked increase in basal PRL (51.7 +/- 11.1 vs 5.7 +/- 1.0 ng/ml) and the PRL and TSH responses to DOM were abolished. The PRL response to TRH was also reduced. In the hyperprolactinaemic subjects, metyrosine had no effect on basal PRL nor on the virtually non-existent PRL response to DOM, whereas it abolished t

In vivo inhibition of dopamine-beta-hydroxylase in rat adrenals during exposure to carbon disulphide.

Male rats were exposed for a maximum of 4 h to carbon disulphide at atmospheric levels of 1.0-4.0 mg/l and the turnover rates of adrenal dopamine was determined by injecting alpha-methyl-p-tyrosine and measuring the rate at which dopamine disappears. Although the level of exposure was significantly higher than the 30.0 micrograms/l permissible limit, or the average occupational exposure, similar or even higher peak exposure values were reported from the viscose rayon industry. After inhibition of tyrosine hydroxylase by alpha-methyl-p-tyrosine, adrenal dopamine contents declined at a slower ra

Brain catecholamine metabolism changes and hypothermia in intoxication by anticholinesterase agents.

Sublethal doses of physostigmine, paraoxon and soman induce a short-lasting fall in rat core temperature potentiated by alpha-methyl-para-tyrosine (alpha-MT) (early effects). When the own hypothermic effect of the anticholinesterase agent has disappeared (late effects), alpha-MT induces a new decrease in temperature. Parallel biochemical studies of catecholamine levels and turnover were performed in several brain areas. The norepinephrine (NE) turnover is generally increased particularly in the hypothalamus, suggesting that NE hypothalamic changes might be linked to a latent perturbation of th

Inhibition of breathing after stimulation of muscle is mediated by endogenous opiates and GABA.

Stimulation of receptors in hindlimb muscles activates a supraspinal mechanism which causes a prolonged, post-stimulatory depression of breathing (Waldrop, T.G., F.L. Eldridge and D.E. Millhorn, 1982, Respir. Physiol. 50: 239-254). In the present study phrenic nerve responses to stimulation of hindlimb muscles were studied in anesthetized, paralyzed cats whose vagi and carotid sinus nerves had been cut. Body temperature and end-tidal PCO2 were kept constant with servocontrollers. The post-stimulatory depression of breathing was greatly attenuated in cats pretreated with an opiate antagonist (n

Contributions of alpha-adrenoreceptor blockade to extrapyramidal effects of neuroleptic drugs.

Prazosin, phenoxybenzamine and clozapine, but not sulpiride, abolished the increase in flexor reflex activity induced by clonidine and they accelerated the noradrenaline utilization in the brain of rats. These findings indicate that the first three drugs block central alpha-adrenoreceptors. The alpha-methyltyrosine-induced disappearance of dopamine in the corpus striatum and the limbic system was decelerated by prazosin and phenoxybenzamine, was accelerated by sulpiride and was not significantly changed by clozapine. Prazosin and phenoxybenzamine almost completely reversed the sulpiride-induce

Central effects of CGP 37849 and CGP 39551, competitive NMDA receptor antagonists, in mice.

Central effects of CGP 37849 and CGP 39551, competitive NMDA receptor antagonists, were studied in male Albino-Swiss mice. CGP 37849--at high doses only--increased the locomotor activity, while CGP 39551 decreased it. CGP 37849 and CGP 39551 did not change the locomotor activity, in monoamine-depleted mice (treated with reserpine + alpha-methyltyrosine). However, when administered together with clonidine, both those compounds produced a distinct hyperactivity. That antiakinetic effect was antagonized by haloperidol, but not by prazosin or idazoxan. In monoamine-depleted mice both the CGP compo

[An autopsy case of extra-adrenal malignant pheochromocytoma].

An autopsy case of an extra-adrenal malignant pheochromocytoma in a 34-year-old woman is reported. On laparotomy, many advanced stage malignant tumors originating from the paraganglia along the abdominal aorta were found to have invaded the lumbar vertebrae. After a partial resection, Co60 radiation therapy of the paraganglia was instituted, as well as of the metastatic lesions, with little effect. It was found that alpha-methyl-tyrosine was effective in controlling the plasma catecholamine, but had to be discontinued because of an untoward effect (anxiety). The patient subsequently developed

Use of metyrosine in the anaesthetic management of patients with catecholamine-secreting tumours. A case report.

Metyrosine 1.5 g daily for days decreased 24-h urine metanephrine concentration by about 60% in a patient with multiple catecholamine-secreting paragangliomas. Despite the considerable inhibition of catecholamine synthesis, this patient exhibited stress-induced sympathetic overactivity, indicated by increases in arterial pressure and serum catecholamine and urine metanephrine concentrations. It is concluded that metyrosine should be introduced early to the preoperative regimen. In this way, optimal inhibitory effect on catecholamine synthesis can be obtained and maintained for a sufficient tim

Effects of alpha-methyl-para-tyrosine (AMPT) in drug-free depressed patients.

A variety of biologic studies have demonstrated abnormal regulation of the norepinephrine (NE) system in patients with major depression, suggesting a role for NE in the etiology of depression. Brain NE and dopamine levels can be rapidly reduced by blocking synthesis with the tyrosine hydroxylase inhibitor alpha-methyl-para-tyrosine (AMPT). In the current investigation, AMPT was administered to drug-free depressed patients to evaluate the effect on mood of diminished catecholamine levels. Seventeen drug-free patients meeting DSM-III-R criteria for major depressive episode were tested with AMPT

Neuroleptic malignant syndrome caused by dopamine-depleting drugs in a patient with Huntington disease.

Prior reports of neuroleptic malignant syndrome (NMS) concerned patients with psychiatric disorders, usually schizophrenia, who were taking dopamine receptor blocking agents. We report the syndrome in a patient with Huntington disease who was treated with dopamine-depleting agents. He had a negative evaluation for malignant hyperthermia (MH), and we suggest that NMS differs from MH. The occurrence of NMS caused by dopamine-depleting agents suggests that anticholinergic properties of phenotiazines are not the only cause. Central dopaminergic systems probably participate in thermoregulation, and
