Generation of controllable gaseous H2S concentrations using microfluidics.

Hydrogen sulfide (H2S) plays an important role as an intercellular and intracellular signaling molecule, yet its targets are not well understood. As a molecule it easily evaporates and it is hard to acquire stable concentration for in vitro studies, constituting a major problem for the field to identify its downstream targets and function. Here we develop a microfluidic system that can provide consistent and controllable H2S levels in contrast to the current method of delivering large bolus doses to cells. The system relies on the permeability of H2S gas through a polydimethylsiloxane thin mem

Live Imaging of Grass Roots Growth Dynamics Using Microfluidics.

Microfluidic tools enable manipulation of microliter-scale volumes within a stable microenvironment. Microfluidics has been widely adopted in various biological fields for, among other applications, controlled delivery of sensory, chemical, and mechanical stimuli to the observed organisms. In this chapter, we describe the design and usage of a custom-made double-layered PDMS microfluidic device, named BUGchip, developed for the purpose of studying plant root growth and metabolic responses in rapidly changing conditions with a high time-scale resolution. We demonstrate the utility of this devic

Microfluidics to assess the impact of transfused blood components.

Assessing the function of transfused blood components within the dynamic environment of the vascular system remains challenging, as conventional assays are largely static and poorly represent the dynamic mechanical and biochemical interactions between blood cells and the vessel wall. Microfluidic approaches, which study blood flow within small-scale channels, provide an opportunity to bridge this gap by introducing physiologically relevant flow and shear rates into experimental models. This review synthesises the current literature describing the application of microfluidic technologies to the

Low-Frequency Segmented-Flow Microfluidics for Biphasic Chemical Sensing.

Droplet microfluidics with droplet generation frequencies of several hundred hertz and droplet volumes below 1 nL has previously been reported for biphasic chemical sensing based on oil sensors. However, tracking fluorescence from these rapidly moving subnanoliter droplets typically requires highly sophisticated optical instrumentation, such as laser-scanning confocal microscopy. In this work, we implement oil-based chemical sensing in segmented-flow microfluidics with operating frequencies below 20 Hz and segment volumes of tens to hundreds of nanoliters. The lower segment generation frequenc

Engineering organs-on-a-chip via multi-channel microfluidics.

Conventional in vitro physiological models, relying on animal studies and 2D/3D cell cultures, are fundamentally limited by interspecies biological discrepancies, ethical constraints, or inadequate replication of human physiology. Organ-on-a-chip (OoC) technology overcomes these challenges through emulating organ-specific microphysiological systems. The transformative power of this innovation lies in multi-channel microfluidic chips. These chips facilitate the formation of 3D cellular organizations and tissue interfaces via integrated porous membranes, micropillar arrays, or perfusable vascula

Improving ARTs: Microfluidics as a Tool for Sperm Selection.

Sperm selection is a critical step in assisted reproductive technology (ART), as sperm quality directly influences embryo quality. In vivo, the female reproductive tract imposes several physical and physiological barriers that favors only the most optimal spermatozoa reaching the fertilization site. Such barriers are absent in in vitro fertilization (IVF) and intracytoplasmic sperm injection (ICSI) treatments, which has driven the continuous development of strategies for sperm selection. This review provides a comprehensive overview of physiological sperm selection within the female reproducti

High-throughput generic single-entity sequencing using droplet microfluidics.

Single-cell sequencing has revolutionized our understanding of cellular heterogeneity by providing a micro-level perspective in the past decade. While heterogeneity is fundamental to diverse biological communities, existing platforms are primarily designed for eukaryotic cells, leaving significant gaps in the study of other single biological entities, such as viruses and bacteria. Current methodologies for single-entity sequencing remain limited by low throughput, inefficient lysis, and highly fragmented genomes. Here, we present the Generic Single-Entity Sequencing (GSE-Seq), a versatile and

The Application of Microfluidics in Traditional Chinese Medicine Research.

Microfluidics enables precise manipulation of scarce Traditional Chinese Medicine (TCM) samples while accelerating analysis and enhancing sensitivity. Device-level structures explain these gains: staggered herringbone and serpentine mixers overcome low-Reynolds-number constraints to shorten diffusion distances and reduce incubation time; flow-focusing or T-junction droplet generators create one-droplet-one-reaction compartments that suppress cross-talk and support high-throughput screening; "Christmas-tree" gradient generators deliver quantitative dosing landscapes for mechanism-aware assays;

Large-scale perfused tissues via synthetic 3D soft microfluidics.

The vascularization of engineered tissues and organoids has remained a major unresolved challenge in regenerative medicine. While multiple approaches have been developed to vascularize in vitro tissues, it has thus far not been possible to generate sufficiently dense networks of small-scale vessels to perfuse large de novo tissues. Here, we achieve the perfusion of multi-mm3 tissue constructs by generating networks of synthetic capillary-scale 3D vessels. Our 3D soft microfluidic strategy is uniquely enabled by a 3D-printable 2-photon-polymerizable hydrogel formulation, which allows for precis

Microfluidics: The future of sperm selection in assisted reproduction.

