Viruses Teaching Immunology: Role of LCMV Model and Human Viral Infections in Immunological Discoveries.

Virology has played an essential role in deciphering many immunological phenomena, thus shaping our current understanding of the immune system. Animal models of viral infection and human viral infections were both important tools for immunological discoveries. This review discusses two immunological breakthroughs originally identified with the help of the lymphocytic choriomeningitis virus (LCMV) model; immunological restriction by major histocompatibility complex and immunotherapy using checkpoint blockade. In addition, we discuss related discoveries such as development of tetramers, viral es

AI-Driven BCR Modeling for Precision Immunology.

The B cell receptor (BCR) repertoire captures an individual's immunological history and antigen-driven evolution within a vast, high-dimensional sequence space. Although bulk and single-cell adaptive immune receptor repertoire sequencing (AIRR-seq) now enables deep profiling of BCR diversity, interpreting these datasets remains challenging due to strong inter-individual heterogeneity, nonlinear sequence-structure-function relationships, dynamic clonal evolution, and the rarity of functionally relevant clones. Artificial intelligence (AI) provides a conceptual and computational framework for ad

Immunological Analysis of Cryptococcal Meningoencephalitis in a Murine Model.

Cryptococcal meningoencephalitis (CM), caused by the fungal pathogen Cryptococcus neoformans species complex, can lead to high mortality or severe neurological sequelae in survivors that are associated with excessive inflammation in the central nervous system (CNS), especially in those who develop immune reconstitution inflammatory syndrome (IRIS) or postinfectious immune response syndrome (PIIRS). While the means to establish a cause-and-effect relationship of a specific pathogenic immune pathway during CM by human studies are limited, mouse models allow dissection of the potential mechanisti

Acute on chronic bronchitis: A model of mucosal immunology.

Acute bronchitis has been studied as a model of disturbed mucosal immunoregulation. A new hypothesis relating to the pathogenesis of acute bronchitis has been developed, based on altered host response as the prime mover. Infection-prone subjects had low levels of lysozyme. Effective oral immunization, especially if early, reduced levels of bacterial colonization. Future attention focuses on intra bronchial inflammation and its link to the host-parasite relationship.

A rat model of Immunologic and hypertensive kidney injury.

Non-surgical rodent chronic kidney disease (CKD) models for both glomerular and tubular injuries are currently limited. The current study aimed to develop a rat model of CKD by combining anti-Fx1A with N(ω)-Nitro-L-Arginine Methyl Ester (L-NAME) administrations. Rats were assigned to groups receiving L-NAME, anti-Fx1A, anti-Fx1A + L-NAME, or vehicle. Renal function, stiffness, renal injury biomarkers, histopathology and renal genome-wide transcriptomic changes were evaluated. Protein and renal injury biomarker levels in urine were elevated in the anti-Fx1A alone and combination group. Shear wa

Immunological phenotype and skin modeling of Netherton syndrome.

Netherton syndrome (NTS) is a syndromic ichthyosis characterized by scaling, erythroderma, and proneness towards allergies and infections. It is caused by mutations in the SPINK5 gene, encoding the lympho-epithelial Kazal-type 5 related inhibitor (LEKTI), which regulates the activity of kallikreins in the skin. Treatment options are limited, insufficient, time-consuming and a financial burden for the patients. In the last years, genotype-phenotype correlations in NTS and immunological phenotypes have been delineated. Psoriasiform patterns have been described. Moreover, the literature reports o

Animal models of the immunology and pathogenesis of human babesiosis.

Animal models of human babesiosis have provided a basic understanding of the immunological mechanisms that clear, or occasionally exacerbate, Babesia infection and those pathological processes that cause disease complications. Human Babesia infection can cause asymptomatic infection, mild to moderate disease, or severe disease resulting in organ dysfunction and death. More than 100 Babesia species infect a wide array of wild and domestic animals, and many of the immunologic and pathologic responses to Babesia infection are similar in animals and humans. In this review, we summarize the knowled

Maximizing the immunological output of the cervicovaginal explant model.

In the field of sexually transmitted infections (STI), the cervicovaginal explant (CVEx) model, not only provides the opportunity to study the different immunological arms present in these tissues under steady state conditions, but also their response against ex vivo infection with relevant pathogens. The methodology associated to the establishment of the HIV infection model in the cervicovaginal tissue was described in detail by Grivel et al. earlier (Grivel and Margolis, 2009). With this model as a foundation, we illustrate different approaches to obtain a large number of immunological reado

Immunological control of congenital toxoplasmosis in the murine model.

Toxoplasmosis is a serious disease in humans where it can cause abortion or congenital infection if a women is exposed to disease for the first time during pregnancy. Infection prior to pregnancy normally results in immunity and which is capable of protecting the foetus. Similar observations have been made in the BALB/c mouse indicating the potential of mice for studying congenital disease. Consequently, the mouse has been used to study how mammals balance the opposing needs of maintaining an immunological environment conducive to successful pregnancy while attempting to control a dangerous pa

Immunological dynamics in orthotopic compared with subcutaneous murine models of HPV-positive oropharyngeal cancer.

