[Proliferation of smooth muscle cells in hypertension].

The aortic intima and media isolated from hypertensives showed a significantly larger number (up to 20%) of smooth muscle cells (SMC) with tetraploid DNA content. The similar process was shown to be also a part of normal human vessel maturation. Normotensive human and rat aortic SMCs were found to accumulate 3H-thymidine, have a lower proliferative ability and they were apt to polyploidize in the primary culture. Such a population could not be detected in the aorta of spontaneously hypertensive rats. It was ascertained that 10 microM of noradrenaline significantly increased (approximately by 2

Skeletal Muscle Stem Cells: Aging and Rejuvenation.

Stem cells are central for tissue homeostasis and regeneration after stress, and their function depends on interactions with neighboring niche cells. In skeletal muscle, the resident stem cells (also called satellite cells [SCs]) decline in number and function with age, leading to loss of regenerative capacity and, eventually, deterioration of muscle integrity and physiology. Aged SCs lose the capacity to preserve quiescence or to activate and expand upon injury, in part due to changes in the local and systemic environment. Here, we focus on the age-associated intrinsic changes of SCs and the

Macrophage-like smooth muscle cells in atherosclerosis.

The structural and functional homeostasis of the vascular wall critically depends on vascular smooth muscle cells (VSMCs) and their secreted extracellular matrix. As the primary cellular component of the vascular tunica media, VSMCs work in concert with elastic fiber networks to maintain vascular tension through their contractile properties. However, the most remarkable characteristic of VSMCs lies in their exceptional phenotypic plasticity. When subjected to mechanical injury or biochemical stimulation, differentiated contractile VSMCs undergo dedifferentiation, transforming into an intermedi

Role of Filamin C in Muscle Cells.

Filamin C (FLNC) is a member of a high-molecular weight protein family, which bind actin filaments in the cytoskeleton of various cells. In human genome FLNC is encoded by the FLNC gene located on chromosome 7 and is expressed predominantly in striated skeletal and cardiac muscle cells. Filamin C is involved in organization and stabilization of thin actin filaments three-dimensional network in sarcomeres, and is supposed to play a role of mechanosensor transferring mechanical signals to different protein targets. Under mechanical stress FLNC can undergo unfolding that increases the risk of its

Smooth muscle-like cells in pulmonary lymphangioleiomyomatosis.

Proliferation, migration, and differentiation of smooth muscle (SM)-like lymphangioleiomyomatosis (LAM) cells in the lungs are pathologic manifestations of pulmonary LAM, a rare lung disease predominantly afflicting women and exacerbated by pregnancy. LAM cells form nodules throughout the lung without any predominant localization, but can also form small cell clusters dispersed within lung parenchyma. LAM cells have the appearance of "immature" SM-like cells, irregularly distributed within the nodule in contrast to organized SM cell layers in airways and vasculature. Progressive growth of LAM

Intracellular calcium signalling in striated muscle cells.

Calcium signalling in cells is dependent on a communication between channels/ transporters in two membrane structures: the cell membrane and the membranes of endo- and sarcoplasmic reticula (ER/SR). In general, cytosolic Ca2+ can be raised by influx of calcium over the cell membrane through three types of channels: voltage-, receptor-, and store-operated channels (VOCs, ROCs and SOCs). This small Ca2+ influx is most often amplified by a Ca2+ release from the ER/SR through two types of channels: the IP3-receptor and the ryanodine receptor (RyR), which are huge proteins identified and cloned in

Smooth muscle differentiation in scleroderma fibroblastic cells.

Using antibodies to alpha-smooth muscle actin and desmin on paraffin-embedded formalin-fixed tissue sections, the authors demonstrate that fibroblastic cells of localized and systemic scleroderma lesions express features of smooth muscle differentiation. Eleven of eleven skin specimens of systemic sclerosis patients and two of four skin specimens of localized scleroderma displayed the presence of fibroblasts expressing alpha-smooth muscle actin, a cell population that predominated in areas of prominent collagen deposition. A similar fibroblastic phenotype was found in the esophagus, the liver,

Acetylcholine receptors in cultured human muscle cells.

Primary cultures prepared from human muscle biopsies were examined for the presence and distribution of acetylcholine receptors, as measured by the binding of 125iodine-labelled alpha-bungarotoxin. The toxin bound to the human muscle cultures with a similar time dependence and specificity as found in muscle cultures from other species. The amount of toxin bound was lower than that obtained for neonatal mouse muscle under similar conditions. The distribution of receptors was similar in cultures derived from the muscles of patients with a variety of neuromuscular disorders. The toxin was located

A muscle stem cell for every muscle: variability of satellite cell biology among different muscle groups.

The human body contains approximately 640 individual skeletal muscles. Despite the fact that all of these muscles are composed of striated muscle tissue, the biology of these muscles and their associated muscle stem cell populations are quite diverse. Skeletal muscles are affected differentially by various muscular dystrophies (MDs), such that certain genetic mutations specifically alter muscle function in only a subset of muscles. Additionally, defective muscle stem cells have been implicated in the pathology of some MDs. The biology of muscle stem cells varies depending on the muscles with w

Pro-elastogenic effects of bone marrow mesenchymal stem cell-derived smooth muscle cells on cultured aneurysmal smooth muscle cells.

