From midsummer to fall, cyanobacterial harmful algal blooms (cHABs) producing microcystins (MCs), a class of hepatotoxic cyclic heptapeptides, plague Lake Erie's western basin. Since MC congener toxicities vary over 2 orders of magnitude, identification and characterization of the microcystin congener profile can be crucial information when balancing between public health and local economic resilience. Herein, five hydroxyproline (Hyp)-containing MCs were discovered in a Lake Erie cHAB. High-resolution liquid chromatography electrospray ionization mass spectrometry (LC-ESI-MS), tandem mass spe

Microcystins (MCs), natural hepatotoxic compounds produced by cyanobacteria, pose significant risks to water quality, ecosystem stability, and the well-being of animals, plants, and humans when present in elevated concentrations. The escalating contamination of irrigation water with MCs presents a growing threat to terrestrial plants. The customary practice of irrigating crops from local water sources, including lakes and ponds hosting cyanobacterial blooms, serves as a primary conduit for transferring these toxins. Due to their high chemical stability and low molecular weight, MCs have the po

Cyanobacterial harmful algal blooms have increased globally, likely in part due to climate change and anthropogenic eutrophication, elevating exposure of humans and other terrestrial and aquatic animals to cyanotoxins through drinking water, food chains, and recreational activities. Microcystin-LR (MC-LR), the most prevalent microcystin congener, is well known for its hepatotoxicity, yet accumulating evidence indicates that it also exerts reproductive and developmental toxicity in mammals. This review summarizes recent experimental and epidemiological studies indicating that MC-LR disrupts rep

The cyanobacterial blooms produced by eutrophic water bodies have become a serious environmental issue around the world. After cellular lysing or algaecide treatment, microcystins (MCs), which are regarded as the most frequently encountered cyanobacterial toxins in fresh water, are released into water. Among all the variants of MCs, MC-LR has been widely studied due to its severe hepatotoxicity. Since 1992, various studies have identified the important roles of MC-LR in the origin and progression of primary liver cancers (PLCs), although few reviews have focused on it. Therefore, this review a

Microcystin-LR (MC-LR) is an emerging environmental pollutant produced by cyanobacteria that poses a threat to wild life and human health. In recent years, the reproductive toxicity of MC-LR has gained widespread attention, a large number of toxicological studies have filled the gaps in past research and more molecular mechanisms have been elucidated. Hence, this paper reviews the latest research advances on MC-LR-induced reproductive toxicity. MC-LR can damage the structure and function of the testis, ovary, prostate, placenta and other organs of animals and then reduce their fertility. Meanw

Acute cyanobacterial hepatotoxicosis in a wild roe deer (Capreolus capreolus) from Norway is reported. The diagnosis was based upon the demonstration of typical liver lesions and high liver concentrations of microcystins. The liver was markedly enlarged and histopathological examination revealed diffuse hepatocellular dissociation, degeneration and necrosis and perisinusoidal haemorrhage. Liquid chromatography-mass spectrometry demonstrated the presence of 1361 ng microcystin-YR, -LR and -RR per gram liver (wet weight). This is believed to be the first report of cyanobacterial intoxication in

Microcystins (MCs) and nodularins (NODs) are groups of toxins produced by cyanobacteria, and occur in surface and drinking water, recreational waters, dietary supplements and seafood. The presence of these toxins in drinking water and food may cause poisoning in humans and animals. More than 300 MCs and 10 NODs have been reported, and this large number of variants, combined with complex sample matrices, makes their analysis challenging for both LC-MS and ELISA. We developed immunoaffinity chromatography (IAC) columns using antibodies with broad specificity for MCs and NODs originally produced

BACKGROUND: Hemodialysis is a common but potentially hazardous procedure. From February 17 to 20, 1996, 116 of 130 patients (89 percent) at a dialysis center (dialysis center A) in Caruaru, Brazil, had visual disturbances, nausea, and vomiting associated with hemodialysis. By March 24, 26 of the patients had died of acute liver failure.
METHODS: A case patient was defined as any patient undergoing dialysis at dialysis center A or Caruaru's other dialysis center (dialysis center B) during February 1996 who had acute liver failure. To determine the risk factors for and the source of the outbrea

Dimefluthrin (DIM), a widely used pyrethroid insecticide, and microcystin-LR (MC-LR), a potent cyanotoxin produced by harmful algal blooms, are both frequently detected in aquatic environments. However, the potential combined effects of these two contaminants, particularly regarding intergenerational toxicity, remain largely unexplored. In this study, we aimed to evaluate the intergenerational effects of chronic parental co-exposure to DIM and MC-LR in zebrafish. Adult zebrafish were exposed to DIM and MC-LR, alone or in combination, for 160 days. A panel of phenotypic, histological, biochemic

Thirteen bacterial isolates from lake sediment, capable of degrading cyanobacterial hepatotoxins microcystins and nodularin, were characterized by phenotypic, genetic and genomic approaches. Cells of these isolates were Gram-negative, motile by means of a single polar flagellum, oxidase-positive, weakly catalase-positive and rod-shaped. According to phenotypic characteristics (carbon utilization, fatty acid and enzyme activity profiles), the G+C content of the genomic DNA (66.1-68.0 mol%) and 16S rRNA gene sequence analysis (98.9-100% similarity) the strains formed a single microdiverse genosp

