Lentinula edodes polysaccharides are natural immunomodulators. SeLe30, analyzed in this study, is a new mixture of selenium-enriched linear 1,4-α-glucans and 1,3-β- and 1,6-β-glucans isolated from L. edodes mycelium. In the present study, we evaluated its immunomodulatory properties in human T cells. Peripheral blood mononuclear cells (PBMCs) and T cells were isolated from healthy donors' buffy coats. The effects of SeLe30 on CD25, CD366, and CD279 expression, the subsets of CD8+ T cells, and IFN-γ, IL-6, and TNF-α production were analyzed. SeLe30 downregulated CD25, CD279, and CD366 expressio

The galactomannan is a major cell wall molecule of Aspergillus fumigatus. This molecule is composed of a linear mannan with a repeating unit composed of four α1,6 and α1,2 linked mannose with side chains of galactofuran. To obtain a better understanding of the mannan biosynthesis in A. fumigatus, it was decided to undertake the successive deletion of the 11 genes which are putative orthologs of the mannosyltransferases responsible for establishing α1,6 and α1,2 mannose linkages in yeast. These deletions did not lead to a reduction of the mannan content of the cell wall of the mycelium of A. fu

Stillage is a byproduct of distilleries which is rich in organic matter, minerals, and acidic components. It is commonly used as animal feed and has high potential for use as an alternative substrate for microorganisms. Filamentous fungi are saprophytes that can utilize stillage solids to grow as threadlike mycelium. The structure and composition of the filamentous mycelium has shown promise to produce pure mycelium materials (PMM), which might have potential to serve as leather-like or other novel materials with improved environmental impact profiles. Basidiomycota fungi (including true mushr

Paracoccidioides brasiliensis is a thermodimorphic fungus associated with paracoccidioidomycosis (PCM), a systemic mycosis prevalent in South America. In humans, infection starts by inhalation of fungal propagules, which reach the pulmonary epithelium and transform into the yeast parasitic form. Thus, the mycelium-to-yeast transition is of particular interest because conversion to yeast is essential for infection. We have used a P. brasiliensis biochip carrying sequences of 4,692 genes from this fungus to monitor gene expression at several time points of the mycelium-to-yeast morphological shi

A novel heteropolysaccharide named HEP-S was separated from the mycelium of Hericium erinaceus using hot water extraction. The structure of HEP-S (2.45 × 106 Da) is mainly composed of →4,6)-α-D-Glcp(1→, →2)-α-D-Manp(1→, →4)-α-D-Glcp(1→, →3,6)-β-D-Glcp(1→, T-β-D-Galp(1→, and →4)-β-D-Galp(1→. In addition, we investigated the anti-inflammatory activity and mechanism of HEP-S in lipopolysaccharide-stimulated RAW264.7 cells. HEP-S (20, 40, and 80 µg/mL) could suppress the production of inflammatory cytokines by reducing vital transcription factors in the nuclear factor-κB signaling (NF-κB) pathway.

Living materials, owing to their inherent capacities for growth, self-healing, sensing, and adaptation, have attracted widespread attention in recent years. However, controlling growth of living matter to achieve robust and tunable material properties remains a challenge. Here, we report a co-culture strategy that integrates mycelial microfibers with bacterial cellulose nanofibers into hierarchical hydrogels and transparent films. The biological crosslinking between fungal cell wall mannans and cellulose chains yields an interpenetrating micro-nano network with enhanced interfacial hydrogen bo

Mycelium-based composites are bio-derived structural materials produced by cultivating filamentous fungi through lignocellulosic substrates under solid-state fermentation conditions. Solid-state fermentation remains difficult to control because fungal growth is highly sensitive to local gradients in temperature, humidity, and gas composition. This study presents the design, construction, and experimental validation of a modular tray bioreactor for the controlled cultivation of mycelium-based composites. The system combines closed-loop feedback control of temperature and relative humidity with

Thioacetamide (TAA), usually used as a fungicide to control the decay of citrus products, itself is not toxic to the liver, but its intermediates are able to increase oxidative stress in livers and further cause fibrosis. Ophiocordyceps sinensis mycelium (OSM) which contains 10% polysaccharides and 0.25% adenosine decreased (P < 0.05) the lipid accumulation and increased (P < 0.05) antioxidative capacity in livers of thioacetamide (TAA) injected rats. Meanwhile, the increased (P < 0.05) liver sizes, serum alanine transaminase (AST) and aspartate transaminase (ALT) values in thioacetamide (TAA)

Given its degradative capacity, mycelium may valorize okara, an underutilized agro-industrial by-product. This study aimed to conduct solid-state fermentation of okara and mixtures (okara with sawdust or wheat bran) using Pleurotus ostreatus mycelium and explore the potential of fermented okara as an alternative food, focusing on its taste characteristics. The fastest mycelial growth with dense cultures was achieved on non-dried okara, followed by wheat bran (11-12 days). Fermentation enhanced the nitrogen content in okara, indicating an improved protein level. Following biotransformation, sub

Paracoccidioidomycosis is an important systemic mycosis caused by thermodimorphic fungi of the Paracoccidioides genus. During the infective process, the cell wall acts at the interface between the fungus and the host. In this way, the cell wall has a key role in growth, environment sensing and interaction, as well as morphogenesis of the fungus. Since the cell wall is absent in mammals, it may present molecules that are described as target sites for new antifungal drugs. Despite its importance, up to now few studies have been conducted employing proteomics in for the identification of cell wal

Rifamycin B production in batch culture of Nocardia mediterranei was compared with mycelium morphological changes. Rheological behaviour of batch culture medium was identified and various rheological parameters were evaluated in order to characterize quantitatively the morphological changes. Rifamycin B production kinetic parameters in the batch culture were also calculated and the parameters were compared with the rheological parameters. Consequently, specific rifamycin B production rate (qrif) was closely related to yield shear stress (tau 0) and to morphological factor (delta*). Both parame

Germination and outgrowth of spores of Streptomyces alboniger, Streptomyces scabies, and Streptomyces violaceus-ruber in the presence of intercalating dyes resulted in a high frequency (2 to 20%) of occurrence of aerial mycelium-negative (Amy-) isolates. Coincident with the appearance of the Amy- trait was the loss of several differentiated functions, including the characteristic pigments and earthy odor of the wild types. All S. alboniger, 27% of S. scabies, and 39% of the S. violaceus-ruber Amy- isolates were arginine auxotrophs. The missing enzyme step was identified as argininosuccinate sy

An in vivo whole-plant bi-dimensional experimental system has been devised and tested with different host plants to obtain extraradical mycelium (ERM) produced by different isolates of arbuscular mycorrhizal fungi (AMF). In this system, a plantlet is inoculated with AMF to establish mycorrhizal symbiosis and, after colonization, newly formed extraradical hyphae and spores are removed. Then, the mycorrhizal root system is wrapped in a nylon net and placed between two membranes in a Petri dish, allowing ERM to grow on the membrane surfaces. Such extraradical hyphae may be used for in situ morpho

Phylloporia lonicerae is an annual fungus that specifically parasitizes living Lonicera plants, offering significant potential for developing new resource food and medicine. However, wild resources and mycelium production of this fungus is limited, and its anti-tumor active ingredients and mechanisms remain unclear, hampering the development of this fungus. Thus, we optimized the fermentation medium of P. lonicerae and studied the anti-tumor activity of its mycelium. The results indicated that the optimum fermentation medium consisted of 2% sucrose, 0.2% peptone, 0.1% KH2PO4, 0.05% MgSO4·7H2O,
