The structure and function of the mouse tyrosinase locus.

Tyr is the mouse gene that encodes tyrosinase, an enzyme that triggers the first and rate-limiting step in the biosynthesis of melanin. Mutations in Tyr might result in non-functional Tyr protein and, consequently, loss of pigment production. This is a rare genetic condition, known as albinism, described for most animal species and one of the most obvious and simple phenotypes to investigate in model organisms. Mutations in the orthologous human TYR gene are associated with oculocutaneous albinism type 1 (OCA1). Over the last thirty years, the mouse Tyr locus has been studied as a paradigm for

The integration of cyanide hydratase and tyrosinase catalysts enables effective degradation of cyanide and phenol in coking wastewaters.

The aim of this study was to design an effective method for the bioremediation of coking wastewaters, specifically for the concurrent elimination of their highly toxic components - cyanide and phenols. Almost full degradation of free cyanide (0.32-20 mM; 8.3-520 mg L(-1)) in the model and the real coking wastewaters was achieved by using a recombinant cyanide hydratase in the first step. The removal of cyanide, a strong inhibitor of tyrosinase, enabled an effective degradation of phenols by this enzyme in the second step. Phenol (16.5 mM, 1,552 mg L(-1)) was completely removed from a real coki

Albinism and the associated ocular defects.

Several types of hypopigmentation in humans are called albinism. The phenotype for different types of albinism varies according to the amount of pigment in the hale, skin and iris, the reduction in visual acuity and the degree of nystagmus and strabismus. Cutaneous and ocular melanin pigment can range from complete absence throughout the lifetime of the individual to the development of nearly normal levels, including the ability to tan. Visual acuity ranges from 20/40 to 20/400, and visual development in an affected infant is slower than normal. Foveal hypoplasia and altered routing of the opt

Hair melanins and hair color: ultrastructural and biochemical aspects.

The color variants of mammalian hair, including spotting and albinism, are the result of melanocyte activity and have been shown to be determined by the action of multiple genes, some of which operate through the milieu in which the pigment cell resides; others appear to act intracellularly to control the type of melanogenesis. Although there has been much descriptive work on the mode of action of these genes, it has only been with the recent advances in the chemistry and molecular biology of melanin pigmentation that some progress is being made in understanding the nature and origin of hair c

Boophilus microplus: characterization of enzymes introduced into the host.

A number of enzymes, presumably secreted by larvae of B. microplus under natural feeding conditions, have been investigated in the skin of previously unexposed calves 4 h after infestation at the attachment site. Carboxylic ester hydrolase activity was demonstrated in the dermis, immediately adjacent to the mouthparts, or in the attachment cone, depending on substrate and reaction pH. The carboxylic ester hydrolase acting on naphthol AS-D acetate (2-acetoxy-3-naphthoic-O-toluidide) at pH 7-1 was characteristically found in the dermis and not in the attachment cone. The use of specific inhibito

Study on the synthesis and biological activity assessment of kojic acid sulfide schiff base.

In this study, twenty-six novel kojic acid derivatives were designed and synthesized based on the strategy of preserving kojic acid's active site for interaction with tyrosinase and the principle of active fragment assembly. Among all the synthesis compounds, compound 6a exhibited the most powerful inhibitory effect with an IC50 value of 1.43 ± 0.39 µM, over 18-fold more potent than the parent compound kojic acid (IC50 = 26.09 ± 0.05 µM). The findings from the mechanism of action study suggested that the compound 6a functions as a competitive inhibitor, exhibiting a Ki value of 0.84 µM. The co

Drug design of tyrosinase inhibitors.

This copper-containing enzyme catalyzes the rate-limiting step for the melanin skin pigment bioproduction. Tyrosinase inhibitors can be exploited as skin whitening agents and food preservatives, opening new scenarios in food, cosmetics, agriculture and medicine. Despite the availability of natural inhibitors (hydroquinone, α-arbutin, kojic acid, retinoids, azelaic acid, resveratrol, caftaric acid, valonea tannin, chrysosplenetin and phenylethyl resorcinol), several synthetic compounds were proposed to overcome side effects and to improve the efficacy of natural agents. Skin Pigmentation Types, Causes and Treatment-A Review.

Human skin pigmentation and melanin synthesis are incredibly variable, and are impacted by genetics, UV exposure, and some drugs. Patients' physical appearance, psychological health, and social functioning are all impacted by a sizable number of skin conditions that cause pigmentary abnormalities. Hyperpigmentation, where pigment appears to overflow, and hypopigmentation, where pigment is reduced, are the two major classifications of skin pigmentation. Albinism, melasma, vitiligo, Addison's disease, and post-inflammatory hyperpigmentation, which can be brought on by eczema, acne vulgaris, and

A Comprehensive Study to Explore Tyrosinase Inhibitory Medicinal Plants and Respective Phytochemicals for Hyperpigmentation; Molecular Approach and Future Perspectives.

