Degradation of sulfonamide antibiotics by Microbacterium sp. strain BR1 - elucidating the downstream pathway.

Microbacterium sp. strain BR1 is among the first bacterial isolates which were proven to degrade sulfonamide antibiotics. The degradation is initiated by an ipso-substitution, initiating the decay of the molecule into sulfur dioxide, the substrate specific heterocyclic moiety as a stable metabolite and benzoquinone imine. The latter appears to be instantaneously reduced to p-aminophenol, as that in turn was detected as the first stable intermediate. This study investigated the downstream pathway of sulfonamide antibiotics by testing the strain's ability to degrade suspected intermediates of th

Oral ingestion of Microbacterium nematophilum leads to anal-region infection in Caenorhabditis elegans.

Microbacterium nematophilum is a gram positive bacterium that colonizes the Caenorhabditis elegans rectal region causing swelling and constipation. This interaction has been exploited as a model system to identify and study genes important in host-pathogen interactions and innate immunity. During attempts to inhibit the host-pathogen interaction, it became important to clarify the route of infection. Using bacteria labeled with the fluorescent dye Cy3, we show that infection is via the oral route only and that infection follows a clear pattern of ingestion, plug formation, and bump development

The strategy of transesterification and degradation improved the resistance of Microbacterium sp. S1037 to antiseptic parabens.

Microorganisms are constantly evolving to resist the toxicity of artificial antibiotics and preservatives released into the environment. Parabens, a group of alkyl and aryl esters of p-hydroxybenzoate, are a class of widely used antimicrobial preservatives, particularly effective against Gram-positive bacteria and fungi. Previous studies have reported the degradation of parabens by numerous Gram-negative strains and activated sludge, with degradation processes often involving transesterification. However, the physiological significance of transesterification has received little attention. Here

Isolation and characterization of efficient isoxaben-transforming Microbacterium sp strains from four European soils.

Nutrient-agar plates containing isoxaben (500 mg litre(-1)) were used to isolate isoxaben-metabolising bacteria from four European soils incubated with the herbicide under laboratory conditions. In flask experiments, inoculation of a basal salts medium containing nitrogen and [phenyl-U-14C]isoxaben with an isolate (B2b) resulted in 33% recovery of the initial radioactivity as [14C]carbon dioxide after 2 weeks. A major metabolite identified by GC-MS and NMR analysis as 3-(1-ethyl-1-methylpropyl)isoxazol-5-ylamine accumulated both in basal salts and nutrient broth media. 2,6-Dimethoxybenzoic aci

Bacteraemia due to Microbacterium paraoxydans in a patient with chronic kidney disease, refractory hypertension and sarcoidosis.

INTRODUCTION: Microbacterium spp. are yellow-pigmented Gram-positive coryneform rods found in various environmental sources, such as soil and water samples. They rarely cause human infection, mostly infecting immunocompromised patients and catheter insertion sites, making them challenging to identify in clinical settings. CASE PRESENTATION: We report a case of a 61-year-old female on long-term prednisone therapy for sarcoidosis with minimal exposure to environmental sources, who presented with an overtly infected Hickman catheter site and presyncope. The patient had a central venous catheter

Acceleration of perchloroethylene dechlorination by extracellular secretions from Microbacterium in a mixed culture containing Desulfitobacterium.

The study was conducted to demonstrate the influence of extracellular secretions from Microbacterium on the reductive dechlorination of tetrachloroethene (PCE). A series of mixed cultures were established from a paddy soil sample. In the mixed cultures amended with extracellular secretions from Microbacterium, PCE was rapidly and completely converted into cis-1,2-dichloroethene (cis-DCE) and trans-1,2-dichloroethene (trans-DCE) within 40 days. The unamended mixed cultures showed weak signs of dechlorination after a pronounced lag phase, and trichloroethene (TCE) was accumulated as a major end

Hydrolysis of fenamiphos and its toxic oxidation products by Microbacterium sp. in pure culture and groundwater.

A bacterium with an exceptional ability to hydrolyse fenamiphos and its toxic oxidation products fenamiphos sulfoxide and fenamiphos sulfone, all possessing POC bond was isolated from soil. Based on 16S rRNA gene determination, this bacterium was putatively identified as Microbacterium esteraromaticum. The phenols (fenamiphos phenol, sulfoxide phenol and sulfone phenol) formed during bacterial hydrolysis resisted further degradation in mineral salts medium and sterile groundwater, but were transitory in non-sterile groundwater due to the catabolism of native microorganisms. Also, the cell-free

Bioprospecting of the novel isolate Microbacterium proteolyticum LA2(R) from the rhizosphere of Rauwolfia serpentina.

