Pulmonary arterial hypertension (PAH) is marked by elevated vascular resistance, right ventricular (RV) failure, and poor clinical outcomes. Current therapies primarily target pulmonary vascular hemodynamics, necessitating novel strategies to address RV remodeling. In this study, we investigated the role of lncRNA FGD5-AS1 and its micropeptides in PAH and RV remodeling, focusing on their effects on hyaluronic acid (HA) synthesis and extracellular matrix organization. FGD5-AS1 expression was significantly reduced in peripheral blood mononuclear cells of PAH patients and inversely correlated wit

Genomic rearrangements can drive cancer through mechanisms that extend beyond classical oncogenic fusions such as BCR-ABL A substantial fraction of these events involve long noncoding RNAs (lncRNAs), yet their functional impact on tumorigenesis has remained largely opaque. The lncRNA plasmacytoma variant translocation 1 (PVT1), positioned adjacent to MYC at chromosome 8q24, is among the most frequently altered loci in MYC-driven (MYC+) cancers. We recently showed that PVT1 translocations produce a characteristic asymmetric architecture that preserves the 5'-PVT1 region, generating a circular R

Influenza A virus (IAV) remains a major threat to human and animal health, highlighting the need for efficient approaches to identify host factors and antivirals. Reporter viruses are critical tools for these efforts, but incorporating large reporter genes often compromises viral fitness and genetic stability. Here, we developed a replication competent reporter IAV by fusing an 11-amino acid HiBiT tag into the nonstructural protein 1 (NS1) of the A/WSN/1933 (H1N1) backbone. The modified virus retained parental virion morphology, comparable replication kinetics, and in vivo tissue tropism, whil

BACKGROUND: The activation of anti-tumor immunity by tumor antigens is central to cancer immunotherapy. Cryptic peptides from non-coding regions represent potential antigens, with long non-coding RNAs (lncRNAs) emerging as a novel source of such peptides. This study aims to demonstrate that specific lncRNAs overexpressed in immunologically "cold" tumors-breast cancer, encode immunogenic peptides capable of activating anti-tumor immune responses.
METHODS: We identified elevated colorectal neoplasia differentially expressed (CRNDE) expression in breast cancer via database mining and polymerase

Micropeptides encoded by small open reading frames (smORFs) within long noncoding RNAs (lncRNAs) have emerged as important regulators of various biological processes. However, whether micropeptides mitigate lung tissue damage in chronic obstructive pulmonary disease (COPD) remains poorly understood. In COPD, the epithelial-mesenchymal transition (EMT) of type II alveolar epithelial (ATII) cells plays a crucial role in lung injury, contributing to fibrosis and airway remodeling. Modulating EMT could offer a promising therapeutic strategy for COPD. In this study, we identified a micropeptide, PE

Immune checkpoint blockade has achieved remarkable success in cancer treatment; however, enhancing its efficacy remains a challenge. Here we identified an immunoregulatory micropeptide encoded by the long noncoding RNA USP30-AS1 gene, highly expressed in tumor-associated macrophages. The so-designated UEIS (USP30-AS1-encoded immune suppressor) drives macrophages toward a protumorigenic phenotype, thereby inhibiting antitumor T cell immunity. Mechanistically, UEIS is induced in macrophages by cGAS-STING-type I interferon signaling at a relatively late stage following tumoral DNA stimulation, an

OBJECTIVE: While carboplatin remains a cornerstone chemotherapeutic agent for ovarian carcinoma, the emergence of resistance constitutes a major contributor to therapeutic failure and compromised clinical outcomes. This study elucidates molecular mechanisms driving carboplatin resistance and identifies novel therapeutic targets for carboplatin-refractory malignancies through characterization of a long non-coding RNA (lncRNA)-encoded micropeptide.
MATERIAL AND METHODS: Ribosome profiling analysis delineated translational alterations induced by carboplatin in ovarian cancer models. Mechanistic

Micropeptides are endogenous peptides translated from alternative open reading frames (alt-ORFs) within coding or non-coding genes. Emerging evidence suggests that some micropeptides play critical roles in both physiological and pathological processes. Multiple myeloma (MM), a haematological malignancy, remains incurable due to frequent relapses and a limited understanding of its underlying mechanisms. In this study, we sought to investigate the function and molecular mechanism of a novel micropeptide in MM pathogenesis. We identified a novel micropeptide, altH19, encoded by the lncRNA H19, wh

In most cells, Malat1 long noncoding RNA localizes to the nucleus where it affects splicing and chromatin function. In mouse neurons, Malat1 is exported to the cytoplasm where it is translated to generate the M1 micropeptide. Here, we characterize an internal ribosome entry site (IRES) required for M1 translation that is sufficient to induce cap-independent expression of a downstream open reading frame. In vivo chemical probing and structural modeling defined a three-stem-loop RNA secondary structure that encompasses the active IRES segment and coincides with a mouse B1 SINE (short intersperse

