Quaking Regulates Neurofascin 155 Expression for Myelin and Axoglial Junction Maintenance.

UNLABELLED: RNA binding proteins required for the maintenance of myelin and axoglial junctions are unknown. Herein, we report that deletion of the Quaking (QKI) RNA binding proteins in oligodendrocytes (OLs) using Olig2-Cre results in mice displaying rapid tremors at postnatal day 10, followed by death at postnatal week 3. Extensive CNS hypomyelination was observed as a result of OL differentiation defects during development. The QKI proteins were also required for adult myelin maintenance, because their ablation using PLP-CreERT resulted in hindlimb paralysis with immobility at ∼30 d after 4-

Copper in fetal and neonatal development.

The essentially of copper for normal fetal and neonatal development has been well documented, although copper metabolism during this period is poorly understood. The dietary requirement for copper is influenced by genetic background. The neurological phenotypic characteristics of the mutant gene quaking (qk) in mice resemble in part those of copper-deficient animals. Supplementation of the maternal diet with copper during pregnancy and lactation, or during lactation alone, greatly reduced the frequency of tremors characteristic of these mutants, and brought the otherwise low copper concentrati

The fetal hydantoin syndrome: answers from a mouse model.

In spite of a large body of literature suggesting that prenatal exposure to hydantoin anticonvulsant drugs can be teratogenic in humans, resulting in offspring with congenital malformations, the wide phenotypic variability of this syndrome has led many clinicians to question its very existence. Because solutions to major teratologic questions may be impossible to find within the constraints of human clinical or epidemiologic investigations, we have developed an animal model for the fetal hydantoin syndrome. With this model, we have been able to demonstrate the dose-related teratogenic effects

QKI regulates adipose tissue metabolism by acting as a brake on thermogenesis and promoting obesity.

Adipose tissue controls numerous physiological processes, and its dysfunction has a causative role in the development of systemic metabolic disorders. The role of posttranscriptional regulation in adipose metabolism has yet to be fully understood. Here, we show that the RNA-binding protein quaking (QKI) plays an important role in controlling metabolic homeostasis of the adipose tissue. QKI-deficient mice are resistant to high-fat-diet (HFD)-induced obesity. Additionally, QKI depletion increased brown fat energy dissipation and browning of subcutaneous white fat. Adipose tissue-specific depleti

The Resistin/TLR4/miR-155-5p axis: a novel signaling pathway in the onset of hypothalamic neuroinflammation.

Hypothalamic neuroinflammation plays a pivotal role in the development of metabolic disorders, contributing to obesity and insulin resistance. Hypercaloric diets rich, particularly high-fat diets (HFDs) induce hypothalamic neuroinflammation, which has been shown to precede peripheral inflammation, even after short-term exposure. However, the mechanisms initiating this central inflammatory response, especially the mediators involved, remain incompletely understood. In this study, we demonstrate that HFD consumption induces the expression of resistin in the hypothalamus. Resistin, an adipokine k

Comparison of conventional, amplification and bio-assay detection methods for a chronic wasting disease inoculum pool.

Longitudinal studies of chronic wasting disease (CWD) in the native host have provided considerable understanding of how this prion disease continues to efficiently spread among cervid species. These studies entail great cost in animal, time and financial support. A variety of methods have emerged including transgenic mouse bioassay, western blot, enzyme-linked immunoassay (ELISA), immunohistochemistry (IHC), serial protein misfolding cyclic amplification (sPMCA) and real time quaking-induced conversion (RT-QuIC), that deepen our understanding of this and other protein misfolding disorders. Recombinant prion protein vaccination of transgenic elk PrP mice and reindeer overcomes self-tolerance and protects mice against chronic wasting disease.

Chronic wasting disease (CWD) is a fatal neurodegenerative disease that affects cervids in North America and now Europe. No effective measures are available to control CWD. We hypothesized that active vaccination with homologous and aggregation-prone recombinant prion protein (PrP) could overcome self-tolerance and induce autoantibody production against the cellular isoform of PrP (PrPC), which would be protective against CWD infection from peripheral routes. Five groups of transgenic mice expressing elk PrP (TgElk) were vaccinated with either the adjuvant CpG alone or one of four recombinant

LncRNA Airn preserves cardiomyocyte fatty acid oxidation to ameliorate diabetic cardiomyopathy by modulating the QKI-PPARα axis.

