Genetic damage in human cells exposed to non-ionizing radiofrequency fields: a meta-analysis of the data from 88 publications (1990-2011).

Based on the 'limited' evidence suggesting an association between exposure to radiofrequency fields (RF) emitted from mobile phones and two types of brain cancer, glioma and acoustic neuroma, the International Agency for Research on Cancer has classified RF as 'possibly carcinogenic to humans' in group 2B. In view of this classification and the positive correlation between increased genetic damage and carcinogenesis, a meta-analysis was conducted to determine whether a significant increase in genetic damage in human cells exposed to RF provides a potential mechanism for its carcinogenic potent

Persistent increase in the rates of apoptosis and dicentric chromosomes in surviving V79 cells after X-irradiation.

In parallel to delayed reproductive death and delayed micronuclei in surviving V79 cells that have been reported, the frequency of apoptosis and dicentrics was investigated in the same cell line, 1-2 weeks after X-ray doses between 1 and 12 Gy. Apoptosis was assessed by morphological criteria after staining with Giemsa and using the Apoptag method in parallel. The frequency of apoptosis peaked 48 h after irradiation at about 35% as a function of dose and then decreased to about 10% on day 3. It remained 10 times higher than in unirradiated cells until day 14. The frequency of dicentric chromos

Sensitivity to DNA cross-linking chemotherapeutic agents in mismatch repair-defective cells in vitro and in xenografts.

Together with tolerance to killing induced by methylating agents, loss of mismatch repair (MMR) has previously been found to be associated with hypersensitivity to the DNAcross-linking agent 1-(2-chloroethyl)-3-cyclohexyl-nitrosourea(CCNU) in several human tumor cell lines (Aquilina et al., 1998). Here, we have investigated whether MMR might act as an efficient repair pathway and provide protection against the clastogenicity induced by CCNU and whether the hypersensitivity of MMR-defective cells is extended to other cross-linking agents. An increase in cell killing and in the frequency of micr

Molecular mechanisms by which in vivo exposure to exogenous chemical genotoxic agents can lead to micronucleus formation in lymphocytes in vivo and ex vivo in humans.

The purpose of this review is to summarise current knowledge on the molecular mechanisms by which in vivo exposure to exogenous chemical genotoxins in humans induces micronuclei (MNi) and other nuclear anomalies in lymphocytes in vivo and ex vivo after nuclear division in vitro. MNi originate from acentric chromosome fragments and/or whole chromosomes that are unable to engage with the mitotic spindle and/or fail to segregate properly to the daughter nuclei during anaphase. The lagging fragments or whole chromosomes are surrounded by membrane and become MNi. Acentric fragments are caused by fa

Mutagenic activity of acrylamide in the rat thyroid cells under conditions of a subacute experiment.

Experimental mutagenic effect of acrylamide on thyroid gland cells was studied by an extended micronucleus test. Acrylamide in doses corresponding to 0.004-0.1 LD(50) increased the incidence of thyroid follicular cells (A-cells) with micronuclei and other karyological parameters in exposed rats after hemithyroidectomy. This cytogenetic effect allows regarding acrylamide as a mutagen for the thyroid gland and as a carcinogen for this organ.

Molecular mechanisms of micronucleus, nucleoplasmic bridge and nuclear bud formation in mammalian and human cells.

Micronuclei (MN) and other nuclear anomalies such as nucleoplasmic bridges (NPBs) and nuclear buds (NBUDs) are biomarkers of genotoxic events and chromosomal instability. These genome damage events can be measured simultaneously in the cytokinesis-block micronucleus cytome (CBMNcyt) assay. The molecular mechanisms leading to these events have been investigated over the past two decades using molecular probes and genetically engineered cells. In this brief review, we summarise the wealth of knowledge currently available that best explains the formation of these important nuclear anomalies that

Aneuploidies and micronuclei in the germ cells of male mice of advanced age.

The objective of this research was to determine whether the frequencies of chromosomally defective germ cells increased with age in male laboratory mice. Two types of chromosomal abnormalities were characterized: (1) testicular spermatid aneuploidy (TSA) as measured by a new method of multi-color fluorescence in situ hybridization (FISH) with DNA probes specific for mouse chromosomes X, Y and 8, and (2) spermatid micronucleus (SMN) analyses using anti-kinetochore antibodies. B6C3F1 mice (aged 22.5 to 30.5 months, heavier than controls but otherwise in good health) showed significant approximat

Chromothripsis as an on-target consequence of CRISPR-Cas9 genome editing.

