Switchable photoluminescence of CdTe nanocrystals by temperature-responsive microgels.

In the present study, we report a method for preparing a fluorescent thermosensitive hybrid material based on monodisperse, thermosensitive poly( N-isopropyl acrylamide) (PNIPAM) microgels covered with CdTe nanocrystals of 3.2 nm diameter. The CdTe nanocrystals were covalently immobilized on the surface of PNIPAM microgels. The chemical environment around the CdTe nanocrystals was modified by changing the temperature and inducing the microgel volume-phase transition. This change provoked a steep variation in the nanocrystal photoluminescence (PL) intensity in such a way that when the temperatu

Unexpected behavior of ultra-low-crosslinked microgels in crowded conditions.

Ultra-low-crosslinked (ULC) microgels are among the softest colloidal particles nowadays routinely synthesized experimentally. Despite a growing literature of experimental results, their microscopic behavior under crowded conditions is yet to be revealed. To this aim, we resort to realistic monomer-resolved computer simulations to investigate their structural, mechanical, and dynamical properties across a wide range of packing fractions. Using particle-resolved analyses, we unveil the role of outer chains in the ULCs, which manifest in peculiar behaviors, utterly different from those of regula

Viscoelasticity-dependent, deformation-based sorting of microgels using sequential bifurcations.

Droplet-based microfluidics have accelerated the discovery of rare variants in large combinatorial protein libraries by enabling high-throughput screening in miniaturized reaction systems. However, throughput limitations and assay compatibility constrain the accessible sequence space and the diversity of enzyme classes amenable to droplet-based screening. Passive, deformability-based sorting systems offer scalability via microfluidic parallelization, but existing platforms lack the sensitivity and throughput required for efficiently exploring vast fitness landscapes. In this work, we present a

Pickering emulsions stabilized by microgels: Three regimes of surface coverage.

Oil-in-water Pickering emulsions can be stabilized by poly-N-Isopropylacrylamide based microgels that adsorb, deform and entangle at the droplets interface. The surface coverage Γemulsion, defined as the mass of microgels per unit interfacial area, likely plays a key role in the emulsion properties as stability and responsiveness. The objective of the present study is to link Γemulsion to the concentration of microgels used during the emulsification process. Γemulsion was monitored by combining droplet size analysis, UV-visible quantification of non-adsorbed microgels and Cryo-SEM visualizatio

Clickable PEG-norbornene microgels support suspension bioprinting and microvascular assembly.

The development of perfusable and multiscale vascular networks remains one of the largest challenges in tissue engineering. As such, there is a need for the creation of customizable and facile methods to produce robustly vascularized constructs. In this study, secondarily crosslinkable (clickable) poly(ethylene glycol)-norbornene (PEGNB) microbeads were produced and evaluated for their ability to sequentially support suspension bioprinting and microvascular self-assembly towards the aim of engineering hierarchical vasculature. Smart chitosan-based microgels for enhanced photothermal-assisted antibacterial activity.

During recent years, antibiotic-resistant bacteria have rapidly emerged owing to the irrational use of antibiotics, rendering a global problem. Currently, few studies introduce customized antibacterial nanoplatforms to overcome antibiotic-resistance according to specific characteristic of bacteria, rather than abuse of antibiotic. Herein, with regard to personalized antibacterial nanoplatform, we design a novel antibiotic delivery nanocarrier composed of polyaniline-grafted-chitosan, presenting pH-responsive, conductive, photothermal, and biodegradable properties. After treatment with divalent

Emulsion-free chitosan-genipin microgels for growth plate cartilage regeneration.

The growth plate is a cartilage tissue near the ends of children's long bones and is responsible for bone growth. Injury to the growth plate can result in the formation of a 'bony bar' which can span the growth plate and result in bone growth abnormalities in children. Biomaterials such as chitosan microgels could be a potential treatment for growth plate injuries due to their chondrogenic properties, which can be enhanced through loading with biologics. They are commonly fabricated via an emulsion method, which involves solvent rinses that are cytotoxic. Here, we present a high throughput, no

Multifunctional Microgels: From Material Design to Skin Wound Healing Applications.

Skin wound healing is a complex and dynamic biological process that requires the suppression of excessive inflammation and oxidative stress, control of microbial invasion, and promotion of tissue regeneration. Conventional wound dressings often fail to modulate the wound microenvironment, limiting their effectiveness in chronic and infected wounds. In recent years, microgels have emerged as a promising candidate for wound healing due to their injectability, high surface area, tunable physicochemical properties, adaptability to irregular wound surface, and ability to encapsulate diverse therape

Granular polyrotaxane microgels as injectable hydrogels for corneal tissue regeneration.

Corneal diseases, a leading cause of global vision impairment, present challenges in treatment due to corneal tissue donor scarcity and transplant rejection. Hydrogel biomaterials in the form of corneal implants for tissue regeneration, while promising, have faced obstacles related to cellular and tissue integration. This study develops and investigates the potential of granular polyrotaxane (GPR) hydrogels as a scaffold for corneal keratocyte growth and transparent tissue generation. Employing host-guest driven supramolecular interactions, we developed injectable, cytocompatible hydrogels. By

Porous Anisometric PNIPAM Microgels: Tailored Porous Structure and Thermal Response.

The porous structure of microgels significantly influences their properties and, thus, their suitability for various applications, in particular as building blocks for tissue scaffolds. Porosity is one of the crucial features for microgel-cell interactions and significantly increases the cells' accumulation and proliferation. Consequently, tailoring the porosity of microgels in an effortless way is important but still challenging, especially for nonspherical microgels. This work presents a straightforward procedure to fabricate complex-shaped poly(N-isopropyl acrylamide) (PNIPAM) microgels wit

Near-Infrared-Light-Modulated Lubricating Coating Enabled by Photothermal Microgels.

