The larval Drosophila mushroom body balances lateralized sensing and interhemispheric integration.

Bilaterian animals must integrate sensory input from both sides of their bodies to make coherent perceptual decisions. In Drosophila larvae, olfactory receptor neurons target the ipsilateral brain hemisphere. Using calcium imaging, unilateral sensory perturbations, connectome analysis, and optogenetics, we identified the mushroom body (MB) as a key substrate for interhemispheric integration. Kenyon cell odor responses were almost entirely ipsilateral, indicating minimal coupling between the two MBs at the input level. In contrast, modulatory MB input neurons (MBINs) responded symmetrically to

Biased connectivity shapes food odor categorization in the Drosophila mushroom body.

Animals rely on neural representations of sensory stimuli to rapidly recognize familiar environments and precisely identify salient cues. In the Drosophila mushroom body, odors are represented by the activity of Kenyon cells (KCs), whose random wiring with olfactory projection neurons (PNs) is generally thought to promote odor discrimination. Here, we investigated how deviations from this random wiring shape odor representations. By recording the responses of individual KC types to odors, we found that food-related odor representations formed by αβ and, in part, α'β' KCs were separated from re

Lipophorin receptors regulate mushroom body development and complex behaviors in Drosophila.

BACKGROUND: Drosophila melanogaster lipophorin receptors (LpRs), LpR1 and LpR2, are members of the LDLR family known to mediate lipid uptake … patterns, which seem to reflect anatomical defects found in a critical brain association area, the mushroom bodies (MB). Moreover, cultured MB neurons respond to mammalian Reelin by increasing the complexity of their neurite arborization. This effect depends on LpRs and Dab, the Drosophila ortholog of the Reelin signaling adaptor protein Dab1. In vitro, two of t …

Circuit reorganization in the Drosophila mushroom body calyx accompanies memory consolidation.

The formation and consolidation of memories are complex phenomena involving synaptic plasticity, microcircuit reorganization, and the formation of multiple representations within distinct circuits. To gain insight into the structural aspects of memory consolidation, we focus on the calyx of the Drosophila mushroom body. In this essential center, essential for olfactory learning, second- and third-order neurons connect through large synaptic microglomeruli, which we dissect at the electron microscopy level. Focusing on microglomeruli that respond to a specific odor, we reveal that appetitive lo

Nutrient and Chemical Analysis of Fruiting Bodies of a Cultivar of the Chinese Caterpillar Mushroom, Ophiocordyceps sinensis (Ascomycetes).

A cultivar of fruiting bodies of Ophiocordyceps sinensis (FBOS; OCS02) was analyzed for nutrients, bioactive compounds, and heavy metal content to showcase its potential as a competitive, sustainable, and safe alternative to wild types and other cultivars. A previous 28-day subacute toxicity study showed that doses up to 1 g · kg-1 did not cause any adverse effects in Sprague-Dawley rats. The OCS02 cultivar contained large amounts of cordycepin, polysaccharides, and essential and semi-essential amino acids (0.66, 482.80, 99.02, and 101.04 g · kg-1, respectively) compared with levels reported i

Learning performance and GABAergic pathway link to deformed wing virus in the mushroom bodies of naturally infected honey bees.

Viral infections can be detrimental to the foraging ability of the western honey bee, Apis mellifera. The deformed wing virus (DWV) is the m … DWV load performed better in the reversal learning phase. DWV is able to replicate in the honey bee mushroom bodies, where the GABAergic signalling pathway has an antagonistic effect on associative learning but is crucial for reversal learning. Hence, we assessed the pattern of expression of several GABA-related genes in bees with different learning responses. In …

Antibacterial and Antifungal Activities of Lectin Extracted from Fruiting Bodies of the Korean Cauliflower Medicinal Mushroom, Sparassis latifolia (Agaricomycetes).

In this article we describe the isolation and characterization of a novel lectin from fruiting bodies of the mushroom Sparassis latifolia. The antibacterial activity of the purified lectin against Escherichia coli and resistant strains of Staphylococcus aureus and Pseudomonas aeruginosa as well as the antifungal activity against Candida and Fusarium species were determined. Circular dichroism spectroscopy and the tryptophan blue shift assay indicated that the lectin interacts with microbial surfaces. This suggests the potential of the lectin isolated from S. latifolia, a valuable source of bio

Using local anaesthetics to block neuronal activity and map specific learning tasks to the mushroom bodies of an insect brain.

The formation of a stable olfactory memory requires activity within several brain regions. The honeybee provides a valuable model to map complex olfactory learning tasks onto certain brain areas. To this end, we used injections of the local anaesthetic procaine to reversibly block spike activity in a specific brain region, the mushroom body (MB). We first investigated the physiological effects of procaine on cultured MB neurons from adult honeybee brains. Using the whole-cell configuration of the patch-clamp technique, we show that procaine blocks voltage-gated Na+ and K+ currents in a dose-de

Precise control of fasciclin II expression is required for adult mushroom body development in Drosophila.

Fasciclin II (FASII) is a cell adhesion molecule that participates in axonal pathfinding, fasciculation and divergence in the Drosophila nervous system. Here, we examined spatio-temporal control of fasII expression during the development of adult mushroom body (MB) and found that suppression of fasII in alpha'/beta' neurons is essential for the formation of adult alpha'/beta' and alpha/beta lobes. Of gamma, alpha'/beta' and alpha/beta neurons, which are derived sequentially from the same four MB neuroblasts, only gamma and alpha/beta neurons expressed fasII. When fasII was misexpressed in deve

Search for neural correlates of object and panorama learning in mushroom body extrinsic neurons of bumblebees.

