Skeletal muscle tissue has the critical function of mechanical support protecting the body. In addition, its functions are strongly influenced by the balanced synthesis and degradation processes of structural and regulatory proteins. The inhibition of protein synthesis and/or the activation of catabolism generally determines a pathological state or condition called muscle atrophy, a reduction in muscle mass that results in partial or total loss of function. It has been established that many pathophysiological conditions can cause a decrease in muscle mass. Skeletal muscle innervation involves

BACKGROUND: Denervation occurs as a consequence of disease or injury and is typically accompanied by skeletal muscle atrophy. Although the relationship between nerves and muscle atrophy has been studied, the direct molecular contributions of nerves remain unclear. METHODS: We used the axolotl (Ambystoma mexicanum), a vertebrate with robust regenerative capacity and optically accessible musculature, to investigate the role of nerve-derived signals in muscle maintenance. RESULTS: Forelimb denervation produced significant muscle atrophy in axolotls. Quantitative imaging showed that reduced muscle

Although electromyography is the gold standard for diagnosing muscle denervation, the role of muscle magnetic resonance (MR) imaging in the diagnosis of muscle denervation has gained clinical relevance. The MR imaging examination of newly denervated musculature shows edema. Conventional and quantitative muscle MR imaging techniques allow for the detection of subclinical disease progression as well as amelioration of the secondary features of nerve damage. In this review article, we present and discuss recent data on the role of conventional and quantitative MR imaging in the diagnosis and moni

Cynomolgus monkeys underwent unilateral panretinal scatter photocoagulation (PRP) and/or nasal and temporal horizontal retinal meridional photocoagulation (HRMP) with xenon arc or argon or krypton laser light. Shortly thereafter, in the PRP-treated eyes, accommodative responsiveness to topical eserine and electrical stimulation of the Edinger-Westphal nucleus (EWN) was diminished, accommodative responsiveness to intramuscular (i.m.) pilocarpine was enhanced, and the number of muscarinic receptors in the ciliary muscle was reduced compared to the contralateral controls. Cynomolgus monkeys underwent unilateral PRP with xenon arc or argon or krypton laser light, employing burn intensity, size, spacing, and topography analogous to standard clinical (eg, Diabetic Retinopathy Study) treatment. Shortly thereafter, accommodative responsiveness to topical eserine and electrical stimulation of the EWN was diminished, accommodative responsiveness to systemic pilocarpine was enhanced, and the number of muscarinic receptors in the ciliary muscle was reduced in the PRP-treated eyes compared to the contralateral controls. In most instances, these parameters returned to nor

Denervated muscle atrophy is a severe neurological complication that significantly impacts patients' quality of life. Currently, there is a lack of effective treatment methods. This study aims to investigate the molecular mechanisms associated with denervated muscle atrophy and explore potential therapeutic targets. In this study, we assessed the severity of denervated muscle atrophy by measuring the wet-weight ratio of the calf muscles. We conducted Western blot and immunofluorescence experiments to observe the morphology and cross-sectional area of muscle fibers following sciatic nerve trans

BACKGROUND: Damage to lower motor neuron causes denervation and degeneration of the muscles affected. Experimental and clinical studies of muscle denervation in lower extremities demonstrated that direct electrical stimulation (ES) of muscle can prevent denervation atrophy and restore contractility. The aim of this study was to identify possible myogenic effect of ES on denervated forearm and hand muscles in persons with spinal cord injury (SCI) and tetraplegia.
METHODS: This prospective interventional study with repeated measurement design included 22 patients aged 48·6 (± 15·7), 0·25 (0·1/4

BACKGROUND: Skeletal muscle exhibits remarkable plasticity under both physiological and pathological conditions. One major manifestation of this plasticity is muscle atrophy that is an adaptive response to catabolic stimuli. Because the heterogeneous transcriptome responses to catabolism in different types of muscle cells are not fully characterized, we applied single-nucleus RNA sequencing (snRNA-seq) to unveil muscle atrophy related transcriptional changes at single nucleus resolution.
METHODS: Using a sciatic denervation mouse model of muscle atrophy, snRNA-seq was performed to generate si