The study aimed to assess the proficiency of secondary metabolites (SMs) synthesized by actinobacteria isolated from the rhizospheric soil of Rauwolfia serpentina for its antimicrobial and anti-biofilm activity. After morphological and biochemical identification of actinobacteria, primary and secondary screening was done for specific metabolite production. The secondary metabolites were then tested for their antioxidant, antibacterial, and antibiofilm potential. Out of 29 bacterial colonies isolated, only one emerged as a novel isolate, Microbacterium LA2(R). Partial 16S rRNA gene sequence of

Isolation and Aflatoxin B1-Degradation Characteristics of a Microbacterium proteolyticum B204 Strain from Bovine Faeces.

Aflatoxin B1 (AFB1) is one of the most harmful mycotoxins, raising serious global health and economic problems. Searching for biological approaches for effective and safe AFB1 degradation is imminent. In our study, Microbacterium proteolyticum B204 isolated from bovine faeces degraded 77% of AFB1 after 24 h, becoming the first reported bacteria from the Microbacterium family to possess AFB1 degradation characteristics. Temperature variation showed little effect on its degradation ratio, demonstrating high thermostability of 75% and 79% after boiling and sterilization, respectively. We suppose

Structural insights into the γ-lactamase activity and substrate enantioselectivity of an isochorismatase-like hydrolase from Microbacterium hydrocarbonoxydans.

(+)-γ-lactamase catalyzes the specific hydrolysis of (+)-γ-lactam out of the racemic γ-lactam (2-Azabicyclo[2.2.1]hept-5-en-3-one) to leave optically pure (-)-γ-lactam, which is the key building block of antiviral drugs such as carbovir and abacavir. However, no structural data has been reported on how the enzymes bind the γ-lactams and achieve their enantioselectivities. We previously identified an isochorismatase-like hydrolase (IHL, Mh33H4-5540) with (+)-γ-lactamase activity, which constitutes a novel family of γ-lactamase. Here, we first discovered that this enzyme actually hydrolyzed both

Mos1 mutagenesis reveals a diversity of mechanisms affecting response of Caenorhabditis elegans to the bacterial pathogen Microbacterium nematophilum.

A specific host-pathogen interaction exists between Caenorhabditis elegans and the gram-positive bacterium Microbacterium nematophilum. This bacterium is able to colonize the rectum of susceptible worms and induces a defensive tail-swelling response in the host. Previous mutant screens have identified multiple loci that affect this interaction. Some of these loci correspond to known genes, but many bus genes [those with a bacterially unswollen (Bus) mutant phenotype] have yet to be cloned. We employed Mos1 transposon mutagenesis as a means of more rapidly cloning bus genes and identifying new

Isolation and characterization of multi-drug-resistant Microbacterium esteraromaticum bacteriophage: assessment of antibacterial efficacy and genomic insights.

Microbacterium esteraromaticum, a common bacterium utilized in the degradation of organic pollutants, is prevalently found in the wastewater environments of rural areas. However, the excessive use of antibiotics in recent years has endowed M. esteraromaticum with a broad spectrum of antibiotic resistance, transforming it into a potential high-risk contaminant capable of disseminating antibiotic resistance genes (ARGs) within the environment. Lytic bacteriophages, due to their characteristic ability to lysogenize and specifically target host bacteria, have emerged as potent biocontrol agents. I

Dual mechanisms of Microbacterium algeriense C14 in alleviating cadmium toxicity during Zinnia elegans seed germination and early seedling growth.

Cadmium (Cd) contamination, a critical type of heavy metal pollution, exerts severe adverse effects on plant growth and poses substantial risks to food safety. Although plant growth-promoting rhizobacteria (PGPR) have demonstrated great potential in mitigating metal-induced stress in plants, a critical limitation remains: most PGPR strains only provide protective effects to plants, failing to reduce the bioavailability of toxic metals in the environment. We herein report the isolation and characterization of Microbacterium algeriense strain C14, which exhibits dual functionality: indole-3-acet

Predictive microbiology and transcriptomic analysis of heat-resistant and spoilage-associated traits in Microbacterium species isolated from dairy products.

Microbacterium spp. are frequently isolated from dairy products and are implicated in spoilage. In this research, we studied the growth and thermal resistance of two strains, Microbacterium oxydans and Microbacterium laevaniformans. Their growth behaviors were analyzed at 4, 10, 15, and 28 °C using the Baranyi & Roberts and Ratkowsky models. All temperatures supported growth, with the time for a 1 log increase ranging from 9.17 h to 71.42 h, indicating these spoilage-associated strains can proliferate across a broad temperature range. Furthermore, both strains were confirmed as protease-produc