Diabetic cardiomyopathy (DbCM) is a major complication of diabetes characterized by metabolic dysregulation in the heart. This study investigated the role of the long non-coding RNA Airn in the regulation of cardiac fatty acid metabolism during DbCM. Airn expression was markedly reduced in the hearts of diabetic mice. Cardiomyocyte-specific Airn overexpression improved cardiac structure and function, enhanced fatty acid oxidation (FAO), and reduced myocardial lipid accumulation, whereas Airn knockdown induced cardiac remodeling and dysfunction even under non-diabetic conditions. Long noncoding RNA MIR100HG regulates inflammatory response by interacting with RNA-binding protein Quaking in periodontitis.

BACKGROUND: Long noncoding RNAs (lncRNAs) are emerging regulators of periodontal inflammation. This study investigates the role of lncRNA MIR100HG in periodontitis and its interaction with the RNA-binding protein Quaking (QKI). METHODS: In vitro, human gingival fibroblasts (HGFs) were stimulated with Porphyromonas gingivalis lipopolysaccharide (Pg.LPS, 200 ng/mL, 24 h). In vivo, ligature-induced periodontitis was established in mice. RNA sequencing, RNA immunoprecipitation (RIP), fluorescence in situ hybridization (FISH), and Western blotting were employed to dissect molecular mechanisms. RE

The m7G methyltransferase METTL1 promotes acute kidney inflammation by disrupting mitochondrial function.

INTRODUCTION: RNA modifications regulate the progression of inflammatory diseases. However, the role of N7-methylguanosine (m7G) modification and its regulatory enzyme, methyltransferase-like 1 (METTL1), in inflammatory kidney diseases remains poorly understood. Here, we aimed to investigate the function and underlying mechanisms of METTL1-mediated m7G modification in acute kidney injury (AKI). METHODS: Kidney tubular epithelial cell (TEC)-specific METTL1 conditional knockout mice were used to evaluate the functional role of METTL1 in multiple AKI mouse models (cisplatin, ischemia/reperfusion

Novel Insights into circRNA Saga Coming from Spermatozoa and Epididymis of HFD Mice.

Obesity is a pathophysiological disorder associated with adiposity accumulation, oxidative stress, and chronic inflammation state that is progressively increasing in younger population worldwide, negatively affecting male reproductive skills. An emerging topic in the field of male reproduction is circRNAs, covalently closed RNA molecules produced by backsplicing, actively involved in a successful spermatogenesis and in establishing high-quality sperm parameters. However, a direct correlation between obesity and impaired circRNA cargo in spermatozoa (SPZ) remains unclear. In the current work, u

Prion seeding activity and infectivity in skin samples from patients with sporadic Creutzfeldt-Jakob disease.

Sporadic Creutzfeldt-Jakob disease (sCJD), the most common human prion disease, is transmissible through iatrogenic routes due to abundant infectious prions [misfolded forms of the prion protein (PrPSc)] in the central nervous system (CNS). Some epidemiological studies have associated sCJD risk with non-CNS surgeries. We explored the potential prion seeding activity and infectivity of skin from sCJD patients. Autopsy or biopsy skin samples from 38 patients [21 sCJD, 2 variant CJD (vCJD), and 15 non-CJD] were analyzed by Western blotting and real-time quaking-induced conversion (RT-QuIC) for Pr

Genetic diseases of copper metabolism.

There are several known examples of mutations which influence copper homeostasis in humans and animals. Pleiotropic effects are observed when the mutant gene disturbs copper flux. In some cases, the mutation alters the level of a specific copper ligand (enzyme) and the clinical consequences are unique. The two most widely studied genetic maladies in humans are Menkes' and Wilson's diseases. Menkes' disease is an X-linked fatal disorder in which copper accumulates in some organs (intestine and kidney) and is low in others (liver and brain). Wilson's disease is an autosomal recessive disorder in

RNA-editing enzyme ADAR1 attenuates rheumatoid arthritis via regulating fibroblast-like synoviocytes-derived exosomal circFTO.

