High-pressure homogenization (HPH) is a non-thermal technology widely studied to replace, partially or in total, the conventional thermal preservation processes used in the food industry, thus minimizing undesirable changes in the nutritional and sensory characteristics of liquid products. The main effect of HPH is the size reduction of dispersed particles thus affecting physical stability of the products, despite also inactivating microorganisms, preserving bioactive compounds, and maintaining sensory characteristics. During the process, the fluid is driven under high-pressure through a micro

The pathogen Listeria monocytogenes causes listeriosis, a severe foodborne disease associated with high mortality. Rapid and sensitive methods are required for specific detection of this pathogen during food production. Bioluminescence-based reporter bacteriophages are genetically engineered viruses that infect their host cells with high specificity and transduce a heterologous luciferase gene whose activity can be detected with high sensitivity to indicate the presence of viable target cells. Here, we use synthetic biology for de novo genome assembly and activation as well as CRISPR-Cas-assis

Many studies report that copper can be used to control microbial growth, including that of viruses. We determined the rates of copper-mediated inactivation for a wide range of bacteriophages. We used two methods to test the effect of copper on bacteriophage survival. One method involved placing small volumes of bacteriophage lysate on copper and stainless steel coupons. Following exposure, metal coupons were rinsed with lysogeny broth, and the resulting fluid was serially diluted and plated on agar with the corresponding bacterial host. The second method involved adding copper sulfate (CuSO(4)

Burkholderia pseudomallei, a gram-negative bacillus found in soil and water, is the causative agent of melioidosis. It can produce a biofilm, which increases resistance to antibacterial agents. Bacteriophages (phages) have been suggested as alternative antibacterial agents. In this study, the ability of six phages (family Myoviridae) to lyse B. pseudomallei isolates was examined using a microplate phage virulence assay. The six phages were more efficient in lysing soil than clinical B. pseudomallei isolates. Phage ST79 had the highest lytic capability, independent of inoculating phage quantity

Helicobacter pylori is globally recognized for its role in chronic gastritis, peptic ulcer disease, and gastric cancer. Yet despite decades of research and standardized eradication protocols, treatment failures and disease recurrence remain frustratingly common. While antibiotic resistance has been a central focus, emerging data suggest that H. pylori employ additional, underappreciated survival strategies that extend its pathogenic potential beyond the stomach. This review redefines H. pylori as a versatile pathogen capable of persisting through underappreciated survival strategies: the cocco

BACKGROUND: Bacteria are known to live inside architectural structures called biofilms. Though standard biofilms have been studied extensively for more than 50 years, little is known about dry-surface biofilms (DSBs). Since 2012, DSBs have been described in several scientific papers, but basic knowledge about the viability and culturability of bacteria remains limited.
AIM: To conduct a systematic review to determine whether bacteria inside DSBs are viable, culturable, and enumerable.
METHODS: Eligible articles had to deal with DSBs containing at least one bacterial species involved in healt

Rabies virus is a notifiable pathogen that must be handled in high containment facilities where national and international guidelines apply. For the effective inactivation of rabies virus, a number of reagents were tested. Virkon S (1%) solution caused more than 4log reduction of rabies virus in culture medium supplemented with 10% foetal calf serum within 1min. Isopropyl alcohol (70%) treatment resulted in >3log reduction of rabies virus within 20s when applied at a ratio of 19:1, making it a suitable agent for surface decontamination whereas 70% ethanol was ineffective. Rabies virus (from 10

OBJECTIVES: Water mist is usually generated using equipment directly connected to the water tap, as its installation is relatively easy. However, there is no legal regulation regarding the maintenance of this equipment, and the quality of the mist has not been sufficiently well investigated. In this study, we sought to establish methods that allow the hygienic maintenance of this equipment.
METHODS: We monitored the use of the mist generating equipment in five of the 61 institutions in the jurisdiction of Ichinomiya Health Center, examined the resulting water quality, and tested for Legionell

PCR is effective in detecting bacterial DNA in samples, but it is unable to differentiate viable bacteria from inactivated cells or free DNA fragments. New PCR-based analytical strategies have been developed to address this limitation. Molecular viability testing (MVT) correlates bacterial viability with the ability to rapidly synthesize species-specific rRNA precursors (pre-rRNA) in response to brief nutritional stimulation. Previous studies demonstrated that MVT can assess bacterial inactivation by chlorine, serum, and low-temperature pasteurization. Here, we demonstrate that MVT can detect

Polymicrobial urinary tract infections (UTIs) pose significant treatment challenges due to interspecies interactions within biofilms. To address this, we developed a static, triple-species biofilm model comprising Escherichia coli, Enterococcus faecalis, and Candida albicans to simulate recurrent UTI communities. Biofilms were treated with ampicillin, ciprofloxacin, fluconazole, ethylenediaminetetraacetic acid (EDTA), and N-acetylcysteine (NAC), and evaluated for biomass (crystal violet assay) and viability (CFU/mL). Ampicillin and ciprofloxacin reduced bacterial viability by > 90%, but had li

UNLABELLED: Botrytis cinerea is a significant postharvest fungal pathogen responsible for substantial losses in a variety of fresh produce. Ultraviolet-C (UV-C) irradiation is a potential intervention that could reduce postharvest losses due to fungal disease. However, the diversity of fungal reproductive forms and lack of standardization in antifungal assessment methods complicate the evaluation of UV-C efficacy. This study evaluated the in vitro antifungal efficacy of UV-C against B. cinerea conidia and hyphal fragments (HF) treated in liquid suspension and on agar surfaces. B. cinerea viabi

The role of curli, amyloid extracellular fibers, in the tolerance of Escherichia coli PHL628 to Hg(II) was examined. Our findings indicate that by sorbing Hg(II) extracellularly, curli protect the cells. To our knowledge, this is the first time a protective role of curli against toxic metals has been demonstrated.

Microorganisms are predominantly organized in biofilms, where cells live in dense communities and are more resistant to external stresses than are their planktonic counterparts. With in vitro experiments, the susceptibility of Candida albicans biofilms to a nonthermal plasma treatment (plasma source, kINPen09) in terms of growth, survival, and cell viability was investigated. C. albicans strain SC5314 (ATCC MYA-2876) was plasma treated for different time periods (30 s, 60 s, 120 s, 180 s, 300 s). The results of the experiments, encompassing CFU, fluorescence Live/Dead, and 2,3-bis-(2-methoxy-4

Oral microbial communities are extremely complex biofilms with high numbers of bacterial species interacting with each other (and the host) to maintain homeostasis of the system. Disturbance in the oral microbiome homeostasis can lead to either caries or periodontitis, two of the most common human diseases. Periodontitis is a polymicrobial disease caused by the coordinated action of a complex microbial community, which results in inflammation of tissues that support the teeth. It is the most common cause of tooth loss among adults in the United States, and recent studies have suggested that it

Streptococcus pneumoniae (SP) and nontypeable Haemophilus influenzae (NTHi) are common commensals of the human airway and major bacterial pathogens of otitis media (OM) and other upper airway infections. The interaction between them may play an important role in the pathogenesis of polymicrobial infections. Although previous studies suggested NTHi could promote pneumococcal survival and biofilm formation, how NTHi affects pneumococcal activities has not been defined. Our data in the present studies indicated that the outcome of the interaction between SP and NTHi was in a cell-density-dependen