Genome editing has therapeutic potential for treating genetic diseases and cancer. However, the currently most practicable approaches rely on the generation of DNA double-strand breaks (DSBs), which can give rise to a poorly characterized spectrum of chromosome structural abnormalities. Here, using model cells and single-cell whole-genome sequencing, as well as by editing at a clinically relevant locus in clinically relevant cells, we show that CRISPR-Cas9 editing generates structural defects of the nucleus, micronuclei and chromosome bridges, which initiate a mutational process called chromot

Transient hypothermia induces micronuclei in mice.

We investigated the effect of hypothermia on micronucleus induction in mouse bone marrow cells. To induce hypothermia, we administered chlorpromazine, which was negative in an in vitro chromosome aberration test, at 3.13, 6.25, 12.5, 25, 50, and 100 mg/kg intraperitoneally. Doses of 12.5-100 mg/kg decreased rectal temperature transiently to less than 33 degrees C. The temperature depression following 25-100 mg/kg lasted for 11 h before returning to normal 24 h later except for the 100 mg/kg treatment group. Doses of 25-100 mg/kg produced a statistically significant (p < 0.01) increase in micro

[Telomeric chromosome fusion in cells with micronuclei under the combined action of hyperthermia and hypothermia and halogenated analogs of thymidine].

The reaction of cells with micronuclei in respect of the induction of specific dicentric chromosomes with halogenated analogs of thymidine at various temperatures was studied. The positive correlation between the temperature and frequency of dicentrics was shown for all halogenated analogs of thymidine. The minimum frequency of dicentrics was found in the case when used 5-iododeoxyuridine and hypothermia (34 degrees C). The using of 5-bromodeoxyuridine at different temperatures displayed the intermediate results. The maximum level of dicentrics discovered under action of 5-chlorodeoxyuridine a

Relationship between radiation induced dicentric chromosome aberrations and micronucleus formation in human lymphocytes.

Chromosome damage measured by the chromosome aberration technique is a reliable method to assess the radiation dose absorbed by cells. However, this technique has some disadvantages. Scoring is difficult and requires skill and experience which of these lead low number of cell counts. The micronucleus (MN) technique which also measures chromosome losses has easy scoring criteria leading high numbers of cell counts and therefore holds more statistical power. In this study, the relationship between the results of the micronucleus technique and those obtained by the chromosome aberration technique

[Chromosomal aberrations, sister chromatid exchanges and micronuclei induced in human lymphocytes by sodium bisulfite (sulfur dioxide)].

The frequencies of chromosomal aberrations (CA), sister chromatid exchanges (SCE) and micronuclei (MN) in human blood lymphocytes exposed to sodium bisulfite (sulfur dioxide) at varies concentrations ranging from 5 x 10(-5) to 2 x 10(-3) mol/L in vitro were studied. It was shown that sodium bisulfite (NaHSO3 and Na2SO3, 1:3 mol/L) caused an increase in SCE and MN of human blood lymphocytes in a dose-dependent manner, and also induced mitotic delays and decreased mitotic index of the lymphocytes. For CA, our results indicated that sodium bisulfite induced an increase of chromatid-type aberratio

In vitro cytogenetic effects of 2450 MHz waves on human peripheral blood lymphocytes.

Cytogenetic analyses were performed on human peripheral blood lymphocytes exposed to 2450 MHz microwaves during 30 and 120 min at a constant temperature of 36.1 degrees C (body temperature). The temperature was kept constant by means of a temperature probe put in the blood sample which gives feedback to a microcomputer that controls the microwave supply. We found a marked increase in the frequency of chromosome aberrations (including dicentric chromosomes and acentric fragments) and micronuclei. On the other hand the microwave exposure did not influence the cell kinetics nor the sister chromat

Observation of frequencies of lymphocytes with micronuclei in human peripheral blood cultures from workers in a sulphuric acid factory.

