Molecular design of hydrogen sulfide probes and their bioimaging applications.

Hydrogen sulfide (H2S) has garnered increasing attention in recent years, not only as a hazardous environmental pollutant but also as the third endogenous gas signaling molecule, alongside nitric oxide and carbon monoxide. At elevated concentrations, it poses serious threats to human health by irritating the respiratory tract and nervous system, with high-level exposure potentially leading to coma or even death. Conversely, at physiological levels, H2S participates in a variety of essential biological processes, including cardiovascular regulation, neuroprotection and cellular metabolism. Give

Molecular Tailoring of Fluorescent Agents Toward SICTERS-Active Raman Probes.

Self-stacked small molecules relying on the SICTERS mechanism have emerged as a promising platform for ultrasensitive in vivo Raman imaging. However, only a limited number of SICTERS‑active molecules have been reported to date, constraining their broader applications. Herein, we reported a rational strategy to transform conventional fluorophores into SICTERS probes based on two fundamental findings. One is that sulfur-to-selenium substitution is capable of suppressing radiative decay for fluorescence quenching, distinct from its conventional role in red-shifting emission. The other is that N-a

Nitroreductase-Activated Fluorescent Probes: From Molecular Design to Disease Diagnosis.

Nitroreductases (NTRs), a class of flavin-dependent oxidoreductases, are highly upregulated in hypoxic microenvironments and are closely associated with cancer, bacterial infections, and other hypoxia-driven diseases. Precise and real-time visualization of NTR activity is therefore critical for understanding disease mechanisms and enabling accurate diagnosis and targeted therapy. Fluorescent probes have emerged as powerful and versatile chemical biology tools for NTR sensing owing to their high sensitivity, selectivity, and spatiotemporal resolution. Recent advances in two-photon fluorescence

Small Molecular Fluorescent Probes for Alzheimer's Disease Associated Active Species.

Alzheimer's disease (AD) is a progressive neurodegenerative disorder and is the main cause of dementia worldwide. As the pathogenesis of AD is quite complicated, there is continuous attention to AD-associated active species, such as amyloid-β plaques, neurofibrillary tangles, metal ions, reactive oxygen/nitrogen/sulphur species, cholinesterase, viscosity, formaldehyde and so on. To this end, a series of small molecular fluorescent probes for these active species have been explored for early diagnosis and even remedy of AD. Herein, we systematacially summarize the versatile fluorescent probes m

Use of molecular probes in the study of endocrine diseases.

DNA hybridization techniques are now commonly used in studies of endocrine disease. Although solution and filter hybridization have been more widely used in endocrine studies, ISH offers many advantages to the pathologist. Individual cells can be visualized directly with the microscope to study the expression of specific gene products. ISH is somewhat similar to immunohistochemistry and can be set up rapidly in most histology laboratories, especially when nonradioactive biotinylated probes are used. The ability to combine ISH with immunochemical techniques offers to the pathologist the use of

[Molecular Conjugation for the "Diazido Probe" Method].

Target identification of screening hit compounds with unknown mechanisms of action obtained from chemical libraries by phenotypic assays has played an important role in the development of innovative drugs that work based on novel mechanisms. To improve the usability of the target identification based on the photoaffinity labeling method, we have studied the "diazido probe" method, wherein a photoreactive aromatic azido and relatively photostable aliphatic azido groups are sequentially used for photoreaction and introduction of a latent detectable tag via the Staudinger ligation or click reacti

Using molecular rotors to probe gelation.

A series of fluorescent probes, including a number of molecular rotors, have been used to follow the self-assembly of dipeptide-based low molecular weight gelators. We show that these probes can be used to gain an insight into the assembly process. Thioflavin T, a commonly used stain for β-sheets, appears to act as a molecular rotor in these gelling systems, with the fluorescence data closely matching that of other rotors. The molecular rotor was incorporated into an assay system with glucose oxidase to enable glucose-concentration specific gelation and hence generating a fluorescent output. A

A molecular epidemiological probe for pig microchimerism.

We have cloned and characterized a single-copy DNA sequence from the porcine alpha-1,3-galactosyltransferase gene that corresponds to a 547-base pair intron separating exons 3 and 4 of the protein coding domain. Polymerase chain reaction amplification of this sequence from flanking oligonucleotides generates a species-specific DNA probe (pgt34) capable of recognizing 50 pg chimeric template DNA at a pig to human cellular ratio of 1/10,000. Homologous DNA sequence is not identified in the macaque, baboon, or human genome by Southern hybridization. Analysis of a discordant model of pig to baboon

Inverse Mixed-Solvent Molecular Dynamics for Visualization of the Residue Interaction Profile of Molecular Probes.

To ensure efficiency in discovery and development, the application of computational technology is essential. Although virtual screening techniques are widely applied in the early stages of drug discovery research, the computational methods used in lead optimization to improve activity and reduce the toxicity of compounds are still evolving. In this study, we propose a method to construct the residue interaction profile of the chemical structure used in the lead optimization by performing "inverse" mixed-solvent molecular dynamics (MSMD) simulation. Contrary to constructing a protein-based, ato

Molecular Probes for Pancreatic Cancer Diagnosis: Recent Advances and Further Perspectives.