Abdominal aortic aneurysms (AAAs) involve slow proteolysis and loss of structural matrix components (collagen and elastin), which lead to wall thinning, weakening and ultimate rupture. At this time, no established non-surgical therapy is available to slow or arrest AAA growth. Inhibiting matrix metalloproteases (MMPs; e.g. MMP2 and -9) overexpressed within AAAs is insufficient to arrest AAA growth, since resident smooth muscle cells (SMCs) are poorly elastogenic and cannot overcome elastolysis to reinstate a healthy elastic matrix. Towards overcoming this limitation, this first study sought to

Normal inflammation and regeneration of muscle following injury require osteopontin from both muscle and non-muscle cells.

BACKGROUND: Osteopontin is secreted by skeletal muscle myoblasts and macrophages, and its expression is upregulated in muscle following injury. Osteopontin is present in many different structural forms, which vary in their expression patterns and effects on cells. Using a whole muscle autograft model of muscle injury in mice, we have previously shown that inflammation and regeneration of muscle following injury are delayed by the absence of osteopontin. The current study was undertaken to determine whether muscle or non-muscle cells provide the source of osteopontin required for its role in mu

Muscle satellite cells are functionally impaired in myasthenia gravis: consequences on muscle regeneration.

Myasthenia gravis (MG) is a neuromuscular disease caused in most cases by anti-acetyl-choline receptor (AChR) autoantibodies that impair neuromuscular signal transmission and affect skeletal muscle homeostasis. Myogenesis is carried out by muscle stem cells called satellite cells (SCs). However, myogenesis in MG had never been explored. The aim of this study was to characterise the functional properties of myasthenic SCs as well as their abilities in muscle regeneration. SCs were isolated from muscle biopsies of MG patients and age-matched controls. We first showed that the number of Pax7+ SCs

Implantation of muscle satellite cells overexpressing myogenin improves denervated muscle atrophy in rats.

This study evaluated the effect of muscle satellite cells (MSCs) overexpressing myogenin (MyoG) on denervated muscle atrophy. Rat MSCs were isolated and transfected with the MyoG-EGFP plasmid vector GV143. MyoG-transfected MSCs (MTMs) were transplanted into rat gastrocnemius muscles at 1 week after surgical denervation. Controls included injections of untransfected MSCs or the vehicle only. Muscles were harvested and analyzed at 2, 4, and 24 weeks post-transplantation. Immunofluorescence confirmed MyoG overexpression in MTMs. The muscle wet weight ratio was significantly reduced at 2 weeks aft

VAMP2 is expressed in muscle satellite cells and up-regulated during muscle regeneration.

Membrane trafficking is one of the most important mechanisms involved in the establishment and maintenance of the forms and functions of the cell. However, it is poorly understood in skeletal muscle cells. In this study, we have focused on vesicle-associated membrane proteins (VAMPs), which are components of the vesicle docking and fusion complex, and have performed immunostaining to investigate the expression of VAMPs in rat skeletal muscle tissue. We have found that VAMP2, but not VAMP1 or VAMP3, is expressed in satellite cells. VAMP2 is also expressed in myofibers in the soleus muscle and n

P2Y12 signaling in muscle satellite cells contributes to masseter muscle contraction-induced pain.

The mechanism behind masseter muscle pain, a major symptom of temporomandibular disorder (TMD), has remained poorly understood. Previous report indicates that adenosine triphosphate (ATP) is involved in the masseter muscle pain development, but the role of its hydrolysis product, adenosine diphosphate (ADP), remains uncertain. Consequently, this study aimed to elucidate the ADP role derived from the sustained masseter muscle contraction in the masseter muscle pain development. The right masseter muscle was electrically stimulated daily by placing electrodes on the muscle fascia, inducing stron

Camk2a suppresses denervated muscle atrophy by maintaining the Ca2+ homeostasis in muscle cells.

Denervated muscle atrophy is a severe neurological complication that significantly impacts patients' quality of life. Currently, there is a lack of effective treatment methods. This study aims to investigate the molecular mechanisms associated with denervated muscle atrophy and explore potential therapeutic targets. In this study, we assessed the severity of denervated muscle atrophy by measuring the wet-weight ratio of the calf muscles. We conducted Western blot and immunofluorescence experiments to observe the morphology and cross-sectional area of muscle fibers following sciatic nerve trans

Twist1 Activation in Muscle Progenitor Cells Causes Muscle Loss Akin to Cancer Cachexia.

Cancer cachexia is characterized by extreme skeletal muscle loss that results in high morbidity and mortality. The incidence of cachexia varies among tumor types, being lowest in sarcomas, whereas 90% of pancreatic ductal adenocarcinoma (PDAC) patients experience severe weight loss. How these tumors trigger muscle depletion is still unfolding. Serendipitously, we found that overexpression of Twist1 in mouse muscle progenitor cells, either constitutively during development or inducibly in adult animals, caused severe muscle atrophy with features reminiscent of cachexia. Using several genetic mo

Bioinspired NO release coating enhances endothelial cells and inhibits smooth muscle cells.

Thrombus and restenosis after stent implantation are the major complications because traditional drugs such as rapamycin delay the process of endothelialization. Nitric oxide (NO) is mainly produced by endothelial nitric oxide synthase (eNOS) on the membrane of endothelial cells (ECs) in the cardiovascular system and plays an important role in vasomotor function. It strongly inhibits the proliferation of smooth muscle cells (SMCs) and ameliorates endothelial function when ECs get hurt. Inspired by this, introducing NO to traditional stent coating may alleviate endothelial insufficiency caused
