Unveiling the therapeutic potential of octreotide in treating morphine dependence.

The potential of octreotide (OCT), an octapeptide synthetic somatostatin agonist, to mitigate addiction, specifically morphine dependence, was evaluated in a comprehensive approach involving behavioral, molecular, histological, and docking studies. OCT decreased addiction-related behaviors in Wistar rats previously exposed to morphine, normalizing their exploratory activities and reducing drug-seeking behaviors. Histological assessments revealed octreotide-induced reductions in opioid-induced neuronal damage, suggesting a neuroprotective function. Octreotide's behavioral and histopathological

A Novel Morphine Drinking Model of Opioid Dependence in Rats.

An animal model of voluntary oral morphine consumption would allow for a pre-clinical evaluation of new treatments aimed at reducing opioid intake in humans. However, the main limitation of oral morphine consumption in rodents is its bitter taste, which is strongly aversive. Taste aversion is often overcome by the use of adulterants, such as sweeteners, to conceal morphine taste or bitterants in the alternative bottle to equalize aversion. However, the adulterants' presence is the cause for consumption choice and, upon removal, the preference for morphine is not preserved. Thus, current animal

Protracted manifestations of acute dependence after a single morphine exposure.

RATIONALE: Acute opiate exposure produces a state of dependence in humans and animals, which is revealed by signs and symptoms of withdrawal precipitated by opioid receptor antagonists. The physiological changes that underlie this state of acute dependence develop rapidly and can persist long after the end of chronic opiate exposure. OBJECTIVES: The purpose of this investigation was to determine the persistence of acute dependence after a single morphine exposure in rodents, focusing on changes in behavior thought to reflect the negative emotional consequences of withdrawal. METHODS: The aco

[Slow-release morphine hydrochloride for maintenance therapy of opioid dependence].

INTRODUCTION: In Austria, methadone, buprenorphine as well as oral slow-release morphine are used for the treatment of opioid dependence. This controlled examination marks the first time that oral slow-release morphine hydrochloride is applied for maintenance therapy in opioid dependent subjects. METHODS: In order to evaluate the effectiveness of this psychopharmacological medication, we examined patients over a three-week period. Outcome measures were retention rate, additional consumption and the evaluation of opioid withdrawal 24 hours after the last oral medication. RESULTS: Sixty-seven

Effects of tizanidine on morphine physical dependence: attenuation and intensification.

It has previously been shown that subchronic and acute administration of L-asparaginase and glutaminase inhibitors D-Aspartic acid (D-ASP) and prolyl-leucyl-glycinamide (PLG) intensifies and attenuates morphine (M) physical dependence, respectively, by the inhibition of ASP and glutamic acid (GLU) production, and subsequently their normal releases. Tizanidine (TIZ) has long been known to be an alpha 2-adrenoceptor agonist and inhibitor of ASP and GLU release. Therefore, in this study TIZ has been administered subchronically during the development of M physical dependence to rats in which M-con

Dopamine-dependent responses to morphine depend on glucocorticoid receptors.

Previous work has shown that glucocorticoid hormones facilitate the behavioral and dopaminergic effects of morphine. In this study we examined the possible role in these effects of the two central corticosteroid receptor types: mineralocorticoid receptor (MR), and glucocorticoid receptor (GR). To accomplish this, specific antagonists of these receptors were infused intracerebroventricularly and 2 hr later we measured: (i) locomotor activity induced by a systemic injection of morphine (2 mg/kg); (ii) locomotor activity induced by an infusion of morphine (1 microg per side) into the ventral tegm

Mechanisms of intestinal dysmotility in morphine dependence: whether central or peripheral?

Changes of intestinal motility and transit due to morphine administration are attributed to its central and peripheral action, however coexistence of both central and peripheral mu receptors in morphine dependence hasn't been clearly demonstrated so far. Our purpose was the evaluation of the effect of either CNS or ENS mu receptor blockade on intestinal motility in morphine dependent rats. Twenty male rats were subjected to chronic subcutaneous morphine infusions for 72 hrs. On each day motility indices (MI) were analysed in the animals' duodenum and ascending colon before and after either int

Previous chronic blockade of NMDA receptors intensifies morphine dependence in rats.

Chronic exposure of receptors to antagonists generally results in upregulation and/or supersensitivity. On the other hand, the noncompetitive NMDA receptor antagonists ketamine (K) and dextromethorphan (DM) suppress opiate abstinence syndrome by blocking NMDA receptors. Therefore, 40 mg/kg ketamine (K), 5 mg/kg dextromethorphan (DM), 5 mg/kg morphine (M) and 2 mg/kg naloxone (NL) alone or in combination with NL were IP administered to the rats five times during the daytime only for five days to see whether they would intensify abstinence syndrome through upregulation and/or supersensitivity of

Production of physical dependence in rats by drinking a morphine solution.

The plasma concentrations of morphine and glucose, the body weight, and the severity of the naloxone-precipitated withdrawal syndrome were studied in female rats in which morphine dependence was induced by administration of the opiate, with or without sucrose, in their drinking water. It was found that sucrose encouraged the animals to consume more morphine and that the initial plasma concentrations of the opiate, as well as the rate of development of physical dependence, were higher than the group not given sucrose. Plasma glucose concentrations, maximum plasma morphine levels and the maximum

Effects of serine on morphine-dependent mice.

Single doses of serine (250 mg/kg i.p.) failed to elevate central glycine in mice (1 h after injection), or to compensate for losses caused by implantation of morphine base (70 mg/animal), and reduced the jumping response to nalorphine (100 mg/kg i.p.). In contrast, sustained treatment with 250-mg/kg doses of serine at 24-hour intervals produced increases in brain and medullar glycine (1 h after 4th injection: 46-50% in normal, 39-75% in morphine-dependent mice, and enhanced the jumping response (40% over controls).