Rheumatoid arthritis (RA) is a chronic debilitating autoimmune disorder and a predominant cause of potentially treatable functional disability. Adenosine deaminase acting on RNA 1 (ADAR1), as an RNA-editing enzyme, can regulate circular RNAs (circRNAs) and fibroblast-like synoviocytes (FLSs)-derived exosomal circFTO has been proposed as a contributor to RA. This study was dedicated to elaborate the role of ADAR1 in RA and the association between ADAR1 and circFTO. RT-qPCR examined circFTO, fat mass and obesity-associated protein (FTO), DExH-box helicase 9 (DHX9), ADAR1 and Quaking (QKI) expres

The quakingviable mutation affects qkI mRNA expression specifically in myelin-producing cells of the nervous system.

The genetic lesion in the quakingviable (qk(v)) mutant mice is a deletion 5' to the qkI gene, resulting in severe hypomyelination. qkI produces several QKI protein isoforms via alternative splicing of the C-terminal coding exons. In the qk(v)/qk(v) brain, immunostaining of QKI proteins is diminished in an isoform-differential manner with undefined mechanisms. We examined the expression of QKI protein isoforms and qkI mRNA isoforms in the qk(v)/qk(v) mutants and the non-phenotypic wt/qk(v) littermates. Our results indicated significant reduction of all qkI mRNA isoforms in the central and perip

Bank Vole Prion Protein As an Apparently Universal Substrate for RT-QuIC-Based Detection and Discrimination of Prion Strains.

Prions propagate as multiple strains in a wide variety of mammalian species. The detection of all such strains by a single ultrasensitive assay such as Real Time Quaking-induced Conversion (RT-QuIC) would facilitate prion disease diagnosis, surveillance and research. Previous studies have shown that bank voles, and transgenic mice expressing bank vole prion protein, are susceptible to most, if not all, types of prions. Here we show that bacterially expressed recombinant bank vole prion protein (residues 23-230) is an effective substrate for the sensitive RT-QuIC detection of all of the differe

Reaching for the stars: Linking RNA binding proteins to diseases.

The prototype STAR (Signal Transduction and Activation of RNA) protein is Sam68, the Src-associated substrate during mitosis of 68 kDa. Sam68, like all other STAR proteins, belongs to the large class of heteronuclear ribonucleoprotein particle K (hnRNP K) homology (KH) domain family of RNA-binding proteins. The KH domain is an evolutionarily conserved RNA binding domain that consists of 70-100 amino acids. The KH domain is one of the most prevalent RNA binding domains that directly contacts single-stranded RNA with a signature topology. Sam68 contains a single KH domain that harbors additional

Mutations in humans and animals which affect copper metabolism.

Various inherited disorders of copper metabolism in man and animals are reviewed. Emphasis is placed on the use of cultured cells from mutants to determine the primary molecular defects and to acquire basic knowledge of normal copper metabolism. This allows better diagnostic tests and possible treatment of the disorders. Menkes' disease in humans and mottled mouse mutants are discussed in detail, as they illustrate these approaches.

FUS driven circCNOT6L biogenesis in mouse and human spermatozoa supports zygote development.

Circular RNA (circRNA) biogenesis requires a backsplicing reaction, promoted by inverted repeats in cis-flanking sequences and trans factors, such as RNA-binding proteins (RBPs). Among these, FUS plays a key role. During spermatogenesis and sperm maturation along the epididymis such a molecular mechanism has been poorly explored. With this in mind, we chose circCNOT6L as a study case and wild-type (WT) as well as cannabinoid receptor type-1 knock-out (Cb1-/-) male mice as animal models to analyze backsplicing mechanisms. Our results suggest that spermatozoa (SPZ) have an endogenous skill to ci
