Katanin-mediated severing generates microtubules during neurite outgrowth.

Neurons contain a polarized and staggered microtubule array that is important for supporting intracellular transport. How these microtubules are generated and oriented during development is not fully understood. Here we have taken advantage of the low microtubule density in the C. elegans PVD neuron to observe in-vivo microtubule dynamics during neurite outgrowth. We found that individual microtubules are added to outgrowing neurites via microtubule severing events, which generate new microtubules while maintaining existing polarity. We further show that the Katanin enzyme complex is specifica

Posttranslational modification of microtubules by the MATCAP detyrosinase.

The detyrosination-tyrosination cycle involves the removal and religation of the C-terminal tyrosine of α-tubulin and is implicated in cognitive, cardiac, and mitotic defects. The vasohibin-small vasohibin-binding protein (SVBP) complex underlies much, but not all, detyrosination. We used haploid genetic screens to identify an unannotated protein, microtubule associated tyrosine carboxypeptidase (MATCAP), as a remaining detyrosinating enzyme. X-ray crystallography and cryo-electron microscopy structures established MATCAP's cleaving mechanism, substrate specificity, and microtubule recognition

Alveolar proteins stabilize cortical microtubules in Toxoplasma gondii.

Single-celled protists use elaborate cytoskeletal structures, including arrays of microtubules at the cell periphery, to maintain polarity and rigidity. The obligate intracellular parasite Toxoplasma gondii has unusually stable cortical microtubules beneath the alveoli, a network of flattened membrane vesicles that subtends the plasmalemma. However, anchoring of microtubules along alveolar membranes is not understood. Here, we show that GAPM1a, an integral membrane protein of the alveoli, plays a role in maintaining microtubule stability. Degradation of GAPM1a causes cortical microtubule disor

Polarity sorting of axonal microtubules: a computational study.

We present a computational model to test a "polarity sorting" mechanism for microtubule (MT) organization in developing axons. We simulate the motor-based axonal transport of short MTs to test the hypothesis that immobilized cytoplasmic dynein motors transport short MTs with their plus ends leading, so "mal-oriented" MTs with minus-end-out are transported toward the cell body while "correctly" oriented MTs are transported in the anterograde direction away from the soma. We find that dynein-based transport of short MTs can explain the predominately plus-end-out polarity pattern of axonal MTs bu

Spin probe analysis of microtubules structure and formation.

Microtubules (MTs) control cell replication, material transport and motion in eukaryotic cells, but MT role in several pathologies is still unknown. These functions are related to the MT physico-chemical properties and MT formation mode starting from tubulin molecules. This study describes a new method, based on the computer aided analysis of the electron paramagnetic resonance (EPR) spectra of selected spin probes to obtain structural and dynamical information on tubulins and MTs and the kinetics of MTs formation promoted by guanosine-5'-triphosphate (GTP). It was found that tubulin and MTs a

Alcoholic hyalin, microfilaments and microtubules in alcoholic hepatitis.

The alcoholic hyalin which is composed of light and dark occasionally swollen and conglomerating filaments, is found to be surrounded by proliferated RER, hypertrophied Golgi apparatus and mitochondria containing enlarged matrical granules. Besides, microfilaments and some microtubules are seen in relation to the hyaline bodies. In biopsies where hyaline bodies are scarce, hepatocytes without alcoholic hyalin present similar changes and the Golgi apparatus is seen to contain very low density lipoprotein-like particles. These ultrastructural changes are suggested to be related to an early stage

A polarized multicomponent foundation upholds ciliary central microtubules.

Cilia's back-and-forth beat pattern requires a central pair (CP) of microtubules. However, the mechanism by which the CP is upheld above the transition zone (TZ) remains unclear. Here, we showed that a rod-like substructure marked by Cep131 and ciliary Centrin serves as a polarized CP-supporting foundation. This CP-foundation (CPF) was assembled independently of the CP during ciliogenesis in mouse ependymal cells. It protruded from the distal end of the basal body out of the TZ to enwrap the proximal end of the CP. Through proximity labeling, we identified 26 potential CPF components, among wh

Microtubules orchestrate local translation to enable cardiac growth.

Hypertension, exercise, and pregnancy are common triggers of cardiac remodeling, which occurs primarily through the hypertrophy of individual cardiomyocytes. During hypertrophy, stress-induced signal transduction increases cardiomyocyte transcription and translation, which promotes the addition of new contractile units through poorly understood mechanisms. The cardiomyocyte microtubule network is also implicated in hypertrophy, but via an unknown role. Here, we show that microtubules are indispensable for cardiac growth via spatiotemporal control of the translational machinery. We find that th

Microtubules self-repair in response to mechanical stress.

Microtubules--which define the shape of axons, cilia and flagella, and provide tracks for intracellular transport--can be highly bent by intracellular forces, and microtubule structure and stiffness are thought to be affected by physical constraints. Yet how microtubules tolerate the vast forces exerted on them remains unknown. Here, by using a microfluidic device, we show that microtubule stiffness decreases incrementally with each cycle of bending and release. Similar to other cases of material fatigue, the concentration of mechanical stresses on pre-existing defects in the microtubule latti

Spastin and microtubules: Functions in health and disease.

