Comparison of 6 Injectable Anesthetic Regimens and Isoflurane in Gray Short-tailed Opossums (Monodelphis domestica).

Gray short-tailed opossums are used in a wide variety of research in the areas of developmental biology, oncology, immunology, and comparative biology. Despite many frequent experimental manipulations of these animals under anesthesia, few studies to date have characterized the effects of anesthesia in this species. Our aim was to identify safe and effective injectable anesthetic combinations using ketamine and xylazine or ketamine and dexmedetomidine at doses of 40 mg/kg to 100 mg/kg for ketamine, 5 mg/kg to 10 mg/kg for xylazine, and 0.05 mg/kg to 0.1 mg/kg for dexmedetomidine. Pyometra with Concurrent Endometrial Adenocarcinoma in Laboratory Opossums (Monodelphis domestica): A Case Series of Four Animals.

The use of laboratory opossums (Monodelphis domestica) in biomedical research has increased since the 1980s, when the first colonies were established in the United States, due to their relatively low maintenance requirements and cyclical breeding patterns resulting in reproductive efficiency. Despite their growing use, reproductive disorders in this species remain poorly characterized. A 2.5-year-old female laboratory opossum was euthanized for humane reasons after presenting lethargy, inappetence, and a severely distended abdomen. Gross necropsy revealed marked uterine enlargement. Cytologic

Comparative genomic analysis and evolution of the T cell receptor loci in the opossum Monodelphis domestica.

BACKGROUND: All jawed-vertebrates have four T cell receptor (TCR) chains: alpha (TRA), beta (TRB), gamma (TRG) and delta (TRD). Marsupials appear unique by having an additional TCR: mu (TRM). The evolutionary origin of TRM and its relationship to other TCR remain obscure, and is confounded by previous results that support TRM being a hybrid between a TCR and immunoglobulin locus. The availability of the first marsupial genome sequence allows investigation of these evolutionary relationships. RESULTS: The organization of the conventional TCR loci, encoding the TRA, TRB, TRG and TRD chains, in

The role of androgens in development of the scrotum of the grey short-tailed Brazilian opossum ( Monodelphis domestica).

In eutherian mammals, sex differentiation is initiated by expression of the testis-determining gene on the Y chromosome. Subsequent phenotypic development of the reproductive tract and genitalia depends on the production of hormones by the differentiated testis. In marsupials the mechanisms of phenotypic development may vary from this pattern, as differentiation of the scrotal primordia has been shown to occur before that of the gonad. Thus, the development of the scrotum in the marsupial has been regarded as an androgen-independent process. We have sought to clarify the ontogeny of scrotal de

Behavioural observations of singly-housed grey short-tailed opossums (Monodelphis domestica) in standard and enriched environments.

The grey short-tailed opossum (Monodelphis domestica) has been used in biomedical research for over three decades. It is normally housed in standard rat cages and appears to have adapted well to captivity. Owing to their aggressive behaviour towards each other, adult males are normally housed singly and may spend considerable periods of time in social isolation. We wanted to carry out a preliminary study on the behaviour of singly-housed male short-tailed opossums in two different settings: a standard rat cage and an enriched floor pen. Five male opossums aged between 10 and 12 weeks were hous

Morphometric analysis of the postnatally developing marsupial lung: the gray short-tailed opossum (Monodelphis domestica).

The postnatal lung development of the gray short-tailed opossum (Monodelphis domestica) was investigated to assess the morphofunctional status in this immature born marsupial. Lung volume, surface densities and surface areas, parenchymal (air spaces and septa) and non-parenchymal (airways, blood vessels, connective tissue) volume proportions were determined in Micro-CT images using morphometry. The lung of the neonate was at the canalicular stage and consisted of large terminal air spaces and a rudimentary bronchial tree. The absolute lung volume was only 0.002 cm3 with an air space surface de

H-ras oncogene activation in invasive UVR-induced corneal sarcomas of the opossum Monodelphis domestica.

Chronic exposure to ultraviolet radiation (UVR) induces corneal sarcomas in the South American opossum Monodelphis domestica. Cell lines are readily established from these tumors. Northern blotting of mRNA from six such cell lines revealed high expression of the H-ras oncogene. H-ras cDNA from an eye tumor cell line was cloned and characterized; the germline sequence of codons 12, 13, and 61 was confirmed by examination of H-ras sequences amplified from liver DNA by the polymerase chain reaction. The Monodelphis H-ras coding sequence is 84-89% identical to that of other vertebrates at the nucl

Ultraviolet radiation-induced skin lesions in laboratory opossums (Monodelphis domestica) exposed from the weanling stage.

After repeated exposure to ultraviolet radiation, adult laboratory opossums (Monodelphis domestica) may develop hyperplasia and neoplasia of the shaved skin. We exposed opossums from the weanling stage (8-10 weeks after birth) and compared the incidence of lesions at designated time points with those observed following introduction of adults (around 5 months old) to the same irradiation protocol (125 J/m2 of UVB three times each week for up to 40 weeks). The overall incidence of hyperplasia and neoplasia among opossums exposed from the weanling stage was markedly lower than among animals first

Efferent connections of the main olfactory bulb in the opossum (Monodelphis domestica): a characterization of the olfactory entorhinal cortex in a marsupial.

