The delta2 glutamate receptor: a key molecule controlling synaptic plasticity and structure in Purkinje cells.

The orphan glutamate receptor delta2 (GluRdelta2) is predominantly expressed in cerebellar Purkinje cells and plays a crucial role in cerebellar functions; mice that lack the GluRdelta2 gene display ataxia and impaired motor-related learning tasks. However, when expressed alone or with other glutamate receptors, GluRdelta2 does not form functional glutamate-gated ion channels nor does it bind to glutamate analogs. Therefore, the mechanisms by which GluRdelta2 participates in cerebellar functions have been elusive. Studies of mutant mice, such as lurcher, hotfoot, and GluRdelta2 knockout mice,

A de novo deafwaddler mutation of Pmca2 arising in ES cells and hitchhiking with a targeted modification of the Pparg gene.

We observed severe ataxia in mice homozygous for modification of the Pparg locus. Genetic analysis and nucleotide sequencing revealed that ataxia is caused by a T692K substitution in plasma membrane calcium ATPase 2 (Pmca2), which is tightly linked to Pparg, but not by modified PPARgamma itself. We traced this mutation and found that it arose spontaneously during clonal expansion of the targeted embryonic stem (ES) cells. Consistent with the deafwaddler phenotype in other Pmca2 mutants, homozygous T692K Pmca2 mutants exhibit severe balance disorder, impaired neurologic reflexes, and motor coor

Hydrocephalus with hop gait (hyh): a new mutation on chromosome 7 in the mouse.

Hydrocephalus with hop gait (hyh) is a new lethal recessive mouse mutation that arose in the C57BL/10J strain at The Jackson Laboratory. It has been mapped to the proximal end of Chromosome 7 close to the Gpi-1 locus. This homozygous mutant is characterized clinically by a domed head and a hopping gait observable at 2 weeks of age and death between 4 and 10 weeks of age. The affected mice have dilated lateral ventricles and a large third ventricular cyst, patent through narrowed rostral cerebral aqueduct, cystic caudal aqueduct and no communication of the aqueduct with the fourth ventricle. Reduction of Microvessel Number and Length in the Cerebellum of Purkinje Cell Degeneration Mice.

Degenerative effects of nerve tissues are often accompanied by changes in vascularization. In this regard, knowledge about hereditary cerebellar degeneration is limited. In this study, we compared the vascularity of the individual cerebellar components of 3-month-old wild-type mice (n = 8) and Purkinje cell degeneration (pcd) mutant mice, which represent a model of hereditary cerebellar degeneration (n = 8). Systematic random samples of tissue sections were processed, and laminin was immunostained to visualize microvessels. A computer-assisted stereology system was used to quantify microvessel

The effects of mild closed head injuries on tauopathy and cognitive deficits in rodents: Primary results in wild type and rTg4510 mice, and a systematic review.

In humans, the majority of sustained traumatic brain injuries (TBIs) are classified as 'mild' and most often a result of a closed head injury (CHI). The effects of a non-penetrating CHI are not benign and may lead to chronic pathology and behavioral dysfunction, which could be worsened by repeated head injury. Clinical-neuropathological correlation studies provide evidence that conversion of tau into abnormally phosphorylated proteotoxic intermediates (p-tau) could be part of the pathophysiology triggered by a single TBI and enhanced by repeated TBIs. However, the link between p-tau and CHI in

Separation of activation and pattern in grooming development of weaver mice.

The effects of environmental conditions and age on grooming behavior were examined in weaver mutant mice and control littermates. Due to deficits in both the cerebellum and the dopaminergic system, weaver mice provide an opportunity to investigate how both of these systems are involved in grooming. Although homozygous weaver (wv/wv mice display deficiencies in grooming behavior, our results indicate that these effects are both context and age dependent. Overall wv/wv mice spent less time grooming than did controls. However, during the post-swim period wv/wv, after day 13, reached the grooming

Insights from mouse models into the molecular basis of neurodegeneration.