A capillary electrophoretic (CE) method incorporating sodium dodecyl sulphate (SDS)-organic modifier solvents in the CE buffer was developed for the detection of toxic cyclic heptapeptide toxins (microcystins) produced by blue-green algae (cyanobacteria). The applicability of these run buffers for the analysis of microcystins was evaluated and optimum conditions for separation were determined. The migration times, elution order and selectivity of the toxic peptides were influenced by modifying the composition of the electrophoretic buffer with organic solvents [0 to 20% (v/v)]. At maximum addi

As cyanobacterial blooms intensify globally, persistent microcystins (MCs) in surface waters present a growing water-quality challenge. Engineered solutions exist but are often costly and impractical for large-scale or natural settings. In contrast, wetlands, with their diverse microbial assemblages and habitats, offer a promising but underexplored pathway for MC removal. We tested this potential through in situ mesocosm experiments in two prairie wetlands; a hypereutrophic system receiving livestock and urban wastewater, and a less-impacted meso‑eutrophic site. Microcystin degradation was mea

Microcystin-LR (MC-LR) is a potent hepatotoxin, but a few studies suggested that it might also induce nephrotoxicity. However, nephrotoxicity induced by prolonged oral exposure to MC-LR is unknown. The aim of this study was to evaluate the potential influence of MC-LR on the kidney in mice following chronic exposure to MC-LR. In this study, we evaluated the nephrotoxicity of MC-LR in mice drinking water at different concentrations (1, 30, 60, 90, and 120 μg/L) for 6 months for the first time. The results showed that the kidney weights and the kidney indexes of mice were not altered in the MC-L

BACKGROUND: After a drought in February, 1996, all 126 patients in a haemodialysis unit in Caruaru, north-east Brazil, developed signs and symptoms of acute neurotoxicity and subacute hepatotoxicity following the use of water from a lake with massive growth of cyanobacteria (blue-green algae). 60 patients died.
METHODS: Besides recording clinical details and outcome at follow-up, we arranged laboratory, radiological, and histological investigations on the patients and toxicological studies of serum and haemodialysis water filters.
FINDINGS: The acute presentation was with malaise, myalgia an

Microcystins (MCs) are a class of monocyclic heptapeptide natural toxins produced by freshwater cyanobacteria. They can enter the human body via contaminated water or food. Currently, MCs are recognized by the World Health Organization as health risk factors, with microcystin-leucine-arginine (MC-LR) being the most prevalent and toxic. Our previous research demonstrated that chronic MC-LR exposure reduced dopamine (DA) release from dopaminergic neurons in the substantia nigra, causing Parkinson's disease (PD)-like motor dysfunction in mice. Motor dysfunction is closely linked to reduced DA lev

BACKGROUND AND AIMS: Biliary atresia is a fibrosing cholangiopathy affecting neonates that is thought to be caused by a prenatal environmental insult to the bile duct. Biliatresone, a plant toxin with an α-methylene ketone group, was previously implicated in toxin-induced biliary atresia in Australian livestock, but is found in a limited location and is highly unlikely to be a significant human toxin. We hypothesized that other molecules with α-methylene ketone groups, some with the potential for significant human exposure, might also be biliary toxins.

Although it is well known that microcystin-LR (MC-LR) may cause male reproductive toxicity, less is known on its potential impact on the development of prostate. In this study, from the 12th day in the embryonic period to the 21st day after birth, 4 randomly assigned groups of pregnant mice were exposed to 0, 1, 10, and 50μg/L of MC-LR through drinking water followed by the analyses of their 30- and 90-day-old male offspring. The result showed that MC-LR could enter and be accumulated in the offsprings prostate. Using serological, morphological, and immunohistochemical analysis, we explored th

Microcystins are cyclic peptide toxins. Chronic intoxication with well-known members of the microcystin family--microcystins-LR--induces liver tumour formation, injury of kidney and heart. Despite worldwide distribution in the environment, the effects of microcystins-YR have not been studied extensively. The aim of the study was to evaluate whether microcystins-YR, in relatively low doses, have a toxic effect on cardiomyocytes of chronically treated rats. Male adult Wistar rats were treated every second day for 8 months with microcystins-YR (10 microg/kg i.p., N = 5). Control groups were treat

Microcystins (MC) are a group of cyanobacterial toxins that comprises MC-LF and other cyclic heptapeptides, best known as potent hepatotoxicants. Cell culture and epidemiological studies suggest that MC might also affect the nervous system when there is systemic exposure, e.g., via drinking water or food. We asked whether in vitro studies with human neurons could provide estimates on the neurotoxicity hazard of MC-LF. First, we used LUHMES neurons, a well-established test system for neurotoxicants and neuropathological processes. These central nervous system cells express OATP1A2, a presumed c
