Histochemical study of urethral striated musculature in the dog.

The striated musculature of the dog urethra was studied histochemically. Two main groups of muscle fibers could be identified: 1 with slow twitch oxidative fibers, which are fatigue resistant (type 1), and 1 with fast twitch fibers (type 2). The fast twitch fibers were subdivided into glycolytic (fatiguable) and oxidative glycolytic (fatigue resistant) fibers: the latter constitute 20 per cent of all fast twitch fibers. Type 1 constitutes of 35 per cent of the whole musculature and its proportion tends to decrease toward the distal end of the external urethral sphincter. From these observation

Morphologic and functional aspects of pelvic floor muscles in patients with pelvic relaxation and genuine stress incontinence.

Thirty women (mean age 49 years; range 33-74) with mild to moderate pelvic relaxation and genuine stress incontinence underwent preoperative urodynamic studies. Biopsy specimens from the levator ani muscles during vaginal hysterectomy and repair were evaluated for the number and the individual diameters of type I (slow-twitch) and type II (fast twitch) fibers. Biopsies from 11 women revealed striated muscle tissue, whereas no muscle tissue was observed in the other 19. Compared with patients without muscle tissue, subjects with positive muscle findings showed significantly higher transmission

Muscle fiber selectivity of Trichinella spiralis and Trichinella pseudospiralis (1983).

The biceps, semimembranosus, biceps femoris, and soleus muscles of female Rockland Wistar mice infected with either 1,000 Trichinella spiralis or 1,000 Trichinella pseudospiralis larvae were removed on days 12, 14, 16, and 18 post-infection (PI), sectioned and stained histochemically for their myosin ATPase activity. Light microscopic examination of the sections revealed that larvae of T. spiralis invade only the slow twitch muscle fibers, and those of T. pseudospiralis invade both the fast twitch and the slow twitch fibers. In sections obtained from mice infected with either parasite and kill

Inhibiting Myozenin 1 Attenuated Muscular Dystrophy Pathology in mdx Mice by Enhancing Calcineurin Activity.

Myozenin 1 (MYOZ1) is expressed in fast-twitch muscle fibers and functions as a calcineurin (CaN)-interacting protein. The deletion of MYOZ1 was reported to enhance the exercise capacity of mice. It would be prospective to explore the exact role of MYOZ1 in Duchenne muscular dystrophy pathology. The transfection of an adenoviral MYOZ1 shRNA in mdx mice was used to knock down MYOZ1 expression. Forelimb grip strength test, hanging wire test, and run-to-exhaustion test were conducted for assessing muscle strength and exercise ability of mice. Muscle tissue pathology was detected by HE and Masson

[Clinico-pathological significance of the immunostaining of myosin heavy chain isoforms in pathological human skeletal muscle].

Expression of the four myosin heavy chain isoforms (fast-twitch, slow-twitch, neonatal and embryonal isoforms) was immunohistochemically observed in 500 biopsied limb muscles of neuromuscular disorders. Fast-twitch isoform was expressed in type 2A, 2B and pathologic 2C fibers. Slow-twitch isoform was expressed in type 1 and 2C fibers. Embryonic isoform was expressed in regenerating type 2C fibers of active myopathies such as Duchenne dystrophy and polymyositis. Expression of neonatal isoform, which was longer positive than that of embryonic isoform, was noted in regenerating fibers, denervated

Expression profiles and transcript properties of fast-twitch and slow-twitch muscles in a deep-sea highly migratory fish, Pseudocaranx dentex.

Fast-twitch and slow-twitch muscles are the two principal skeletal muscle types in teleost with obvious differences in metabolic and contractile phenotypes. The molecular mechanisms that control and maintain the different muscle types remain unclear yet. Pseudocaranx dentex is a highly mobile active pelagic fish with distinctly differentiated fast-twitch and slow-twitch muscles. Meanwhile, P. dentex has become a potential target species for deep-sea aquaculture because of its considerable economic value. To elucidate the molecular characteristics in the two muscle types of P. dentex, we genera

Activity of mu- and m-calpain in regenerating fast and slow twitch skeletal muscles.