A laboratory-based setup was developed that allowed us to train bumblebees to a rewarding object (local cue) in the context of a panorama and simultaneously to record extracellularly the neural activity of units interpreted as mushroom body extrinsic neurons (MBENs). This setting simulates conditions flower visiting bees face when foraging excluding the compass related conditions. The bumblebees navigated between the colony and the test/feeding arena on their own motivation, starting from a fully functional colony with a queen. They learned by sucrose reward a specific feeding place marked by

Self-avoidance alone does not explain the function of Dscam1 in mushroom body axonal wiring.

Alternative splicing of Drosophila Dscam1 into 38,016 isoforms provides neurons with a unique molecular code for self-recognition and self-avoidance. A canonical model suggests that the homophilic binding of identical Dscam1 isoforms on the sister branches of mushroom body (MB) axons supports segregation with high fidelity, even when only a single isoform is expressed. Here, we generated a series of mutant flies with a single exon 4, 6, or 9 variant, encoding 1,584, 396, or 576 potential isoforms, respectively. Surprisingly, most of the mutants in the latter two groups exhibited obvious defect

The Unc13A isoform is important for phasic release and olfactory memory formation at mushroom body synapses.

The cellular analysis of mushroom body (MB)-dependent memory forming processes is far advanced, whereas, the molecular and physiological understanding of their synaptic basis lags behind. Recent analysis of the Drosophila olfactory system showed that Unc13A, a member of the M(Unc13) release factor family, promotes a phasic, high release probability component, while Unc13B supports a slower tonic release component, reflecting their different nanoscopic positioning within individual active zones. We here use STED super-resolution microscopy of MB lobe synapses to show that Unc13A clusters closer

Anti-MRSA Activity of Fruiting Body Extracts of Spectacular Rustgill Mushroom, Gymnopilus junonius (Agaricomycetes).

Despite the great advances in chemotherapeutics, infectious diseases are still one of the leading causes of death worldwide. Among some of the clinically relevant pathogens, methicillin-resistant Staphylococcus aureus (MRSA) ranks as one of the most difficult bacteria to treat. It is a common cause of skin, soft-tissue, and endovascular infections, as well as pneumonia, septic arthritis, endocarditis, osteomyelitis, and sepsis. The research on Basidiomycota is extensive; many species show a broad spectrum of pharmacological activities, including antimicrobial activity. The vast majority of the

Bisphenol F affects neurodevelopmental gene expression, mushroom body development, and behavior in Drosophila melanogaster.

Bisphenol F (BPF) is a potential neurotoxicant used as a replacement for bisphenol A (BPA) in polycarbonate plastics and epoxy resins. We investigated the neurodevelopmental impacts of BPF exposure using Drosophila melanogaster as a model. Our transcriptomic analysis indicated that developmental exposure to BPF caused the downregulation of neurodevelopmentally relevant genes, including those associated with synapse formation and neuronal projection. To investigate the functional outcome of BPF exposure, we evaluated neurodevelopmental impacts across two genetic strains of Drosophila- w1118 (co

Chemical Composition and Ultraviolet Absorption Activity of an Aqueous Alkali Extract from the Fruiting Bodies of the Tinder Conk Mushroom, Fomes fomentarius (Agaricomycetes).

White rot mushroom Fomes fomentarius is a medicinal fungus with great potential to be explored. This work focused on the chemical composition of a basic aqueous extract from F. fomentarius fruiting bodies. The extract was mostly composed of phenolics, carbohydrates, minerals, and crude fat with a low amount of proteins and chitin. One-third of the total carbohydrates were in the form of beta-glucans with minor amounts of alpha-glucans. The most valuable essential part of the extract was composed of an acid-resistant ultraviolet (UV)-absorbing mixture of phenolic compounds such as melanins, lig

Amyloid precursor protein ortholog Appl acts with Vnd during mushroom body axon growth in Drosophila.

The amyloid precursor protein (APP) is associated with Alzheimer's disease. Appl is the single Drosophila APP ortholog and is expressed in all neurons throughout development. Appl was previously shown to cell-autonomously modulate axon outgrowth in the mushroom bodies (MBs), the fly olfactory memory center. However, we found that Appld, the only reported null allele, affects the normal function of vnd, the gene just proximal to Appl. To decipher developmental and memory defects specifically due to a loss of only Appl function, we generated a precise Appl null allele (ApplC2.1) by CRISPR/Cas9 g…

Regulation of axon outgrowth during mushroom body development by acetylated α-tubulin lysine 394 and Tau.

Disruptions in the microtubule cytoskeleton play a role in various neurological diseases that afflict a large fraction of the population. Microtubule function is regulated by post-translational modifications like acetylation, and one consistently identified acetylation site in mammals and Drosophila melanogaster is α-tubulin lysine 394 (K394). Our previous research demonstrated that an acetylation-blocking point mutation-K394R-causes a decrease in microtubule stability in axon terminals at the developing neuromuscular junction. Here, we asked whether K394 acetylation regulates the development