The necessity of reliable preclinical models for evaluating the efficacy of novel therapeutic strategies is imperative. Nevertheless, the degree to which tumor-bearing murine models represent the immunological characteristics of human papillomavirus (HPV)-positive oropharyngeal squamous cell carcinoma (OPSCC) has largely been unexplored. By utilizing single-cell RNA sequencing technology, our research elucidated that subcutaneous (SC) murine models more accurately reflect the early immunogenic phase of human HPV-positive OPSCC, marked by a stage-dependent increase in effector T-cell infiltrati

Therapeutic efficacy and immunological response of CCL5 antagonists in models of contact skin reaction.

Skin-infiltrating T-cells play a predominant role in allergic and inflammatory skin diseases such as atopic dermatitis, psoriasis and allergic contact dermatitis. These T-cells are attracted by several chemotactic factors including the chemokine CCL5/RANTES, a CC chemokine inducing both the migration and activation of specific leukocyte subsets. CCL5 has been found to be associated with various cell-mediated hypersensitive disorders such as psoriasis, atopic dermatitis and irritant contact dermatitis. We have used two antagonists, the first, Met-CCL5, a dual CCR1/CCR5 antagonist and the second

Engineering autoimmune disease models using organoids: Harnessing microenvironmental engineering for precision medicine and immunological recapitulation.

Autoimmune diseases-including systemic sclerosis (SSc), systemic lupus erythematosus (SLE), and rheumatoid arthritis (RA)-are increasingly understood as programmable microenvironmental states, wherein evolving changes in matrix mechanics, barrier integrity, interferon and cytokine networks, immune-complex deposition, and stromal-immune reciprocity progressively reshape tissue behavior. These shifting axes generate nonlinear trajectories of fibrosis, vascular injury, and joint destruction that static or reductionist in vitro systems fail to recapitulate. Organoid and organ-on-a-chip platforms n

Review of immunological plasma markers for longitudinal analysis of inflammation and infection in rat models.

Disease or trauma of orthopedic tissues, including osteomyelitis, osteoporosis, arthritis, and fracture, results in a complex immune response, leading to a change in the concentration and milieu of immunological cells and proteins in the blood. While C-reactive protein levels and white blood cell counts are used to track inflammation and infection clinically, controlled longitudinal studies of disease/injury progression are limited. Thus, the use of clinically-relevant animal models can enable a more in-depth understanding of disease/injury progression and treatment efficacy. Though longitudin

Dual Immunological Prognostic Models for Risk Stratification and Treatment Insights in Triple-Negative Breast Cancer.

Triple-negative breast cancer (TNBC) represents the most aggressive breast cancer subtype, with its highly heterogeneous tumor microenvironment posing substantial challenges for precision diagnosis and therapy. To address this, we aim to construct a novel prognostic framework based on tumor-immune interactions. Through integrative analysis of single-cell RNA sequencing data from 30 TNBC samples (106,132 cells), we identify key tumor expression metaprograms and uncover their interaction with an immunosuppressive dendritic-cell subset, a process associated with the NECTIN1-NECTIN4 axis. Waldenström macroglobulinemia: clinical and immunological aspects, natural history, cell of origin, and emerging mouse models.

Waldenström macroglobulinemia (WM) is a rare and currently incurable neoplasm of IgM-expressing B-lymphocytes that is characterized by the occurrence of a monoclonal IgM (mIgM) paraprotein in blood serum and the infiltration of the hematopoietic bone marrow with malignant lymphoplasmacytic cells. The symptoms of patients with WM can be attributed to the extent and tissue sites of tumor cell infiltration and the magnitude and immunological specificity of the paraprotein. WM presents fascinating clues on neoplastic B-cell development, including the recent discovery of a specific gain-of-function

Immunologic deviations in recurrent spontaneous abortion mouse model.

BACKGROUND: Recurrent spontaneous abortion (RSA) or recurrent pregnancy loss (RPL) is an abnormality that has a great impact on women's quality of life. RSA is defined as at least three unexplained abortions (without prior live birth) occurring before the 20th week of pregnancy. AIM: The present review attempts to discuss immunologic deviations in mouse models of RSA. CONTENT: The mating of DBA/2J males with CBA/J female mice has provided specialists with a homologous model of RSA. Much of the research using the CBA/J × DBA/2J mouse model has shown immune system alteration results in rejecti

Structural and immunological effects of skin cryoablation in a mouse model.

Cryoablation is therapeutically applied for various disorders in several organs, and skin diseases are typical targets as this cryotherapy has been widely used for viral warts, benign tumors, and actinic keratosis. The main mechanisms of cryoablation consist of direct freezing effect on skin constituents, thrombosis formation in microcirculation, and subsequent immunological responses. Among them, however, the immunological mechanism remains unelucidated, and it is an issue how the direct freezing injury induces immunological consequences. We established a mouse cryoablation model with liquid