Denervation of skeletal muscle is a debilitating consequence of injury of the peripheral nervous system, causing skeletal muscle to experience robust atrophy. However, the molecular mechanisms controlling the wasting of skeletal muscle due to denervation are not well understood. Here, we demonstrate that transection of the sciatic nerve in Sprague-Dawley rats induced robust skeletal muscle atrophy, with little effect on the neuromuscular junction (NMJ). Moreover, the following study indicates that all three arms of the unfolded protein response (UPR) are activated in denervated skeletal muscle

INTRODUCTION: It is not clear if electrical stimulation (ES) can affect muscle reinnervation. This study aimed to verify if ES affects neuromuscular recovery after nerve crush injury in rats.
METHODS: Denervated muscles were electrically stimulated daily for 6 or 14 days. Neuromuscular performance and excitability, and muscle morphology were determined. Muscle trophism markers (atrogin-1, MuRF-1, and myoD), as well as neuromuscular junction (NMJ) organization (muscle-specific receptor tyrosine kinase [MuSK], cytoplasmic protein downstream of kinase-7 [Dok-7], nicotinic ACh receptor [nAChR], a

We previously reported double innervation of rat mimetic muscles with labeling of facial nuclei. However, whether denervated mimetic muscles are affected after such nerve repair is not known. Rats were divided into five groups: Group A, controls; Group B, complete facial palsy; Group C, complete facial palsy with repair using end-to-end neurorrhaphy; Group D, incomplete facial palsy; and Group E, incomplete facial palsy with repair using end-to-side neurorrhaphy. Preoperatively and postoperatively, facial palsy and myogenin (Myog) expression in mimetic muscles were evaluated. Expression peaked

This research is to study the electrical stimulation characteristics of orbicularis oculi muscle and the characteristics of the mechanical contraction. We observed the stimulus current diffusion regularity and its relationship with mechanical contraction in the orbicularis oculi muscle using an electrode gathering line. Under different stimulus intensities of 2 or 4 mA, the closer the recording electrodes were to the stimulating electrode, the larger was the amplitude. When the recording electrode and stimulating electrode distance increased, the amplitude declined linearly with decreasing fun

The purpose of this study was to investigate the hypothesis that electrical stimulation regulates the levels of gene expression related to apoptosis in denervated muscle and prevents muscle atrophy after denervation.Nineteen rats were used in this study. To denervate soleus muscle, sciatic nerve was resected under aseptic condition. Electrical stimulation with 4 mA rectangular pulses of 0.5 ms duration at 2 Hz lasting for 1 hour was delivered to lower limb including the soleus muscle using two surface electrodes. After the stimulation periods of 4 weeks, the levels of gene expression related t

For many years there has been a dearth of effective treatment options for the severe wasting and secondary consequences of motor nerve injury. In recent years, however, an intensive regime of electrical stimulation has been shown to have considerable therapeutic benefits. This article reviews the results of an extensive study designed to address the clinically relevant issues in an appropriate animal model. The study reveals both the benefits and the limitations of the technique, but strongly endorses the therapeutic advantages of introducing a program of stimulation during the initial, nondeg

OBJECTIVE: To clarify the character of nerve growth factor (NGF) in the denervated red and white muscles and the relationship between the amount of NGF and sensitive neurotrophism of denervated red and white muscles.
METHODS: The model of the denervated gastrocnemius and soleus was made by clipping the sciatic nerve of Wister rats. The immunohistochemistry was taken to measure the amount of NGF in muscles, and the neurotrophism of extracts of muscles was tested with culture of dorsal root ganglions at the 1st, 3rd, 7th and 14th days after injury.
RESULTS: The amount of NGF in denervated gast

Denervation results in a series of changes in skeletal muscle. To elucidate the molecular basis underlying these changes, it is important to identify the profile of altered gene expression in skeletal muscle following nerve injury. In the present study, we have examined the differentially expressed genes in denervated gastrocnemius muscle using RNA fingerprinting by arbitrarily primed PCR. Eight differentially expressed mRNA transcripts have been identified. A bilateral regulatory profile can be observed for the up-regulated genes in both denervated and contralateral control muscle following u