Frequencies of lymphocytes with micronuclei (MNF) in human peripheral blood cultures from workers in a sulphuric acid factory (the SO2 concentrations in the workplace ranged from 0.34 mg/m3 to 11.97 mg/m3 air) were investigated. It was shown that among 40 workers, the MNF of 28 workers was more than 0.1%, and the MNF of 7 workers was greater than the upper limit of the normal range (0.2%). The mean MNF of the worker and the control group was 0.168% and 0.071%, respectively. The difference between them was statistically significant (P less than 0.001). It implies that SO2 is a clastogenic and g

Micronuclei, inflammation and auto-immune disease.

Auto-immune diseases (AUD) are characterized by an immune response to antigenic components of the host itself. The etiology of AUD is not well understood. The available evidence points to an interaction between genetic, epigenetic, environmental, infectious and life-style factors. AUD are more prevalent in women than in men; sex hormones play a crucial role in this sex bias. Micronuclei (MN) emerged as a new player in the induction of AUD, based on the capacity of DNA-sensors to detect self-DNA that leaks into the cytoplasm from disrupted MN and induce the cGAS-STING pathway triggering an inna

Evaluation of the potential of mobile phone specific electromagnetic fields (UMTS) to produce micronuclei in human glioblastoma cell lines.

Some epidemiological studies indicate that mobile phones cause glioblastomas in humans. Since it is known that genomic instability plays a key role in the etiology of cancer, we investigated the effects of the universal mobile telecommunications system radiofrequency (UMTS-RF) signal, which is used in "smart" phones, on micronucleus (MN) formation and other anomalies such as nuclear buds (NBUDs) and nucleoplasmatic bridges (NPBs). MN are formed by structural and numerical aberrations, NBs reflect gene amplification and NPBs are formed from dicentric chromosomes. The experiments were conducted

Effect of sulfur dioxide hydrates on cell cycle, sister chromatid exchange, and micronuclei in barley.

The effects of sulfur dioxide (SO(2)) hydrates exposure on cell cycle, sister chromatid exchange (SCE), and micronuclei (MN) were investigated in barley (Hordeum vulgare) roots. A mixture of sodium bisulfite and sodium sulfite (1:3), at various concentrations from 1x10(-5) to 3x10(-2)M, was used for the treatments. The results showed that the mixture induced the formation of SCE and MN in barley root cells with different effective concentrations and with different trends as treatment concentrations increased. At high concentrations of 0.5-30.0mM, SO(2) hydrates inhibited the mitotic activity a

Micronuclei frequency in peripheral blood lymphocytes and cancer risk: evidence from human studies.

Over a century ago, Theodor Boveri paved the way to mechanistic studies linking chromosomal abnormalities to cancer pathogenesis. Since then, theoretical and empirical evidence has been accumulated, supporting a causal role of these events in the aetiology of human cancer. A powerful tool for measurement of chromosomal abnormalities is the cytokinesis-block micronucleus cytome (CBMNcyt) assay. The validation of the micronucleus (MN) as marker of phenotypic susceptibility to cancer has received decisive support from mutagens sensitivity studies, particularly from a recent case-control study on

The micronucleus assay as a biological dosimeter of in vivo ionising radiation exposure.

Biological dosimetry, based on the analysis of micronuclei (MN) in the cytokinesis-block micronucleus (CBMN) assay can be used as an alternative method for scoring dicentric chromosomes in the field of radiation protection. Biological dosimetry or Biodosimetry, is mainly performed, in addition to physical dosimetry, with the aim of individual dose assessment. Many studies have shown that the number of radiation-induced MN is strongly correlated with dose and quality of radiation. The CBMN assay has become, in the last years, a thoroughly validated and standardised technique to evaluate in vivo

The bystander effect-induced formation of micronucleated cells is inhibited by antioxidants, but the parallel induction of apoptosis and loss of viability are not affected.

X-rays induce various DNA damages including strand breaks that lead to formation of micronuclei and chromosomal aberrations as well as increased number of apoptotic cells. Similar effects appear when non-irradiated cells are treated with medium collected from cultures of irradiated cells (irradiation conditioned medium - ICM). This phenomenon was termed "bystander effect". A number of studies suggest that bystander effect appears to be associated with up-regulation of oxidative metabolism. We thus compared the effects of antioxidant Vitamins C and E on the frequency of micronuclei and apoptoti