Thanks largely to cloning the genes for several neurodegenerative diseases over the past decade and the existence of mouse mutants, the molecular basis of neurodegeneration is finally beginning to yield some of its secrets. We discuss what has been learned about the pathogenesis of "triplet repeat" diseases through mouse models for spinocerebellar ataxia types 1 and 3 and Huntington disease, including the roles of nuclear aggregates and protein cleavage. We also discuss the neurologic phenotypes that arise from mutations in neurotransmitter receptors (lurcher mice) and ion channels (weaver, le

Alzheimer disease: mouse models pave the way for therapeutic opportunities.

Research into the molecular mechanisms of Alzheimer disease (AD) continues to clarify important issues in aberrant protein processing while seeking to identify therapeutic targets. Mutations of genes on chromosomes 1, 14 (presenilins 1 and 2), and 21 (the amyloid-beta [Abeta] amyloid precursor protein [APP]) cause the familial forms of AD that often begin before age 65. An allelic polymorphism on chromosome 19 (apolipoprotein E ) affects the age of onset of the more common forms of sporadic AD. Multiple studies in transgenic mice provide strong evidence to support the view that Abeta amyloid f

Deformed optic nerves in coisogenic albino and pigmented C57BL/6J--c2j mice.

The incidence of deformed optic nerve (DON) in C57 BL/6J--c2J mice is similar to that of a pigmented substrain of Long-Evans rats and much less than that of an albino substrain of Sprague Dawley rats. C57BL/6J--c2J albinos have no more DON than do coisogenic black mice. This suggests that albinism is not a factor in DON and that observed DON in Sprague Dawley rats are strain or substrain characteristics.

Lurcher Mouse as a Model of Cerebellar Syndromes.

Cerebellar extinction lesions can manifest themselves with cerebellar motor and cerebellar cognitive affective syndromes. For investigation of the functions of the cerebellum and the pathogenesis of cerebellar diseases, particularly hereditary neurodegenerative cerebellar ataxias, various cerebellar mutant mice are used. The Lurcher mouse is a model of selective olivocerebellar degeneration with early onset and rapid progress. These mice show both motor deficits as well as cognitive and behavioral changes i.e., pathological phenotype in the functional domains affected in cerebellar patients. T

[A morphological study on the brain in wriggle mouse Sagami, a new neurological mutant with dystonic disorders, with special reference to the abnormality of the cerebellum].

The 'Wriggle Mouse Sagami (WMS)' is a new neurological mutation that was discovered in an inbred strain of BALB/c mice at the Ohmura Institute for Laboratory Animals (Address: Zama 228, Kanagawa, Japan) in 1984. The affected characteristics are transmitted by an autosomal recessive gene (wri). The clinical symptoms are characterized by dystonic involuntary movements, such as fine tremors of the forelimbs, hypertonic musculature of the extremities, difficulty in maintaining an upright posture, writhing of the trunk, wriggling of the neck up-and-down and from side to side without any coordinatio

Defective development of the thymus and immunological abnormalities in the neurological mouse mutation "staggerer".

The autosomal recessive mouse mutation "staggerer" (sg/sg), located on chromosome 9, has been recognized as a neurological mutant because of movement abnormalities and defective cerebellar development. We show here that the sg/sg mutation not only affects the development of the cerebellum, but also causes developmental and regulatory changes of the immune system: We observed, on gross inspection, a marked delay in the development of sg/sg thymus, generally enlarged lymph nodes, and undersized spleens. When immunized with SRBC, the sg/sg mouse generated, in normal proportions, helper T-cells in

Expression of mutant huntingtin in mouse brain astrocytes causes age-dependent neurological symptoms.