Calpains--non-lysosomal intracellular calcium-activated neutral proteinases, form a family consisting of several distinct members. Two of the isoenzymes: mu (calpain I) and m (calpain II) responded differently to the injury during complete regeneration of Extensor digitorum longus (EDL) muscle and partial regeneration of Soleus muscle. In the crushed EDL the level of m-calpain on the 3rd and 7th day of regeneration was higher than in non-operated muscles, whereas the activity of this calpain in injured Soleus decreased. The level of mu-calpain in EDL oscillated irregularly during regeneration

Skeletal muscle fiber size and fiber type distribution in human cancer: Effects of weight loss and relationship to physical function.

BACKGROUND & AIMS: Cancer patients frequently experience weight loss, with negative consequences for functionality and prognosis. The extent to which muscle atrophy contributes to weight loss, however, is not clear, as few studies have directly measured muscle fiber morphology in cancer patients. METHODS: Whole body and regional tissue composition were measured, along with the cross-sectional area (CSA) and fiber type of mechanically-isolated, single muscle fibers, in 19 cancer patients (8 with a history of weight loss, 11 weight-stable) and 15 non-diseased controls. RESULTS: Whole body fat

Muscle type and fiber type specificity in muscle wasting.

Muscle wasting occurs in a variety of conditions, including both genetic diseases, such as muscular dystrophies, and acquired disorders, ranging from muscle disuse to cancer cachexia, from heart failure to aging sarcopenia. In most of these conditions, the loss of muscle tissue is not homogeneous, but involves specific muscle groups, for example Duchenne muscular dystrophy affects most body muscles but spares extraocular muscles, and other dystrophies affect selectively proximal or distal limb muscles. In addition, muscle atrophy can affect specific fiber types, involving predominantly slow ty

Slow or fast: Implications of myofibre type and associated differences for manifestation of neuromuscular disorders.

Many neuromuscular disorders can have a differential impact on a specific myofibre type, forming the central premise of this review. The many different skeletal muscles in mammals contain a spectrum of slow- to fast-twitch myofibres with varying levels of protein isoforms that determine their distinctive contractile, metabolic, and other properties. The variations in functional properties across the range of classic 'slow' to 'fast' myofibres are outlined, combined with exemplars of the predominantly slow-twitch soleus and fast-twitch extensor digitorum longus muscles, species comparisons, and

Fiber-type-specific architecture and pathophysiology of the neuromuscular junction.

The neuromuscular junction (NMJ) is a specialized synapse essential for translating neuronal signals into muscle contraction. This review examines the complex structural, functional, and molecular differences in NMJs that innervate fast- and slow-twitch skeletal muscle fibers. Fast-twitch fibers, optimized for rapid and powerful contractions, possess elaborate NMJs with deep folds, high neurotransmitter turnover, and greater vulnerability to synaptic fatigue and degeneration. In contrast, slow-twitch fiber NMJs exhibit simpler but more stable architectures that support sustained, fatigue-resis

Physiological effects of diaphragm muscle denervation and disuse.

From our studies, it is clear that diaphragm muscle neuromotor control is responsive to alterations in innervation and activation. These adaptations to altered use appear to be most pronounced among fast-twitch motor units composed of type II muscle fibers. Because the plasticity involves diminished contractile strength and a slowing of shortening velocity, it might be considered maladaptive with respect to diaphragm functional demands; however, because ventilatory behaviors of the diaphragm most likely require the recruitment of only type S motor units (type I muscle fibers) that appear to be

Combating muscle atrophy: emerging therapeutic targets that are fiber-type-specific.

Skeletal muscle is essential for life as it enables physical movement, maintains posture, is crucial for breathing, and serves as a major site for energy and carbohydrate metabolism. Pathological conditions that reduce skeletal muscle mass and function-such as muscular dystrophies, motor-neuron diseases, cancer, type-2 diabetes, or aging-have detrimental effects on human health, reducing quality of life and survival. Currently, exercise is the only validated treatment for increasing muscle mass and function, but it is impractical for bedridden patients or the frail elderly. Significant advance

Loss of STARD7 Impairs Mitochondrial Phospholipid Homeostasis and Contributes to Mitochondrial Myopathy.