As a micro-/nano-sized hydrogel, polymeric microgel not only has three-dimensional (3D) molecular networks but also displays the small-size effect, which has been widely used in various fields, such as drug nanocarrier, photonic crystal, functional coating, and aqueous lubrication. In this work, a photothermal lubricating coating was prepared using polymeric/inorganic hybrid microgels and its surficial friction was deliberately modulated by the remote irradiation of near-infrared (NIR) light. Specifically, a photothermal hybrid microgel, Fe3O4@poly(N-isopropylacrylamide-co-polyacrylic acid) (F

MicroGelzymes: pH-Independent Immobilization of Cytochrome P450 BM3 in Microgels.

Microgels are an emerging class of "ideal" enzyme carriers because of their chemical and process stability, biocompatibility, and high enzyme loading capability. In this work, we synthesized a new type of permanently positively charged poly(N-vinylcaprolactam) (PVCL) microgel with 1-vinyl-3-methylimidazolium (quaternization of nitrogen by methylation of N-vinylimidazole moieties) as a comonomer (PVCL/VimQ) through precipitation polymerization. The PVCL/VimQ microgels were characterized with respect to their size, charge, swelling degree, and temperature responsiveness in aqueous solutions. P45

Encapsulation of Porphyrins in Polymer Microgels and Microbeads for Heterogeneous Photooxidation.

Using porphyrin-mediated photocatalytic energy transfer to generate singlet oxygen is a premier strategy for green, selective chemical synthesis. However, such catalytic systems are mostly conducted in organic solvents. This work aims to explore the generation and utilization of singlet oxygen for photochemical oxidation in aqueous media. To this end, we prepared two porphyrins with reactive primary amines or charged quaternized ammonium groups for covalent and ionic crosslinking, producing TPP-loaded hydrophilic microgels and microbeads, respectively. We explored catalyst formulations to opti

Chitosan-gelatin-based microgel for sustained drug delivery.

We describe here the proof-of-concept of synthesizing microgels via cross-linking of chitosan and gelatin with succinimide-end polyethylene glycol (PEG). The introduction of PEG provided thermo-responsiveness to microgels, and the size of microgels significantly changed with increasing temperature. Furthermore, the microgel was biocompatible as confirmed in vitro by MTT assay with NIH 3T3 fibroblast and degradable. Chitosan-based microgels exhibited strong affinity to hydrophobic drug and prolonged release of folic acid was observed at physiological temperature with varying pH. The proposed co

Interfacial and emulsifying behavior of plant-based microgels via complex coacervation.

In recent years, rising global protein demand and increasing awareness of sustainability, climate change, and animal welfare issues have accelerated the shift toward plant-based foods. Despite their promise as sustainable animal-protein alternatives, plant proteins are often limited by inherent astringency associated with high friction and poor lubrication. In this work, pH-responsive microgels derived from liquid-liquid phase separation (LLPS) of deamidated β-conglycinin (D-β-CG) and chitosan oligosaccharide (COS) were engineered to modulate interfacial behavior and lubrication in dairy syste

Synthesis of Biodegradable Cell-Laden Microgels Assembly by Stop-Flow Lithography.

This study presents a novel approach for the synthesis of biodegradable cell-laden microgels using stop-flow lithography (SFL), addressing critical challenges in the field of tissue engineering. Traditional methods for creating 3D cell cultures often rely on non-biodegradable materials, which limit their application and raise concerns about cell viability. In this work, we successfully replace poly(ethylene glycol) diacrylate (PEGDA) with dextran-2-hydroxyethyl methacrylate (dex-HEMA), a biocompatible and biodegradable alternative. Furthermore, we introduce a technical solution for sterile cel

Microfluidics-Inspired Microgels with Intrinsic Bioactivity for Advanced Burn Wound Therapy.

Functionalized hydrogel microspheres hold great promise in tissue regeneration, yet their clinical translation is often hindered by high production costs, complex fabrication processes, and reliance on external bioactive additives. To overcome these challenges, multifunctional gelatin-tea polyphenol (GTP) microgels using a simplified, cost-effective, microfluidics-inspired platform are reported, which is composed of an injection unit, a reciprocating apparatus, and a collection substrate. Using a microinjection pump, uncrosslinked microspheres with precisely controlled diameters (125-570 µm) a

FasL microgels induce immune acceptance of islet allografts in nonhuman primates.

Islet transplantation to treat insulin-dependent diabetes is greatly limited by the need for maintenance immunosuppression. We report a strategy through which cotransplantation of allogeneic islets and streptavidin (SA)-FasL-presenting microgels to the omentum under transient rapamycin monotherapy resulted in robust glycemic control, sustained C-peptide levels, and graft survival in diabetic nonhuman primates for >6 months. Surgical extraction of the graft resulted in prompt hyperglycemia. In contrast, animals receiving microgels without SA-FasL under the same rapamycin regimen rejected islet

Metabolomic Profiling and Mechanotransduction of Single Chondrocytes Encapsulated in Alginate Microgels.

Articular cartilage is comprised of two main components, the extracellular matrix (ECM) and the pericellular matrix (PCM). The PCM helps to protect chondrocytes in the cartilage from mechanical loads, but in patients with osteoarthritis, the PCM is weakened, resulting in increased chondrocyte stress. As chondrocytes are responsible for matrix synthesis and maintenance, it is important to understand how mechanical loads affect the cellular responses of chondrocytes. Many studies have examined chondrocyte responses to in vitro mechanical loading by embedding chondrocytes in 3-D hydrogels. Howeve
