Visualization of neuritic plaques in Alzheimer's disease by polarization-sensitive optical coherence microscopy.

One major hallmark of Alzheimer's disease (AD) and cerebral amyloid angiopathy (CAA) is the deposition of extracellular senile plaques and vessel wall deposits composed of amyloid-beta (Aβ). In AD, degeneration of neurons is preceded by the formation of Aβ plaques, which show different morphological forms. Most of them are birefringent owing to the parallel arrangement of amyloid fibrils. Here, we present polarization sensitive optical coherence microscopy (PS-OCM) for imaging mature neuritic Aβ plaques based on their birefringent properties. Formalin-fixed, post-mortem brain samples of advanc

Nanoparticle size determination via through-focus scanning polarized microscopy.

Accurate qualitative and quantitative characterization of sub-micron nanoparticles with a high efficiency is highly desirable across a wide range of scientific and industrial applications. In this study, we introduce through-focus scanning polarized microscopy (TSPM) for the estimation of nanoparticle parameters, where physical properties of nanoparticles are related to in-focus and out-of-focus polarization-resolved images. The feasibility of TSPM is rigorously demonstrated through a combination of numerical simulation and experimental measurements. Owing to its practical advantages, TSPM is

A CNN-GNN Approach for Polarity Vectors Prediction in 3D Microscopy Images.

The polarity between nuclei and Golgi is an important aspect of cellular division, migration and signaling. For example, nucleus-Golgi polarity significantly impacts angiogenesis, the physiological process in which new blood vessels develop from pre-existing vessels. Therefore, estimating polarity between nuclei and Golgi is crucial to understanding complex cellular mechanisms.Previous approaches for the prediction of nucleus-Golgi polarity vectors use simple versions of bipartite matching algorithms. These methods, while effective, fail to consider other important interactions between nuclei

Hair abnormality in Netherton syndrome observed under polarized light microscopy.

BACKGROUND: Trichorrhexis invaginata, the main diagnostic feature of Netherton syndrome, is often difficult to detect, especially in adult patients. OBJECTIVE: We sought to describe a characteristic feature of hairs in Netherton syndrome using a polarized light microscope and the underlying histopathologic changes. METHODS: Hairs obtained from 8 patients with Netherton syndrome were observed under polarized light, and we evaluated the correlation between number of band-like patterns and disease severity. RESULTS: Under polarized microscopy, the hair shafts of 8 patients showed a characteris

Pathological crystal imaging with single-shot computational polarized light microscopy.

Pathological crystal identification is routinely practiced in rheumatology for diagnosing arthritis disease such as gout, and relies on polarized light microscopy as the gold standard method used by medical professionals. Here, we present a single-shot computational polarized light microscopy method that reconstructs the transmittance, retardance and slow-axis orientation of a birefringent sample using a single image captured with a pixelated-polarizer camera. This method is fast, simple-to-operate and compatible with all the existing standard microscopes without extensive or costly modificati

Collagen reorganization in cartilage under strain probed by polarization sensitive second harmonic generation microscopy.

Type II collagen fibril diameters in cartilage are beneath the diffraction limit of optical microscopy, which makes the assessment of collagen organization very challenging. In this work we use polarization sensitive second harmonic generation (P-SHG) imaging to map collagen organization in articular cartilage, addressing in particular its behaviour under strain and changes which occur in osteoarthritis. P-SHG yields two parameters, molecular order and orientation, which provide measures of the degree of organization both at the molecular scale (below the diffraction limit) and above a few hun

In vivo optical microscopy of peripheral nerve myelination with polarization sensitive-optical coherence tomography.

Assessing nerve integrity and myelination after injury is necessary to provide insight for treatment strategies aimed at restoring neuromuscular function. Currently, this is largely done with electrical analysis, which lacks direct quantitative information. In vivo optical imaging with sufficient imaging depth and resolution could be used to assess the nerve microarchitecture. In this study, we examine the use of polarization sensitive-optical coherence tomography (PS-OCT) to quantitatively assess the sciatic nerve microenvironment through measurements of birefringence after applying a nerve c

Polarized Versus Diffractive Microscopy in Distinguishing Scarring From Non-Scarring Alopecia.

BACKGROUND: The histologic diagnosis of alopecia can be challenging. Polarized light microscopy and diffractive microscopy can be performed without added cost or the need for special stains. We sought to determine if they can be of value in the diagnosis of alopecia biopsy specimens. METHODS: We retrospectively reviewed 152 deidentified alopecia scalp biopsy slides (80 non-scarring and 72 scarring) with polarized and diffractive microscopy and analyzed the performance of each method. RESULTS: Fibrous tracts were evaluable in 150 of the 152 specimens. One was excluded as the dermis was comple

Nanoscale Membrane Budding Induced by CTxB and Detected via Polarized Localization Microscopy.

For endocytosis and exocytosis, membranes transition among planar, budding, and vesicular topographies through nanoscale reorganization of lipids, proteins, and carbohydrates. However, prior attempts to understand the initial stages of nanoscale bending have been limited by experimental resolution. Through the implementation of polarized localization microscopy, this article reports the inherent membrane bending capability of cholera toxin subunit B (CTxB) in quasi-one-component-supported lipid bilayers. Membrane buds were first detected with <50 nm radius, grew to >200 nm radius, and extended

Optical Density and Polarized Light Microscopy to confirm calcification of Intra-ocular Lens.