Long considered transcriptional noise, noncoding RNAs (ncRNAs), including microRNAs and long noncoding RNAs (lncRNAs), are now recognized as central regulators of cellular function, acting as scaffolds, structural elements, and regulators of gene expression. This expanding functional landscape is reshaping our understanding of cancer biology, immune regulation, and the limits of translation itself. Beyond gene regulation, lncRNAs may be implicated in the earliest stages of oncogenic transformation, orchestrating molecular reprogramming and remodeling of the immune microenvironment in premalign

Long noncoding RNAs (lncRNA) have been implicated in many diseases, including cancer. Although these disease-associated effects have been mostly attributed to the ability of lncRNAs to function as regulatory noncoding transcripts, there is growing evidence that lncRNAs may also encode functional micropeptides. In the current issue of Cancer Research, Wu and colleagues report a micropeptide encoded by a Y chromosome-linked lncRNA that may explain the higher incidence of esophageal cancer in male smokers. Furthermore, this report provides broader insights related to the molecular epidemiology of

Metastasis and therapeutic resistance remain the principal causes of cancer-related mortality, reflecting the failure of current therapies to eradicate disseminated and treatment-refractory tumor cells. Long non-coding RNAs, once considered transcriptional noise, have now emerged as pivotal regulators of cancer progression, acting through diverse mechanisms to modulate signaling pathways, transcriptional programs, tumor metabolism, and the tumor microenvironment. Accumulating evidence demonstrates that lncRNAs orchestrate epithelial-mesenchymal transition, metabolic reprogramming, and immune e

Marine biofouling plagues all maritime industries at vast economic and environmental cost. Previous and most current methods to control biofouling have employed highly persistent toxins and heavy metals, including tin, copper, and zinc. These toxic methods are resulting in unacceptable environmental harm and are coming under immense regulatory pressure. Eco-friendly alternatives are urgently required to effectively mitigate the negative consequence of biofouling without causing collateral harm. Amphiphilic micropeptides have recently been shown to exhibit excellent broad-spectrum antifouling a

Breast cancer continues to pose a public health problem and is the second leading cause of cancer-related mortality among women globally. Despite advances in our understanding of cancer biology and the development of treatment strategies, the therapeutic response is not often satisfactory as treatment efficacy is limited by severe side effects and drug resistance. Therefore, there is an urgent need for innovative approaches to improve diagnosis, prognosis, and treatment of breast cancer. Recent advances in bioinformatics and proteomics have enabled the discovery of a novel class of small prote

Microproteins are small polypeptides translated from short open reading frames (sORFs) that typically encode < 100 amino acids. Advances in ribosome profiling, mass spectrometry, and computational prediction have revealed a growing number of microproteins that play important roles in cellular metabolism, organelle function, and stress adaptation; however, these were considered non-coding or functionally insignificant. At the mitochondrial level, microproteins, such as MTLN (also known as mitoregulin/MOXI) and BRAWNIN, contribute to lipid oxidation, oxidative phosphorylation efficiency, and res

To investigate the roles of long non-coding RNAs (lncRNAs) and potential micropeptides encoded by these lncRNAs in influenza A virus (IAV) replication, we analyzed the data obtained from RNA-seq and Ribo-seq. We found that the lncRNA zinc finger antisense 1 (ZFAS1) was significantly up-regulated by IAV infection. This finding was confirmed by in vitro experiments, which showed that IAV infection caused a significant increase in the expression of ZFAS1, with the effect exhibiting a dose- and time-dependent relationship in response to the viral infection. This up-regulation was also observed in

Parkinson's disease (PD) is the second most common neurodegenerative disorder and mainly occurs in people above the age of 60 years. It is defined by the progressive degeneration of dopaminergic neurons of the substantia nigra pars compacta, which results in the classic motor symptoms. Though aggregation of alpha-synuclein and Lewy body formation are still the core of the disease pathogenesis, PD pathogenesis is complex with mitochondrial dysfunction, oxidative stress, neuroinflammation, impaired autophagy, and endoplasmic reticulum (ER)-Golgi stress. Of these, mitochondrial dysfunction has be

Recent advances in genomics uncovered a large number of microproteins, which are peptides of less than 100 amino-acids encoded by small open reading frames. In contrast to their identification, the validation of the functions of microproteins remains challenging. Especially, what are their biological functions in the cell and how this relates to disease conditions are still largely unknown. Although microproteins ensure a plethora of cellular functions, recent evidence demonstrate that they may disproportionately affect cellular metabolism. In this review, we will address the roles of mitochon