Tyrosinase is a copper-containing key substance in the pigmentation of mammalian hair and skin. Melanin synthesis is influenced by a variety of extrinsic and internal variables, including hormone fluctuations, inflammation, ageing, and subsequent ultraviolet light exposure. Melasma, senile lentigines, freckles, and diminished colour are all undesirable side effects of excessive melanin production. The current review provides the pursuit of effective and safe tyrosinase inhibitors derived from medicinal plants and ascribes updated inferences on current practices. Toxin detection using a tyrosinase-coupled oxygen electrode.

An enzyme-based "electrochemical canary" is described for the detection of cyanide. The sensing system imitates cyanide's site of toxicity in the mitochondria. The terminal sequence of electron transfer in aerobic respiration is mimicked by mediator coupling of tyrosinase catalysis to an electro-chemical system. An enzyme-coupled oxygen electrode is created which is sensitive to selective poisoning. Biocatalytic reduction of oxygen is promoted by electrochemically supplying tyrosinase with electrons. Thus, ferrocyanide is generated at a cathode and mediates the enzymatic reduction of oxygen to

Identification of fungal melanin production and the role of anti-melanin agents.

Accumulation of melanin pigment in some specific areas of human skin leads to various skin disorders. This study includes melanin production and extraction from non-dermatophyte, filamentous fungi (Aspergillus niger). The effect of carbon, nitrogen, pH and temperature on melanin production was also observed. Qualitative and Quantitative analysis was performed to confirm melanin pigment from A. niger by using solubility test, precipitation test and reaction with oxidizing and reducing agent. Fungus gave maximum melanin production with dextrose, peptone, pH 5.6 at 37oC. To avoid hyper pigmentati

Selective protein self-deprivation by Mormon crickets following fungal attack.

Immune responses to infection result in behavioral changes that affect resource acquisition, such as general starvation and compensatory feeding to offset changes in resource allocation. Mormon crickets aggregate and march in bands containing millions of insects. Some bands are comprised of insects seeking proteins. They are also low in circulating phenoloxidase (PO) and more susceptible to fungal attack, as we have demonstrated in the lab. Here, we ask: Do Mormon crickets elevate PO and consume protein in response to infection by the pathogenic fungus Beauveria bassiana? B. bassiana was appli

Mechanism of melanogenesis inhibition by Keggin-type polyoxometalates.

Abnormal melanin overproduction can result in hyperpigmentation syndrome in human skin diseases and enzymatic browning of fruits and vegetables. Recently, our group found that Keggin-type polyoxometalates (POMs) can efficiently inhibit tyrosinase activity. However, it remains unclear whether Keggin-type POMs exhibit optimal effects in vivo. Additionally, the inhibitory effect and mechanism of action of POMs on cellular tyrosinase activity and melanogenesis have been rarely reported. Here we demonstrate that our screened and synthesised PMo11Zn and GaMo12 show superior inhibitory effects on mel

Inhibition of Melanogenesis by Some Well-Known Polyphenolics: A Review.

Melanogenesis is simply defined as the production of melanin in melanosomes by melanocytes through a complex process. Melanin, a pigment derived from L-tyrosine, comes into two forms, namely eumelanin (brownish to black) and pheomelanin (red to yellow). Melanin synthesis starts via the hydroxylation of L-tyrosine to L-3,4-Dihydroxyphenylalanine (DOPA) catalyzed by the enzyme known as Tyrosinase (TYR), which triggers further conversion reaction to DOPAquinone and then to DOPAchrome. Additionally, this process is also related to two more proteins, i.e., oxygenase TYR-related Protein 1 (TYRP1), a

Insights on the Inhibitory Power of Flavonoids on Tyrosinase Activity: A Survey from 2016 to 2021.

Tyrosinase is a multifunctional copper-containing oxidase enzyme that initiates melanin synthesis in humans. Excessive accumulation of melanin pigments or the overexpression of tyrosinase may result in skin-related disorders such as aging spots, wrinkles, melasma, freckles, lentigo, ephelides, nevus, browning and melanoma. Nature expresses itself through the plants as a source of phytochemicals with diverse biological properties. Among these bioactive compounds, flavonoids represent a huge natural class with different categories such as flavones, flavonols, isoflavones, flavan-3-ols, flavanone

Fluorescence copolymer-based dual-signal monitoring tyrosinase activity and its inhibitor screening via blue-green emission transformation.

Tyrosinase (TYR) is a crucial enzyme in melanin metabolism and catecholamine production, its abnormal overexpression is closely associated with many human diseases involving melanoma cancer, vitiligo, Parkinson's disease and so on. Herein, a dual-signal fluorescence sensing system for monitoring TYR activity is constructed depending on the transformation of blue-green fluorescence emission of copolymer. The developed sensing system is based on TYR catalyzing the hydroxylation of mono-phenol to o-diphenol and the conversion of fluorescence copolymer (FCP) blue emission (430 nm) and green emissi