BACKGROUND: Obtaining functional sperm cells is the first step to treat infertility. With the ever-increasing trend in male infertility, clinicians require access to effective solutions that are able to single out the most viable spermatozoa, which would max out the chance for a successful pregnancy. The new generation techniques for sperm selection involve microfluidics, which offers laminar flow and low Reynolds number within the platforms can provide unprecedented opportunities for sperm selection. Previous studies showed that microfluidic platforms can provide a novel approach to this chal

Microfluidics for rapid cytokeratin immunohistochemical staining in frozen sections.

Frozen sections (FS) of tumor samples represent a cornerstone of pathological intraoperative consultation and have an important role in the microscopic analysis of specimens during surgery. So far, immunohistochemical (IHC) stainings on FS have been demonstrated for a few markers using manual methods. Microfluidic technologies have proven to bring substantial improvement in many fields of diagnostics, though only a few microfluidic devices have been designed to improve the performance of IHC assays. In this work, we show optimization of a complete pan-cytokeratin chromogenic immunostaining pro

Microfluidics-Based Approaches to the Isolation of African Trypanosomes.

African trypanosomes are responsible for significant levels of disease in both humans and animals. The protozoan parasites are free-living flagellates, usually transmitted by arthropod vectors, including the tsetse fly. In the mammalian host they live in the bloodstream and, in the case of human-infectious species, later invade the central nervous system. Diagnosis of the disease requires the positive identification of parasites in the bloodstream. This can be particularly challenging where parasite numbers are low, as is often the case in peripheral blood. Enriching parasites from body fluids

Analysis of platelet function: role of microfluidics and nanodevices.

Platelet aggregation is essential for vascular haemostasis and thrombosis. To improve the therapy of arterial thrombotic disorders and identify novel therapeutic targets it is imperative to study basic mechanisms of platelet thrombus formation. To date most data on biology, physiology and pharmacology of platelet aggregation have been obtained by studying this phenomenon under static or quasi-dynamic conditions at the macroscale level. There is a widespread recognition for the need of new technologies that will help to further elucidate the role of platelets in physiological and pathological t

Lattice Slot Waveguide for Terahertz Microfluidics Biomedical Trace Analysis.

Electromagnetic metasurface integrated microfluidic chips enable a real-time, label-free platform for terahertz trace analysis of volume-limited biomedical samples with suppressed water absorption noise. However, conventional metal-insulator-metal (MIM) metasurface resonators exhibit inherently limited Q-factor and sensitivity due to radiative leakage through open side boundaries. Here, a lattice slot waveguide based on MIM configuration is designed to effectively confine energy within the microfluidic channel and mitigate radiative loss. This trapped mode achieves enhanced sensitivity and Q-f

Yield stress fluids in microfluidics: research, applications and opportunities.

Microfluidic technology enables scalable solutions in precision medicine, diagnostics, drug delivery, organ-on-a-chip models, single-cell analysis, high-throughput screening, and environmental monitoring. In fact, many of these applications process fluids with complex rheology, e.g., blood, mucus, bioinks, and polymer or particle suspensions, exhibiting yield stress, viscoelasticity, and shear-thinning that govern transport, mixing, and interfacial dynamics at small scales. However, the interplay of such rheology with microscale confinement, roughness, and surface patterning remains underexplo

Orbital Electrowetting: From Continuous Droplet Transport to Programmable Microfluidics.

Droplet transport on solid surfaces underpins applications in microfluidics, thermal management, and water harvesting. Conventional electrowetting (CEW) has enabled digital droplet control, but reliance on numerous individually addressed electrodes, complex driving circuitry, and step-wise motion has limited scalability and practical deployment. Orbital electrowetting (OEW) has recently been introduced as a complementary paradigm, where asymmetric electrowetting forces and electrostatic energy gradients drive droplets into continuous, high-speed motion along predefined orbital pathways using o

Capillary Microfluidics for Bioanalysis: Design Principles and Application Perspectives.

Capillary flow-driven microfluidic devices enable autonomous fluid transport for bioanalytical applications by using surface tension and wettability in microscale channels and porous structures, eliminating the need for external pumps. This review develops a coherent physical framework for capillary devices, based on the Young-Laplace equation, Jurin's law, the Lucas-Washburn equation, and Darcy's law, that relates interfacial curvature, capillary pressure, viscous dissipation, and permeability to fluid behavior in both hollow channels and porous media. From this framework, we derive practical

Integration of microfluidics in animal in vitro embryo production.

The in vitro production of livestock embryos is central to several areas of animal biotechnology. Further, the use of in vitro embryo manipulation is expanding as new applications emerge. ARTs find direct applications in increasing genetic quality of livestock, producing transgenic animals, cloning, artificial insemination, reducing disease transmission, preserving endangered germplasm, producing chimeric animals for disease research, and treating infertility. Whereas new techniques such as nuclear transfer and intracytoplasmic sperm injection are now commonly used, basic embryo culture proced