Pancreatic cancer (PC) is a highly aggressive malignancy of the digestive system, characterized by an annual mortality rate nearly equivalent to its incidence. Early surgical resection remains the most promising curative approach; however, approximately 90% of patients are diagnosed at an advanced stage due to its insidious onset and rapid metastasis. Currently, anatomical imaging techniques such as endoscopic ultrasound, computed tomography, and magnetic resonance imaging (MRI) have limited sensitivity and specificity for detecting small and early-stage lesions of PC. Molecular probes, which

Organic Activatable Afterglow Imaging Probes: From Molecular Design to Biomedical Translation.

Organic activatable afterglow probes have developed into a distinct class of optical imaging tools. They allow for a temporal separation between signal emission and external irradiation, thereby enabling high-contrast imaging. We have summarized the latest advances in the design of organic activated afterglow probes, with a particular focus on their excitation mechanisms, performance modulation, and activation strategies, including energy transfer, intramolecular regulation, and singlet oxygen(1O2)-mediated processes. The roles of various external energy sources, including light, ultrasound, a

A molecular trap inside microtubules probes luminal access by soluble proteins.

The uniquely hollow structure of microtubules (MTs) confers characteristic mechanical and biological properties. Although most regulatory processes take place at the outer surface, molecular events inside MTs, such as α-tubulin acetylation, also play a critical role. However, how regulatory proteins reach the site of action remains obscure. To assess luminal accessibility, we first identified luminally positioned residues of β-tubulin that can be fused to a protein of interest. We then developed a chemically inducible technique with which cytosolic proteins can be rapidly trapped at the lumen

Raman Probes for In Situ Molecular Analyses of Peripheral Nerve Myelination.

The myelinating activity of living Schwann cells in coculture with neuronal cells was examined in situ in a Raman microprobe spectroscope. The Raman label-free approach revealed vibrational fingerprints directly related to the activity of Schwann cells' metabolites and identified molecular species peculiar to myelinating cells. The identified chemical species included antioxidants, such as hypotaurine and glutathione, and compartmentalized water, in addition to sphingolipids, phospholipids, and nucleoside triphosphates also present in neuronal and nonmyelinating Schwann cells. PET Molecular Probes for Neuroinflammation in Neurodegenerative Diseases: Progress and Prospects.

Neuroinflammation is a central pathological process underlying neurodegenerative diseases, including Alzheimer disease (AD), Parkinson disease (PD), and amyotrophic lateral sclerosis. Positron emission tomography (PET) molecular probes now enable the noninvasive, quantitative visualization of neuroinflammatory processes in the living brain. This review surveys recent advances in PET probes targeting microglial activation markers─including the 18 kDa translocator protein (TSPO), the purinergic P2X7 receptor (P2X7R), colony-stimulating factor 1 receptor (CSF1R), and sphingosine-1-phosphate recep

Anomalous translational dynamics of molecular probes near the polymer glass transition.

While single-molecule (SM) experiments using fluorescent probes have provided critical insight for molecular and polymeric glass formers, the microscopic mechanism of probe transport remains largely unexplored in such systems at the molecular length scale. Here, we report SM translational dynamics of molecular probes in high molecular weight polystyrene at three temperatures near the glass transition temperature (T_{g}). The probes exhibit quasistationary position fluctuations, non-Gaussian displacement distributions, subdiffusive transport with anticorrelated displacements, and a characterist

Molecular Imaging Probes for Early Detection and Staging of Liver Fibrosis.

Liver fibrosis, a progressive condition marked by excessive extracellular matrix deposition and activation of hepatic stellate cells, often develops asymptomatically in its early stages, leading to delayed clinical intervention. Conventional imaging techniques typically fail to detect mild fibrosis, resulting in diagnosis only at advanced stages such as cirrhosis, when therapeutic efficacy is significantly compromised. Recent advances in molecular imaging have facilitated the development of targeted contrast agents that enhance diagnostic sensitivity by selectively binding to fibrosis-specific

Safirinium Fluorescent "Click" Molecular Probes: Synthesis, CuAAC Reactions, and Microscopic Imaging.

Fluorescent labeling utilizing Cu(I)-catalyzed azide-alkyne cycloaddition reactions (CuAAC) is among the leading applications of the "click" chemistry strategy. Fluorescent probes for this approach can be constructed by linking an azide or alkyne group to a fluorophore, such as the recently developed Safirinium derivatives. These compounds are water-soluble, highly fluorescent heterocycles based on 1,2,4-triazolium, with significant potential for various labeling applications, although they have not yet been converted to azide or alkyne probes. Herein, we report the synthesis of Safirinium-bas

Molecular Imaging of Nitric Oxide Surrogates with Organelle-Specific Fluorescent Probes.

Nitric oxide is an important signalling molecule responsible for maintaining body's homeostasis. Any dysregulation in NO can lead to many pathological conditions like atherosclerosis, cancers, neurodegenerative disorders, hypertension and inflammation. Several, sensing technologies are used for sensing NO. Among these, fluorescent imaging is considered to be one of the most efficient. Till date, approximately 123 fluorescent probes are reported related to nitric oxide (NO) sensing fluorescent probes for the sensitive, selective, and real-time detection of NO at both the cellular and subcellula

Chemical probes for the identification of the molecular targets of honokiol.

Honokiol is a natural product with an interesting array of biological effects, including significant anti-tumor properties. However, full exploration of its therapeutic potential is hampered by its modest pharmacokinetic profile and by the lack of synthetic methods that allow to obtain specifically designed derivatives with improved properties. In addition, the specific molecular targets of honokiol remain poorly understood, a fact that limits the search of alternative hits for subsequent optimization programs. In this work we describe an optimized series of synthetic routes that allow to acce