SPG4, the gene encoding for spastin, a member of the ATPases associated with various cellular activities (AAA) family, is mutated in around 40% of cases of autosomal dominant hereditary spastic paraplegia (AD-HSP). This group of neurodegenerative diseases is characterized by a progressive spasticity and lower limb weakness with degeneration of terminal axons in cortico-spinal tracts and dorsal columns. Spastin has two main domains, a microtubule interacting and endosomal trafficking (MIT) domain at the N-terminus and the C-terminus AAA domain. Early studies suggested that spastin interacts wit

Axons regulate the distribution of Schwann cell microtubules.

It is well established that axons regulate Schwann cell phenotype. The purpose of the present study was to determine whether axons influence the arrangement of Schwann cell microtubules (MTs). Using double-labeling immunocytochemistry and confocal microscopy, we show that MTs in undifferentiated Schwann cells are nucleated from and attached to a single MT organizing center (MTOC) that is associated with the centrosome. Physical contact with appropriate axons initiates a myelin-forming phenotype that disperses MT minus ends and induces multiple MT-nucleating sites in Schwann cell perinuclear cy

Linking axonal degeneration to microtubule remodeling by Spastin-mediated microtubule severing.

Mutations in the AAA adenosine triphosphatase (ATPase) Spastin (SPG4) cause an autosomal dominant form of hereditary spastic paraplegia, which is a retrograde axonopathy primarily characterized pathologically by the degeneration of long spinal neurons in the corticospinal tracts and the dorsal columns. Using recombinant Spastin, we find that six mutant forms of Spastin, including three disease-associated forms, are severely impaired in ATPase activity. In contrast to a mutation designed to prevent adenosine triphosphate (ATP) binding, an ATP hydrolysis-deficient Spastin mutant predicted to rem

Ultrastructural studies of microtubules and microtubule organizing centers of the vertebrate olfactory neuron.

The olfactory neuron is specialized along its length into highly determined morphological regions. These regions include the dendritic cilia, dendritic vesicle, dendritic shaft proper, perikaryon, axon, zone of transition where the axon widens as it approaches its termination, and the axon terminal. Except for the zone of transition and the terminal, characteristic populations of microtubules occur in these compartments. In the olfactory vesicle, three discrete microtubule organizing centers (MTOCs) nucleate microtubules: the basal body, the lateral foot associated with the body, and dense mas

The microtubule cytoskeleton at the synapse.

In neurons, microtubules (MTs) provide routes for transport throughout the cell and structural support for dendrites and axons. Both stable and dynamic MTs are necessary for normal neuronal functions. Research in the last two decades has demonstrated that MTs play additional roles in synaptic structure and function in both pre- and postsynaptic elements. Here, we review current knowledge of the functions that MTs perform in excitatory and inhibitory synapses, as well as in the neuromuscular junction and other specialized synapses, and discuss the implications that this knowledge may have in ne

Parasite microtubule arrays.

Microtubules are a key component of eukaryotic cell architecture. Regulation of the dynamic growth and shrinkage of microtubules gives cells their shape, allows cells to swim, and drives the separation of chromosomes. Parasites have developed intriguingly divergent biology, seemingly expanding upon and reinventing microtubule use in fascinating ways. These organisms affect life on the planet at scales that are often overlooked: there are likely more parasitic than free-living organisms on Earth, and they have a sizeable influence across ecosystems. As parasites can cause devastating diseases,

Chimeric microtubule disruptors.

A chimeric approach is used to discover microtubule disruptors with excellent in vitro activity and oral bioavailability; a ligand-protein interaction with carbonic anhydrase that enhances bioavailability is characterised by protein X-ray crystallography. Dosing of a representative chimera in a tumour xenograft model confirms the excellent therapeutic potential of the class.

How short and dense are axonal microtubules? Approaches for analyzing microtubule organization and dynamics.

Axons are cable-like extensions of neurons that conduct nerve impulses and relay information to subsequent cells at synapses. Their length varies dramatically, from micrometers in small invertebrates to over a meter in some neurons of large vertebrates. The cytoskeleton is the most important internal structure that determines neuronal morphology. Microtubules, in particular, play a crucial role in maintaining axonal structure, transport, and function. The classical view of axonal microtubules as long, continuous, and stable polymers extending over vast distances is being challenged by evidence

Scaling of microtubule spirals driven by dyneins: collective effect of microtubule and motor mechanics.

Molecular motor-driven spirals of actin filaments pinned at one end while driven along their length were seen to be scale-independent. The spiral radius and frequency dependence on force was predicted to follow a universal law, validated by actin-myosin experiments. However, the theory was primarily compared to acto-myosin spirals, assuming no effect of filament length and that motor velocity is additive with density. Here, we reconstitute microtubule spirals driven by dynein transport and end-pinning and find that spiral size scales with force with an exponent α approximately -1/3, consistent

Coordination of Zika Virus Infection and Viroplasm Organization by Microtubules and Microtubule-Organizing Centers.

Zika virus (ZIKV) became a global health concern in 2016 due to its links to congenital microcephaly and other birth defects. Flaviviruses, including ZIKV, reorganize the endoplasmic reticulum (ER) to form a viroplasm, a compartment where virus particles are assembled. Microtubules (MTs) and microtubule-organizing centers (MTOCs) coordinate structural and trafficking functions in the cell, and MTs also support replication of flaviviruses. Here we investigated the roles of MTs and the cell's MTOCs on ZIKV viroplasm organization and virus production. We show that a toroidal-shaped viroplasm form