Possible involvement of the total amount of morphine infused in the development of acute morphine dependence in rats.

The severity of naloxone-precipitated withdrawal in rats infused intravenously with morphine at the rates of 2.5, 5 and 10 mg/kg/hr over various time periods was investigated. Plasma morphine concentration reached a constant and rate-dependent level at 1 hr after the start of morphine infusion, and this level was maintained until the termination of infusion. Naloxone (2.0 mg/kg, s.c.) was challenged 18 hr after infusion was stopped, and the withdrawal was evaluated by plasma corticosterone (PCS) increase, diarrhea and body weight loss. The incidence of naloxone-precipitated withdrawal signs wa

Environmental Enrichment Ameliorates Psychological Dependence Symptoms and Voluntary Morphine Consumption in Morphine Withdrawn Rats Under Methadone Maintenance Treatment.

INTRODUCTION: Previous studies have shown that physical and psychological dependence and the vulnerability to relapse are still present during MMT. Thus, this study examined whether Enriched Environment (EE) would attenuate anxiety, depressive, and obsessive-compulsive-like behaviors, as well as voluntary morphine consumption following Methadone Maintenance Treatment (MMT) in morphine withdrawn rats. METHODS: The rats were injected bi-daily doses (10 mg/kg, 12-h interval) of morphine for 14 days. Then, the rats were reared in a Standard Environment (SE) or EE for 30 more days during morphine

Self administration of heroin and cocaine in morphine-dependent and morphine-withdrawn rhesus monkeys.

RATIONALE: Dependence can develop during chronic opioid use, and the emergence of withdrawal might promote drug taking. OBJECTIVE: This study examined how chronic morphine administration or withdrawal modified self administration of heroin or cocaine. METHODS: Four monkeys responded under a fixed ratio 10 schedule to receive i.v. infusions of heroin (0.56-560 microg/kg/infusion) or cocaine (1-100 microg/kg/infusion). Monkeys received morphine twice daily; the final dose was 10 mg/kg/12 h. Dose-effect curves for heroin or cocaine were determined in 150-min sessions throughout morphine adminis

Morphine-induced μ-opioid receptor isoform MOR-1X amplifies dependence-related signaling.

Clinically used opioids, such as morphine, primarily act through the μ-opioid receptor (MOR), encoded by the OPRM1 gene, which undergoes extensive alternative splicing. More than 20 OPRM1 isoforms have been identified, yet functional characterization has largely focused on the canonical MOR-1 variant. With the emergence of three-dimensional human cerebral organoids (hCOs) derived from induced pluripotent stem cells (iPSCs), it is now possible to model human-specific neuronal responses to opioids more accurately. In this study, we established hCOs as a functional platform to investigate the imp

Spontaneous daily sleep disruptions associated with morphine dependence and withdrawal in rats.

Sleep disturbances are prominent symptoms of opioid withdrawal that contribute to relapse in individuals with opioid use disorder (OUD). Although preclinical studies often examine protracted withdrawal after sustained abstinence, few address the repeated, spontaneous withdrawal cycles that model human patterns of use. Moreover, targeting sleep disruptions provides a novel strategy to reduce relapse risk. The present studies characterized the effects of repeated spontaneous daily withdrawal on sleep and quantitative electroencephalography (qEEG) after once-daily morphine administration in male

How are testicular functions affected after morphine dependence and withdrawal in rats?

OBJECTIVE: The effects of morphine addiction on testicular activity are significant. This study aimed to examine testicular contraction and tissue changes that may occur in the testicles of rats with experimentally induced morphine addiction. METHOD: Thirty-two Wistar albino rats were divided into four groups: Control (C), Morphine Dependence (M), Morphine Dependence + Morphine Withdrawal (MN), and Morphine Withdrawal (N). Morphine dependence was induced in the M and MN groups. Before decapitation, the rats were evaluated for behavioral changes and scored according to the Geller-Holtzman scal

Involvement of the lateral hypothalamic peptide orexin in morphine dependence and withdrawal.

The lateral hypothalamus (LH) is implicated in the behavioral actions of drugs of abuse, but the cellular and molecular basis of this role is unclear. Recent identification of neuropeptides localized in LH neurons has allowed for more specific studies of LH function. The LH-specific peptide orexin (hypocretin) has been shown to be important in arousal and sleep regulation. However, orexin cells of the LH project broadly throughout the brain such that orexin may influence other behaviors as well. In this study, we show that orexin neurons, and not nearby LH neurons expressing melanin-concentrat

Molecular mechanisms of morphine tolerance and dependence; novel insights and future perspectives.

Drug addiction is a devastating condition that poses a serious burden on the society. The use of some drugs like morphine for their tremendous analgesic properties is also accompanied with developing tolerance, dependence and the withdrawal symptoms. These symptoms are frequently severe enough to reinforce the person in recovery to start over the use of drug again and hinder the clinical use of drugs like morphine for chronic pain. Research into opioid receptors and related molecular pathways has seen resurgence in the wake of the growing opioid epidemic. The current study provides a comprehen

Effects of glucosamine against morphine-induced antinociceptive tolerance and dependence in mice.

BACKGROUND: The most important limitations of morphine in pain therapy are its tolerance and dependence. In this study, we evaluated the protective effect of glucosamine against morphine-induced tolerance and dependence in mice. METHODS: Mice received twice daily morphine (20 mg/kg, s.c.) alone, or along with orally administered glucosamine (500, 1000 and 2000 mg/kg), for 9 continuous days. To assess antinociceptive effect of morphine, percentage of maximal possible effect (%MPE) of animals exposed to thermal stimulus was measured in the hot plate test, 30 min after morphine administration.