Olfactory projections have been investigated for decades, but few reports using modern, sensitive neural tracers are available. In marsupials, only lesion-degeneration studies exist and they are restricted to the genera Didelphis and Trichosurus. Some of the territories described as olfactory-recipient such as the upper portion of the rhinal fissure and the vomeronasal amygdala are, however, controversial. Also, the characterization of the olfactory portion of the entorhinal cortex is far from clear in acallosal mammals. The present report investigates, using biotinylated dextran-amine, the ol

Perinatal exposure to progesterone, estradiol, or mifepristone affects sexual differentiation of behavior in opossums (Monodelphis domestica).

The effects of perinatal exposure to progesterone (P) and estradiol (E) on sexual differentiation of behavior and morphology were examined by treating male and female gray short-tailed opossums on postnatal day 8 with progesterone alone (P), P plus estradiol (E) (PE), the P receptor antagonist mifepristone/RU486 (MIF), or corn oil control (C) and gonadectomizing them before puberty. When given female hormone replacement therapy in adulthood and tested with intact stimulus males, MIF animals showed less female-typical aggressive threat behavior than animals in other treatment groups. Development of olfactory marker protein and N-CAM expression in chemosensory systems of the opossum, Monodelphis domestica.

Using olfactory marker protein (OMP) and neural cell adhesion molecule (N-CAM) immunohistochemistry, the present study describes development of olfactory and vomeronasal systems in pre- and postnatal opossums, Monodelphis domestica. As revealed by OMP expression, development of the main olfactory system precedes that of the vomeronasal system by 1-2 weeks. OMP is expressed throughout (homogeneously) the nerve and glomerular layers of the main (MOB) but is expressed more strongly (heterogeneously) in the anterior as compared to the posterior accessory (AOB) olfactory bulb. N-CAM expression is h

Species-specific, multifaceted venom resistance in Monodelphis domestica reveals novel physiological behavior of von Willebrand Factor under flow.

Interactions between predators and prey are often characterized by strong selection pressures that shape extreme physiological adaptations. Venom resistance in large-bodied South American opossums (Clade Didelphini) is a striking example, as these marsupials prey on venomous snakes and exhibit remarkable resistance to their venom. While resistance is well documented in Didelphini, relatively little is known about venom resistance in the smaller, more diverse members of Didelphidae, which inhabit the same regions and encounter the same predators. Here, we investigate venom resistance in the sma

Age-related differences in the local cellular and molecular responses to injury in developing spinal cord of the opossum, Monodelphis domestica.

Immature spinal cord, unlike adult, has an ability to repair itself following injury. Evidence for regeneration, structural repair and development of substantially normal locomotor behaviour comes from studies of marsupials due to their immaturity at birth. We have compared morphological, cellular and molecular changes in spinal cords transected at postnatal day (P)7 or P14, from 3 h to 2 weeks post-injury, in South American opossums (Monodelphis domestica). A bridge between severed ends of cords was apparent 5 days post-injury in P7 cords, compared to 2 weeks in P14. The volume of neurofilame

Postnatal Development of the Gray Short-Tailed Opossum (Monodelphis domestica): Implications for Metatherian Decline at the K-Pg.

Marsupials give birth to extremely altricial offspring which must be reared externally for an extended period, often in a pouch. Despite often being considered a defining feature of marsupials, around a third of living species lack a pouch. Here, we describe the postnatal development of the gray short-tailed opossum, Monodelphis domestica, a small pouchless South American didelphid and consider its implications for life history evolution within Metatheria. We find that at birth, ossification and chondrogenesis in neonatal M. domestica is more extensive than in basal pouched Australidelphian ma

Changes in the epithelium of the vaginal complex during the estrous cycle of the marsupial Monodelphis domestica. 1. Transmission electron microscopy study.

The vaginal complex of marsupials differs from that of eutherians. Cervices open separately in a sinus vaginalis or cul-de-sac. Two lateral vaginae adjoin the sinus vaginalis and fuse at the level of the urethra opening and form the sinus urogenitalis. During the estrous cycle the vaginal epithelium undergoes a number of specified morphological changes. This paper is the first to describe these changes on an ultrastructural level in a marsupial. Investigations in Monodelphis vagina reveal that a cyclic switch exists between a keratinized and a stratified nonkeratinized epithelium. Keratinizati

Cortical perineuronal nets in the gray short-tailed opossum (Monodelphis domestica): a distribution pattern contrasting with that shown in placental mammals.

Extracellular matrix proteoglycans accumulated in perineuronal nets and in certain neuropil zones have been shown to influence the immediate neuronal microenvironment, and to contribute to the chemoarchitectonic characteristics of neuronal networks. Studies in different placental mammals, including the human, have suggested that the major principles of extracellular matrix distribution remained constant during phylogenesis of the different mammalian strains. However, the comparison of matrix distribution between various species also indicates that striking deviations from the basic pattern may

Development of the terminal air spaces in the gray short-tailed opossum (Monodelphis domestica)- 3D reconstruction by microcomputed tomography.

Marsupials are born with structurally immature lungs when compared to eutherian mammals. The gray short-tailed opossum (Monodelphis domestica) is born at the late canalicular stage of lung development. Despite the high degree of immaturity, the lung is functioning as respiratory organ, however supported by the skin for gas exchange during the first postnatal days. Consequently, the majority of lung development takes place in ventilated functioning state during the postnatal period. Microcomputed tomography (μCT) was used to three-dimensionally reconstruct the terminal air spaces in order to re