Mitochondria are composed of phospholipid bilayers rich in phosphatidylcholine (PC). StAR-related lipid transfer domain-containing protein 7 (STARD7) functions as a lipid transfer protein that plays a crucial role in maintaining mitochondrial PC homeostasis. In this study, we investigated the physiological role of STARD7 in skeletal muscle using muscle-specific knockout (mKO) mice. STARD7 expression was markedly higher in the soleus, a mitochondria-dense slow-twitch muscle, compared with fast-twitch fibers. Although muscle fibers from mKO mice exhibited no apparent structural abnormalities, th

A modelling perspective on torque-frequency trade-offs in multifunctional lever systems driven by antagonist muscle pairs.

Rapid cyclic movements are generated by antagonistic muscle pairs contracting in an alternating pattern. The highest frequencies can be generated in balanced torque-producing systems with specialized muscle fibers. The system's frequency output is expected to change when it becomes more adapted to functions with conflicting mechanical demand, such as increased static torque production in one direction. This study first conceptualized how distinct factors (fiber type, muscle cross-sectional area, moment arm and inertial properties) could influence this torque-frequency trade-off. Special attent

Slow twitch paraspinal muscle dysregulation in adolescent idiopathic scoliosis exhibiting HIF-2α misexpression.

BACKGROUND: Adolescent idiopathic scoliosis (AIS) refers to a three-dimensional spinal deformity which has a typical onset during adolescence. In most cases, the cause of the deformity cannot be clearly identified. Unbalanced paraspinal muscle activity in AIS patients was reported and hypoxia was implicated to regulate myogenesis. This study aims to investigate the association between myogenesis/muscle toning and HIF-αs activity in the pathogenesis of AIS. METHODS: HIF-αs expression was examined by enzyme-linked immunosorbent assay and western blot in paraspinal myoblasts isolated from 18 sub

Dystrophin-negative slow-twitch soleus muscles are not susceptible to eccentric contraction induced injury over the lifespan of the mdx mouse.

Duchenne muscular dystrophy (DMD) is the second most common fatal genetic disease in humans and is characterized by the absence of a functional copy of the protein dystrophin from skeletal muscle. In dystrophin-negative humans and rodents, regenerated skeletal muscle fibers show abnormal branching. The number of fibers with branches and the complexity of branching increases with each cycle of degeneration/regeneration. Previously, using the mdx mouse model of DMD, we have proposed that once the number and complexity of branched fibers present in dystrophic fast-twitch EDL muscle surpasses a st

Responses of neuromuscular systems under gravity or microgravity environment.

Hindlimb suspension of rats induces induces fiber atrophy and type shift of muscle fibers. In contrast, there is no change in the cell size or oxidative enzyme activity of spinal motoneurons innervating muscle fibers. Growth-related increases in the cell size of muscle fibers and their spinal motoneurons are inhibited by hindlimb suspension. Exposure to microgravity induces atrophy of fibers (especially slow-twitch fibers) and shift of fibers from slow- to fast-twitch type in skeletal muscles (especially slow, anti-gravity muscles). In addition, a decrease in the oxidative enzyme activity of s

Muscle fibre capillarization is a critical factor in muscle fibre hypertrophy during resistance exercise training in older men.

BACKGROUND: Adequate muscle fibre perfusion is critical for the maintenance of muscle mass; it is essential in the rapid delivery of oxygen, nutrients and growth factors to the muscle, stimulating muscle fibre growth. Muscle fibre capillarization is known to decrease substantially with advancing age. However, whether (relative) low muscle fibre capillarization negatively impacts the muscle hypertrophic response following resistance exercise training in older adults is unknown. METHODS: Twenty-two healthy older men (71 ± 1 years) performed 24 weeks of progressive resistance type exercise train

A histochemical study of the biceps brachii muscle cross-innervated by intercostal nerves. 6 cases of brachial plexus injuries operated with nerve-crossing.

Direct nerve-crossing of intercostal nerves from the lateral thorax to the musculocutaneous nerve was performed in 6 patients after spinal nerve root avulsion with brachial plexus palsy. Elbow flexion power was regained well enough to move against gravity and some resistance in all cases. The muscles were examined histochemically 4 (1-9) years after the operation. The intercostally-innervated biceps brachii muscle showed motor predominance of slow-twitch Type 1 fiber regeneration much more than that of fast-twitch Type 2 fiber in 5 of our patients. Our study suggests that the motor nerves of s