INTRODUCTION: Intra-ocular lens (IOL) calcification is a rare yet serious complication, presenting as clouding within the optics of IOL and necessitating IOL exchange. PATIENT AND CLINICAL FINDINGS: In this case, a 77-year-old male experienced gradual vision loss in his left eye (LE) over four years post-cataract surgery a decade before. His best corrected visual acuity (BCVA) was 3/60 in the LE, with the anterior segment displaying a clear cornea but an opacified IOL within the capsular bag. Ultrasound biomicroscopy (UBM) revealed hyperechoic reflections from the IOL in the LE. ImageJ softwa

Automated measurement of lamellar thickness in human bone using polarized light microscopy.

Lamellar bone formed in individuals with moderate and severe osteogenesis imperfecta (OI) is often composed of lamellae that are structurally abnormal. Measuring the thickness of these lamellae can be helpful in assessing the effect of specific collagen and collagen-related mutations on OI bone synthesis. Manual measurement of lamellar thicknesses in large quantities is very time consuming. The method for automated measurement described in this article utilizes an image processing script to identify the average thickness of multiple lamellae automatically from histologic images of bone. This a

Theoretical mathematics, polarized light microscopy and computational models in healthy and pathological bone.

The needs of everyday life, such as counting and measuring, are roots of theoretical mathematics. I believe these roots are why mathematical ideas ground research so amazingly well within many scientific fields. Initially trained as a theoretical mathematician and having collaborated with non-mathematicians in the field of bone research, I address the advantages and challenges of collaborations across fields of research among investigators trained in different disciplines. I report on the mathematical ideas that have guided my research on the mechanics of bone tissue. I explain how the mathema

Discovery of a novel fibrous tissue in the spinal pia mater by polarized light microscopy.

Abstract It is very well known that spinal meninges are composed of three layers, dura, arachnoid and pia mater, and that the main components of pia mater are collagen and reticular fibers. However, the distribution of those fibers has not been extensively investigated but just described as a mesh of fibers. In this study, we detected novel structures, which are composed of unidirectionally arranged fibers, in a rat spinal pia mater by using a polarized light microscope. They were seen as three parallel lines, one of which ran along a posterior spinal vein and the rest two of which ran along a

Collagen fibres in the wall of odontogenic keratocysts: a study with picrosirius red and polarizing microscopy.

The collagen in the walls of 15 keratocysts was studied histochemically by staining sections with picrosirius red and examining them with polarizing microscopy. This was compared to 15 cases of dentigerous cyst and 15 cases of radicular cyst. Polarization colours of the collagen fibres were recorded according to their width. No differences were found between the polarization colours of thin fibres (<0.8 microm) in all three lesions; the polarization colours of thick fibres (1.6-2.4 microm) in keratocysts were significantly more greenish-yellow when compared with those of dentigerous cysts and

Wavelength and polarization optimization through Stokes parameters for second harmonic generation microscopy in corneal tissue.

Second harmonic generation (SHG) microscopy has experienced a research expansion from fundamental collagen studies in tendon and skin to a growing body of work on ocular tissues, where the label-free and high-resolution aspects have offered unique possibilities. Nevertheless, polarization states and excitation wavelength have not been investigated for corneal samples. Here, we present a comprehensive investigation of SHG microscopy applied to fibrillar collagen from murine corneal tissue. Emphasis is given to polarization control, cross-polarization effects, and excitation wavelength optimizat

Heterogeneity and Birefringence of Soft Tissues Probed by the Polarization-Sensitive Terahertz Solid Immersion Microscopy.

The polarization-sensitive THz solid immersion (SI) microscope is applied to quantify the heterogeneity and birefringence of freshly-excised rat tissues: the abdominal and tongue muscles, tendons of the flexor digitorum and extensor digitorum of the forelimb, Achilles tendon, abdominal aorta, and Corpus callosum of the brain. THz refractive indices of tissues are retrieved for the two orthogonal linear polarizations of the incident beam, which are collinear and transverse toward the fibrous structures. The observed THz birefringence, as high as a few percent, is attributed to tissue morphology

Polychromatic Polarization Microscopy Differentiates Collagen Fiber Signatures in Benign Pancreatic Tissue and Pancreatic Ductal Adenocarcinoma.

The orientation of collagen fibers in relation to malignant epithelium is known to carry prognostic information in a variety of tissues. The data are the strongest for breast and pancreatic ductal adenocarcinoma. However, information inherent in collagen fiber topology in malignant tissues remains untapped in daily surgical pathology practice, largely because collagen fibers within areas of desmoplasia cannot be resolved with standard diagnostic microscopy. The methodologies used to visualize collagen fiber orientation are either of insufficient resolution to consistently capture collagen fibe

Polarized Light Microscopy for Quantitative Assessment of Sarcomere-Scale Structural Order in Cardiac Tissue.

Polarization-resolved imaging provides a powerful, label-free means of visualizing anisotropic structural order in biological tissue, yet its quantitative exploitation remains limited in routine microscopy. Here, we present a polarization-resolved imaging approach based on polarized light microscopy (PLM) that enables automated, quantitative extraction of sarcomere-scale structural metrics from unstained cardiac tissue sections. Using experimental rat models of acute cardiorespiratory arrest as controlled test systems, we demonstrate that polarization-derived intensity profiles encode reproduc