Huntington disease (HD) is an inherited neurological disorder caused by a polyglutamine expansion in the protein huntingtin and is characterized by selective neurodegeneration that preferentially occurs in striatal medium spiny neurons. Because the medium spiny neurons are innervated abundantly by glutamatergic axons from cortical neurons, the preferential degeneration in the striatal neurons supports the glutamate excitotoxicity theory for HD pathogenesis. Thus, glutamate uptake by glia may be particularly important for preventing glutamate excitotoxicity in HD. Although mutant huntingtin is

Laforin and malin deletions in mice produce similar neurologic impairments.

Lafora disease is a progressive myoclonus epilepsy caused by mutations in the EPM2A gene encoding laforin or in the EPM2B gene encoding malin. It is characterized by the presence of polyglucosan intracellular inclusion bodies (Lafora bodies) in brain and other tissues. Targeted disruption of Epm2a or Epm2b genes in mice produced widespread neuronal degeneration and accumulation of Lafora bodies in neuronal and nonneuronal tissues. Here we analyzed the neurologic alterations produced by disruption of the laforin gene in Epm2a mice and compared them to those in malin-deficient mice. Both Epm2a a

Scrapie agent proteins do not accumulate in grey tremor mice.

The grey tremor mouse is an autosomal recessive mutant characterized by a phenotype of unusual pigmentation, neurological abnormalities and early death. These mice have a spongiform encephalopathy similar to scrapie and Creutzfeldt-Jakob disease. Although the disease is clearly heritable, the grey tremor mouse spongiform pathology has also been transmitted by inoculation of genetically normal mice with diseased brain homogenates. The possibility that a scrapie-like agent is involved has been proposed. We examined brain homogenates from grey tremor mice, scrapie-affected mice and normal mice fo

Pogo: a novel spontaneous ataxic mutant mouse.

The Pogo (pogo/pogo) mouse is a naturally occurring neurological mutant from a Korean wild-type mouse characterized by loss of balance and motor coordination due to dysfunction of the cerebellum. The Pogo mutation is believed to be an allele of P/Q-type calcium channel mutants such as tottering, leaner, and rolling mouse Nagoya. These mutants have been served as mouse models for a group of neurodegenerative diseases. The overall aim of this minireview is to summarize our current understanding of the ataxic Pogo mouse. To address this issue, we first describe the discovery of Pogo mouse and its

[Behavioral effects of ceruletide in Wriggle Mouse Sagami--in comparison with in Rolling Mouse Nagoya].

The behavioral effects of ceruletide in Wriggle Mouse Sagami (WMS) and Rolling Mouse Nagoya (RMN) were observed in an open-field study. Ceruletide decreased the movements of both types of mice, improving the ataxic gait in WMS; however, no such improvement was observed in RMN. There may be different mechanisms underlying the ataxic manifestations in WMS and RMN.

Depressed convulsions by diazepam and its effects on brain monoamines and amino acids in E1 mice.

The effect of diazepam on inbred mutant E1 mice, which develop convulsive seizures after repeated sessions of being tossed up, was examined. Acute administration of diazepam (32 mg/kg, i.p.) completely inhibited the convulsions. At that time, the dopamine level was increased in the cortex and hippocampus, and the norepinephrine level in the cerebellum was decreased. 5-Hydroxytryptamine levels were not changed. As for amino acids, the glutamine level increased and the levels of GABA, glutamic acid, aspartic acid, alanine and other amino acids were not changed.

Effect of Purkinje cell loss on cerebellar gangliosides in nervous mutant mice.

The distribution of cerebellar gangliosides was studied in adult (73 +/- 2 days) nervous (nr/nr) mutant mice which lose 50-90% of their Purkinje cells. This neuronal loss is associated with significant reductions in cerebellar weight and ganglioside concentration. The cerebellar dry weights (mg) and the ganglioside concentrations (microgram N-acetylneuraminic acid per 100 mg dry weight) in nr/nr mice and age-matched normal littermates (+/?) are 7.4 +/- 0.3 mg and 13.2 +/- 0.4 mg; and 411.7 +/- 4.8 micrograms and 438.5 +/- 2.1 micrograms, respectively.
